Ectodysplasin receptor-mediated signaling is essential for embryonic submandibular salivary gland development.

Hypohidrotic (anhidrotic) ectodermal dysplasia (HED), the most common of the approximately 150 described ectodermal dysplasias, is a disorder characterized by abnormal hair, teeth, sweat glands, and salivary glands. Mutations in the EDA (ectodysplasin-A) and EDAR (ectodysplasin-A receptor) genes are responsible for X-linked and autosomal HED, respectively. Abnormal phenotypes similar to HED are seen in Tabby (Eda(Ta)) and downless (Edar(dl)) mutant mice. Although recent studies have focused on the role of Eda/Edar signaling during hair and tooth development, very little is known about its role

Thyroid hormone deiodinases during embryonic development of the saltwater crocodile (Crocodylus porosus).

All tissues of the embryonic saltwater crocodile (Crocodylus porosus) gradually increased in weight during development except for lung tissue, which had a peak weight of 1.09 g at day 67, thereafter decreasing in weight. The brain was a relatively large organ. Deiodinase activities in liver, kidney, lung, heart, gut, and brain from day 29 to day 77 of development of the saltwater crocodile were investigated. High-K(m) reverse triiodothyronine (rT(3)) outer ring deiodination (ORD) activity was present in all tissues except the brain. Activity ranged from 559 +/- 51.3 pmol rT(3) deiodinated/mg p

Fibrillin-1 and fibrillin-2 in human embryonic and early fetal development.

The extracellular glycoproteins fibrillin-1 and fibrillin-2 are major components of connective tissue microfibrils. Mutations in the fibrillin-1 and fibrillin-2 genes are responsible for the phenotypical manifestations of Marfan syndrome and congenital contractural arachnodactyly respectively, which emphasizes their essential roles in developmental processes of various tissues. Consistent with this last notion, organ culture experiments have indirectly suggested morphogenic roles for fibrillins in lung and kidney development. In order to contribute to the understanding of the roles of fibrilli

Role of laminin-nidogen complexes in basement membrane formation during embryonic development.

Laminin and nidogen (entactin) are major glycoprotein components of basement membranes. At least seven different isoforms of laminin have been identified. Laminin and nidogen form high affinity complexes in basement membranes by specific binding between the laminin gamma 1 chain and the G3 globule of nidogen. Additional interactions between nidogen and collagen IV, perlecan and other basement membrane components result in the formation of ternary complexes between these matrix components. Nidogen is highly susceptible to proteolytic cleavage, and binding to laminin protects nidogen from degrad

Cells Expressing mRNA for Neurotrophins and their Receptors During Embryonic Rat Development.

In situ hybridization analysis of cells expressing messenger RNAs (mRNAs) for the neurotrophins nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF) and neurotrophin-3 (NT-3) and their high-affinity receptors (trk, trkB and trkC) in the rat embryo revealed a complex but specific expression pattern for each of these mRNAs. For all mRNAs a developmentally regulated expression was seen in many different tissues. BDNF and NT-3 mRNAs were expressed in the sensory epithelia of the cochlea and vestibule macula of the sacculus and utricle, and both trkB and trkC mRNA were expressed in t

Role of zygotic arrest genes in female meiosis and early embryonic development.

Zygotic arrest genes play an indispensable role in regulating the critical transition from maternal to zygotic control during early embryonic development, commonly referred to as zygotic genome activation (ZGA). These genes orchestrate fundamental processes such as maternal mRNA clearance, chromatin remodelling, epigenetic reprogramming, and transcriptional activation, ensuring the developmental competence of embryos. Key genes, including ZAR1, ZAR2, BTG4, NPM2, PADI6, and KHDC3, have been identified as essential regulators of oocyte maturation, meiotic progression, and cleavage-stage developm

Histone Deacetylases Are Required for Aedes aegypti Female Reproduction and Embryonic Development.

Histone acetylation and deacetylation are essential for chromatin modifications that regulate various cellular functions. This study identified nine histone deacetylase (HDAC) genes in the yellow fever mosquito, Aedes aegypti, and investigated their roles in female reproduction and embryonic development using RNA interference (RNAi). RNAi-mediated knockdown of HDAC1 (Histone deacetylase 1), HDAC3 (Histone deacetylase 3), HDAC4 (Histone deacetylase 4), and SIRT2 (Sirtuin 2) resulted in severe defects in oocyte development and reduced egg-laying compared with control insects injected with double

Origin and differentiation of lymph node and spleen progenitors during embryonic development.

Secondary lymphoid tissues, including the spleen and lymph nodes, play an essential role in supporting immune responses. These organs are structurally organized into specialized compartments in which the interactions between hematopoietic and stromal cells are crucial for immune cell function. In this Review, we examine the cellular and molecular mechanisms that control spleen and lymph nodes, primarily in mice, with a particular emphasis on the embryonic origins of the different cell types involved. We also discuss the processes that determine the acquisition of lymphoid and mesenchymal cell

Exploring hyperglycemia's impact on embryonic development: Insights from the chicken embryo model.

Hyperglycemia during pregnancy is a growing health concern due to its association with congenital anomalies. While rodent models have been used to study this condition, their variability and inability to control the embryonic microenvironment is a limitation. Having this in mind, this study explores the chicken embryo (Gallus gallus) as an alternative model for studying the effects of hyperglycemia on early embryonic development. For that purpose, fertilized chicken eggs were exposed to glucose (0.2 and 0.4 mmol) from embryonic day 1 (E1) onwards. On embryonic day 5 (E5), glucose levels, devel

Expression analysis of the adaptor proteins Sh2b family during zebrafish embryonic development.

The SH2B family, which includes SH2B1, SH2B2, and SH2B3, consists of adaptor proteins that possess conserved Src homology 2 (SH2) and pleckstrin homology (PH) domains, playing essential roles as signaling mediators. However, the gene expression patterns of this family during embryonic development are still mostly unclear. In this study, we first investigated the evolutionary conservation of SH2B across multiple species using phylogenetic analysis, which revealed high sequence homology between zebrafish Sh2b and its orthologs in other vertebrates. Subsequently, we examined the expression patter

Noninvasive Tracking of Embryonic Cardiac Dynamics and Development with Volumetric Optoacoustic Spectroscopy.

Noninvasive monitoring of cardiac development can potentially prevent cardiac anomalies in adulthood. Mouse models provide unique opportunities to study cardiac development and disease in mammals. However, high-resolution noninvasive functional analyses of murine embryonic cardiac models are challenging because of the small size and fast volumetric motion of the embryonic heart, which is deeply embedded inside the uterus. In this study, a real time volumetric optoacoustic spectroscopy (VOS) platform for whole-heart visualization with high spatial (100 µm) and temporal (10 ms) resolutions is de

Embryonic Development and Cranial Ossification Sequence in Two Heremites Species (Squamata: Scincidae).

Although embryological studies of squamates have a long history, most groups in this large clade remain poorly studied. One such group is the family Scincidae, which consists of morphologically and ecologically diverse lizards. In this study, we describe several stages of embryonic development based on cleared and stained specimens of Heremites auratus and Heremites vittatus. Our analysis indicates that the pterygoid and frontal are the first bones to be ossified at stage 34 in the skull of H. auratus. At stage 37, which was examined in both studied species, the ossified bones include the nasa

Knock-down of YME1L1 induces mitochondrial dysfunction during early porcine embryonic development.

YME1L1, a mitochondrial metalloproteinase, is an Adenosine triphosphate (ATP)-dependent metalloproteinase and locates in the mitochondrial inner membrane. The protease domain of YME1L1 is oriented towards the mitochondrial intermembrane space, which modulates the mitochondrial GTPase optic atrophy type 1 (OPA1) processing. However, during embryonic development, there is no report yet about the role of YME1L1 on mitochondrial biogenesis and function in pigs. In the current study, the mRNA level of YME1L1 was knocked down by double strand RNA microinjection to the 1-cell stage embryos. The expre

Low-dose lipopolysaccharide exposure during oocyte maturation disrupts early bovine embryonic development.

Gram-negative bacteria release of lipopolysaccharide (LPS) endotoxin elicits robust immune responses capable of disrupting normal ovarian function contributing to female infertility. However, effects of subclinical or non-detectable infections on oocyte competence and subsequent embryo development remain to be fully elucidated. The aim of this study was to investigate the effects of exposing bovine oocytes to low LPS doses on oocyte and embryo competence. Bovine oocytes were collected from slaughterhouse-derived ovaries and matured with vehicle-control or increasing doses of LPS (0.01, 0.1, an

What Does Intracytoplasmic Sperm Injection Change in Embryonic Development? The Spermatozoon Contribution.

The intracytoplasmic sperm injection (ICSI) technique was invented to solve severe male infertility due to altered sperm parameters. Nowadays, it is applied worldwide for the treatment of couple infertility. ICSI is performed with any available spermatozoon from surgery or ejaculated samples, whatever are the sperm motility, morphology or quantity. The aim of the present review was to study if embryo development and kinetics would be modified by (1) ICSI under the technical aspects, (2) the micro-injected spermatozoa in connection with male infertility. From published data, it can be seen that

Genome-wide screening reveals the genetic basis of mammalian embryonic eye development.

BACKGROUND: Microphthalmia, anophthalmia, and coloboma (MAC) spectrum disease encompasses a group of eye malformations which play a role in childhood visual impairment. Although the predominant cause of eye malformations is known to be heritable in nature, with 80% of cases displaying loss-of-function mutations in the ocular developmental genes OTX2 or SOX2, the genetic abnormalities underlying the remaining cases of MAC are incompletely understood. This study intended to identify the novel genes and pathways required for early eye development. Additionally, pathways involved in eye formation

Fmr1 exon 14 skipping in late embryonic development of the rat forebrain.

BACKGROUND: Fragile X syndrome, the major cause of inherited intellectual disability among men, is due to deficiency of the synaptic functional regulator FMR1 protein (FMRP), encoded by the FMRP translational regulator 1 (FMR1) gene. FMR1 alternative splicing produces distinct transcripts that may consequently impact FMRP functional roles. In transcripts without exon 14 the translational reading frame is shifted. For deepening current knowledge of the differential expression of Fmr1 exon 14 along the rat nervous system development, we conducted a descriptive study employing quantitative RT-PCR

Point Mutation in Prkra Alters miRNA Expression During Embryonic External Ear Development.

Microtia is a congenital malformation of the external ear that can lead to conductive hearing impairment. In this study, we investigated the role of the Prkra gene in external ear development. We used advanced sequencing techniques to evaluate the differential expression of microRNAs (miRNAs) involved in external ear development in mouse embryos after point mutation in the Prkra gene. The Prkra Little ear mouse model was used to obtain mouse embryos at the E15.5 and E17.5 developmental stages, and changes in miRNA expression profiles were detected. Di-butyl phthalate (DBP) induces defects during embryonic eye development in zebrafish.

Di-butyl phthalate (DBP) is a phthalate ester (PAEs) added during the manufacturing of plastics to make them stronger, yet more pliable. DBP is noncovalently bound to plastics resulting in leaching into the environment. Concerning concentrations of DBP have been noted in surface and groundwater, aquatic ecosystems, soil and atmospheric environments globally. Global production of phthalates and thus concomitant exposure has increased over the years making studies on the ecological and environmental safety needed. Most of the literature on DBP focuses on the endocrine disrupting properties of ph
