Improved micro-sampling methodology enables accurate platelet function analysis.

Micro-sampling for platelet function assays offers a minimally invasive alternative to venipuncture that may facilitate point-of-care testing. We have developed a new micro-sampling methodology for human and mouse and evaluated these methods for platelet function analysis. Blood was collected by venepuncture or our new finger-prick sampling method in humans, and by terminal bleeding or a modified tail vein bleeding method in mice. Platelet activation was assessed by flow cytometry, measuring fibrinogen binding and P-selectin exposure in response to ADP, CRP-XL or TRAP-6 in humans (n = 14) and

New Strategies and Artificial Intelligence Methods for the Mitigation of Toxigenic Fungi and Mycotoxins in Foods.

The proliferation of toxigenic fungi in food and the subsequent production of mycotoxins constitute a significant concern in the fields of public health and consumer protection. This review highlights recent strategies and emerging methods aimed at preventing fungal growth and mycotoxin contamination in food matrices as opposed to traditional approaches such as chemical fungicides, which may leave toxic residues and pose risks to human and animal health as well as the environment. The novel methodologies discussed include the use of plant-derived compounds such as essential oils, classified as

Three-dimensional (3D) liver cell models - a tool for bridging the gap between animal studies and clinical trials when screening liver accumulation and toxicity of nanobiomaterials.

Despite the exciting properties and wide-reaching applications of nanobiomaterials (NBMs) in human health and medicine, their translation from bench to bedside is slow, with a predominant issue being liver accumulation and toxicity following systemic administration. In vitro 2D cell-based assays and in vivo testing are the most popular and widely used methods for assessing liver toxicity at pre-clinical stages; however, these fall short in predicting toxicity for NBMs. Focusing on in vitro and in vivo assessment, the accurate prediction of human-specific hepatotoxicity is still a significant c

[The development of mucosal vaccines].

The live oral polio vaccine was the first mucosal vaccine accepted for general use. Since then, similar vaccines have been developed against typhoid fever, cholera and rotavirus infection, and a nasal vaccine against influenza has recently been registered in the USA. The only non-living mucosal vaccine on the market today is an oral cholera vaccine consisting of inactivated Vibrio cholerae and the B subunit of cholera toxin. Several groups of scientists are at present working on the development of other mucosal vaccines based on inactivated microbes or parts of them. Results from animal trials

Anti-VEGF antibody in experimental hepatoblastoma: suppression of tumor growth and altered angiogenesis.

BACKGROUND: Hepatoblastoma is the most common primary hepatic malignancy of childhood, frequently presenting as advanced disease. Vascular endothelial growth factor (VEGF) is an endothelial mitogen and survival factor critical to growth and angiogenesis in many human cancers. Inhibition of VEGF effectively suppresses tumorigenesis in multiple experimental models. The authors hypothesized that anti-VEGF antibody would alter vascular architecture and impede tumor growth in experimental hepatoblastoma. METHODS: The Institutional Animal Care and Use Committee of Columbia University approved all p

Evaluating the effect of γ-oryzanol on MASLD pathology using a medaka fish model.

Metabolic dysfunction-associated steatotic liver disease (MASLD) encompasses liver conditions not caused by alcohol, including metabolic dysfunction-associated steatohepatitis (MASH), which involves inflammation and fibrosis, with rising global prevalence. While research to explore MASLD/MASH prevention or cure has largely relied on rodent models, ethical concerns regarding animal welfare have prompted the exploration of alternatives. This study examines the application of a recently developed medaka fish MASH/MASLD model to assess the impact of food components on liver pathology. Medaka were

Investigating Neuroprotective Effects of Berberine on Mitochondrial Dysfunction and Autophagy Impairment in Parkinson's Disease.

Parkinson's disease (PD) is a common neurodegenerative disorder with substantial global impact. Although current therapies can provide symptomatic relief, they are often associated with high costs and adverse effects. Natural compounds with a history of traditional medicinal use have emerged as promising alternatives. In this study, we investigated the therapeutic potential and underlying mechanisms of berberine in both cellular and animal models of PD. In vitro, SH-SY5Y cells exposed to 6-hydroxydopamine (6-OHDA) exhibited decreased viability and increased oxidative stress, both of which were

Integrating Concentration-Dependent Toxicity Data and Toxicokinetics To Inform Hepatotoxicity Response Pathways.

Failure of animal models to predict hepatotoxicity in humans has created a push to develop biological pathway-based alternatives, such as those that use in vitro assays. Public screening programs (e.g., ToxCast/Tox21 programs) have tested thousands of chemicals using in vitro high-throughput screening (HTS) assays. Developing pathway-based models for simple biological pathways, such as endocrine disruption, has proven successful, but development remains a challenge for complex toxicities like hepatotoxicity, due to the many biological events involved. To this goal, we aimed to develop a comput

Natural Polyphenols for Prevention and Treatment of Urinary Tract Infections.

Urinary tract infections (UTIs) are the second most common type of bacterial infection worldwide. UTIs are gender-specific diseases, with a higher incidence in women. This type of infection could occur in the upper part of the urogenital tract, leading to pyelonephritis and kidney infections, or in the lower part of the urinary tract, leading to less serious pathologies, mainly cystitis and urethritis. The most common etiological agent is uropathogenic E. coli (UPEC), followed by Pseudomonas aeruginosa and Proteus mirabilis. Conventional therapeutic treatment involves the use of antimicrobial

Therapeutic use of mesenchymal stem cell-derived extracellular vesicles in acute lung injury.

Acute respiratory distress syndrome is a major cause of respiratory failure in critically ill patients. Despite extensive research into its pathophysiology, mortality remains high. No effective pharmacotherapy exists. Based largely on numerous preclinical animal studies, administration of mesenchymal stem or stromal cell (MSC) as a therapeutic for acute lung injury (ALI) holds great promise, and Phase I and II clinical trials are currently under way internationally. However, concern for the use of stem cells, specifically the risk of iatrogenic tumor formation, as well as the prohibitive cost

A comparative study of mandibular growth patterns in seven animal models.

The use of nonhuman primate experimental models has helped make significant contributions toward the clinical and surgical management of patients with craniofacial disorders. With concerns such as increasing cost, however, alternative models will have to be identified. The present comparative study describes baseline, age-related changes in mandibular growth patterns for seven commonly used animal models. The data was obtained from 144 serial and cross-sectional lateral head radiographs of unoperated control animals from a number of previous craniofacial growth studies. The sample consisted of

Antimicrobial Peptides: Bringing Solution to the Rising Threats of Antimicrobial Resistance in Livestock.

Antimicrobial therapy is the most applied method for treating and preventing bacterial infection in livestock. However, it becomes less effective due to the development of antimicrobial resistance (AMR). Therefore, there is an urgent need to find new antimicrobials to reduce the rising rate of AMR. Recently, antimicrobial peptides (AMPs) have been receiving increasing attention due to their broad-spectrum antimicrobial activity, rapid killing activities, less toxicity, and cell selectivity. These features make them potent and potential alternative antimicrobials to be used in animals. Here, we

Improved ex vivo/in vitro lymph node cell proliferation assay in guinea pigs for a screening test of contact hypersensitivity of chemical compounds.

Simple and efficient ex vivo/in vitro screening systems for contact allergens are developed for alternative to conventional animal tests. We have previously proposed an ex vivo/in vitro proliferation assay as a first stage screening method with advantages over existing alternatives, using lymph node cells (LNC) from sensitized guinea pigs of the Hartley strain. In this study, we have first confirmed, by histochemical analysis using in vivo bromodeoxyuridine (BrdU) and pyronin staining, that the ex vivo/in vitro LNC proliferation reflects in vivo response of lymph nodes to contact allergens. A review of sperm cryopreservation in the domestic dog and cat: part I, from science to clinic.

Semen cryopreservation is widely applied in the breeding of several domestic animal species. In the domestic dog, artificial insemination with cryopreserved semen is now routinely performed, whereas in the domestic cat the technique is more challenging. Achieving acceptable pregnancy rates requires high post-thaw semen quality. Unfortunately, sperm cells are susceptible to damage caused by temperature reduction. At temperatures below - 130 °C harmful reactions that can damage spermatozoa are essentially halted. Therefore, spermatozoa can be stored in liquid nitrogen at -196 °C for virtually un

Antibacterial activity and mode of action of acetone crude leaf extracts of under-investigated Syzygium and Eugenia (Myrtaceae) species on multidrug resistant porcine diarrhoeagenic Escherichia coli.

BACKGROUND: Diarrhoea, a global economically important disease burden affecting swine and, especially piglets, is commonly caused by infection with entero-toxigenic E. coli (ETEC). Adherence of ETEC to porcine intestinal epithelial cells following infection, is necessary for its pathogenesis. While antimicrobials are commonly given as therapy or as feed additives for prophylaxis against microbial infections, the concern over increased levels of antimicrobial resistance necessitate the search for safe and effective alternatives in livestock feed. Attention is shifting to natural products includ

Major bone defect treatment with an osteoconductive bone substitute.

A bone defect can be provoked by several pathological conditions (e.g. bone tumours, infections, major trauma with bone stock loss) or by surgical procedures, required for the appropriate treatment. Surgical techniques currently used for treating bone defects may count on different alternatives, including autologous vascularized bone grafts, homologous bone graft provided by musculoskeletal tissue bank, heterologous bone graft (xenograft), or prostheses, each one of them dealing with both specific advantages and complications and drawbacks. The main concerns related to these techniques respect

Veterinary Forensic Pathology of Animal Sexual Abuse.

Animal sexual abuse (ASA) involves harm inflicted on animals for the purposes of human sexual gratification and includes such terms as bestiality, zoophilia, zoosadism, animal sexual assault, and others. The prevalence of ASA is not known, although it may be more common than is currently perceived. Veterinarians have the skills required to identify and document cases of ASA. This article reviews the terminology, legal definitions and forms of ASA, and its social and psychological context. An investigative approach is outlined, including an alternate light source examination; collection of swab

Antivenom Evaluation by Electrophysiological Analysis.

Scorpion stings on humans are medically relevant because they may contain toxins that specifically target ion channels. During antivenom production, pharmaceutical companies must use a large number of experimental animals to ensure the antivenom's efficacy according to pharmacopeia methods. Here we present an electrophysiological alternative for the evaluation of horse antivenoms produced against two species of Moroccan scorpions: Buthus mardochei and Androctonus mauretanicus. Human sodium and potassium channels and acetylcholine nicotinic receptors were analyzed by standard patch-clamp techni

Epigenetic studies in insects and the valproic acid perspective.

Valproic acid in association with sodium valproate (VPA) is an important anticonvulsant drug used for decades to treat neurological disorders. VPA also acts as an epigenetic modulator by inhibiting histone deacetylases, permitting histone acetylation, affecting the DNA and histone methylation status and gene expression, and inducing chromatin remodeling. Insects represent an important animal model for studies in several areas of science. Their high phenotypic plasticity makes them alternative models for epigenetic studies. This brief review emphasizes recent reports on insect epigenetics and t
