Association between circadian syndrome and frailty progression among middle-aged and older adults in China: A longitudinal study based on CHARLS.

BACKGROUND: Frailty is a significant predictor of adverse outcomes. Circadian syndrome (CircS), a cluster of cardiometabolic risk factors linked to circadian disruption, may interact with frailty; however, evidence for this relationship remains limited. METHODS: A total of 7330 participants from the China Health and Retirement Longitudinal Study (CHARLS) were included. CircS was defined by seven components. Frailty was measured by the frailty index (FI). Group-based trajectory modeling (GBTM) was employed to identify different frailty trajectories. Multinomial logistic regression models were

An Automated Home-Cage System to Assess Learning and Performance of a Skilled Motor Task in a Mouse Model of Huntington's Disease.

Behavioral testing is a critical step in assessing the validity of rodent models of neurodegenerative disease, as well as evaluating the efficacy of pharmacological interventions. In models of Huntington's disease (HD), a gradual progression of impairments is observed across ages, increasing the need for sensitive, high-throughput and longitudinal assessments. Recently, a number of automated systems have been developed to perform behavioral profiling of animals within their own home-cage, allowing for 24-h monitoring and minimizing experimenter interaction. However, as of yet, few of these hav

Comparative utility of instruments for monitoring sleepiness-related performance decrements in the operational environment.

As both military and commercial operations increasingly become continuous, 24-h-per-day enterprises, the likelihood of operator errors or inefficiencies caused by sleep loss and/or circadian desynchrony also increases. Avoidance of such incidents requires the timely application of appropriate interventions--which, in turn, depend on the ability to measure and monitor the performance capacity of individuals in the operational environment. Several factors determine the potential suitability of candidate measures, including their relative sensitivity, reliability, content validity, intrusiveness

Circadian rhythm disruption induces myopia in mice.

Emerging evidence suggests a link between circadian rhythm and myopia, with studies showing that retinal-specific knockout of the clock gene Bmal1 induces myopia in mice. This study aims to elucidate the relationship between circadian rhythm disruption (CRD) and myopia, and to investigate the potential mechanisms underlying CRD-mediated myopia development. Three-week-old C57BL/6J mice were entrained to a 12 h light/12 h dark (12L/12D) photoperiod for one week. Mice were then randomly assigned to the 12L/12D control group and the CRD groups. CRD groups were divided into two subgroups: Chronic j

Neuroregulation of histamine of circadian rhythm disorder induced by chronic intermittent hypoxia.

Obstructive sleep apnea (OSA) is characterized by intermittent hypoxemia, sleep fragmentation, and excessive daytime sleepiness. OSA patients are at an elevated risk for circadian rhythm disturbances. Histamine is known to regulate the sleep-wake cycle predominantly via histamine H1 receptors. We utilized a C57BL/6 mouse model exposed to chronic intermittent hypoxia (CIH) for three weeks to assess alterations in circadian rhythmicity. Sleep architecture and voluntary wheel-running activity were evaluated. Additionally, c-fos expression and mPer2 levels in the frontal cortex (FC) and the suprac

Rest-activity disturbances in children with septo-optic dysplasia characterized by actigraphy and 24-hour plasma melatonin profiles.

INTRODUCTION: A trial of melatonin treatment in children with septo-optic dysplasia (SOD) and sleep disruption is accepted clinical practice in many centers. However, no objective measurements of sleep/activity patterns with 24-h melatonin profiles have been published for these individuals, and the pathophysiological basis underlying sleep disorders in SOD remains largely unknown. METHODS: We studied six children with rest-activity disturbances and SOD. All wore an Actiwatch-Mini (a noninvasive method of detecting and recording movement intensity) for 2 wk and were admitted to hospital for a

Diurnal physiology: core principles with application to the pathogenesis, diagnosis, prevention, and treatment of myocardial hypertrophy and failure.

The circadian system has been shown to be fundamentally important in human health and disease. Recently, there have been major advances in our understanding of daily rhythmicity, and its relevance to human physiology, and to the pathogenesis and treatment of cardiac hypertrophy and heart failure. Cardiovascular tissues, such as heart and blood vessels, show remarkable daily variation in gene expression, metabolism, growth, and remodeling. Moreover, synchrony of daily molecular and physiological rhythms is integral to healthy organ growth and renewal. Disruption of these rhythms adversely affec

Long-term dysregulation of circadian and 17-beta estradiol-induced LH, prolactin and corticosterone secretion after dimethylbenz (a) anthracene administration in the Sprague-Dawley female rat.

A single intragastric administration of 7,12-dimethylbenz (a) anthracene (DMBA) has been shown, when given at 55-60 days of age, to induce mammary tumors in young cycling female Sprague-Dawley rats. The appearance of the tumors is preceded by a series of neuroendocrine disturbances of the hypothalamo-pituitary-gonadal (HPG) axis, including attenuation of the preovulatory Luteinizing Hormone (LH) and Gonadotropin-Releasing Hormone (GnRH) release and amplification of the preovulatory 17beta-Estradiol (E(2)) surge. In this study, we examined the hypothesis that a single administration of DMBA cou

Circadian neuroendocrine functions in disorders of eating behavior.

Eating behavior is a complex function determined by regulatory mechanisms characterized by bioperiodic fluctuations. It involves the hypothalamus as well as the related higher centers in the central nervous system (CNS). Many hormones, neurotransmitters and neuropeptides play an important role in the synchronization of food intake. Our study therefore sets out to evaluate the circadian rhythms of several endocrine functions in women with eating disorders, to clarify the pathophysiology of the limbic-hypothalamic system. We measured the circadian rhythms of plasma melatonin, serum cortisol, gro

Association between sleep disturbance and metabolic dysfunctions in adipose tissue: Insights into melatonin's role.

The increased prevalence of sleep disturbances in modern society is frequently linked to various metabolic disorders, including insulin resistance, obesity, hypertension, fatty liver disease, and cardiometabolic complications. Melatonin, a pineal gland-secreted neurohormone, plays a pivotal role in maintaining the circadian rhythm. It is involved in regulating adipose tissue development, lipid accumulation, browning of white adipose tissue, and activation of brown adipose tissue. The adipose tissue is a dynamic endocrine organ that secretes hormones and cytokines. Recent research has highlight
