C-reactive protein and chitinase 3-like protein 1 as biomarkers of spatial redistribution of retinal blood vessels on digital retinal photography in patients with diabetic retinopathy.

The aim of the study was to investigate the correlation between the levels of C-reactive protein (CRP) and chitinase 3-like protein 1 (YKL-40) in blood samples with morpohometric parameters of retinal blood vessels in patients with diabetic retinopathy. Blood laboratory examination of 90 patients included the measurement of glycemia, HbA1C, total cholesterol, LDL-C, HDL-C, triglycerides and CRP. Levels of YKL-40 were detected and measured in serum by ELISA (Micro VueYKL-40 EIA Kit, Quidel Corporation, San Diego, USA). YKL-40 correlated positively with diameter and negatively with number of ret

The chitinolytic enzymes from the entomopathogenic fungus Metarhizium anisopliae: GH family distribution, domain architecture, and protein regulation.

Metarhizium anisopliae is an entomopathogenic fungus that is widely used in the biological control of agricultural pests. During host infection, M. anisopliae secretes an arsenal of hydrolytic enzymes such as proteases and chitinases that allow for cuticle penetration and host colonization. The degradation of chitin into N-acetylglucosamine (GlcNAc) monomers is carried out by chitinases and β-N-acetylglucosaminidases (NAGases). Chitin-degrading enzymes of M. anisopliae have been the subject of extensive research; however, these studies have not been consolidated into a comprehensive review. Th

Eosinophilic globules (syn. hyaline inclusions) and protein crystalloids in the nasal mucosa of rats treated with synthetic amorphous silica, an unspecific chitinase-like-protein-positive background lesion.

In a 13-week inhalation toxicity study with three recovery periods (3, 6, and 12 months), Crl: WI rats were allocated to nine groups, each containing 25 animals per sex. Eight groups were treated daily by inhalation with the test items at concentrations of 0.5, 1.0, 2.5, or 5.0 mg/m3 (SAS 1 groups 2, 3, 4, or 5, respectively; SAS 2 groups 6, 7, 8, or 9, respectively). Controls (group 1) were treated with air only. In nasal cavities, the major lesions consisted of increased eosinophilic globules and chitinase-3-like-protein-positive crystalloids* in the nasal mucosa, mainly in nasal cavity leve

Exploring the properties of chitinolytic Bacillus isolates for the pathogens biological control.

Plant fungal diseases generate serious losses in the agriculture. The bacteria producing biologically active substances that inhibit the growth of fungal pathogens can be an alternative to the chemicals. The chitinolytic bacteria were isolated from the rhizosphere of wheat (Triticum aestivum L.) and their physiological properties which may be useful in the promotion of plant growth have been investigated. Their chitinases and antifungal activity were studied. The isolates were also tested for indirect growth-promoting traits such as ammonia production, siderophore production, hydrogen cyanide

Glycosylhydrolase genes control respiratory tubes sizes and airway stability.

Tight barriers are crucial for animals. Insect respiratory cells establish barriers through their extracellular matrices. These chitinous-matrices must be soft and flexible to provide ventilation, but also tight enough to allow oxygen flow and protection against dehydration, infections, and environmental stresses. However, genes that control soft, flexible chitin-matrices are poorly known. We investigated the genes of the chitinolytic glycosylhydrolase-family 18 in the tracheal system of Drosophila melanogaster. Our findings show that five chitinases and three chitinase-like genes organize the

Asymmetry in Herbivore Effector Responses: Caterpillar Frass Effectors Reduce Performance of a Subsequent Herbivore.

Multiple species of phytophagous insects may co-occur on a plant and while plants can defend themselves from insect herbivory, plant responses to damage by different species and feeding guilds of insects may be asymmetric. Plants can trigger specific responses to elicitors/effectors in insect secretions altering herbivore performance. Recently, maize chitinases present in fall armyworm (FAW, Spodoptera frugiperda) frass were shown to act as effectors suppressing caterpillar-induced defenses in maize while increasing caterpillar performance. We investigated the effect of frass chitinase-mediate

Structural insights into the catalytic mechanism of SsChiA from Sporisorium scitamineum.

Sugarcane smut, caused by the biotrophic fungus Sporisorium scitamineum, represents a devastating global threat to sugarcane production. During fungal infection, secreted chitinases are involved not only in cell wall remodeling and nutrient acquisition, but also in the suppression of host immune recognition, making chitinases attractive targets for antifungal drug development. Despite their importance, the catalytic and inhibitory mechanisms underlying chitinases from S. scitamineum remain poorly understood. Here, we report the crystal structure of the S. scitamineum chitinase SsChiA at a reso

Multifunctional transcription factor CytR of Vibrio cholerae is important for pathogenesis.

Vibrio cholerae, the Gram-negative facultative pathogen, resides in the aquatic environment and infects humans and causes diarrhoeagenic cholera. Although the environment differs drastically, V. cholerae thrives in both of these conditions aptly and chitinases play a vital role in their persistence and nutrient acquisition. Chitinases also play a role in V. cholerae pathogenesis. Chitinases and its downstream chitin utilization genes are regulated by sensor histidine kinase ChiS, which also plays a significant role in pathogenesis. Recent exploration suggests that CytR, a transcription factor

Effects of disease-modifying drugs on serum neurofilament light chain, chitinase-3-like-1 protein levels, and selected plasmacytoid dendritic cell biomarkers in relapsing-remitting multiple sclerosis.

BACKGROUND: Multiple sclerosis (MS) is a neurodegenerative central nervous system disorder causing axonal damage and disability. Relapses develop over hours or days and then subside over weeks. Disease-modifying drugs (DMDs) influence disease activity. Interferon beta-1A (IFN-b-1A) is a widely used first-line treatment for relapsing remitting MS (RRMS) that reduces central nervous system inflammation. Fingolimod affects lymphocyte trafficking. B-cell therapy (rituximab) depletes circulating CD20+ B cells in cerebrospinal fluid, but their specific effects in RRMS remain limited. OBJECTIVE: The

Unique posttranslational modifications of chitin-binding lectins of Entamoeba invadens cyst walls.

Entamoeba histolytica, which causes amebic dysentery and liver abscesses, is spread via chitin-walled cysts. The most abundant protein in the cyst wall of Entamoeba invadens, a model for amebic encystation, is a lectin called EiJacob1. EiJacob1 has five tandemly arrayed, six-Cys chitin-binding domains separated by low-complexity Ser- and Thr-rich spacers. E. histolytica also has numerous predicted Jessie lectins and chitinases, which contain a single, N-terminal eight-Cys chitin-binding domain. We hypothesized that E. invadens cyst walls are composed entirely of proteins with six-Cys or eight-

Chitinase 3-like protein 2 (CHI3L2, YKL-39) activates phosphorylation of extracellular signal-regulated kinases ERK1/ERK2 in human embryonic kidney (HEK293) and human glioblastoma (U87 MG) cells.

Human cartilage chitinase 3-like protein 2 (CHI3L2, YKL-39) is secreted by articular chondrocytes, also synoviocytes, lung, and heart. Increased levels of YKL-39 have been demonstrated in synovial fluids of patients with rheumatoid or osteoarthritis as well as in some other pathologies and in malignant tumors, particularly in glioblastomas. It belongs to glycosyl hydrolase family 18 and the most closely related to human cartilage glycoprotein 39 (HC gp-39 or chitinase 3-like protein 1, CHI3L1 or YKL-40), which as it was shown previously, promotes the growth of human synovial cells as well as s

Laterally Transferred Gene Recruited as a Venom in Parasitoid Wasps.

Parasitoid wasps use venom to manipulate the immunity and metabolism of their host insects in a variety of ways to provide resources for their offspring. Yet, how genes are recruited and evolve to perform venom functions remain open questions. A recently recognized source of eukaryotic genome innovation is lateral gene transfer (LGT). Glycoside hydrolase family 19 (GH19) chitinases are widespread in bacteria, microsporidia, and plants where they are used in nutrient acquisition or defense, but have previously not been known in metazoans. In this study, a GH19 chitinase LGT is described from th

The role of the chi1 gene from the endophytic bacteria Serratia proteamaculans 336x in the biological control of wheat take-all.

Take-all, a disease caused by the fungus Gaeumannomyces graminis var. tritici, is the most important root disease of wheat and causes severe yield losses worldwide. Using microorganisms as biological agents to control the disease is important because no resistant cultivars or effective chemical fungicides are available. In this study, we tested the biological control capability of a chitinase produced by the endophytic bacterium Serratia proteamaculans 336x against wheat take-all. The chitinase gene chi1 of S. proteamaculans 336x was cloned and heterologously expressed in Escherichia coli. The

High levels of human chitotriosidase hinder the formation of peritrophic membrane in anopheline vectors.

In the Anopheles midgut, Plasmodium falciparum produces a specific chitinase able to penetrate the blood meal surrounding the chitin-containing peritrophic membrane (PM). High levels of an analogous chitinase, chitotriosidase (CHIT), may be found in human blood, being the markers of macrophage activation. To verify the hypothesis that CHIT present in malaria patient blood could help parasite to overcome PM, we carried out a bioassay by feeding Anopheles stephensi females on an artificial apparatus that contained human blood from four different sources and with different chitinase concentration

Expression pattern of glycoside hydrolase genes in Lutzomyia longipalpis reveals key enzymes involved in larval digestion.

The sand fly Lutzomyia longipalpis is the most important vector of American Visceral Leishmaniasis. Adults are phytophagous (males and females) or blood feeders (females only), and larvae feed on solid detritus. Digestion in sand fly larvae has scarcely been studied, but some glycosidase activities putatively involved in microorganism digestion were already described. Nevertheless, the molecular nature of these enzymes, as the corresponding genes and transcripts, were not explored yet. Catabolism of microbial carbohydrates in insects generally involves β-1,3-glucanases, chitinases, and digesti

Comparison of local and systemic induction of acquired disease resistance in cucumber plants treated with benzothiadiazoles or salicylic acid.

The accumulation of chitinase and its involvement in systemic acquired disease resistance was analyzed using acibenzolar-S-methyl and salicylic acid (SA). Resistance against scab (pathogen: Cladosporium cucumerinum) and the accumulation of chitinase were rapidly induced in cucumber plants after treatment with acibenzolar-S-methyl. In contrast, SA protected the plants from C. cucumerinum and the accumulation of chitinase was induced only on the treated leaves. The accumulation of chitinase in response to inoculation with the pathogen was induced more rapidly in cucumber plants previously treate

Fungal chitin from asthma-associated home environments induces eosinophilic lung infiltration.

Development of asthma and allergic inflammation involves innate immunity, but the environmental contributions remain incompletely defined. Analysis of dust collected from the homes of asthmatic individuals revealed that the polysaccharide chitin is environmentally widespread and associated with β-glucans, possibly from ubiquitous fungi. Cell wall preparations of Aspergillus isolated from house dust induced robust recruitment of eosinophils into mouse lung, an effect that was attenuated by enzymatic degradation of cell wall chitin and β-glucans. Mice expressing constitutively active acidic mamm

Fungal endo and exochitinase production, characterization, and application for Candida biofilm removal.

Chitinases are promising enzymes for a multitude of applications, including chitooligosaccharide (COS) synthesis for food and pharmaceutical uses and marine waste management. Owing to fungal diversity, fungal chitinases may offer alternatives for chitin degradation and industrial applications. The rapid reproduction cycle, inexpensive growth media, and ease of handling of fungi may also contribute to reducing enzyme production costs. Thus, this study aimed to identify fungal species with chitinolytic potential and optimize chitinase production by submerged culture and enzyme characterization u

Effects of dietary transgenic poplar leaf pellets on performance and tissues in rabbits.

BACKGROUND: Various studies have been conducted to evaluate the effect of Bt crops on animals. Insect-resistant bioassays have revealed that the chitinase-BmkIT combination could be used as a new pest-resistant gene source and might be a complementary alien gene source to the Bt toxin gene. So it is necessary to assay the effects of chitinase-BmkIT plants on animals. RESULTS: Forty 40-day old New Zealand White Rabbits (Oryctolagus cuniculus) were fed for 60 consecutive days with formulated feed containing dehydrated poplar (Populus cathayana Rehd) leaves harbouring chitinase-BmkIT gene combin

Intraspecies variation in the Kanzawa spider mite differentially affects induced defensive response in lima bean plants.

The Kanzawa spider mite, Tetranychus kanzawai, is a polyphagous herbivore that feeds on various plant families, including the Leguminacae. Scars made by the mite on lima bean leaves (Phaseolus lunatus) were classified into two types: white and red. We obtained two strains of mites--"White" and "Red"--by selecting individual mites based on the color of the scars. Damage made by the Red strain induced the expression of genes for both basic chitinase, which was downstream of the jasmonic acid (JA) signaling pathway, and acidic chitinase, which was downstream of the salicylic acid (SA) signaling p
