Inclusion Complexation of Native and Functionalized α-, β-, and γ-Cyclodextrins with PFAS: An Experimental and Molecular Simulation Study.

β-Cyclodextrin (β-CD)-based polymers have shown high adsorption capacities for removing per- and polyfluoroalkyl substances (PFAS) from drinking water. PFAS capture by these materials involves many physical and chemical processes, such as adsorption and inclusion complexation. Quantifying the underlying host-guest binding between CDs and PFAS nevertheless remains essential because it governs the primary inclusion step. Here, we investigated native and linker-modified CD-PFAS inclusion complexes in aqueous solution using isothermal titration calorimetry (ITC) and molecular dynamics (MD) simulat

Encapsulation Efficiency and Functional Stability of Cinnamon Essential Oil in Modified β-cyclodextrins: In Vitro and In Silico Evidence.

Essential oils (EOs) have good natural antioxidant and antimicrobial properties; however, their volatility, intense aroma, poor aqueous solubility, and chemical instability limit their applications in the food industry. The encapsulation of EOs in β-cyclodextrins (β-CDs) is a widely accepted strategy for enhancing EO applications. The complexation of cinnamon essential oil (CEO) with five types of β-CDs, containing different substituent groups (β-CD with primary hydroxyl, Mal-β-CD with maltosyl, CM-β-CD with carboxymethyl, HP-β-CD with hydroxypropyl, and DM-β-CD with methyl), inclusion process

A sensitive and visual molecularly imprinted fluorescent sensor incorporating CaF2 quantum dots and β-cyclodextrins for 5-hydroxymethylfurfural detection.

Calcium fluoride (CaF2) quantum dots have many applications in various fields. But there is no report on fluorescent characteristics of CaF2 quantum dots (CaF2 QDs). Here, a synthesis of multiple-color emission CaF2 QDs by changing the temperature, time and raw ratio is reported, by which the CaF2 QDs with purple, blue, green, and yellow emission can be obtained, respectively. They were characterized by X-ray diffraction (XRD) and transmission electron microscope (TEM). On this basis, a novel molecular imprinting ratiometric fluorescence sensor (MIR sensor) had been constructed based on the pr

Effects of cholesterol-loaded cyclodextrins on the rate and the quality of motility in frozen and thawed rabbit sperm.

The motility of sperm after freezing and thawing is critical for effective cryopreservation. It is known that supplementation with cholesterol-loaded cyclodextrin (CLC) improves cryosurvival of sperm in various animals. To clarify the effects of supplementation with CLC on rabbit sperm motility after freezing and thawing, rabbit sperm motility was analyzed using a computer-assisted sperm analysis system. Sperm motility with CLC supplementation was 29.4 ± 9.6% (mean ± SD), which was significantly higher than that of controls (20.8 ± 7.1%, P<0.05). The curvilinear velocity of sperm with CLC exce

Behavior of alpha-, beta-, and gamma-cyclodextrins and their derivatives on an in vitro model of blood-brain barrier.

Cyclodextrins (CDs) can be envisaged to cure some diseases related to the brain, but the behavior of these compounds toward the blood-brain barrier (BBB) remains largely unexplored to envisage such clinical applications. To fulfill this gap, the toxicity and endothelial permeability for native, methylated, and hydroxypropylated alpha-, beta-, and gamma-CDs have been studied on an in vitro model of BBB. As shown by the endothelial permeability for sucrose and immunofluorescence stainings, the native CDs are the most toxic CDs (alpha- > beta- > gamma-CD). Whereas the chemical modification of bet

Cyclodextrin Nanosponge-PVA Hybrid Networks as Supramolecular Reservoir Systems for Amitriptyline.

Poly-(vinyl alcohol) (PVA) hydrogels incorporating β-cyclodextrin (βCD) nanosponges (CDNSs) were developed as hybrid supramolecular systems for the controlled delivery of amitriptyline hydrochloride (AMT). The βCD-derived nanosponges provide host-guest cavities for drug inclusion, while the PVA network acts as a diffusion-regulating matrix. Initially, adsorption-desorption studies were performed on isolated CDNSs to evaluate their drug-loading capacity prior to incorporation into the hydrogels. The CDNSs exhibited efficient encapsulation (∼60%) and released more than 90% of the drug, confirmin

Cyclodextrin-based in situ gel systems: Their benefits and limitations for drug delivery.

The inherent limitations of conventional dosage forms, particularly for ocular and nasal administration, necessitate advanced drug delivery systems. In situ gelling systems and cyclodextrins (CDs) individually offer significant advantages, while their combination provides a synergistic platform. CD-based in situ gels combine the inclusion complexation ability of CDs with the sol-gel transition properties of stimuli-responsive polymers, enhancing the solubility, stability, and residence time of poorly soluble drugs across various mucosal routes. CDs improve drug availability and protect labile

Evaluation of bitterness in mother tinctures by an electronic tongue: impact of cyclodextrin complexation.

The pronounced bitterness of herbal mother tinctures often limits their palatability, despite their therapeutic value. This study evaluates the effectiveness of different cyclodextrins (CDs) in masking the bitterness of five herbal tinctures (marrubium, propolis, thyme, plantain and eucalyptus) using an α-Astree II electronic tongue equipped with seven potentiometric sensors. The bitterness-masking abilities of β-CD, methylated β-CD, and hydroxypropyl β- and γ-CDs (HP-β-CD and HP-γ-CD) were investigated by applying a Principal Component Analysis (PCA) to the e-tongue data. Caffeine was used as

Cyclodextrin inclusion complex enhances solubility and antimicrobial activity of chlortetracycline hydrochloride.

Chlortetracycline hydrochloride (CTC) is a broad-spectrum tetracycline antibiotic used to prevent or resist different bacterial infections. However, due to its poor aqueous stability and physicochemical instability, its clinical development is limited. To improve such properties, CTC complexation with β-cyclodextrin (β-CD) and 2-hydroxypropanol-β-cyclodextrin (HP-β-CD) was performed using different preparation techniques (Freeze drying and ball milling). Inclusion complexes were comprehensively characterized using different analytical techniques and showed different properties distinct to pure

Innovative in situ drug-releasing hydrogels are emerging as a promising therapeutic strategy for anterior segment ocular diseases, leveraging the unique anatomy of the eye. Rapamycin (RAP) is an effective immunosuppressive agent for organ transplantation; however, high hydrophobicity and low bioavailability have strongly constrained its clinical application. Chitosan (CS) is used as the backbone, and RAP can be loaded through supramolecular host-guest interactions of cyclodextrin (CD) to obtain chitosan-conjugated-(cydodextrin with 3,4-dihydroxyhrocinnamic acid) and loaded with rapamycin (CCH/

Supramolecular cyclodextrin-based reservoir as nasal delivery vehicle for rivastigmine to brain.

The purpose of this study involved the synthesis of supramolecular reservoir (i.e. cyclodextrin metal-organic framework, MOF) using cyclodextrins as building blocks, followed by cross-linking to obtain crosslinked CD framework (CDF) using CD-MOF as template and functionalized with borneol (BO) to enhance rivastigmine (RIV) permeation and facilitate brain targeting via intranasal administration. Utilizing BO modified CDF (BO-CDF) with cubic shape as a carrier for the encapsulation of RIV, a nasal RIV delivery system (RIV@BO-CDF) was fabricated. The particle size of RIV@BO-CDF was approximately

Engineered extracellular vesicles loaded in boronated cyclodextrin framework for pulmonary delivery.

Extracellular vesicles (EVs) are promising therapeutic carriers for their ideal nano-size and intrinsic biocompatibility, while rapid clearance and limited targeting ability are the major setbacks of EVs. With minimal absorption into the systemic circulation, inhalation for pulmonary disease therapy minimizes off-target toxicity to other organs and offers a safe and effective treatment for respiratory disorders. Herein, a nano-grid carrier made of boronated cyclodextrin framework (BCF) was prepared for pH/H2O2 responsive release of EVs. A novel design of cyclo (Arg-Gly-Asp-D-Tyr-Lys) peptide (

γ-Cyclodextrin hydrogel for the sustained release of josamycin for potential ocular application.

Glaucoma is the leading cause of blindness worldwide. However, its surgical treatment, in particular via trabeculectomy, can be complicated by fibrosis. In current clinical practice, application of the drug, Mitomycin C, prevents or delays fibrosis, but can lead to additional side effects, such as bleb leakage and hypotony. Previous in silico drug screening and in vitro testing has identified the known antibiotic, josamycin, as a possible alternative antifibrotic medication with potentially fewer side effects. However, a suitable ocular delivery mechanism for the hydrophobic drug to the surgic

γ-Cyclodextrin Metal-Organic Frameworks: Do Solvents Make a Difference?

Conventionally, methanol is the solvent of choice in the synthesis of gamma-cyclodextrin metal-organic frameworks (γ-CD-MOFs), but using ethanol as a replacement could allow for a more food-grade synthesis condition. Therefore, the aim of the study was to compare the γ-CD-MOFs synthesised with both methanol and ethanol. The γ-CD-MOFs were characterised by scanning electron microscopy (SEM), surface area and pore measurement, Fourier transform infrared spectroscopy (FTIR) and powder X-ray diffraction (PXRD). The encapsulation efficiency (EE) and loading capacity (LC) of the γ-CD-MOFs were also

OKL-1111, A modified cyclodextrin as a potential universal reversal agent for anticoagulants.

BACKGROUND: Antithrombotic therapy is inevitably associated with a risk for bleeding and these bleeding complications can be life-threatening. Recently, specific reversal agents were developed for the direct factor Xa and thrombin inhibitors (DOACs). However, next to the fact that these agents are relatively expensive, the use of selective reversal agents complicates treatment of bleeding patients in practice. In a series of screening experiments, we discovered a class of cyclodextrins with procoagulant properties. In this study we characterize a lead compound, OKL-1111, and demonstrate its po

Cyclodextrin-Coordinated Liposome-in-Gel for Transcutaneous Quercetin Delivery for Psoriasis Treatment.

Psoriasis is a chronic inflammatory skin disease that is difficult to treat. Quercetin (QT) is a dietary flavonoid known for its anti-inflammatory effects and safe use in humans. However, the topical application of quercetin for psoriasis treatment presents a significant challenge due to its poor water solubility and low stability in semisolid preparations, where it tends to recrystallize. This work presents a novel liposome-in-gel formulation for the quercetin-based topical treatment of psoriasis. The quercetin-loading liposomes are stabilized by hydroxypropyl-β-cyclodextrin (HPCD), which int

Synthesis and Characterization of Cyclodextrin-Based Polyhemiaminal Composites with Enhanced Thermal Stability.

Polyhemiaminal (PHA) polymers are a new class of thermosetting polymers that have recently gained attention owing to their high mechanical strength and excellent recycling behavior. However, low thermal stability is a common issue in PHA polymers due to the thermally labile crosslinked knots. Herein, crosslinked PHA polymer composites were synthesized by reacting formaldehyde with a precursor solution of 4,4'-oxydianiline (ODA) and cyclodextrins (CDs) (α-, β-, and γ-). The material obtained under optimal conditions (ODA:CD molar ratio of 1:0.5, 37% aqueous solution of formaldehyde (formalin))
