Insulin-like growth factor-I stimulates gonadotrophin production from eel pituitary cells: a possible metabolic signal for induction of puberty.

Insulin-like growth factor (IGF)-I has been suggested as a potential signal linking growth and puberty in mammals. Using the juvenile European eel as a model, we employed a long-term, serum-free primary culture of pituitary cells to study the direct effect of IGF-I on gonadotrophin (GtH-II=LH) production. IGF-I increased both cell content and release of GtH-II in a time- and dose-dependent manner. IGF-I and IGF-II had similar potencies but insulin was 100-fold less effective, suggesting the implication of an IGF type 1 receptor. Other growth and metabolic factors, such as basic fibroblast grow

Dietary Scutellaria baicalensis extract enhances growth, intestinal health, and resistance to Aeromonas hydrophila in juvenile Asian swamp eel (Monopterus albus).

INTRODUCTION: Bacterial infection seriously restricts the healthy culture of Monopterus albus. Scutellaria baicalensis extract (SBE) is a natural plant‑derived additive with antioxidant and anti‑inflammatory properties. This study aimed to evaluate the effects of dietary SBE supplementation on growth performance, intestinal health, gut microbiota and resistance against Aeromonas hydrophila infection in juvenile Asian swamp eel. METHODS: A 60‑day feeding trial was performed. Five diets were formulated with 0%, 0.5%, 1%, 2% and 3% SBE (CK, SBE‑A, SBE‑B, SBE‑C, SBE‑D). Growth indices, intestinal

Integrative effects of Bacillus tropicus FG2 on growth performance, immunity, gut microbiota, and metabolome in short-finned eel, Anguilla bicolor pacifica.

This study investigated the effects of dietary Bacillus tropicus FG2 supplementation at 0 (control), 107, 108, and 109 cfu g-1 diet (designated D7, D8, and D9, respectively) on growth, immunity, disease resistance, gut microbiota, and metabolomic profiles in eels. After 56 days, eels receiving the D9 diet exhibited significantly increased final body weight, weight gain, feed efficiency, and total biomass yield compared with the control and lower-dose groups, with no effect on survival. Intestinal protease, amylase and lipase, were markedly enhanced by probiotic supplementation. Immune response

Characterization and Functional Analysis of the 17-Beta Hydroxysteroid Dehydrogenase 2 (hsd17b2) Gene during Sex Reversal in the Ricefield Eel (Monopterus albus).

In mammals, 17-beta hydroxysteroid dehydrogenase 2 (Hsd17b2) enzyme specifically catalyzes the oxidation of the C17 hydroxyl group and efficiently regulates the activities of estrogens and androgens to prevent diseases induced by hormone disorders. However, the functions of the hsd17b2 gene involved in animal sex differentiation are still largely unclear. The ricefield eel (Monopterus albus), a protogynous hermaphroditic fish with a small genome size (2n = 24), is usually used as an ideal model to study the mechanism of sex differentiation in vertebrates. Therefore, in this study, hsd17b2 gene

Not your typical abdominal pain: Case report of a fisherman presenting to the trauma & emergency surgery department with intestinal perforation due to an eel fish.

BACKGROUND: Traumatic intestinal perforation by foreign bodies is rare, with cases involving live fish being exceedingly uncommon, with only one reported case to date. We present a unique case of a 55-year-old fisherman who presented to the Emergency Department with traumatic intestinal perforation due to an eel fish accidentally entering his rectum. Despite initial reluctance to seek medical attention, prompt intervention was crucial to addressing peritonitis. CASE PRESENTATION: The patient presented with severe abdominal pain and signs of peritonitis. X-ray findings confirmed pneumoperitone

RNA-seq analysis revealed the pathogenicity of Vibrio vulnificus to American eel (Anguilla rostrata) and the strategy of host anti-V. vulnificus infection.

Vibrio vulnificus is a commonly pathogenic bacterium in cultivated eels, but its pathogenicity to American eel (Anguilla rostrata) and the molecular mechanism of host anti-V. vulnificus infection remains uncertain. In this study, American eels were infected with different dose of V. vulnificus to determine the LD50. Then, bacterial load in the liver and kidney histopathology were assessed post the LD50 of V. vulnificus infection. Additionally, gene expressions of 18 immune related genes in the liver, spleen and kidney were detected. Furthermore, transcriptome sequencing and enrichment of diffe

Inoculation of Freund's adjuvant in European eel (Anguilla anguilla) revealed key KEGG pathways and DEGs of host anti-Edwardsiella anguillarum infection.

Freund's complete (FCA) and incomplete adjuvants (FIA), generally applied in subunit fishery vaccine, have not been explored on the molecular mechanism of the nonspecific immune enhancement. In this study, we examined the RNA-seq in the spleen of European eel (Anguilla anguilla) inoculated with FCA and FIA (FCIA group) to elucidate the key KEGG pathways and differential expressed genes (DEGs) in the process of Edwardsiella anguillarum infection and A. anguilla anti-E. anguillarum infection using genome-wide transcriptome. After eels were challenged by E. anguillarum at 28 d post the first inoc

Effects of luteinizing hormone-releasing hormone analog and pimozide on the release of luteinizing hormone and ovulation in artificially matured Japanese eel Anguilla japonica.

Pituitary gonadotropins, follicle-stimulating hormone (Fsh) and luteinizing hormone (Lh), control oogenesis in all vertebrates. In particular, Lh plays a key role in stimulating the final oocyte maturation and subsequent ovulation. The biosynthesis and secretion of Lh are regulated by several neurohormones, including gonadotropin-releasing hormone (GnRH) and dopamine. GnRH analogs, also known as Lh releasing hormone analogs (LHRHa), and dopamine antagonists are commonly used to induce sexual maturation in teleosts. However, the effects of these reagents differ among fish species. Therefore, in

Differential impacts of carp and salmon pituitary extracts on induced oogenesis, egg quality, molecular ontogeny and embryonic developmental competence in European eel.

Low egg quality and embryonic survival are critical challenges in aquaculture, where assisted reproduction procedures and other factors may impact egg quality. This includes European eel (Anguilla anguilla), where pituitary extract from carp (CPE) or salmon (SPE) is applied to override a dopaminergic inhibition of the neuroendocrine system, preventing gonadotropin secretion and gonadal development. The present study used either CPE or SPE to induce vitellogenesis in female European eel and compared impacts on egg quality and offspring developmental competence with emphasis on the maternal-to-z

Functional domains and amino acid residues of Japanese eel IRF1, AjIRF1, regulate its nuclear import and IFN/Mx promoter activation.

Interferon regulatory factors (IRFs) are a family of transcriptional factors capable of regulating the expression of distinct subsets of interferon (IFN)-stimulated genes by binding to their promoters. IRF1 was the first member identified for its ability to regulate the IFNβ gene and has now been revealed to exhibit remarkable functional diversity in the regulation of different cellular responses. In the present study, the IRF1 gene was identified and characterized in Japanese eel, Anguilla japonica (AjIRF1). The open reading frame of AjIRF1 was 804 bp in length, encoding a protein of 267 amin

Morpho-histochemical adaptations of the digestive tract in Gangetic mud-eel Ophichthys cuchia (Hamilton 1822) support utilization of mud-dwelling prey.

Freshwater mud-eel, Ophichthys cuchia is nocturnal, carnivorous and economically important fish, yet its digestive physiology is unknown. We therefore studied the gastrointestinal (GI) tract of O. cuchia using morphological, osteological, histological and histochemical approach to understand how the structural adequacy of GI tract helps in acquisition of bottom and mud-dwelling prey and supports utilization of uncommon food resources. Morphologically the GI tract showed typical features of carnivorous fishes in the form of sub-terminal mouth, short muscular esophagus, expendable stomach, short

Heme oxygenase 1 plays a crucial role in swamp eel response to oxidative stress induced by cadmium exposure or Aeromonas hydrophila infection.

Oxidative stress contributes a lot to initiation and progression of pathological conditions. Heme oxygenase 1 (HO1), a cytoprotective enzyme, is usually upregulated to alleviate oxidative stress in vivo. The function of teleost HO1 in the response to oxidative stress induced by heavy metal exposure and in pathogenic bacterial infection remains uncertain. In the present study, both complementary DNA and genomic sequence of a HO1-like gene cloned from the liver of swamp eel (Monopterus albus) are reported. Sequence analysis showed that the putative amino acid sequence contained a conserved heme

Changes to the tropomyosin structure alter the angiotensin-converting enzyme inhibitory activity and texture profiles of eel balls under high hydrostatic pressure.

Changes in the structure of tropomyosin (TM) altered the texture profiles of eel balls and the inhibitory activity of the angiotensin-converting enzyme (ACE). The secondary and tertiary structure of TM was determined after high hydrostatic pressure (HHP) treatment. The correlation between the spatial structure of TM and the texture profiles of eel balls was developed and discussed. The β-sheet was converted to a β-turn and a random coil when treated at HHP (200-400 MPa), meanwhile the α-helix unfolded and was converted into a β-sheet, β-turn and a random coil with treatment at 500 and 600 MPa.

A new gonad-infecting species of Philometra (Nematoda: Philometridae) from the red cusk-eel Genypterus chilensis (Osteichthyes: Ophidiidae) off Chile.

A new nematode species, Philometra genypteri sp. n. (Philometridae), is described from male and female specimens found in the ovary of red cusk-eel, Genypterus chilensis (Guichenot) (Ophidiidae, Ophidiiformes), from the eastern South Pacific Ocean off the coast of northern Chile. Based on light and scanning electron microscopy examination, the new species differs from most other gonad-infecting Philometra spp. in the length of spicules (126-135 μm), a gubernaculum with dorsal lamella-like structures on its distal end, and the number and arrangement of genital papillae in the male. From a few c

Immunocytochemical localization of mammalian GnRH (gonadotropin-releasing hormone) and chicken GnRH-II in the brain of the European silver eel (Anguilla anguilla L.).

Using specific antibodies for the two molecular forms of gonadotropin-releasing hormone (GnRH) present in the European eel, Anguilla anguilla, (mammalian GnRH, mGnRH, and chicken GnRH II, cGnRH-II), we employed immunocytochemistry to determine the distribution of these two peptides in the brain and in the pituitary. The results indicate that mGnRH and cGnRH-II are localized in different neurons: mGnRH-immunoreactive (ir) perikaria were observed in the olfactory bulbs, the junction between olfactory bulbs and telencephalon (nucleus olfactoretinalis), the telencephalon, the preoptic region and t

Biodegradation of titanium implants after long-time insertion used for the treatment of fractured upper and lower jaws through osteosynthesis: element analysis by electron microscopy and EDX or EELS.

Twelve patients underwent an osteosynthesis with titanium to treat upper and lower jaw fractures. Six to 12 months later, the miniplates were removed. Tissue samples were analyzed by light and electron microscopy for detection of a metallosis. The analysis showed new bone formation like callus tissue around the miniplates. In some cases small, rounded deposits and accumulation of colloid-like particles located next to bigger titanium artifacts were detected in the cytoplasm of histiocytes and in the matrix of connective tissue. The titanium was identified by elemental analysis using EDX in SEM

Sex-Specific Marker Development and Identification of an XY Sex Determination System in the Tire Track Eel (Mastacembelus favus) via Whole-Genome Resequencing.

The tire track eel (Mastacembelus favus) is a freshwater fish with high economic value and aquaculture potential. However, its sex determination mechanism remains unclear, which limits the development of monosex culture and sex-controlled breeding. To address this, preliminary female and male reference genomes were generated using second-generation sequencing, followed by whole-genome resequencing of four females and four males. Comparative analyses identified 69 male-specific sequences, with a total length of approximately 44.5 kb. Based on these sequences, two PCR-based sex-specific markers

High temperature induces oxidative damage, immune modulation, and atrophy in the gills and skeletal muscle of the teleost fish black cusk-eel (Genypterus maculatus).

The high temperature associated with heat waves is a relevant abiotic factor that could impact the biology of teleost fish. The innate immune response, muscular growth, and oxidative stress status are relevant functions in fish tissues that could be affected by increased temperature. In this study, black cusk-eel (Genypterus maculatus) juveniles were subjected to increased temperature, to experimentally replicate heat waves registered from the South Pacific Ocean for five days. The results showed that thermal stress modulated the immune response in gills, with up-regulation of antibacterial pe
