Picomolar SARS-CoV-2 Neutralization Using Multi-Arm PEG Nanobody Constructs.

Multivalent antibody constructs have a broad range of clinical and biotechnological applications. Nanobodies are especially useful as components for multivalent constructs as they allow increased valency while maintaining a small molecule size. We here describe a novel, rapid method for the generation of bi- and multivalent nanobody constructs with oriented assembly by Cu-free strain promoted azide-alkyne click chemistry (SPAAC). We used sortase A for ligation of click chemistry functional groups site-specifically to the C-terminus of nanobodies before creating C-to-C-terminal nanobody fusions

High Yield Branched Puromycin Linker Design Enables Efficient cDNA Display and Chemical Modification of Peptides.

Peptide chemical modification is a valuable technique for improving peptide stability and bioactivity, particularly in drug discovery applications. Here, we report the development of a novel linker junction strategy using strain-promoted azide-alkyne cycloaddition (SPAAC) that enables the efficient formation of branched puromycin linkers with an average yield of 97%. This approach represents an improvement over traditional Michael-Addition methods, which typically yield ~15%-20%. The high yield of the SPAAC reaction and the near absence of by-products make the linkage by click reaction easy to

Tagging lipoproteins in mycobacteria via metabolic incorporation of modified fatty acids.

Lipoproteins are lipid-modified proteins and critical components of the mycobacterial cell envelope with diverse functions related to virulence and survival. In Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis, lipoproteins account for ∼3 % of the Mtb proteome, yet their precise functions remain poorly characterized. Prior experimental efforts have addressed the validation and preliminary functional annotation of specific lipoproteins. Whereas these studies elucidated aspects of lipoprotein localization and involvement in host-pathogen interactions, the methodologies emplo

UV- and Visible-Light Photopatterning of Molecular Gradients Using the Thiol-yne Click Reaction.

The rational design of chemical coatings is used to control surface interactions with small molecules, biomolecules, nanoparticles, and liquids as well as optical and other properties. Specifically, micropatterned surface coatings have been used in a wide variety of applications, including biosensing, cell growth assays, multiplexed biomolecule interaction arrays, and responsive surfaces. Here, a maskless photopatterning process is studied, using the photocatalyzed thiol-yne "click" reaction to create both binary and gradient patterns on thiolated surfaces. Nearly defect-free patterns are prod

Recent Advances in Copper-Based Solid Heterogeneous Catalysts for Azide-Alkyne Cycloaddition Reactions.

The copper(I)-catalyzed azide-alkyne cycloaddition (CuAAC) reaction is considered to be the most representative ligation process within the context of the "click chemistry" concept. This CuAAC reaction, which yields compounds containing a 1,2,3-triazole core, has become relevant in the construction of biologically complex systems, bioconjugation strategies, and supramolecular and material sciences. Although many CuAAC reactions are performed under homogenous conditions, heterogenous copper-based catalytic systems are gaining exponential interest, relying on the easy removal, recovery, and reus

Post-Functionalization of Organometallic Complexes via Click-Reaction.

CuAAC (Cu catalyzed azide-alkyne cycloaddition) click-reaction is a simple and powerful method for the post-synthetic modification of organometallic complexes of transition metals. This approach allows the selective introduction of additional donor sites or functional groups to the periphery of the ligand environment. This is especially important if a metalloligand with free donor sites, which are of the same nature as the primary site for the coordination of the primary metal, has to be created. The concept of post-synthetic modification of organometallic complexes by click-reaction is relati

Thymol as an Interesting Building Block for Promising Fungicides against Fusarium solani.

The semisynthesis of 15 new thymol derivatives was achieved through Williamson synthesis and copper-catalyzed azide-alkyne cycloaddition (CuAAC) approaches. The reaction of CuAAC using the "Click Chemistry" strategy, in the presence of an alkynyl thymol derivative and commercial or prepared azides, provided nine thymol derivatives under microwave irradiation. This procedure reduces reaction time and cost. All molecular entities were elucidated by 1H and 13C NMR, IR, and HRMS data. These derivatives were evaluated in vitro for their fungicidal activity against Fusarium solani sp. Among the nine

[Molecular Conjugation for the "Diazido Probe" Method].

Target identification of screening hit compounds with unknown mechanisms of action obtained from chemical libraries by phenotypic assays has played an important role in the development of innovative drugs that work based on novel mechanisms. To improve the usability of the target identification based on the photoaffinity labeling method, we have studied the "diazido probe" method, wherein a photoreactive aromatic azido and relatively photostable aliphatic azido groups are sequentially used for photoreaction and introduction of a latent detectable tag via the Staudinger ligation or click reacti

Exploiting Activated Alkyne Chemistry for the Synthesis of Tailored Amphiphilic Janus Dendrimers with Aggregation-Induced Emission and Their Assemblies for Cell Imaging.

Amphiphilic Janus dendrimers (AJDs) are a promising class of macromolecules capable of self-assembling into well-defined nanostructures. In this study, AJDs were synthesized employing, for the first time, the activated alkyne-hydroxyl "click" chemistry approach, enabling the construction of dendritic architectures under mild conditions. The AJDs were functionalized with a π-extended tetraphenylethylene (TPE) derivative, imparting aggregation-induced emission (AIE) property. These AJDs self-assembled into fluorescent nanostructures, including micelles and dendrimersomes, with tunable dimensions

Development of a Long-Acting Myeloid-Derived Growth Factor via Site-Specific PEGylation.

Extracellular myeloid-derived growth factor (MYDGF) can improve organ repair. However, short in vivo half-life hampers its therapeutic application. Herein, we developed a long-acting MYDGF via site-specific PEGylation at its C-terminus. Bacterially overexpressed human MYDGF carrying a C-terminal Asn-Ala-Leu tripeptide motif was first ligated with a synthetic azido-functionalized Gly-Ile-Gly-Lys(N3) tetrapeptide linker via catalysis of [G238 V]BmAEP1, an engineered bamboo-derived asparaginyl endopeptidase (AEP)-type peptide ligase. Thereafter, the azido-functionalized MYDGF was efficiently conj

Visualization of Respiratory Syncytial Virus RNA Synthesis Sites by Ethynyl Uridine Labeling.

Modified cell-permeable nucleosides such as 5-bromouridine 5'-triphosphate (BrUTP) or 5-ethynyluridine (5-EU), together with the powerful click chemistry, have been widely used to specifically label newly synthetized RNA in cells and tissues in a simple two-step approach. The use of cellular transcription inhibitors has established metabolic RNA labeling as an optimal approach for the precise visualization of viral RNA transcripts and the microscopic analysis of their distribution in infected cells. Targeted delivery of liposomal chemoimmunotherapy for cancer treatment.

Chemoimmunotherapy that utilizes the immunomodulatory effect of chemotherapeutics has shown great promise for treating poorly immunogenic solid tumors. However, there remains a significant room for improving the synergy between chemotherapy and immunotherapy, including the efficient, concurrent delivery of chemotherapeutics and immunomodulators into tumors. Here, we report the use of metabolic glycan labeling to facilitate cancer-targeted delivery of liposomal chemoimmunotherapy. 4T1 triple-negative breast cancer cells can be metabolically labeled with azido groups for subsequently targeted co

Copper(ii) complexes of N-propargyl cyclam ligands reveal a range of coordination modes and colours, and unexpected reactivity.

The coordination chemistry of N-functionalised cyclam ligands has a rich history, yet cyclam derivatives with pendant alkynes are largely unexplored. This is despite the significant potential and burgeoning application of N-propargyl cyclams and related compounds in the creation of diversely functionalised cyclam derivatives via copper-catalysed azide-alkyne 'click' reactions. Herein we describe single crystal X-ray diffraction and spectroscopic investigations of the coordination chemistry of copper(ii) complexes of cyclam derivatives with between 1 and 4 pendant alkynes. The crystal structure

Development of inhibitor-directed enzyme prodrug therapy (IDEPT) for prostate cancer.

Prostate cancer (PCa) is the second most common cause of cancer death among American men after lung cancer. Unfortunately, current therapies do not provide effective treatments for patients with advanced, metastatic, or hormone refractory disease. Therefore, we seek to generate therapeutic agents for a novel PCa treatment strategy by delivering a suicide enzyme (yCDtriple) to a cell membrane bound biomarker found on PCa cells (prostate-specific membrane antigen (PSMA)). This approach has resulted in a new PCa treatment strategy reported here as inhibitor-directed enzyme prodrug therapy (IDEPT)

Imaging-Based High-Content Screening with Clickable Probes Identifies XPB Inhibitors.

High-content screening (HCS) has become a powerful tool in drug discovery; however, its reliance on indirect readouts and surrogate markers limits HCS's ability to directly assess drug-protein interactions at endogenous levels, particularly in subcellular contexts. Here, we report an approach to address these limitations by combining confocal imaging-based HCS and bio-orthogonal labeling with clickable probes. As a proof-of-concept, we synthesized a probe Triptolide-alkyne (TL-alk) that rapidly and specifically labels xeroderma pigmentosum type B (XPB), a critical protein in nucleotide excisio

Pyranthiones/Pyrones: "Click and Release" Donors for Subcellular Hydrogen Sulfide Delivery and Labeling.

Hydrogen sulfide (H2 S), one of the most important gasotransmitters, plays a critical role in endogenous signaling pathways of many diseases. However, developing H2 S donors with both tunable release kinetics and high release efficiency for subcellular delivery has been challenging. Here, we describe a click and release reaction between pyrone/pyranthiones and bicyclononyne (BCN). This reaction features a release of CO2 /COS with second-order rate constants comparable to Strain-Promoted Azide-Alkyne Cycloaddition reactions (SPAACs). Interestingly, pyranthiones showed enhanced reaction rates co

Metalloenzyme-Mediated Thiol-Yne Addition Towards Photoisomerizable Fluorescent Dyes.

Proteins are able to irreversibly assemble biologically active ligands from building blocks bearing complementary reactive functions due their spatial proximity, through a kinetic target-guided synthetic process (also named in situ click chemistry). Although linkages thus formed are mostly passive, some of them have shown to significantly contribute to the protein binding through for instance hydrogen bonding and stacking interactions. Biocompatible reactions and click chemistry are a formidable source of inspiration for developing such new protein-directed ligations. This study reports a prox

Calcium Bistriflimide-Mediated Sulfur(VI)-Fluoride Exchange (SuFEx): Mechanistic Insights toward Instigating Catalysis.

We report a mechanistic investigation of calcium bistriflimide-mediated sulfur(VI)-fluoride exchange (SuFEx) between sulfonyl fluorides and amines. We determine the likely pre-activation resting state─a calcium bistriflimide complex with ligated amines─thus allowing for corroborated calculation of the SuFEx activation barrier at ∼21 kcal/mol, compared to 21.5 ± 0.14 kcal/mol derived via kinetics experiments. Transition state analysis revealed: (1) a two-point calcium-substrate contact that activates the sulfur(VI) center and stabilizes the leaving fluoride and (2) a 1,4-diazabicyclo[2.2.2]octa
