How Neutrophils Shape the Immune Response: Reassessing Their Multifaceted Role in Health and Disease.

Neutrophils are the most abundant of the circulating immune cells and are the first to be recruited to sites of inflammation. Neutrophils are a heterogeneous group of immune cells from which are derived extracellular traps (NETs), reactive oxygen species, cytokines, chemokines, immunomodulatory factors, and alarmins that regulate the recruitment and phenotypes of neutrophils, macrophages, dendritic cells, T cells, and B cells. In addition, cytokine-stimulated neutrophils can express class II major histocompatibility complex and the internal machinery necessary for successful antigen presentati

Programmed Cell Death in the Small Intestine: Implications for the Pathogenesis of Celiac Disease.

The small intestine has a high rate of cell turnover under homeostatic conditions, and this increases further in response to infection or damage. Epithelial cells mostly die by apoptosis, but recent studies indicate that this may also involve pro-inflammatory pathways of programmed cell death, such as pyroptosis and necroptosis. Celiac disease (CD), the most prevalent immune-based enteropathy, is caused by loss of oral tolerance to peptides derived from wheat, rye, and barley in genetically predisposed individuals. Although cytotoxic cells and gluten-specific CD4+ Th1 cells are the central pla

Neuropeptide CGRP Limits Group 2 Innate Lymphoid Cell Responses and Constrains Type 2 Inflammation.

Innate lymphocytes maintain tissue homeostasis at mucosal barriers, with group 2 innate lymphoid cells (ILC2s) producing type 2 cytokines and controlling helminth infection. While the molecular understanding of ILC2 responses has advanced, the complexity of microenvironmental factors impacting ILC2s is becoming increasingly apparent. Herein, we used single-cell analysis to explore the diversity of gene expression among lung lymphocytes during helminth infection. Following infection, we identified a subset of ILC2s that preferentially expressed Il5-encoding interleukin (IL)-5, together with Cal

Interleukin-33: a novel player in osteonecrosis of the femoral head?

Osteonecrosis of the femoral head (ONFH) is a disabling disease affecting young adults, which usually leads to the destruction of the hip joint. It is mainly due to an inadequate blood supply that causes the death of osteocytes and bone marrow cells. Joint salvaging procedures are numerous but relatively inefficient, justifying the need for new therapeutic strategies. In this regard, the recently discovered interleukin (IL)-33 alarmin appears as a possible target. Indeed, IL-33 seems to be specifically released by necrotic cells, and interestingly, is constitutively expressed in human bone, in

Cancer cell death: Cell-autonomous and immunogenic dimensions.

Regulated cell death (RCD) shapes neoplastic transformation, tumor progression, and response to treatment. While apoptosis was long viewed as the only RCD variant, additional modalities, including necroptosis, pyroptosis, and ferroptosis, have been characterized. These interconnected pathways operate in a context-dependent manner to influence the dynamic interplay between malignant and non-malignant cells that governs disease progression or regression, both naturally and during therapy. Major advances stemmed from recognizing immunogenic cell death (ICD) as an RCD type defined by the emission

From stress signals to fertility challenges: the role of damage-associated molecular patterns in male reproduction.

Male infertility is influenced by genetic abnormalities, hormonal imbalances, lifestyle factors, and environmental exposures. Recently, Damage-Associated Molecular Patterns (DAMPs) have emerged as key players in male reproductive health, particularly in regulating inflammatory responses and tissue damage. This review highlights the role of critical DAMPs, such as HMGB1, HSPs, ATP, eCIRP, histones, and cfDNA, in processes like spermatogenesis, sperm maturation, and fertilization. Released through mechanisms like necrosis, apoptosis, pyroptosis, and exosomes, DAMPs significantly influence immune

Role of S100A8/A9 in Platelet-Neutrophil Complex Formation during Acute Inflammation.

Acute respiratory distress syndrome (ARDS) due to pulmonary infections is associated with high morbidity and mortality. Upon inflammation, the alarmin S100A8/A9 is released and stimulates neutrophil recruitment mainly via binding to Toll-like receptor 4 (TLR4). TLR4 is also expressed on platelets, which modulate the immune response through direct interaction with leukocytes. In a murine model of Klebsiella pneumoniae-induced pulmonary inflammation, global S100A9 deficiency resulted in diminished neutrophil recruitment into the lung alveoli and neutrophil accumulation in the intravascular space

Reduced hydration regulates pro-inflammatory cytokines via CD14 in barrier function-impaired skin.

Damage to epidermis results in loss of barrier function and resultant pathological inflammatory signaling, triggering further damage to the skin. Here we investigate transcriptomic datasets generated from varied skin pathologies associated with disrupted epidermis and pinpoint CD14/S100 signaling as a conserved pathway upregulated in dermatopathologies characterized by a compromised epidermal barrier. We show that dermatitic and fibrotic tissues of humans and mouse models, which are associated with compromised epidermal barrier, demonstrate upregulation of CD14 and S100 proteins, damage-associ

Altered lung metabolism and mitochondrial DAMPs in lung injury due to acute kidney injury.

Acute respiratory distress syndrome (ARDS) is a common cause of mortality in patients with acute kidney injury (AKI). Inflammatory crosstalk from the kidney to the lung has been shown to contribute to lung injury after AKI, but anti-inflammatory therapies have not been proven beneficial in human studies. Recently, AKI was shown to alter mitochondria and related metabolic pathways in the heart, but the impact of AKI on lung metabolism has not been investigated to our knowledge. In this study, we evaluated the metabolomic profile of the lung following renal ischemia and reperfusion to identify n

Emerging insights into the presentation, pathophysiology, and management of eosinophilic esophagitis.

Eosinophilic esophagitis (EoE) is a chronic, immune-mediated disease of the esophagus with substantial clinical manifestations including dysphagia, odynophagia, food impaction and failure to thrive in children. New evidence is emerging regarding phenotypic and symptom variations in diverse EoE populations. EoE pathogenesis involves impaired epithelial barrier function, antigen-driven Type 2 inflammation, and eosinophilic infiltration, yet eosinophil and mast cell depletion does not improve patient symptoms. Recent research has unveiled new potential mechanisms of epithelial dysfunction, includ

Treatment of chronic obstructive pulmonary disease: current pipeline and new opportunities.

Chronic obstructive pulmonary disease (COPD) is an inflammatory disorder of the lungs that affects about 10% of the adult population and is currently the third leading global cause of death. COPD is the result of multiple, repeated and dynamic gene-environment interactions, starting early in life, that determine the lung function trajectory that a given individual follows over a lifetime. Increasing understanding of COPD pathogenesis has opened many new opportunities for drug development, including recently approved monoclonal antibodies that reduce inflammatory cytokine signalling by targetin

Migration of Lung Resident Group 2 Innate Lymphoid Cells Link Allergic Lung Inflammation and Liver Immunity.

Group 2 innate lymphoid cells (ILC2s) are tissue resident in the lung and activated by inhaled allergens via epithelial-derived alarmins including IL-33. Activated ILC2s proliferate, produce IL-5 and IL-13, and induce eosinophilic inflammation. Here, we report that intranasal IL-33 or the protease allergen papain administration resulted in increased numbers of ILC2s not only in the lung but also in peripheral blood and liver. Analyses of IL-33 treated parabiosis mice showed that the increase in lung ILC2s was due to proliferation of lung resident ILC2s, whereas the increase in liver ILC2s was

Sterile Inflammation and Cell Death Pathways in Liver Ischemia-Reperfusion Injury: A Review and Perspective.

BACKGROUND: Hepatic Ischemia-Reperfusion Injury (IRI) is a critical complication in liver transplantation and resection, driven by oxidative stress and sterile inflammation mediated by damage-associated molecular patterns (DAMPs). Current therapeutic challenges arise from interconnected cell death pathways and redundant inflammatory mechanisms. OBJECTIVE: This review synthesizes mechanistic insights into DAMP signaling and regulated cell death modalities in IRI, aiming to identify translational gaps and propose precision-targeted therapies. The NF-κB-driven inflammatory cascade in ischemic stroke: Linking DAMPs, inflammasomes, and neurovascular dysfunction.

Ischemic stroke is increasingly recognized as an inflammation-driven disease in which secondary injury extends beyond the initial ischemic insult. Sterile inflammation triggered by endogenous danger signals plays a decisive role in neuronal loss, blood-brain barrier disruption, and neurological outcome. Among the pathways involved, nuclear factor-κB (NF-κB) is a canonical inflammatory transcription factor, but its broader integrative role in post-ischemic inflammation remains insufficiently defined. In this review, we summarize evidence showing how NF-κB links three interconnected dimensions o

The Role of Thermal Immunomodulation in Postoperative Wound Repair with a Focus on Hepatic Surgery.

Controlled local hyperthermia supports postoperative wound healing in liver surgery by stimulating metabolism, angiogenesis, and immune responses through the induction of heat shock proteins (HSPs) and modulation of Damage-Associated Molecular Patterns (DAMPs). This study evaluates the impact of thermal modulation on immune processes during abdominal wound repair, specifically analyzing the role of HSPs and immune activation pathways. A narrative review of the literature from 2010 to 2025 was conducted to summarize molecular mechanisms regarding temperature, HSP activation, cytokine expression

[Inflammasome Activation and Idiosyncratic Drug-induced Liver Injury: Fusion Study of Basic and Real-world Data].

Idiosyncratic drug-induced liver injury (IDILI) is particularly problematic due to its severity and unpredictability. Various pathogenic mechanisms have been proposed for IDILI; however, these mechanisms have yet to be comprehensively elucidated. Although the hapten hypothesis has long been posited as a possible pathogenic mechanism, it is believed that other mechanisms may also elicit a strong immune response. We hypothesized that reactive metabolites generated by drug metabolism cause cellular stress and release damage-associated molecular patterns (DAMPs) that may activate antigen-presentin

The role of cell death pathways in respiratory viral infection and vaccination: two sides of the same coin.

Cell death pathways play contrasting roles in physiological processes such as responses to viral infections and vaccinations, potentially exerting either detrimental or beneficial effects. On one hand, uncontrolled cell death accompanied by the release of damage-associated molecular patterns (DAMPs) can lead to excessive inflammation and tissue damage. On the other hand, when properly regulated, these processes help establish an immunocompetent environment by activating innate immunity, which in turn stimulate antiviral immune responses. These mechanisms have emerged as promising targets for t

A multi-damage-associated molecular pattern targeting opsonic peptide attenuates gut ischemia/reperfusion-induced lung injury.

BACKGROUND: Gut ischemia-reperfusion induces the release of damage-associated molecular patterns (DAMPs), exacerbating inflammation and organ injury and reducing survival. We recently developed a multi-damage-associated molecular pattern scavenging molecule, named opsonic peptide 18, which promotes the clearance of multiple damage-associated molecular patterns by phagocytes. The present study aims to investigate the therapeutic potential of opsonic peptide 18 in gut ischemia-reperfusion-induced lung injury. METHODS: Male C57BL6/J mice underwent superior mesenteric artery occlusion and were tr

Self-DNA driven inflammation in COVID-19 and after mRNA-based vaccination: lessons for non-COVID-19 pathologies.

The coronavirus disease 2019 (COVID-19) pandemic triggered an unprecedented concentration of economic and research efforts to generate knowledge at unequalled speed on deregulated interferon type I signalling and nuclear factor kappa light chain enhancer in B-cells (NF-κB)-driven interleukin (IL)-1β, IL-6, IL-18 secretion causing cytokine storms. The translation of the knowledge on how the resulting systemic inflammation can lead to life-threatening complications into novel treatments and vaccine technologies is underway. Nevertheless, previously existing knowledge on the role of cytoplasmatic

Therapeutic targeting of chronic kidney disease-associated DAMPs differentially contributing to vascular pathology.

Chronic Kidney Disease (CKD) is associated with markedly increased cardiovascular (CV) morbidity and mortality. Chronic inflammation, a hallmark of both CKD and CV diseases (CVD), is believed to drive this association. Pro-inflammatory endogenous TLR agonists, Damage-Associated Molecular Patterns (DAMPs), have been found elevated in CKD patients' plasma and suggested to promote CVD, however, confirmation of their involvement, the underlying mechanism(s), the extent to which individual DAMPs contribute to vascular pathology in CKD and the evaluation of potential therapeutic strategies, have rem
