Regional patterns of bone loss and altered bone remodeling in response to calcium deprivation in laboratory rabbits.

This study explores the effects of a calcium-deficient diet on patterns of bone remodeling, and examines regional differences in the amount of bone lost. Skeletally mature female rabbits (n = 6) were fed a calcium-deficient diet (0.10% Ca2+ and 0.50% P) for 14 weeks. A separate group of rabbits (n = 4) were fed a maintenance diet (1.2% Ca2+ and 0.45% P). Bone mineral content, serum calcium, and serum phosphorus were measured each week during the experimental period. Following sacrifice, the L3 vertebra, femoral head, proximal tibial metaphysis, and tibial midshaft were analyzed histomorphometr

Assessment of mechanical variables best describing bone remodelling responses based on their correlation with bone density.

Density distribution in bones can be estimated using bone remodelling models (BRM) and applying daily normal loads to assess the stress/strain state to which the bone is subjected. These models locally relate a certain mechanical stimulus, derived from the stress/strain state, directly to bone density or to its variation over time. The background of this idea is Frost's Mechanostat Theory, which states that overloading states tend to increase bone density and disuse states tend to decrease it. Different variables have been proposed in the literature to measure the mechanical stimulus. Strain e

Finite-Element Syntheses of Callus and Bone Remodeling: Biomechanical Study of Fracture Healing in Long Bones.

Computational simulations of fracture healing are a valuable tool to improve fracture treatment and implants. Several finite-element models have been established to predict callus formation due to mechanobiological rules. This work provides a comprehensive simulation for virtual implantation through the combination of callus simulation and finite-element structural synthesis (FESS) of (re-)modeling during and after healing based on Pauwel's theory of histogenesis and Wolff's law. The simulation is based on a linear elastic material model and includes generation of fracture hematoma and initial

Proteoglycans as Molecular Regulators of Bone Metastasis: Extracellular Matrix Remodeling, Tumor-Bone Crosstalk, Dormancy, and Therapeutic Opportunities.

Background: Bone metastasis is a frequent and debilitating complication of advanced cancer, particularly in breast and prostate cancer, and is driven by complex interactions among tumor cells, bone-resident cells, immune populations, vascular components, and the extracellular matrix. Within this specialized microenvironment, proteoglycans have emerged as key molecular regulators of tumor-bone crosstalk, matrix remodeling, metastatic niche formation, dormancy, and therapeutic resistance. Methods: We conducted a narrative review using targeted searches of PubMed and Google Scholar for studies pu

Prostaglandin E2 enhances cortical bone mass and activates intracortical bone remodeling in intact and ovariectomized female rats.

To assess the efficacy of prostaglandin E2 (PGE2) in augmenting cortical bone mass, graded doses of PGE2 were subcutaneously administered for 30 days to seven-month old sham-ovariectomized (SHAM) and ovariectomized (OVX) rats. Both groups were operated at three months of age. Histomorphometric analyses of double fluorescent labeled tibial shafts were performed on basal control, OVX, and SHAM rats treated with 0, 0.3, 1, 3, and 6 mg PGE2/kg/d for 30 days. Baseline aging data showed increased cortical tissue and cortical bone area and reduced bone formation parameters at the periosteal and endoc

Naringin attenuates orchidectomy-induced bone loss in male rats: evidence from bone remodeling and telomere-related indices.

Androgen deficiency contributes to secondary osteoporosis in men, yet the effects of naringin in androgen-deficient male bone loss remain unclear. We evaluated whether naringin attenuates orchidectomy (ORX)-induced bone loss in male Sprague-Dawley rats and whether this response is accompanied by changes in bone turnover, the OPG/RANKL axis, TERT expression, and relative telomere length. Forty rats were assigned to Sham, ORX, ORX + Naringin, and ORX + Alendronate (ALN) groups. Treatments were administered by oral gavage for 8 weeks. ORX impaired L4 trabecular microarchitecture, shifted serum bo

Increased proportion of hypermineralized osteocyte lacunae in osteoporotic and osteoarthritic human trabecular bone: implications for bone remodeling.

Hypermineralized osteocyte lacunae (micropetrosis) have received little research attention. While they are a known aspect of the aging human skeleton, no data are available for pathological bone. In this study, intertrochanteric trabecular bone cores were obtained from patients at surgery for osteoporotic (OP) femoral neck fracture (10F, 4M, 65-94 years), for hip osteoarthritis (OA; 7F, 8M, 62-87 years), and femora at autopsy (CTL; 5F, 11M, 60-84 years). Vertebral trabecular bone cores were also obtained from the vertebra of autopsy cases (CVB; 3F, 6M, 53-83 years). Specimens were resin-embedd

Mechanical rejuvenation of senescent stem cells and aged bone via chromatin remodeling.

Bone aging compromises skeletal integrity and increases vulnerability to osteoporosis and other age-related disorders, underscoring the need for new therapeutic strategies. Although pharmacological and genetic approaches have been widely explored, how cellular mechanical remodeling contributes to bone aging remains unclear. Here, we find that senescent bone marrow stem cells show markedly reduced intracellular force and impaired mechanical behavior. Moderate mechanical stimulation in cell culture and in mice restores cellular force, increases chromatin accessibility at the FOXO1 locus, activat

Beyond resorption: targeting osteoclast fusion and polarization to restore balanced bone remodeling.

The prevalence of osteoporosis is increasing worldwide as populations age, creating a growing clinical burden of fragility fractures and highlighting limitations of current antiresorptive therapies. Conventional agents such as bisphosphonates and denosumab effectively reduce fracture risk but suppress osteoclast number and activity indiscriminately, potentially impairing bone remodeling dynamics and silencing osteoclast-derived anabolic and angiogenic coupling signals. Recent advances have redefined osteoclasts as multifunctional cells that not only resorb bone but also orchestrate osteoblast

β-TCP/Collagen Composite Scaffolds Facilitate Bone Remodeling in Vertebral Plate Fusion.

β-tricalcium phosphate (β-TCP) is widely used as a bone substitute because of its biocompatibility, osteoconductivity, and biodegradability; however, its brittleness and limited adaptability to irregular bone surfaces may restrict its performance in spinal fusion. This study aimed to evaluate whether a β-TCP/collagen composite scaffold could enhance osteogenesis and promote sustained bone remodeling in a sheep lumbar interlaminar fusion model. β-TCP/collagen composite scaffolds and β-TCP scaffolds were compared in vitro using MC3T3-E1 osteogenic precursor cells. Tripartite motif-containing 27 negatively regulates NF-κB activation in bone remodeling.

BACKGROUND: Tripartite motif-containing 27 (TRIM27) is highly expressed in the mouse thymus, spleen, and hematopoietic compartment cells and regulates cell proliferation, apoptosis, and innate immune responses. However, the role of TRIM27 in bone remodeling remains unknown. This study aimed to investigate the role of TRIM27 in the differentiation of osteoclasts and osteoblasts. METHODS: We measured the effects of overexpression or knockdown of TRIM27 in osteoclasts and osteoblasts using real-time PCR and Western blot analysis to quantify the mRNA and protein levels of marker genes. Citrate: a key signalling molecule and therapeutic target for bone remodeling disorder.

Bone remodeling is a continuous cyclic process that maintains and regulates bone structure and strength. The disturbance of bone remodeling leads to a series of bone metabolic diseases. Recent studies have shown that citrate, an intermediate metabolite of the tricarboxylic acid (TCA) cycle, plays an important role in bone remodeling. But the exact mechanism is still unclear. In this study, we focused on the systemic regulatory mechanism of citrate on bone remodeling, and found that citrate is involved in bone remodeling in multiple ways. The participation of citrate in oxidative phosphorylatio

M2-exo promote orthodontic bone remodeling via the MeCP2-TCF20-HDAC1 axis.

BACKGROUND: Orthodontic bone remodeling is a complex process involving a dynamic balance between osteogenesis and osteoclast genesis. Although M2 macrophage-derived exosomes (M2-exos) have emerged as key regulators of bone metabolism through immunomodulation and paracrine signaling, their specific mechanisms in orthodontic tooth movement (OTM) remain unclear. This study aimed to investigate the role of the epigenetic MeCP2-TCF20 complex in M2-exo-mediated orthodontic bone remodeling. METHODS: A rat OTM model was established to evaluate bone remodeling dynamics using micro-CT, histomorphometry

On interaction between fatigue of reconstruction plate and time-dependent bone remodeling.

BACKGROUND AND OBJECTIVE: The fibula free flap (FFF) has been extensively used to repair large segmental bone defects in the maxillofacial region. The reconstruction plate plays a key role in maintaining stability and load-sharing while the fibula unites with adjacent bone in the course of healing and remodeling. However, not all fibula flaps would fully unite, and fatigue of prosthetic devices has been recognized as one major concern for long-term load-bearing applications. This study aims to develop a numerical approach for predicting the fatigue life of the reconstruction plate by taking in

Loss of miR-23a cluster in skeletal muscle can suppress bone remodeling.

Muscle-bone interaction might regulate bone remodeling in an endocrine manner, but the exact mediators have not been identified. Previous in vitro studies suggest that exosomal miRNAs are a candidate for this interaction. Here we present an in vivo study to show that targeted knockout of a muscle-specific miR-23a cluster including miR-23a, miR-27, and miR-24-2 in skeletal muscle tissues can suppress bone remodeling in mice. The effect of miR-23a cluster seem to not be related to aging, but can worsen the pathological extent of osteoporosis in mice. Our findings suggest that muscle-derived miRN
