Glandular cell abnormalities in cervical cytology: What has changed in this decade and what has not?

The relative as well as absolute increase in cervical adenocarcinomas has brought the glandular lesions of cervix into attention of the cytologists and molecular biologists alike. Though the cytologic criteria for diagnosis of these lesions have been refined to a great extent through the evolution of the Bethesda System for reporting cervical smears, still some challenges exist regarding an accurate recognition of glandular cell changes in cervical smears and molecular tests to aid their detection. The present review is a narrative compilation of the current status of identification of cervica

Molecular epidemiology of human coronavirus OC43 reveals evolution of different genotypes over time and recent emergence of a novel genotype due to natural recombination.

Although human coronavirus OC43-OC43 (HCoV-OC43) is the coronavirus most commonly associated with human infections, little is known about its molecular epidemiology and evolution. We conducted a molecular epidemiology study to investigate different genotypes and potential recombination in HCoV-OC43. Twenty-nine HCoV-OC43 strains from nasopharyngeal aspirates, collected from 2004 to 2011, were subjected to RNA-dependent RNA polymerase (RdRp), spike, and nucleocapsid gene analysis. Phylogenetic analysis showed at least three distinct clusters of HCoV-OC43, although 10 unusual strains displayed i

Analysis of 4.3 kilobases of divergent locus B of macaque retroperitoneal fibromatosis-associated herpesvirus reveals a close similarity in gene sequence and genome organization to Kaposi's sarcoma-associated herpesvirus.

We previously identified retroperitoneal fibromatosis-associated herpesvirus (RFHV) as a simian homolog of Kaposi's sarcoma-associated herpesvirus (KSHV) in a fibroproliferative malignancy of macaques that has similarities to Kaposi's sarcoma. In this report, we cloned 4.3 kb of divergent locus B (DL-B) flanking the DNA polymerase gene from two variants of RFHV from different species of macaque with a consensus degenerate hybrid oligonucleotide primer approach. Within the DL-B region of RFHV, viral homologs of the cellular interleukin-6, dihydrofolate reductase, and thymidylate synthase genes

A trimethoprim derivative impedes antibiotic resistance evolution.

The antibiotic trimethoprim (TMP) is used to treat a variety of Escherichia coli infections, but its efficacy is limited by the rapid emergence of TMP-resistant bacteria. Previous laboratory evolution experiments have identified resistance-conferring mutations in the gene encoding the TMP target, bacterial dihydrofolate reductase (DHFR), in particular mutation L28R. Here, we show that 4'-desmethyltrimethoprim (4'-DTMP) inhibits both DHFR and its L28R variant, and selects against the emergence of TMP-resistant bacteria that carry the L28R mutation in laboratory experiments. Furthermore, antibio

Evolutionary and molecular characteristics of high-Shannon entropy codons in VP1 of Coxsackievirus A6.

Coxsackievirus A6 (CVA6) has emerged as a predominant global pathogen of hand, foot and mouth disease, yet its evolutionary and molecular characteristics remain poorly understood. This study integrated 10 year molecular surveillance data from central China (2013-2024) with global CVA6 sequences to analyse high-entropy codons in the VP1 capsid protein and their co-evolution patterns. Phylogenetic and Bayesian evolutionary analyses indicated that CVA6 strains in China primarily clustered into sub-genotype D3, with an estimated global VP1 substitution rate of 3.62×10⁻³ substitutions/site/year. Surfactant-Mediated Synthesis of Twisted Cu2-xS Nanobowties with Chirality Continuum toward Enantioselective Catalysis.

The influence of achiral surfactants on synergistically chiral effect of interfacial nucleation, assembly orientation, and morphological evolution in chiral nanostructural synthesis via a chemical solution method is largely ignored and underexplored. Herein, we report a surfactant-mediated continuous evolution of chiral Cu2-xS (0 < x < 1) from layered nanosheets to twisted nanobowties (TNBs) and uncovered a multiscale chirality amplification mechanism driven by tilted stacking of coordination nanosheets. Surfactant identity, including headgroup and alkyl chain length, controls the potential, a

Molecular distinction between true centric fission and pericentric duplication-fission.

Centromere (centric) fission, also known as transverse or lateral centric misdivision, has been defined as the splitting of one functional centromere of a metacentric or submetacentric chromosome to produce two derivative centric chromosomes. It has been observed in a range of organisms and has been ascribed an important role in karyotype evolution; however, the underlying mechanisms remain unknown. We have investigated four cases of apparent centric fission in humans. Two cases show a missing chromosome 22 or 18 that is replaced by two centric ring products, a third case shows two chromosome-

Cell and molecular biology of kidney development.

Abnormalities of kidney and urinary tract development are the most common cause of end-stage kidney failure in childhood in the United States. Over the past 20 years, the advent of mutant and transgenic mice and the manipulation of gene expression in other animal models has resulted in major advances in identification of the cellular and molecular mechanisms that direct kidney morphogenesis, providing insights into the pathophysiology of renal and urologic anomalies. This review focuses on the molecular mechanisms that define kidney progenitor cell populations, induce nephron formation within

Multiple sex chromosome drivers in a mammal with three sex chromosomes.

Eukaryotes with separate males and females display a great diversity in the way they determine sex, but it is still unclear what evolutionary forces cause transitions between sex-determining systems. Rather that the lack of hypotheses, the problem is the scarcity of adequate biological systems to test them. Here, we take advantage of the recent evolution of a feminizing X chromosome (called X∗) in the African pygmy mouse Mus minutoides to investigate one of the evolutionary forces hypothesized to cause such transitions, namely sex chromosome drive (i.e., biased transmission of sex chromosomes

Exceptional Heterogeneity in Viral Evolutionary Dynamics Characterises Chronic Hepatitis C Virus Infection.

The treatment of HCV infection has seen significant progress, particularly since the approval of new direct-acting antiviral drugs. However these clinical achievements have been made despite an incomplete understanding of HCV replication and within-host evolution, especially compared with HIV-1. Here, we undertake a comprehensive analysis of HCV within-host evolution during chronic infection by investigating over 4000 viral sequences sampled longitudinally from 15 HCV-infected patients. We compare our HCV results to those from a well-studied HIV-1 cohort, revealing key differences in the evolu

A novel nitric oxide (NO)-dependent 'molecular switch' mediates LTP in the Octopus vulgaris brain through persistent activation of nitric oxide synthase (NOS).

Cephalopod mollusks provide a striking example of the independent evolution of advanced cognitive abilities in invertebrates. In octopuses, learning and memory are mediated by the vertical lobe (VL), whose visual input synapses undergo robust activity-dependent long-term potentiation (LTP). Here, we used Octopus vulgaris of either sex to show that persistent activation of nitric oxide synthase (NOS) mediates both LTP expression and maintenance, independently of de novo protein synthesis. Contrary to the prevailing view, NO signaling is not required for LTP induction, as LTP can still be induce

Generation of Tailorable Multi-Property Molecules via Multimodal Spectral Fusion Evolution.

Molecular generation stands at the forefront of intelligent molecular design, promising transformative advances in the discovery of molecules that meet drug-likeness or catalysis requirements. However, existing generative models typically focus on optimizing a single target property at a time, hindered by the intrinsic difficulty of simultaneously encoding and evolving multiple structural features. Herein, we developed MUltimodal Spectral fusion Evolution (MUSE)-a spectroscopy-guided generative framework that unifies multimodal spectroscopic descriptors as a universal chemical language for mul

Structural transition from ordered molecular layers to disordered arrangement triggers nonmonotonic friction at n-hexadecane-graphite interfaces.

Friction is generally understood to increase with the normal load according to classical tribological theory; however, a few nanoscale studies have reported deviations from this well-known behavior. Here, we report nonmonotonic friction behavior at the n-hexadecane-graphite interface, characterized by a transition from negative to positive load dependence in single-asperity atomic force microscopy (AFM) measurements. Combined analyses of experiments and molecular dynamics (MD) simulations attribute this unusual trend to load-induced structural evolution of the confined hexadecane ordered layer

Atomistic Simulation of Loading Direction-Dependent Brittle-to-Ductile Transition in a ⟨110⟩-Oriented B2-FeAl Nanowire Under Bending.

Molecular dynamics simulation was employed to investigate the effect of loading direction on the bending behavior of a B2-FeAl nanowire. The simulations were carried out with different loading directions of [001], [112], [111] and [110] at 300 K. The results show that the FeAl nanowire yields by nucleation of dislocations in all cases, which is independent of the loading direction. However, various loading directions result in different mechanical responses and failure mechanisms. Under [001]- and [112]-bending, the nanowire fails in a brittle manner, whereas changing the loading direction to

Recombination Events and Conserved Nature of Receptor Binding Motifs in Coxsackievirus A9 Isolates.

Coxsackievirus A9 (CVA9) is an enterically transmitted enterovirus and one of the most pathogenic type among human enteroviruses. CVA9 isolates use a distinctive RGD (Arg-Gly-Asp) motif within VP1 capsid protein that defines its ability to bind to integrin receptor(s) for cellular entry. To investigate CVA9 evolution and pathogenicity, genetic relationships and recombination events were analyzed between 54 novel clinical isolates of CVA9, as well as 21 previously published full length CVA9 sequences from GenBank. Samples were investigated by partial sequencing of the novel VP1 and 3Dpol genes,

Mutational events in cefotaximase extended-spectrum beta-lactamases of the CTX-M-1 cluster involved in ceftazidime resistance.

CTX-M beta-lactamases, which show a high cefotaxime hydrolytic activity, constitute the most prevalent extended-spectrum beta-lactamase (ESBL) type found among clinical isolates. The recent explosive diversification of CTX-M enzymes seems to have taken place due to the appearance of more efficient enzymes which are capable of hydrolyzing both cefotaxime and ceftazidime, especially among the CTX-M-1 cluster. A combined strategy of in vitro stepwise evolution experiments using bla(CTX-M-1), bla(CTX-M-3), and bla(CTX-M-10) genes and site-directed mutagenesis has been used to evaluate the role of

The spiral evolution of techniques for the laboratory diagnosis of microbial infections.

The laboratory diagnosis of microbial infections has undergone a profound transformation over the past century, evolving from classical phenotypic methods to molecular genetic approaches and now toward ultra-sensitive molecular phenotyping. While this progression is often described as linear, a closer examination reveals a cyclical pattern: an initial reliance on observable biological function, followed by a shift toward genetic abstraction, and, more recently, a return to phenotypic measurement at unprecedented molecular resolution. This "diagnostic circle" reflects a deeper epistemological t

A toughening mechanism of the strain rate-optimal chain length on polymer-modified calcium silicate hydrates (CSH).

Polymers are known to effectively improve the toughness of inorganic matrices; however, the mechanism at the molecular level is still unclear. In this study, we used molecular dynamics simulations to unravel the effects and mechanisms of different molecular chain lengths of polyacrylic acid (PAA) on toughening calcium silicate hydrate (CSH), which is the basic building block of cement-based materials. Our simulation results indicate that an optimal molecular chain length of polymers contributes to the largest toughening effect on the matrix, leading to up to 60.98% increase in fracture energy.
