Generic Protocols for the Analytical Validation of Next-Generation Sequencing-Based Circulating Tumor DNA Assays, Version 2.0.

Circulating tumor DNA (ctDNA) is a promising biomarker for cancer detection, treatment selection, and disease monitoring. As a result, an increasing number of diagnostic developers are pursuing liquid biopsy assays using next-generation sequencing (NGS) to detect ctDNA. To support clinical adoption, these assays must undergo rigorous analytical validation (AV) to facilitate regulatory approval, support market entry, and ultimately improve outcomes for patients with cancer. To this end, the BLOODPAC AV Working Group developed and published a set of generic AV protocols in September 2020. These

A Disposable CRISPR-Nanozyme Electrochemical Biosensor for Rapid and Sensitive Detection of Breast Cancer Circulating Tumor DNA.

Circulating tumor DNA (ctDNA) serves as a promising next-generation biomarker for noninvasive cancer screening and monitoring. In this work, we report an electrochemical biosensor for ctDNA detection. The sensor is constructed based on the synergistic integration of CRISPR/Cas12a and PB-Au NPs. CRISPR/Cas12a provides precise target recognition and triggers trans-cleavage, while the nanozyme enables strong signal amplification through its catalytic activity. Using a disposable carbon-fiber paper as the biosensing interface, we developed this sensitive detection strategy. On this interface, a PB

NeoCircle: pre- and post-operative circulating tumor DNA dynamics predicts survival in neoadjuvant-treated early breast cancer.

Persistent circulating tumor DNA (ctDNA) during neoadjuvant treatment (NAT) of early breast cancer (EBC) indicates high-risk disease. Similarly, detection of ctDNA post-resection indicates molecular residual disease (MRD) and impending relapse. For ctDNA to be integrated into EBC management, accessible and scalable diagnostics are required. Here we apply an ultrasensitive, personalized tumor-informed approach to ctDNA evaluation predicated on analyses of structural variants (SVs) using a novel digital PCR (dPCR) multiplex SV technology. 136 patients eligible for NAT (29.4% TNBC, 44.9% HR+ /HER

Interim assessment by circulating tumor DNA in primary mediastinal large B-cell lymphoma: a multicenter LYSA study.

Primary mediastinal large B-cell lymphoma (PMBL) achieves excellent outcomes with dose-dense immunochemotherapy, yet response assessment by positron emission tomography (PET) remains limited. In this prospective multicenter observational study, we evaluated the clinical relevance of circulating tumor DNA (ctDNA) minimal residual disease (MRD) in patients with newly diagnosed PMBL and assessed whether MRD enhances outcome discrimination beyond PET. Plasma and PET images were collected at baseline and after 2 and 4 cycles. ctDNA and tumor biopsy were analyzed by high-depth, error-corrected seque

Peritoneal Metastases in Stage II-III Colon Cancer Represent a Blind Spot for Circulating Tumor DNA Surveillance.

While circulating tumor DNA (ctDNA) may guide adjuvant therapy in stage II-III colon cancer, detection largely hinges on systemic tumor shedding. In a secondary analysis of three prospective ctDNA studies, peritoneal metastases occurred in up to 9.4% of patients, were predominantly isolated, and were enriched among ctDNA-negative recurrences. Molecular and clinical data demonstrate reduced ctDNA levels in peritoneum-only disease, identifying a surveillance blind spot with direct implications for high-risk patients.

Optimization of bile preparation for liquid biopsy in cholangiocarcinoma focusing on circulating tumor DNA and protein stability.

Liquid biopsy using bile offers a promising non-invasive approach for molecular analysis in cholangiocarcinoma (CCA). However, the stability of key biomarkers, such as proteins and circulating DNA (ctDNA), at room temperature has not been fully elucidated. This study investigates the temporal stability of proteins and ctDNA in bile samples under room temperature conditions to optimize pre-analytical handling for molecular diagnostics. Bile samples were collected from six patients diagnosed with CCA. Protein concentrations, enzyme activity (E-Cadherin and N-Cadherin), and mutant KRAS ctDNA leve

Circulating Tumor DNA in Addition to Fecal Immunochemical Test in a Dual-Test Colorectal Cancer Screening Approach.

BACKGROUND: Early detection is paramount when reducing incidence and mortality of colorectal cancer (CRC). Current population-based screening programs primarily use fecal immunochemical test (FIT) to allocate individuals for colonoscopy although low specificity challenges colonoscopy capacities. We aimed to assess the potential of circulating tumor (ct)DNA markers for early CRC detection in a dual-test CRC screening approach among FIT positive individuals. METHODS: Plasma samples from 774 FIT positive (≥100 ng Hemoglobin/mL) individuals from the Danish CRC screening program were analyzed for

Self-signal electrochemical identification of circulating tumor DNA employing poly-xanthurenic acid assembled on black phosphorus nanosheets.

A self-signal electrochemical identification interface was prepared for the determination of circulating tumor DNA (ctDNA) in peripheral blood based on poly-xanthurenic acid (PXTA) assembled on black phosphorus nanosheets (BPNSs) acquired through simple ultrasonication method. The BPNSs with large surface area could be integrated with the xanthurenic acid (XTA) monomers by right of physisorption, and hence improved the electropolymerization efficiency and was beneficial to the enlargement of the signal response of PXTA. The assembled PXTA/BPNSs composite with attractive electrochemical activit
