Induction of interleukin 8 by American cockroach allergens from human airway epithelial cells via extracellular signal regulatory kinase and jun N-terminal kinase but not p38 mitogen-activated protein kinase.

BACKGROUND: Cockroaches are potent aeroallergens associated with asthma. Several reports suggest that a novel group of G protein-linked receptors, protease-activated receptors (PARs), may be involved in the intracellular signaling pathway induced by aeroallergens of the epithelial cells. OBJECTIVE: To investigate the mechanisms of American cockroach allergens (CraA) on interleukin 8 (IL-8) in human pulmonary epithelial cells. METHODS: Protease activities of CraA were quantified by the Azocoll method. The gene and protein expressions of IL-8 from CraA-stimulated A549 cells were quantified by

Getting the Most Out of Your Zombie: Abdominal Sensors and Neural Manipulations Help Jewel Wasps Find the Roach's Weak Spot.

The parasitoid emerald jewel wasp (Ampulex compressa) subdues the American cockroach (Periplaneta americana) with a sting to the 1st thoracic ganglion, followed by a sting to the roach's brain, causing long-term pacification. The wasp then leads the cockroach to a hole where it lays an egg on the roach middle leg before barricading the entrance and departing. Although many aspects of the wasp's initial attack have been investigated, few studies have detailed the egg-laying process and the subsequent fate of the larvae. Here I show that larval survival depends on precise egg positioning on the

Succinate-driven methyl thiazolyl tetrazolium (MTT) reduction is coupled to mitochondrial electron transport chain (ETC) activity: comparative evidence from subcellular fractionation in the lobster cockroach (Nauphoeta cinerea) and zebrafish (Danio rerio).

The tetrazolium-based MTT assay is widely used to assess cellular metabolic activity, yet the subcellular origin and mechanistic basis of MTT reduction remain incompletely defined. Here, we investigated the localization and biochemical determinants of MTT reductive activity using subcellular fractionation of lobster cockroach Nauphoeta cinerea head tissue and zebrafish Danio rerio brain, combined with substrate specificity, protein-dependence analysis, and mitochondrial electron transport chain (ETC) inhibition. Succinate-driven MTT reduction was predominantly localized to mitochondrial fracti

Harmonia axyridis ladybug hypersensitivity in clinical allergy practice.

The imported Harmonia axyridis ladybug infests homes in northern West Virginia from fall through spring, causing allergic disease. Retrospective single-practice chart reviews were performed: (1) all skin prick tests (1400 included ladybug) in a community allergy practice over 4 years and (2) clinical analysis of 400 randomly chosen patients. The usual adult aeroallergen skin test panel included ladybug and 57 other allergens. Statistics used were contingency table analyses and the kappa-statistic for concordance. Home infestation with ladybugs was most common in rural areas but did not predict

[Blatella germanica as a sensiting factor, and cause of allergic rhinitis in a group of our patients].

Most up-to-data data from the literature, as well as our experience gained during the routine clinical practice, led us to investigate the incidence of sensibilization on the cockroaches, as well as its role in development of allergic rhinitis. The study included 46 patients with chronic rhinitis. Out of that number, 11 were sensibilized to cockroach extract, while in another 8 senzibilized patients, allergic rhinitis, induced by the same allergen, was evidenced as well. According to this study, we have concluded that in our conditions, cockroach is to be considered significant factor of sensi

Indoor insect allergens are potent inducers of experimental eosinophilic esophagitis in mice.

EE is an emerging disease reported in children and adults of urbanized countries, where indoor insect allergens are major health risk factors. Review of our hospital patient database uncovered that a number of EE patients have hypersensitivity to indoor cat, dog, cockroach, and dust mite allergens. We tested the hypothesis whether inhaled indoor insect allergens are effective inducers of experimental EE. We delivered cat, dog, cockroach, and dust mite allergen extracts intranasally to wild-type and eotaxin-1/2-, CCR3-, and IL-5-deficient mice. Interestingly, wild-type mice exposed to cockroach

Determinants of allergen concentrations in apartments of asthmatic children living in public housing.

There is growing evidence linking poor housing conditions and respiratory diseases, including asthma. The association between housing conditions and asthma in the inner city has been attributed in part to cockroach and mouse infestation and the resulting allergen exposures. Multiple social and behavioral factors can influence environmental exposures and health conditions, necessitating a thorough examination of such factors. As part of the Healthy Public Housing Initiative, we evaluated the association between physical and household characteristics and pest-related allergen levels in three pub

Neuron-specific monoclonal antibodies raised against the low molecular weight fraction of a brain homogenate of the pond snail Lymnaea stagnalis immunoreact with neurons in the central nervous system of the cockroach, the guppy, the wall lizard, the rat and man.

Monoclonal antibodies were raised against the small molecular weight fraction (less than 30 kilodaltons) of an extract from 200 central nervous systems (CNS) of the freshwater snail Lymnaea stagnalis. In a first screening step the supernatants of the 297 emerging hybridomas were immunocytochemically tested on sections of the CNS of L. stagnalis. Sixty-six appeared to produce neuron-specific antibodies, five reacted with non-neuronal elements. In a second step the 66 neuron-specific antibodies were tested on sections of the CNS of the guppy. Three reacted positively. In the third step the three

Kappa-bungarotoxin blocks an alpha-bungarotoxin-sensitive nicotinic receptor in the insect central nervous system.

Snake venom kappa-neurotoxins are selective antagonists of nicotinic acetylcholine responses in avian, murine and bovine neurons, and have been used as probes for functionally defined vertebrate neuronal nicotinic receptors. The actions of kappa-bungarotoxin, a kappa-neurotoxin, have now been examined at a central invertebrate nicotinic receptor. kappa-Bungarotoxin is a potent antagonist (IC50 = 100 nM) of nicotinic responses, producing a long-lasting blockade of insect nicotinic acetylcholine receptors. The blockade appears to be competitive, and voltage-clamp experiments on an identified coc

Surveillance study of vector species on board passenger ships, risk factors related to infestations.

BACKGROUND: Passenger ships provide conditions suitable for the survival and growth of pest populations. Arthropods and rodents can gain access directly from the ships' open spaces, can be carried in shiploads, or can be found on humans or animals as ectoparasites. Vectors on board ships may contaminate stored foods, transmit illness on board, or, introduce diseases in new areas. Pest species, ship areas facilitating infestations, and different risk factors related to infestations were identified in 21 ferries. METHODS: 486 traps for insects and rodents were placed in 21 ferries. Archives of

Hemimetabolous genomes reveal molecular basis of termite eusociality.

Around 150 million years ago, eusocial termites evolved from within the cockroaches, 50 million years before eusocial Hymenoptera, such as bees and ants, appeared. Here, we report the 2-Gb genome of the German cockroach, Blattella germanica, and the 1.3-Gb genome of the drywood termite Cryptotermes secundus. We show evolutionary signatures of termite eusociality by comparing the genomes and transcriptomes of three termites and the cockroach against the background of 16 other eusocial and non-eusocial insects. Dramatic adaptive changes in genes underlying the production and perception of pherom
