Dioscin ameliorates cardiac hypertrophy through inhibition of the MAPK and Akt/GSK3β/mTOR pathways.

Cardiac hypertrophy occurs in response to multiple stimuli and develops into congestive heart failure with morbidity and mortality. Dioscin exerts protective effects against tumor growth and ischemia/reperfusion injury. However, whether and how dioscin attenuates angiotensin II (AngII)-induced cardiac hypertrophy is still unknown. In the current study, we found that dioscin attenuated cardiac hypertrophy and restored the impaired cardiac function induced by AngII infusion in vivo. In addition, dioscin blocked the activation of the MAPK and Akt/GSK3β/mTOR pathways and nuclear accumulation of p-

Polyphyllin I alleviates neuroinflammation after cerebral ischemia-reperfusion injury via facilitating autophagy-mediated M2 microglial polarization.

Microglial activation and polarization play a central role in poststroke inflammation and neuronal damage. Modulating microglial polarization from pro-inflammatory to anti-inflammatory phenotype is a promising therapeutic strategy for the treatment of cerebral ischemia. Polyphyllin I (PPI), a steroidal saponin, shows multiple bioactivities in various diseases, but the potential function of PPI in cerebral ischemia is not elucidated yet. In our study, the influence of PPI on cerebral ischemia-reperfusion injury was evaluated. Mouse middle cerebral artery occlusion (MCAO) model and oxygen-glucos

Mechanism investigation of dioscin against CCl4-induced acute liver damage in mice.

The mechanisms of the ameliorating effects of dioscin against CCl(4) induced acute liver damage are investigated in this study. Dioscin significantly inhibited (p<0.01) the increases of serum ALT and AST activities compared with the CCl(4)-treated animals. The hepatic lipid peroxidation formation and, concentrations of TNF-α and IL-6 were also decreased. Liver histopathologic studies and a DNA laddering assay indicated that dioscin protected hepatocytes against CCl(4)-induced apoptosis and necrosis. Furthermore, dioscin decreased the protein expressions of Fas/FasL, increased Bcl-2/Bax ratio,

Protodioscin enhances Methotrexate-induced senescence and senolytic activity in HepG2 liver cancer spheroids by modulating cell cycle regulators and the TGF-β/p-Smad2-4 signaling pathway.

Protodioscin (PD) is a natural saponin with anti-inflammatory, anticancer, and metabolic regulatory properties. Senolytic agents, which selectively eliminate senescent cells, are gaining attention for enhancing cancer therapy. This study evaluated the senolytic potential of PD in 3D HepG2 spheroids subjected to Methotrexate (MTX)-induced senescence. Spheroids were generated using the hanging drop method with a 1:1 mixture of HepG2 medium and HFF1-conditioned medium. Senescence was induced with 6 µM MTX, followed by 24-h PD treatment at 1.6, 4, and 8.14 µM (10%, 25%, and 50% of IC₅₀). Cell viab

A systems biology approach reveals dual neurotherapeutic mechanisms of Dioscorea bulbifera in Alzheimer's disease via estrogen signaling and cholinergic modulation.

Alzheimer's disease (AD) is a multifactorial neurodegenerative disorder marked by cognitive decline, cholinergic dysfunction, synaptic loss, and neuroinflammation. Existing therapies such as Donepezil and estrogen replacement offer only symptomatic relief, failing to address the complexity of the disease due to their reductionist, single-targeted approach. In this study, we employed an integrative systems biology framework to evaluate the neurotherapeutic potential of Dioscorea bulbifera (DB), a core component of the US-patented polyherbal formulation BHD (comprising Bacopa monnieri, Hippophae

Dioscin activates myeloid AMPK to restore Lgr5⁺ stem cell homeostasis and promote mucosal repair in ulcerative colitis.

Ulcerative colitis (UC) is a chronic relapsing inflammatory disease with limited efficacy in achieving durable mucosal healing and preventing dysplasia. Macrophage-intestinal stem cell (ISC) crosstalk plays a central role in epithelial regeneration and tumorigenic transition but remains therapeutically underexplored. Here, we show that Dioscin, a natural steroidal saponin from Dioscoreae Rhizoma, alleviates DSS-induced acute colitis and AOM/DSS-induced chronic colitis-associated dysplasia by reprogramming macrophage-dependent ISC homeostasis. Dioscin significantly reduced disease severity, sup

Identification and characterization of several dietary alkaloids as weak inhibitors of hedgehog signaling.

The Hedgehog (Hh) signaling pathway plays an integral role in the patterning and development of diverse structures in the vertebrate embryo. Aberrations in Hh signaling are associated with a range of developmental defects including failure of interhemispheric division of the embryonic forebrain as well as midline facial dysmorphia including cleft lip/palate and cyclopia, collectively termed holoprosencephaly (HPE). Postnatally, Hh signaling has been postulated to play a pivotal role in healing and repair processes and inappropriate Hh pathway activation has been implicated in several types of

Protective effect of dioscin against thioacetamide-induced acute liver injury via FXR/AMPK signaling pathway in vivo.

Our previous works showed that dioscin, a natural product, could protect liver from acute liver damages induced by dimethylnitrosamine, ethanol, carbon tetrachloride and acetaminophen. However, the effect of dioscin on thioacetamide (TAA)-induced acute liver injury still remained unknown. The purpose of this study was to investigate whether dioscin confers a protective effect against TAA-induced acute liver injury in rats and mice. The results showed that dioscin decreased the serum levels of ALT, AST, and rehabilitated histopathological changes compared with the model groups. In addition, dio
