Microvascular disease not type 2 diabetes is associated with increased cortical porosity: A study of cortical bone microstructure and intracortical vessel characteristics.

INTRODUCTION: Fracture risk is elevated in type 2 diabetes (T2D) despite normal or even high bone mineral density (BMD). Microvascular disease (MVD) is a diabetic complication, but also associated with other diseases, for example chronic kidney disease. We hypothesize that increased fracture risk in T2D could be due to increased cortical porosity (Ct.Po) driven by expansion of the vascular network in MVD. The purpose of this study was to investigate associations of T2D and MVD with cortical microstructure and intracortical vessel parameters. METHODS: The study group consisted of 75 participan

Bone turnover markers are associated with higher cortical porosity, thinner cortices, and larger size of the proximal femur and non-vertebral fractures.

Bone turnover markers (BTM) predict bone loss and fragility fracture. Although cortical porosity and cortical thinning are important determinants of bone strength, the relationship between BTM and cortical porosity has, however, remained elusive. We therefore wanted to examine the relationship of BTM with cortical porosity and risk of non-vertebral fracture. In 211 postmenopausal women aged 54-94 years with non-vertebral fractures and 232 age-matched fracture-free controls from the Tromsø Study, Norway, we quantified femoral neck areal bone mineral density (FN aBMD), femoral subtrochanteric bo

Influence of Screw Diameter on the Pull-Out Resistance of Additively Manufactured Auxetic Bone Screws in Cancellous and Cortical Bone.

This study investigates the effect of screw diameter on the pull-out strength of a bone screw with a loosening-resistant auxetic structure manufactured using additive manufacturing. The variation in mechanical performance depending on screw diameter was examined experimentally and through finite element analysis in cancellous and cortical bone structures. First, screw designs with different diameters incorporating an auxetic cell structure were created in SolidWorks. The designed screws were analyzed using ANSYS Workbench finite element software, confirming that the structures exhibit a negati

Ultrasound Imaging of Cortical Bone: Cortex Geometry and Measurement of Porosity Based on Wave Speed for Bone Remodeling Estimation.

BACKGROUND: It has been suggested that ultrasound (US) imaging can be used to assess cortical bone health, which is of particular interest owing to its major role in bone mechanical stability. Intra-cortical US imaging extends B-mode imaging into bone using a dedicated image reconstruction algorithm that corrects for refraction at the bone-soft tissue interfaces. It has shown promising results in a few healthy, predominantly young adults, providing anatomical images of the cortex (periosteal and endosteal surfaces) along with estimations of US wave speed. However, its reliability in older or o

A soft diet attenuates exercise-induced increases in cortical bone density and formation in long bones in growing mice.

Bone and muscle are closely linked through functional interdependence. Training and regular exercise can improve and maintain optimal bone mass and strength. Here, we investigated the effects of exercise combined with soft versus hard diet (pellet) on long bones in mice. Four groups of male C57BL/6J mice (n = 5 per group) were studied: hard diet (pellet) with exercise, soft diet (powdered) with exercise, hard diet (pellet) without exercise, and soft diet (powdered) without exercise. Femur and tibia were analyzed using radiographic, micro-CT, and histomorphometric techniques. Exercise combined

Global Disruption of α2A Adrenoceptor Barely Affects Bone Tissue but Minimizes the Detrimental Effects of Thyrotoxicosis on Cortical Bone.

Evidence shows that sympathetic nervous system (SNS) activation inhibits bone formation and activates bone resorption leading to bone loss. Because thyroid hormone (TH) interacts with the SNS to control several physiological processes, we raised the hypothesis that this interaction also controls bone remodeling. We have previously shown that mice with double-gene inactivation of α2A- and -adrenoceptors (α2A/2C-AR-/-) present high bone mass (HBM) phenotype and resistance to thyrotoxicosis-induced osteopenia, which supports a TH-SNS interaction to control bone mass and suggests that it involves

Denosumab rapidly increases cortical bone in key locations of the femur: a 3D bone mapping study in women with osteoporosis.

Women with osteoporosis treated for 36 months with twice-yearly injections of denosumab sustained fewer hip fractures compared with placebo. Treatment might improve femoral bone at locations where fractures typically occur. To test this hypothesis, we used 3D cortical bone mapping of postmenopausal women with osteoporosis to investigate the timing and precise location of denosumab versus placebo effects in the hips. We analyzed clinical computed tomography scans from 80 female participants in FREEDOM, a randomized trial, wherein half of the study participants received subcutaneous denosumab 60

Differences in cortical bone in overloaded and underloaded femurs from ovariectomized rats: comparison of bone morphometry with torsional testing.

Osteopenia and mechanical incompetence are the defining features of osteoporosis, yet the effects of changes in bone structure on mechanical properties are not completely understood. The primary objective of this study was to determine if in a rodent model, changes in cortical bone structure, as measured by static morphometry, would correlate with functional properties, as measured by torsional testing. For this, cortical bone structure and stiffness were determined in overloaded and underloaded limbs in rats rendered ovarian hormone-deficient by ovariectomy (OVX). Rats were OVX and six weeks

The long-term effects of protein energy malnutrition in early childhood on bone age, bone cortical thickness and height.

Three groups of Ugandan children, 18 in each group, and one comparison group of 18 children were examined at 11-17 years of age. The three groups had previously been admitted for treatment of protein energy malnutrition between the ages of 8 to 15, 16 to 21 and 22 to 27 months respectively. The comparison group had not been clinically malnourished throughout the period up to 27 months of age. The children came from one tribe and from similar socio-economic background, and were individually matched on age and sex. The bone age was estimated by hand wrist radiography scored for maturity by the T

Human tibial cortical bone with high porosity in type 2 diabetes mellitus is accompanied by distinctive bone material properties.

Diabetes mellitus is a metabolic disease affecting bone tissue at different length-scales. Higher fracture risk in diabetic patients is difficult to detect with common clinical fracture risk assessment due to normal or high bone mineral density in diabetic patients. The observed higher fracture risk despite normal to high areal bone mineral density in diabetic patients points towards impaired bone material quality. Here, we analyze tibial bone from individuals with type 2 diabetes mellitus using a multiscale-approach, which includes clinical and laboratory-based bone quality measures. Tibial c

Melatonin attenuates radiation-induced cortical bone-derived stem cells injury and enhances bone repair in postradiation femoral defect model.

BACKGROUND: The healing of bone defects can be challenging for clinicians to manage, especially after exposure to ionizing radiation. In this regard, radiation therapy and accidental exposure to gamma (γ)-ray radiation have been shown to inhibit bone formation and increase the risk of fractures. Cortical bone-derived stem cells (CBSCs) are reportedly essential for osteogenic lineages, bone maintenance and repair. This study aimed to investigate the effects of melatonin on postradiation CBSCs and bone defect healing. METHODS: CBSCs were extracted from C57BL/6 mice and were identified by flow c

Adult Tph2 knockout mice without brain serotonin have moderately elevated spine trabecular bone but moderately low cortical bone thickness.

Disruption of serotonin synthesis in neurons and the periphery by knockout (KO) of mouse genes for tryptophan hydroxylases (peripheral Tph1 and neuronal Tph2) has been claimed to decrease (Tph2 KO) and increase (Tph1 KO) bone mass. In this report, adult male and female Tph2 KO mice were observed to have elevated spine trabecular bone. Female Tph2 KO mice have reduced midshaft femur cortical bone thickness. Bone mass was normal in male and female Tph1 KO mice examined as part of a Tph1/Tph2 double knockout (DKO) mouse cohort.

Anti-TGF-β improves bone architecture without detectable restoration of cortical bone lamellar nanomechanical organization in Col1a1+/- osteogenesis imperfecta.

Osteogenesis imperfecta (OI) is characterized by impaired bone mechanical integrity and increased fracture risk. Although anti-transforming growth factor beta (anti-TGF-β) treatment improves bone architecture and whole-bone mechanical properties in Col1a1+/- OI mice, its effects on tissue-scale strain behavior and local cortical mechanics remain unclear. Here, digital volume correlation (DVC), atomic force microscopy (AFM), and Raman spectroscopy were used to assess tissue-scale strain distributions, local cortical mechanical organization, and matrix composition following 8 weeks of anti-TGF-β

Type 2 diabetes is associated with higher trabecular bone density but lower cortical bone density: the Vietnam Osteoporosis Study.

UNLABELLED: It is not clear why type 2 diabetes (T2D) has an increased risk of fracture despite higher areal bone mineral density. This study showed that compared with controls, T2D patients had higher trabecular bone density but lower cortical bone density, resulting in a lower bone strength. INTRODUCTION: To define the association between type 2 diabetes and bone architecture and measures of bone strength. METHODS: The study was part of the Vietnam Osteoporosis Study, in which 1115 women and 614 men aged ≥ 30 were randomly recruited from Ho Chi Minh City. HbA1c levels were measured with an
