Cloning, expression and purification of envelope proteins E1 and E2 of western equine encephalitis virus and potential use of them as antigens in immunoassays.

The genes encoding envelope proteins E1 and E2 of western equine encephalitis virus (WEEV) were respectively cloned into a prokaryotic T7 RNA polymerase-regulated expression vector. The recombinant C-terminal 6xHis-tagged WEEV E1 and E2 were expressed in bacteria as inclusion bodies that were subsequently solubilized with 8M urea, purified by immobilized metal ion affinity chromatography and finally refolded using an arginine system. The purified 6xHis-tagged proteins showed 50kDa bands as revealed by sodium dodecyl sulphate-polyacrylamide gel electrophoresis, consistent with the expected size

Natural and experimental arboviral infections in a population of blacktail jackrabbits along the sacramento river in butte county, california (1971-1974).

A serologic survey of the blacktail jackrabbit ( Lepus californicus ) for infections with 10 arboviruses was conducted from 1971 through 1974 along the Sacramento River in Butte County, California. Of 325 animals captured and bled a total of 493 times, 40% were found positive for hemagglutination-inhibiting (HI) antibody to California encephalitis (CE) virus, 34% to western equine encephalomyelitis (WEE) virus, 20% to Buttonwillow virus, 15% to St. Louis encephalitis (SLE) virus and 12% to Main Drain virus. Only 5 and 2% of the animals had HI antibodies to Lokern and Turlock (TUR) viruses, res

Role of plant phytochemicals/extracts for the control of Dermanyssus gallinae in poultry and its zoonotic importance.

Dermanyssus gallinae (poultry red mite) has negative impact on poultry production, causing major economic losses by damaging the health of chickens and other bird species, globally. D. gallinae also acts as vector and reservoir of different viral and bacterial pathogens such as, E. coli, S. enteriditis, P. multocida, S. gallinarum, Newcastle disease, Avian influenza, Erysipelothrix rhusiopathiae, Eastern, Western and Venezuelan equine encephalomyelitis viruses. Some of them are important from zoonotic point of view like, S. enteritidis, Borrelia burgdorferi and avian influenza A virus. It also

Potential effect of global warming on mosquito-borne arboviruses.

If global warming occurs in California, daily mean temperatures may increase by 3 to 5 degrees C, precipitation patterns will change, and sea level may rise 1 m. Studies were done on effect of temperature changes on survival of Culex tarsalis Coquillett, the primary vector of western equine encephalomyelitis (WEE) and St. Louis encephalitis (SLE) viruses, in two regions where temperatures differed by 5 degrees C. Daily mortality of adult vectors increased by 1% for each 1 degree C increase in temperature. At 25 degrees C, only 5% of Cx. tarsalis survived for 8 or more days, the time required f

Persistence of mosquito-borne viruses in Kern County, California, 1983-1988.

The persistence of arboviruses was studied from 1983 to 1988 in mixed agriculture, marsh, riparian, and foothill habitats in Kern County, CA. Western equine encephalomyelitis (WEE) virus was isolated frequently during 1983 from Culex tarsalis and Aedes melanimon and was detected by the seroconversion of sentinel chickens. WEE virus then disappeared, even though vector competence studies during 1984-1986 showed that Cx. tarsalis was able to transmit WEE virus. St. Louis encephalitis (SLE) virus was detected sporadically in 3 of the 6 years of the study by isolation from Cx. tarsalis and/or by s

Chimeric alphavirus vaccine candidates protect mice from intranasal challenge with western equine encephalitis virus.

We developed two types of chimeric Sindbis virus (SINV)/western equine encephalitis virus (WEEV) alphaviruses to investigate their potential use as live virus vaccines against WEE. The first-generation vaccine candidate, SIN/CO92, was derived from structural protein genes of WEEV strain CO92-1356, and two second-generation candidates were derived from WEEV strain McMillan. For both first- and second-generation vaccine candidates, the nonstructural protein genes were derived from SINV strain AR339. Second-generation vaccine candidates SIN/SIN/McM and SIN/EEE/McM included the envelope glycoprote

Phylogenetic analysis of Buggy Creek virus: evidence for multiple clades in the Western Great Plains, United States of America.

We present the first detailed phylogenetic analysis of Buggy Creek virus (BCRV), a poorly known alphavirus with transmission cycles involving a cimicid swallow bug (Oeciacus vicarius) vector and cliff swallows (Petrochelidon pyrrhonota) and house sparrows (Passer domesticus) as the principal avian hosts. Nucleotide sequences of a 2,075-bp viral envelope glycoprotein-coding region, covering the entire PE2 gene, were determined for 33 BCRV isolates taken from swallow bugs at cliff swallow colonies in Nebraska and Colorado in the summer of 2001 and were compared with the corresponding region of B

Intensive early season adulticide applications decrease arbovirus transmission throughout the Coachella Valley, Riverside County, California.

In the Coachella Valley of California the seasonal onset of St. Louis encephalitis virus (SLEV), western equine encephalomyelitis virus (WEEV), and West Nile virus (WNV) has been detected consistently at the shoreline of the Salton Sea near the community of North Shore. The timing and intensity of initial amplification in the Culex tarsalis Coquillett/wild bird cycle at this focus seemed closely linked to the subsequent dispersal of virus to the rest of the Coachella Valley and perhaps southern California. In 2004, an attempt was made to interrupt the amplification and dispersal of WNV using g

Genetic relatedness of Sindbis virus strains from Europe, Middle East, and Africa.

The relatedness of 40 strains of Sindbis virus (SIN) from Europe, the Middle East, and Africa was investigated by limited sequencing within the gene encoding the E2 glycoprotein corresponding to amino acid residues 117 to 229 and encompassing one of the major neutralization epitopes. Phylogenetic analyses using distance matrix and parsimonious methods identified two major genetic clusters of western SIN strains, although the variability was less than that of the corresponding region for Venezuelan equine encephalomyelitis virus with a maximum divergence of 12.4% versus 28.5%, respectively. Inhibition of dengue virus by targeting viral NS4B protein.

We report a novel inhibitor that selectively suppresses dengue virus (DENV) by targeting viral NS4B protein. The inhibitor was identified by screening a 1.8-million-compound library using a luciferase replicon of DENV serotype 2 (DENV-2). The compound specifically inhibits all four serotypes of DENV (50% effective concentration [EC(50)], 1 to 4 μM; and 50% cytotoxic concentration [CC(50)], >40 μM), but it does not inhibit closely related flaviviruses (West Nile virus and yellow fever virus) or nonflaviviruses (Western equine encephalomyelitis virus, Chikungunya virus, and vesicular stomatitis

Persistent West Nile virus transmission and the apparent displacement St. Louis encephalitis virus in southeastern California, 2003-2006.

West Nile virus (family Flaviviridae, genus Flavivirus, WNV) invaded the Colorado Desert biome of southern California during summer 2003 and seemed to displace previously endemic St. Louis encephalitis virus (family Flaviviridae, genus Flavivirus, SLEV, an antigenically similar Flavivirus in the Japanese encephalitis virus serocomplex). Western equine encephalomyelitis virus (family Togaviridae, genus Alphavirus, WEEV), an antigenically distinct Alphavirus, was detected during 2005 and 2006, indicating that conditions were suitable for encephalitis virus introduction and detection. The Western Equine Encephalitis Lyophilized, Inactivated Vaccine: An Update on Safety and Immunogenicity.

BACKGROUND: Western Equine Encephalitis (WEE) is a naturally acquired infection and potentially devastating bioweapon, with no specific human countermeasures. An experimental inactivated Western Equine Encephalitis Vaccine (WEEV; WEE TSI-GSD 210) has been used under an IND (investigational New Drug) protocol at the United States Army Medical Research Institute of Infectious Diseases (USAMRIID) since 1976. METHODS: Over 24 years from 1987 to 2011, 876 subjects received 3 primary vaccine doses under 3 studies with 1,537 booster doses administered (FY87-8, phase 2, laboratory workers, vaccine lo

The amino-terminal domain of alphavirus capsid protein is dispensable for viral particle assembly but regulates RNA encapsidation through cooperative functions of its subdomains.

Venezuelan equine encephalitis virus (VEEV) is a pathogenic alphavirus, which circulates in the Central, South, and North Americas, including the United States, and represents a significant public health threat. In recent years, strong progress has been made in understanding the structure of VEEV virions, but the mechanism of their formation has yet to be investigated. In this study, we analyzed the functions of different capsid-specific domains and its amino-terminal subdomains in viral particle formation. Our data demonstrate that VEEV particles can be efficiently formed directly at the plas

A two-phase innate host response to alphavirus infection identified by mRNP-tagging in vivo.

A concept fundamental to viral pathogenesis is that infection induces specific changes within the host cell, within specific tissues, or within the entire animal. These changes are reflected in a cascade of altered transcription patterns evident during infection. However, elucidation of this cascade in vivo has been limited by a general inability to distinguish changes occurring in the minority of infected cells from those in surrounding uninfected cells. To circumvent this inherent limitation of traditional gene expression profiling methods, an innovative mRNP-tagging technique was implemente

Bionomics of Culex tarsalis (Diptera: Culicidae) in relation to arbovirus transmission in southeastern California.

Population dynamics and bionomics of host-seeking Culex tarsalis Coquillett were studied in the Imperial and Coachella valleys of California during periods in 1991 and 1992 when western equine encephalomyelitis (WEE) and St. Louis encephalitis (SLE) viruses were transmitted to sentinel chickens. Female abundance was greatest during the spring and fall, before and after most virus transmission occurred and was not correlated with temperature, humidity, or rainfall. Parity rates were highest during late summer when virus activity peaked and were lowest during December when females may enter a sh

Infectivity variation and genetic diversity among strains of Western equine encephalitis virus.

Variation in infectivity and genetic diversity in the structural proteins were compared among eight strains of Western equine encephalitis virus (WEEV) to investigate WEEV virulence at the molecular level. A lethal intranasal infectivity model of WEEV was developed in adult BALB/c mice. All eight strains examined were 100 % lethal to adult mice in this model, but they varied considerably in the time to death. Based on the time to death, the eight strains could be classified into two pathotypes: a high-virulence pathotype, consisting of strains California, Fleming and McMillan, and a low-virule

Analysis of the Sarcocystis neurona microneme protein SnMIC10: protein characteristics and expression during intracellular development.

Sarcocystis neurona, an apicomplexan parasite, is the primary causative agent of equine protozoal myeloencephalitis. Like other members of the Apicomplexa, S. neurona zoites possess secretory organelles that contain proteins necessary for host cell invasion and intracellular survival. From a collection of S. neurona expressed sequence tags, we identified a sequence encoding a putative microneme protein based on similarity to Toxoplasma gondii MIC10 (TgMIC10). Pairwise sequence alignments of SnMIC10 to TgMIC10 and NcMIC10 from Neospora caninum revealed approximately 33% identity to both ortholo

Encephalitis virus persistence in California birds: preliminary studies with house finches.

Field-collected house finches of mixed sex and age were infected experimentally with either western equine encephalomyelitis (WEE) or St. Louis encephalitis (SLE) viruses during the summer or fall of 1998 and maintained over the winter under ambient conditions. To detect natural relapse during the spring, 32 birds were bled weekly from February through June 1999, and then necropsied 1 yr after infection to detect chronic infections using a reverse transcription polymerase chain reaction (RT-PCR). After 10 mo, 13/14 surviving birds previously infected with WEE were antibody positive by enzyme i
