The nucleotide sequences encoding the major 25-kDa outer membrane protein (OMP) (omp25 genes) of Brucella ovis 63/290, Brucella melitensis 16M, Brucella suis 1330, Brucella canis RM6/66, and Brucella neotomae 5K33 (all reference strains) were determined and compared with that of Brucella abortus 544 (P. de Wergifosse, P. Lintermans, J. N. Limet, and A. Cloeckaert, J. Bacteriol. 177:1911-1914, 1995). The major difference found was between the omp25 gene of B. ovis and those of the other Brucella species; the B. ovis gene had a 36-bp deletion located at the 3' end of the gene. The corresponding

BACKGROUND: Increased dog relocation can cause dissemination of pathogen and vector populations, and this is being recognised in countries across Northern Europe, including the UK. Data regarding the prevalence of exotic infections in dogs entering the UK would be beneficial to veterinarians to help assess pets entering the UK from abroad and to help calculate the risk of establishment of novel pathogens. This study reports the findings from a group of imported dogs that was seized as part of a Royal Society for the Prevention of Cruelty to Animals (RSPCA)-led animal welfare investigation and

OBJECTIVES: This study aimed to identify limitations in interministerial role allocation and information sharing during the response to the 2025 Brucella canis outbreaks. It also sought to propose practical improvements for the One Health collaboration system through simulation exercises using the joint risk assessment (JRA) tool.
METHODS: A tabletop exercise was conducted with 34 interministerial practitioners using scenarios based on actual B. canis cases and social issues in April and August 2025. The effectiveness of the exercise was evaluated using pre- and post-surveys (on a 5-point Lik

To date, no totally effective antibiotic for the eradication of canine brucellosis has been found. The purpose of this study was to evaluate the efficacy of enrofloxacin in a kennel infected with Brucella canis. Twelve dogs, 2 males and 10 females (including 1 in estrus, 3 pregnant, and 6 in anestrus) infected with B. canis were given 5 mg/kg of enrofloxacin orally every 12 h for 30 days. Females received additional courses of enrofloxacin during the estral and luteal phases of the subsequent cycles (0-2 cycles). They were repeatedly mated by infected males. A serological follow-up was carried

VirB proteins from Brucella spp. constitute the type IV secretion system, a key virulence factor mediating the intracellular survival of these bacteria. Here, we assessed whether a Th1-type immune response against VirB proteins may protect mice from Brucella infection and whether this response can be induced in the dog, a natural host for Brucella. Splenocytes from mice immunized with VirB7 or VirB9 responded to their respective antigens with significant and specific production of gamma interferon (IFN-γ), whereas interleukin-4 (IL-4) was not detected. Thirty days after an intraperitoneal chal

BACKGROUND: Brucella, a Gram-negative bacterium, is classified as a potential bioterrorism agent mainly due to the low dose needed to cause infection and the ability to transmit the bacteria via aerosols. Goats/sheep, cattle, pigs, dogs, sheep and rodents are infected by B. melitensis, B. abortus, B. suis, B. canis, B. ovis and B. neotomae, respectively, the six classical Brucella species. Most human cases are caused by B. melitensis and B. abortus. Our aim was to specifically detect Brucellae with 'smooth' lipopolysaccharide (LPS) using a highly sensitive monoclonal antibody (mAb) based immun

Pre-weaning mortality and stillbirth rates in dogs are relatively high. Hypoxia during prolonged parturition is the main cause of neonatal death, followed by septic infection, predominantly caused by opportunistic bacteria. Several viruses also contribute to neonate mortality. Good breeding management and vaccination are important in improving newborn survival. The purpose of the study was to establish pregnancy outcome rates in a large cohort of Swedish breeding dogs, information that had not been previously documented. Further, we aimed to identify risk factors associated with abortion, cong

Brucellosis is one of the most common bacterial zoonoses worldwide affecting not only livestock and wildlife but also pets. Canine brucellosis is characterized by reproductive failure in dogs. Human Brucella canis infections are rarely reported but probably underestimated due to insufficient diagnostic surveillance. To improve diagnostics, we investigated dogs in a breeding kennel that showed clinical manifestations of brucellosis and revealed positive blood cultures. As an alternative to the time-consuming and hazardous classical identification procedures, a newly developed species-specific i

Discospondylitis can affect dogs of any age and breed and may be seen in cats. Although radiography remains the gold standard, advanced imaging, such as CT and MRI, has benefits and likely allows earlier diagnosis and identification of concurrent disease. Because discospondylitis may affect multiple disk spaces, imaging of the entire spine should be considered. There is a lengthening list of causative etiologic agents, and successful treatment hinges on correct identification. Image-guided biopsy should be considered in addition to blood and urine cultures and Brucella canis screening and as a

An 18,228-bp region containing open reading frames predicted to be derived from the IncP plasmid or phage ancestors is present in the genomes of Brucella suis biovars 1 to 4, B. canis, B. neotomae, and strains isolated from marine mammals, but not in B. melitensis, B. abortus, B. ovis, and B. suis biovar 5. The presence of circular excision intermediates and the results of an analysis of sequenced bacterial genomes suggest that the region downstream of the guaA gene is a hotspot for site-specific integration of foreign DNA mediated by a CP4-like integrase.

BACKGROUND: Brucellosis is a widespread zoonotic disease caused by Gram-negative Brucella bacteria. Immunisation with attenuated vaccine is an effective method of prevention, but it can interfere with diagnosis. Live, attenuated Brucella abortus strain 104M has been used for the prevention of human brucellosis in China since 1965. However, at present, no fast and reliable method exists that can distinguish this strain from field strains. Single nucleotide polymorphism (SNP)-based assays offer a new approach for such discrimination. SNP-based minor groove binder (MGB) and Cycleave assays have b

Brucellosis is a major zoonotic disease caused by Brucella spp., notably B. melitensis, B. abortus, B. canis, and B. suis. It continues to impose a significant public health and economic burden in endemic regions, including parts of Asia, Africa, the Middle East, and Latin America. The clinical presentation of the disease is often nonspecific, and early, accurate diagnosis is further complicated by the susceptibility of conventional serological assays to cross-reactivity. This narrative review covers literature published between 2000 and 2025 regarding the detection of Brucella, indexed in Pub

Brucellosis is a zoonosis, reported for the first time in 1859 by Marston, in Malta. It has also been referred to as Mediterranean Fever, Malta Fever and Bang's disease. Its causative agent was isolated by David Bruce in 1886; this was Brucella melitensis: five different strains were later described: B. abortus, B. suis, B. ovis, B. neotomae and B. canis. Recently, in 1994, the isolation of Brucella was reported in marine mammals, eventually pathogenic to man and unofficially designated "B. maris". Human brucellosis is still a public health problem in countries where the infection has not been

Culture is considered as the reference standard assay for diagnosis of Brucella spp. in humans and animals but it is time-consuming and hazardous. In this study, we evaluated the performances of newly designed real-time PCR assays using TaqMan probes and targeting the 3 following specific genes: (i) the insertion sequence IS711, (ii) bcsp31 and (iii) per genes for the detection of Brucella at genus level. The real-time PCR assays were compared to previously described conventional PCR assays targeting the same genes. The genus-specificity was evaluated on 26 Brucella strains, including all spec

BACKGROUND: Brucellosis is an infectious zoonotic bacterial disease of humans and other animals. In the Republic of South Africa (RSA), animal brucellosis is widespread and the current available data on the prevalence of this disease rely solely on serological testing. The primary limitation of brucellosis serology is the lack of discriminatory powers to differentiate between Brucella species and biovars as well as the cross-reactivity observed with other Gram-negative bacteria.
AIM: The aim of this study was to conduct a retrospective laboratory-based survey on Brucella species and biovars i

INTRODUCTION: Brucella spp. are gram-negative, facultative intracellular bacteria pathogens responsible for brucellosis, a zoonotic disease that can cause abortion, fetal death, and genital infections in animals and undulant fever in humans. Lipopolysaccharide (LPS) is known as a major virulence factor of Brucella spp. The wboA gene is capable of encoding a glycosyltransferase that appears to play a major role in LPS biosynthesis. Hence, the characterization of this gene can help in the clarification of the pathogenicity of Brucella spp.
METHODS: This study was carried out at Razi Vaccine and

Despite Brucella suis biovar 2's (BSB2) active circulation in wildlife, no canine infections have been reported. The present paper is the first to describe two cases of BSB2 infections in French dogs. The first case occurred in 2020 and concerned a 13-year-old male neutered Border Collie with clinical signs of prostatitis. The urine culture revealed the excretion of significant levels of Brucella in the sample. The second case concerned a German Shepherd with bilateral orchitis, in which it was possible to detect Brucella colonies following neutering. HRM-PCR and classical biotyping methods cl
