Disrupted Information Flow in Resting-State in Adolescents With Sports Related Concussion.

Children and youths are at a greater risk of concussions than adults, and once injured, take longer to recover. A key feature of concussion is an increase in functional connectivity, yet it remains unclear how changes in functional connectivity relate to the patterns of information flow within resting state networks following concussion and how these relate to brain function. We applied a data-driven measure of directed effective brain connectivity to compare the patterns of information flow in healthy adolescents and adolescents with subacute concussion during the resting state condition. Dat

Controversies in enhanced recovery after cardiac surgery.

Advances in cardiac surgical operative techniques and myocardial protection have dramatically improved outcomes in the past two decades. An unfortunate and unintended consequence is that 80% of the preventable morbidity and mortality following cardiac surgery now originates outside of the operating room. Our hope is that a renewed emphasis on evidence-based best practice and standardized perioperative care will reduce overall morbidity and mortality and improve patient-centric care. The Perioperative Quality Initiative (POQI) and Enhanced Recovery After Surgery-Cardiac Society (ERAS® Cardiac)

Is not fasting before cardiac catheterisation better than fasting? A systematic integrative literature review.

AIM: To synthesise international evidence about not fasting before cardiac catheterisation. METHODS: We used a systematic, integrative literature review and applied quality assessment criteria. RESULTS: Nine of 1,535 articles met the inclusion criteria. Critique and analysis of the literature revealed three themes: (1) Fasting before coronary angiography and angioplasty is associated with perceived risk management, not actual risk management. (2) Not fasting before coronary angiography and angioplasty is considered safe and beneficial for the patient when compared to the risks of fasting. (3

Dysregulation of lipids in Alzheimer's disease and their role as potential biomarkers.

The brain is highly enriched in lipids, and an intensive study of these lipids may be informative, not only of normal brain function but also of changes with age and in disease. In recent years, the development of highly sensitive mass spectrometry platforms and other high-throughput technologies has enabled the discovery of complex changes in the entire lipidome. This lipidomics approach promises to be a particularly useful tool for identifying diagnostic biomarkers for early detection of age-related neurodegenerative disease, such as Alzheimer's disease (AD), which has till recently been lim

Mitochondrial dysfunction: A promising therapeutic target for liver diseases.

The liver is an important metabolic and detoxification organ and hence demands a large amount of energy, which is mainly produced by the mitochondria. Liver tissues of patients with alcohol-related or non-alcohol-related liver diseases contain ultrastructural mitochondrial lesions, mitochondrial DNA damage, disturbed mitochondrial dynamics, and compromised ATP production. Overproduction of mitochondrial reactive oxygen species induces oxidative damage to mitochondrial proteins and mitochondrial DNA, decreases mitochondrial membrane potential, triggers hepatocyte inflammation, and promotes prog

ATP8A2-related disorders as recessive cerebellar ataxia.

ATP8A2-related disorders are autosomal recessive conditions that associate encephalopathy with or without hypotonia, psychomotor delay, abnormal movements, chorea, tremor, optic atrophy and cerebellar atrophy (CARMQ4). Through a multi-centric collaboration, we identified six point mutations (one splice site and five missense mutations) involving ATP8A2 in six individuals from five families. Two patients from one family with the homozygous p.Gly585Val mutation had a milder presentation without encephalopathy. Expression and functional studies of the missense mutations demonstrated that protein

Revisiting Alzheimer's Disease Through the Somatostatin-Mitochondria Axis.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cognitive decline and pathological hallmarks such as amyloid-beta (Aβ) deposition and tau hyperphosphorylation. Increasing evidence identifies mitochondrial dysfunction as an early and central driver of AD pathogenesis, contributing to impaired bioenergetics, oxidative stress, disrupted calcium homeostasis, and synaptic failure. Notably, somatostatin (SST), a neuropeptide significantly reduced in AD brains, has emerged as a candidate regulator of neuronal metabolism and mitochondrial homeostasis. In this revi

A patient-centric approach to neuro-recovery after acute brain injuries.

BACKGROUND: Patients discharged after acute brain injuries require ongoing medical care to support recovery and treat secondary neurologic complications. Most therapeutic trials for interventions after acute brain injuries use measures of disability (i.e., the Modified Rankin Scale) as primary outcomes, but systematically collecting these outcomes as part of clinical care remains challenging. In addition, understanding patients' perspectives on recovery is critical to providing personalized care and ultimately improving outcomes. METHODS: The Post-ICU Neurorecovery clinic at a tertiary care h

Use of metformin and outcome of patients with newly diagnosed glioblastoma: Pooled analysis.

Metformin has been linked to improve survival of patients with various cancers. There is little information on survival of glioblastoma patients after use of metformin. We assessed the association between metformin use and survival in a pooled analysis of patient data from 1,731 individuals from the randomized AVAglio, CENTRIC and CORE trials. We performed multivariate Cox analyses for overall survival (OS) and progression-free survival (PFS) comparing patients' use of metformin at baseline and/or during concomitant radiochemotherapy (TMZ/RT). Further exploratory analyses investigated the effe

A Survey of the Management of Patients with Temporomandibular Disorders by General Dental Practitioners in Southern Brazil.

PURPOSE: Temporomandibular disorders (TMD) are recognized as one of the most controversial topics in dentistry, despite the fact that both basic science and clinical researchers have currently reached some degree of consensus. This study aimed to conduct a questionnaire-based survey about the management of TMD patients by general dental practitioners (GDPs). MATERIALS AND METHODS: One hundred fifty-one GDPs with a private practice in a city of southern Brazil were included, independent of school of origin, gender, graduation year, and curriculum content. All participants were administered a q

Development of a lead foil crown delineation technique for implant rehabilitations to generate patient specific finite element model of occlusal loading points.

Understanding the clinical biomechanical basis of dental implant supported functional rehabilitation of edentulous jaws improves precision, longevity and overall success of a planned treatment. Stress distribution pattern around dental implants is an important determinant for rate of bone resorption around them. During planning the treatment for most prosthetic rehabilitations, the surgeon uses a software to virtually plan the dimension, position and angulation of the implants considering only the quantity of available bone in the area of interest but does not usually consider the strain gener

Dental wear caused by association between bruxism and gastroesophageal reflux disease: a rehabilitation report.

Bruxism is a pathological activity of the stomatognathic system that involves tooth grinding and clenching during parafunctional jaw movements. Clinical signs of bruxism are mostly related to dental wear and muscular and joint discomforts, but a large number of etiological factors can be listed, as local, systemic, psychological and hereditary factors. The association between bruxism, feeding and smoking habits and digestive disorders may lead to serious consequences to dental and related structures, involving dental alterations (wear, fractures and cracks), periodontal signs (gingival recessi

Interfacial and colloidal drivers of wine drying astringency: the role of wine-saliva aggregates.

Astringency in red wine is associated with lubrication changes driven by tannin-saliva interactions, yet the physicochemical basis of astringency subqualities remains unclear. Focusing on drying, a key quality-related subquality, we examined whether wine-saliva aggregates can contribute to lubrication stability rather than only increasing friction. Red wines spanning non-drying to highly drying profiles were mixed 1:1 with unstimulated human saliva. Film thickness and friction were measured under controlled, oral-inspired sliding conditions, and mixtures were centrifuged to obtain soluble (sup

The regulatory role of m6A RNA methylation in insect sexual development: a review.

N6-methyladenosine (m6A) is the most prevalent internal mRNA modification in eukaryotes and serves as a key post-transcriptional regulator in insect development. This review synthesizes current knowledge on m6A in insect sexual development, beginning with its well-established role in Drosophila melanogaster, where it ensures female fate by promoting the female-specific splicing of the master switch gene Sex-lethal (Sxl). Notably, classical female-lethal genes such as Fl(2)d and Virilizer are now recognized as essential regulatory subunits of the m6A methyltransferase complex, linking classic g

Comparative Analysis of Mouse CSF Proteins in Experimental Autoimmune Encephalomyelitis Model and Cuprizone-Induced Demyelination Model by Olink Proteomics.

Multiple sclerosis (MS) is marked by an autoimmune-driven inflammatory response within the central nervous system (CNS), resulting in demyelination and associated neurological impairment. To investigate the pathways involved in MS-related neuroinflammation, we performed cerebrospinal fluid (CSF) proteomic profiling (Olink®) across two murine models, i.e., the experimental autoimmune encephalomyelitis (EAE) and the cuprizone (CPZ)-induced demyelination models. Out of the 92 proteins analyzed, five (CCL2, CCL3, EDA2R, Fas, and HGF) were consistently dysregulated in both models, highlighting shar

Amygdala-Centered Emotional Processing in Prolonged Grief Disorder: Relationship With Clinical Symptomatology.

BACKGROUND: Prolonged grief disorder (PGD) is a multidimensional condition with adverse health consequences. We hypothesized that enhanced negative emotional bias characterizes this disorder and underlies its key clinical symptoms. METHODS: In a cross-sectional design, chronically grieving older adults (age 61.5 ± 8.9 years) experiencing probable PGD (n = 33) were compared with demographic- and time since loss-equated integrated (adaptive) grief participants (n = 38). To probe generalized negative affective reactivity, participants performed an emotional face-matching task during functional m
