7-Methylxanthine methyltransferase of coffee plants. Gene isolation and enzymatic properties.

Caffeine is synthesized through sequential three-step methylation of xanthine derivatives at positions 7-N, 3-N, and 1-N. However, controversy exists as to the number and properties of the methyltransferases involved. Using primers designed on the basis of conserved amino acid regions of tea caffeine synthase and Arabidopsis hypothetical proteins, a particular DNA fragment was amplified from an mRNA population of coffee plants. Subsequently, this fragment was used as a probe, and four independent clones were isolated from a cDNA library derived from coffee young leaves. Upon expression in Esch

Coffee and Parkinson's disease: associations by CYP1A2 genotype and sex in UK Biobank.

Coffee consumption has been associated with Parkinson's disease (PD), but findings remain inconsistent. We examined whether this association differs by CYP1A2 genotype and sex in 435,551 UK Biobank participants free of PD at baseline. During a median follow-up of 15.7 years, 3319 incident PD cases were identified. In overall analyses, coffee intake was not associated with PD risk. However, coffee intake significantly interacted with CYP1A2 genotype. Among AA carriers, coffee intake below 5 cups/day was associated with lower PD risk (HR 0.83, 95% CI 0.74-0.93), and spline analyses indicated a n

Coffee Consumption is Associated with a High Prevalence of Headache in a Taiwanese Population Study.

The relationship between coffee consumption and headache remains controversial. While caffeine has both analgesic and vasoconstrictive properties, excessive intake is associated with a higher prevalence of headache. This cross-sectional study investigated the association between coffee consumption and headache in a large Taiwanese cohort. Data were obtained from 27,109 participants aged 30-70 years from the Taiwan Biobank. Headache status and coffee consumption patterns, including type, frequency, and daily intake, were assessed via structured questionnaires. Multivariable logistic regression

Chlorantraniliprole: bioactivation in coffee seedlings and control of resistant Leucoptera coffeella populations.

BACKGROUND: The diamide chlorantraniliprole stands out for its high performance in controlling agricultural pests, with high selectivity and reduced environmental impact. In coffee cultivation, this insecticide has been widely employed for the management of the coffee leaf miner (Leucoptera coffeella Guérin-Méneville, 1842). However, intensive use has favored the selection of resistant populations, compromising control efficacy and intensifying biotic stress in plants. This study aimed to evaluate the morphophysiological effects of chlorantraniliprole on coffee seedlings (Coffea arabica L., cv

EDTA enabled chelation for stability and sensory quality in iron enriched coffee.

The research aims to unveil the iron fortification of coffee with chelating agents for reducing iron-polyphenol complexation. Among fortificants, Ferrous Sulphate (FS) exhibited complexation of 1.78 mM FeCl₃/TAE due to Fe2+ and was selected for further evaluation. Chelators considerably decreased complexation by 89-94%; with EDTA as the competitive ligand due to preferential chelation at 1:1 of FS-FC:EDTA ratio (0.18 mM FeCl₃/TAE). FTIR showed attenuated FeO phenolic stretching (500-600 cm-1), dynamic light scattering revealed stable colloidal aggregation (-33.10 to -39.33 mV and 231 to 376 d.

Application of Microbial Fermentation in Caffeine Degradation and Flavor Modulation of Coffee Beans.

Coffee is one of the most widely consumed beverages worldwide, primarily due to the stimulating effects attributed to its caffeine content. However, excessive intake of caffeine results in negative effects, including palpitations, anxiety, and insomnia. Therefore, low-caffeine coffee has captivated growing consumer interest, highlighting its significant market potential. Traditional decaffeination methods often lead to non-selective extraction, resulting in a loss of desirable flavor compounds, thereby compromising coffee quality. In recent years, microbial fermentation has emerged as a promis

Insights into acrylamide and furanic compounds in coffee with a focus on roasting methods and additives.

Roasting is necessary for bringing out the aroma and flavor of coffee beans, making coffee one of the most consumed beverages. However, this process also generates a series of toxic compounds, including acrylamide and furanic compounds (5-hydroxymethylfurfural, furan, 2-methylfuran, 3-methylfuran, 2,3-dimethylfuran, and 2,5-dimethylfuran). Furthermore, not much is known about the formation of these compounds in emerging coffee formulations containing alcohol and sugars. Therefore, this study investigated the effect of roasting time and degree on levels of acrylamide and furanic compounds in ar

Functional properties and sensory profile of coffee prepared by different brewing methods.

Coffee is one of the main sources of antioxidants in the diet of many countries. The purpose of this research was to assess the effect of different brewing methods, particle size, and coffee quality on the total phenolic content, antioxidant capacity (AC), and sensory profile of the beverage. The brewing methods yielded differences in taste with higher bitterness in immersion methods. However, the main factors that influenced coffee extraction and taste were particle size and coffee type. A finer particle size allowed for greater phenolic and caffeine (CA) extraction (2.82 mg GA/mL; 1.01 mg CA
