Association of genetically predicted levels of circulating blood lipids with coronary artery disease incidence.

BACKGROUND AND AIMS: Estimating the genetic risk of coronary artery disease (CAD) is now possible by aggregating data from genome-wide association studies (GWAS) into polygenic risk scores (PRS). Combining multiple PRS for specific circulating blood lipids could improve risk prediction. Here, we sought to evaluate the performance of PRS derived from CAD and blood lipids GWAS to predict the incidence of CAD. METHODS: This study included individuals aged between 40 and 69 from UK Biobank. We conducted GWAS for blood lipids measured by nuclear magnetic resonance in individuals without lipid-lowe

Sex differences in circulating metabolites across glycemic status and risk of coronary heart disease.

BACKGROUND: Women with type 2 diabetes (T2D) have a 50% excess risk of coronary heart disease (CHD) than men with T2D. We compared circulating metabolites and their associations with CHD in men and women across glycemic status. METHODS: We used metabolomic data (lipoproteins, fatty acids, amino acids, glycolysis, ketones, inflammation, and fluid balance) for 87,326 CHD-free UK Biobank participants. We used linear regressions to examine the association of sex and metabolites (log) in newly diagnosed T2D (diagnosis<2 yrs from baseline), prediabetes (A1c 5.7-6.5%), and euglycemia, accounting for

Circulating CD31 and resistin levels reflect different stages of coronary atherosclerosis in patients with psoriasis.

Psoriasis is a skin disease with a complicated pathophysiology that includes an extensive inflammatory cytokine network. Nevertheless, the relationship between psoriasis severity, cytokine levels, and coronary artery atherosclerosis remains poorly understood. Our aim was to find serum markers as potential candidates for cardiovascular disease (CVD) risk monitoring in patients with psoriasis. Therefore, we examined coronary artery atherosclerosis via coronary computed tomography angiography (CCTA), serum cytokine levels via multiple immunoassays, and the patients' psoriasis state. Our findings

Deoxycholic Acid, a Metabolite of Circulating Bile Acids, and Coronary Artery Vascular Calcification in CKD.

BACKGROUND: Vascular calcification is common among patients with chronic kidney disease (CKD), and it is associated with all-cause and cardiovascular disease mortality. Deoxycholic acid, a metabolite of circulating bile acids, is elevated in CKD and induces vascular mineralization and osteogenic differentiation in animal models. STUDY DESIGN: Cohort analysis of clinical trial participants. SETTING & PARTICIPANTS: 112 patients with moderate to severe CKD (estimated glomerular filtration rate, 20-45mL/min/1.73m2) who participated in a randomized controlled study to examine the effects of phosp

Incremental diagnostic value of circulating pentraxin in patients with intermediate risk of coronary artery disease.

OBJECTIVES: Pentraxins are a superfamily of multifunctional conserved proteins, some of which are components of the humoral arm of innate immunity and behave as functional ancestors of antibodies. They are divided into short (C reactive protein) and long pentraxins (pentraxin 3; PTX3). We investigated the diagnostic values of systematic arterial and coronary sinus (PTX3) in developing prediction models for estimating pretest probability of coronary artery disease (CAD) among an intermediate-risk population of patients with chronic stable angina. DESIGN: Cross-sectional analysis. SETTING: Ref

Predictive value of the decrease in circulating dendritic cell precursors in stable coronary artery disease.

DCs (dendritic cells) are present in atherosclerotic lesions leading to vascular inflammation, and the number of vascular DCs increases during atherosclerosis. Previously, we have shown that the levels of circulating DCPs (DC precursors) are reduced in acute coronary syndromes through vascular recruitment. In the present study, we have investigated whether DCP levels are also reduced in stable CAD (coronary artery disease). The levels of circulating mDCPs (myeloid DCPs), pDCPs (plasmacytoid DCPs) and tDCP (total DCPs) were investigated using flow cytometry in 290 patients with suspected stable

Decreased number of circulating plasmacytoid dendritic cells in patients with atherosclerotic coronary artery disease.

BACKGROUND: Dendritic cells are potent antigen-presenting and immune modulating cells that have been implicated in the development of atherosclerosis. In human blood, two distinct lineages are distinguished: plasmacytoid dendritic cells and myeloid dendritic cells. Although dendritic cells have been described in atherosclerotic plaques, no information exists concerning circulating blood dendritic cells in atherosclerosis. This study aims to evaluate the number of circulating dendritic cells in patients with coronary artery disease. The relation with the extent of coronary artery disease, the c

[Serum inflammatory related cytokines and circulating endothelial cells in patients with acute coronary syndrome].

OBJECTIVE: To study the number of CECs in patients with acute myocardial infarction (AMI) and unstable angina (UA), and to investigate its relationship with inflammatory related cytokines. METHODS: 37 patients with AMI, 12 patients with UA, and 42 health controls were studied. CECs were isolated from peripheral blood by using of immunomagnetic beads coated with antibodies against CD146. Their endothelial origin was confirmed by the positive labelling of von Willebrand Factor (vWF), CD31 and electron microscope. Annexin V-FITC/PI kit was used to measure the apoptosis of CECs. Circulating levels of proinflammatory cytokines and neutrophil-platelet aggregates in patients with coronary artery disease.

Several lines of evidence indicate that increased inflammatory activity in peripheral blood is associated with the acute coronary syndrome. Systemic inflammation in clinically stable conditions of coronary artery disease has been less studied. We examined cytokine profiles in 20 patients who had acute coronary syndrome, 45 who had angiographically verified coronary artery disease and stable angina pectoris, and 45 healthy controls. Circulating levels of C-reactive protein, interleukin-1 receptor antagonist, interleukin-2 receptor, interleukin-6, interleukin-10, and interleukin-18 were determin

Clinical and pathologic features of obstructive disease in the predominant right and left coronary circulations in man.

The clinical features and the location and severity of obstructive coronary artery disease are contrasted in 98 patients with predominant left and 99 patients with predominant right coronary circulations. A significantly higher incidence of ventricular conduction disturbances and a greater incidence and severity of obstructive coronary artery disease (greater than or equal to 70% cross-sectional narrowing in the proximal left anterior descending, circumflex and right coronary arteries and their major branches) distinguish the predominant left from the predominant right coronary circulation. TH1/TH2 imbalance, measured by circulating and intracytoplasmic inflammatory cytokines--immunological alterations in acute coronary syndrome and stable coronary artery disease.

To describe how peripheral immune-parameters reflect the inflammatory alterations of the atherosclerotic plaques in coronary atherosclerosis. We measured general inflammatory markers C-reactive protein (CRP) and granulocyte activity, lymphocyte subpopulations and their state of activation, evaluated circulating Th1/Th2-type cytokines, and specific intracytoplasmic cytokines. We investigated the association of immune-parameters with disease outcome and mortality. Thirty-three patients with acute coronary syndrome (ACS), 62 with stable coronary artery disease (CAD) and 58 healthy controls were s

The C-reactive protein-albumin-lymphocyte index is associated with coronary collateral circulation in patients with chronic total occlusion.

OBJECTIVE: Well-developed coronary collateral circulation (CCC) protects the myocardium from ischemia, but its development varies among individuals. The C-reactive protein-albumin-lymphocyte (CALLY) index is a composite marker of nutritional and inflammatory status. This study evaluated its association with well-developed CCC in patients with chronic total occlusion (CTO). METHODS: We included 369 patients with stable angina pectoris and CTO in at least one major epicardial coronary artery. The CALLY index was calculated from serum albumin, lymphocyte, and C-reactive protein levels. Patients

The Relationship Between Collateral Circulation and Electrocardiographic Frontal QRS-T Angle in Patients with Coronary Artery Chronic Total Occlusion.

Background: Ischemic heart disease is the primary contributor to global mortality. The QRS-T angle at the anterior aspect of the heart serves as a significant biomarker of the heterogeneity in myocardial repolarization and the electrophysiological instability of the cardiac myocytes. A wide frontal QRS-T angle is associated with proximal vascular disease, coexistence of three-vessel disease, and increased mortality. Hereby, we aimed to examine the relationship between collateral circulation and frontal QRS-T angle in patients with chronic total occlusion (CTO). Methods: A cohort comprising 120

The relationship between diastolic pressure and coronary collateral circulation in patients with stable angina pectoris and chronic total occlusion.

BACKGROUND: The most important biomechanical source of activation of the coronary collateral circulation (CCC) is increased tangential fluid shear stress at the arterial endothelial surface. The coronary circulation is unique in that most coronary blood flow occurs in diastole. Consequently, the diastolic blood pressure (DBP) may influence the tangential fluid shear stress on the arterial endothelial surface in diastole, therebyaffecting development of the CCC. METHODS: To investigate this, we conducted a study of 222 patients with stable angina pectoris and chronic total occlusion of coronar

Growth factors in the collateral circulation of chronic total coronary occlusions: relation to duration of occlusion and collateral function.

BACKGROUND: Despite extensive animal experimental evidence, there are few data on the relation of growth factors and collateral function in humans. METHODS AND RESULTS: In 104 patients with a chronic total coronary occlusion (CTO; >2 weeks' duration), collateral function was assessed invasively during recanalization by intracoronary Doppler and pressure recordings. A collateral resistance index, R(Coll), was calculated. Blood samples were drawn from the distal coronary bed supplied by the collaterals and from the aortic root to measure basic fibroblast growth factor (bFGF), monocytic chemotac

Cryosurgical ablation of left parietal wall accessory atrioventricular connections through the coronary sinus without the use of extracorporeal circulation.

A simplified technique for surgical treatment of preexcitation syndrome caused by left-sided accessory atrioventricular connections without the use of extracorporeal circulation has been elaborated experimentally and used in 21 patients with tachycardia refractory to medical treatment. Cryoablation of the accessory bundles was performed by means of local cryothermia using a specially designed cryoinstrument introduced into the coronary sinus through the right atrial wall. The operations were performed after thoracotomy on the working closed heart. The signs of preexcitation and paroxysms of ta
