Metabolomics analysis of the 3D L-02 cell cultures revealing the key role of metabolism of amino acids in ameliorating hepatotoxicity of perfluorooctanoic acid.

To simulate the real cell status and morphology in the living systems is substantial for using cell models to address the detrimental effects of toxic contaminants. In this study, the comparative profiles of metabolites in three-dimensional (3D) human normal liver (L-02) cell spheroids with perfluorooctanoic acid (PFOA) treatment were analyzed using a metabolomic approach. The uniform 3D cell spheroids were well formed in 3 days (e.g., sphericity index >0.9) and stably maintained over the subsequent 11 days. The cytotoxicity of PFOA to the 3D L-02 cell spheroids was highly dependent on both ex

Perfluoroalkyl Chemicals and Male Reproductive Health: Do PFOA and PFOS Increase Risk for Male Infertility?

Poly- and perfluoroalkyl substances (PFAS) are manmade synthetic chemicals which have been in existence for over 70 years. Though they are currently being phased out, their persistence in the environment is widespread. There is increasing evidence linking PFAS exposure to health effects, an issue of concern since PFAS such as perfluorooctane sulfonate (PFOS) and perfluorooctanoic acid (PFOA) bioaccumulate in humans, with a half-life of years. Many epidemiological studies suggest that, worldwide, semen quality has decreased over the past several decades. One of the most worrying effects of PFOS

Comparative Hepatotoxicity of Novel PFOA Alternatives (Perfluoropolyether Carboxylic Acids) on Male Mice.

As novel alternatives to perfluorooctanoic acid (PFOA), perfluoropolyether carboxylic acids (multiether PFECAs, CF3(OCF2) nCOO-, n = 2-4) have been detected in various environmental matrices; however, public information regarding their toxicities remains unavailable. To compare the hepatotoxicity of multiether PFECAs (e.g., PFO2HxA, PFO3OA, and PFO4DA) with PFOA, male mice were exposed to 0.4, 2, or 10 mg/kg/d of each chemical for 28 d, respectively. Results demonstrated that PFO2HxA and PFO3OA exposure did not induce marked increases in relative liver weight; whereas 2 and 10 mg/kg/d of PFO4D

Photodegradation of fluorotelomer carboxylic 5:3 acid and perfluorooctanoic acid using zinc oxide.

Occurrence of per- and poly-fluoroalkyl substances (PFASs) in the environment and biota has raised a great concern to public health because these compounds are persistent, bioaccumulative, and toxic. Biodegradation of polyfluoroalkyl substances, particularly long-chain fluorotelomer-based products, can lead to production of various short-chain PFASs, with 5:3 fluorotelomer carboxylic acid (referred as 5:3 FTCA hereafter) as a dominant polyfluoroalkyl metabolite. Perfluoroalkyl acids, particularly perfluorooctanoic acid (PFOA), are toxic and current removal methods are not cost-effective. This

Toxicity of perfluorooctanoic acid to Pseudomonas putida in the aquatic environment.

Perfluorinated compounds are potential persistent organic pollutants which attracted much concerns in recent years. Thus relevant toxicity data of perfluorooctanoic acid (PFOA) are vitally important for identification of possible risk in the aquatic environment. In the present study, the acute toxic effect of PFOA in the absence and presence of either chromium (III) or tetra butyl ammonium (TBA) towards Pseudomonas putida in the aquatic environment was investigated by microcalorimetry. The thermokinetic parameters including growth rate constant (k), inhibitory ratio, and half inhibitory concen

Kinetic model of 1,3-specific triacylglycerols alcoholysis catalyzed by lipases.

A new model of enzymatic 1,3-specific alcoholysis of triacylglycerols has been developed. The irreversibility of the acyl bounds cleavage in glycerides, a reversible monoglycerides isomerization and an irreversible enzyme deactivation have been assumed. The Ping Pong Bi Bi mechanism with competitive inhibition by alcohol has been applied to describe rates of acyl bonds cleavage. The enzymatic propanolysis and iso-propanolysis of triacetin and tricaprylin catalyzed by immobilized lipase B from Candida antarctica (Novozym 435) have been investigated to verify the model. Good agreement between ex

Dissolution of bile duct stones.

Mono-octanoin can be used either alone or as an adjunct to other techniques to dissolve cholesterol bile duct stones. This solvent can be administered through an existing T tube, through the nasobiliary route, or percutaneously through the liver. Unlike basket extraction, which requires a mature T-tube sinus tract, mono-octanoin can be used immediately postoperatively or for home dissolution therapy. The endoscopic extraction of bile duct stones has a 1 percent mortality rate and a 5 to 7 percent complication rate. Special mixtures of mono-octanoin, bile acids, and ethylene diaminetetraacetic

Effect of octanoate on blood-brain barrier permeability using L-dopa as a marker.

Short-chain fatty acid injection into animals produces coma, seizures, and hyperventilation. One mechanism of coma production may be through alterations in membrane permeability characteristics. The SPG histofluorescence technique was used to evaluate changes in blood-brain barrier permeability to injected L-dopa in rats after intraperitoneal injections of the short-chain fatty acid, sodium octanoate. Diffusion of intravascular fluorescence was observed around brain capillaries in the octanoate-injected rats suggesting an alteration in capillary permeability to L-dopa. Diffusion of fluorescenc

"Endotoxicity" of the Legionnaires' disease bacterium.

The Legionnaires' disease (LD) bacterium is a gram-negative organism whose "endotoxicity" appears to differ in several respects from the classic endotoxicity generally associated with gram-negative bacteria. Discrepancies were noted between the high activity of LD bacteria in gelating limulus lysate in vitro and their low pyrogenicity in rabbits. Further in-vivo biologic tests indicated that LD bacteria were relatively weak in "endotoxicity". Analysis of LD bacterial cells and in their cellular fractions by gas-liquid chromatography indicated that LD bacteria did not contain hydroxy fatty acid

Impact of perfluorooctanoic acid on sperm function and early development in Prochilodus magdalenae: implications for aquatic species reproduction.

Given the well-documented capacity of perfluorooctanoic acid (PFOA) to induce endocrine alterations, this study aimed to investigate the toxicity of PFOA on sperm kinetics and larval viability of Prochilodus magdalenae (bocachico), Colombia's most economically significant freshwater fish species. Gametes were obtained through hormonal induction of P. magdalenae broodstock, and in vitro fertilization was performed to obtain larvae. The semen was activated either with water or water containing different PFOA concentrations (10-500 µM). Sperm motility (both total and specific types), progressivit

Per- and polyfluoroalkyl substances and the risk of thyroid and breast cancer: Evidence from a meta-analysis and computational toxicology.

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental pollutants with endocrine-disrupting and carcinogenic effects, but epidemiological evidence linking PFAS exposure to cancer remains inconsistent. We conducted a meta-analysis to systematically evaluate associations between PFAS exposure and risks of thyroid and breast cancer. Relevant literature published between January 2003 and May 2025 was retrieved from Web of Science and PubMed. The analysis showed that the pooled odds ratios (ORs) for perfluorooctanoic acid (PFOA) and perfluorohexane sulfonate (PFHxS) in relation to

Comparative transcriptomic analysis of perfluoroalkyl substances-induced responses of exponential and stationary phase Escherichia coli.

BACKGROUND: Per- and polyfluoroalkyl substances (PFAS) are highly stable chemical contaminants of emerging concern for human and environmental health due to their non-natural chemistry, widespread use, and environmental persistence. Despite conventional metrology, mitigation strategies, and removal technologies, the complexity of this growing problem necessitates alternative approaches to tackle the immense challenges associated with complex environmental PFAS contamination. Recently, biology has emerged as an alternative approach to detect and mitigate PFAS and understand the molecular-level

Unraveling the molecular mechanisms of PFOA in clear cell renal cell carcinoma through network toxicology and molecular docking strategies.

BACKGROUND: As Perfluorooctanoic Acid (PFOA) has been extensively utilized as a processing aid in the manufacture of non-stick coatings, waterproof materials, and other products, concerns regarding its adverse health effects have emerged. Epidemiological data revealed a strong correlation between renal cell carcinoma (RCC) and PFOA concentration, while animal experimental results also demonstrate the association between PFOA and RCC. However, the key targets and mechanisms underlying PFOA-induced RCC remain elusive. This study utilized network toxicology to elucidate the critical target genes

Changing the structure of PFOA and PFOS: a chemical industry strategy or a solution to avoid thyroid-disrupting effects?

BACKGROUND: The family of perfluoroalkyl and polyfluoroalkyl substances (PFAS) raised concern for their proven bioaccumulation and persistence in the environment and animals as well as for their hazardous health effects. As a result, new congeners of PFAS have rapidly replaced the so-called "old long-chain PFAS" (mainly PFOA and PFOS), currently out-of-law and banned by most countries. These compounds derive from the original structure of "old long-chain PFAS", by cutting or making little conformational changes to their structure, thus obtaining new molecules with similar industrial applicatio

Evaluation of ecological impacts with ferrous iron addition in constructed wetland under perfluorooctanoic acid stress.

In this study, ferrous ion (Fe(II)) had the potential to promote ecological functions in constructed wetlands (CWs) under perfluorooctanoic acid (PFOA) stress. Concretely, Fe(II) at 30 mg/L and 20-30 mg/L even led to 11.37% increase of urease and 93.15-243.61% increase of nitrite oxidoreductase respectively compared to the control. Fe(II) promotion was also observed on Nitrosomonas, Nitrospira, Azospira, and Zoogloea by 1.00-6.50 folds, which might result from higher expression of nitrogen fixation and nitrite redox genes. These findings could be explanation for increase of ammonium removal by

Selective pressure of PFOA on microbial community: Enrichment of denitrifiers harboring ARGs and the transfer of ferric-electrons.

Perfluorooctanoic acid (PFOA), a class of permanent organic pollutants, is frequently detected in surface and ground water, with the latter made up primarily of porous media (such as soils, sediments, and aquifers) that harbor microbial communities. Therefore, we investigated the effects of PFOA on water ecosystems and found that, under stimulation by 2.4 μM PFOA, denitrifiers were significantly enriched due to their hosting antibiotic resistant genes (ARGs), which were 1.45 times more abundant than the control. Furthermore, denitrifying metabolism was enhanced by Fe(II) electron donation. Spe

Investigation on removal of perfluorooctanoic acid (PFOA), perfluorooctane sulfonate (PFOS), perfluorohexane sulfonate (PFHxS) using water treatment sludge and biochar.

This work assessed the adsorption performance of three common PFAS compounds (PFOA, PFOS and PFHxS) on two water treatment sludges (WTS) and two biochars (commercial biomass biochar and semi-pilot scale biosolids biochar). Of the two WTS samples included in this study, one was sourced from poly-aluminium chloride (PAC) and the other from alum (Al2(SO4)3). The results of experiments using a single PFAS for adsorption reinforced established trends in affinity - the shorter-chained PFHxS was less adsorbed than PFOS, and the sulphates (PFOS) were more readily adsorbed than the acid (PFOA). Adverse effects of gestational exposure to hexafluoropropylene oxide trimer acid (HFPO-TA) homologs on maternal, fetal, and placental health in mice.

In an effort to identify and develop potential alternatives for perfluorooctanoic acid (PFOA), PFDMO2HpA and PFDMO2OA have been engineered by reducing the -CF2 content in the molecular structure of hexafluoropropylene oxide trimer acid (HFPO-TA). Yet, despite their subsequent presence in environmental samples, there is a paucity of information regarding their toxicity, particularly on pregnancy. Here, pregnant CD-1 mice were exposed to PFDMO2HpA (0, 0.04, 0.16, 0.63, 2.5, or 10 mg/kg/day) or PFDMO2OA (0, 0.01, 0.04, 0.16, 0.63, or 2.5 mg/kg/day) via oral gavage from gestational days 2 (GD2) to
