Regulatory effects of winter Morchella esculenta cultivation on the summer maize phyllosphere microbiome and plant health-related traits.

In response to the intensifying demand for reduced fertilizer use and increased climate instability, the strategic incorporation of cover crops to improve the maize (Zea mays L.) phyllosphere microenvironment and leaf functional traits has emerged as an important approach for sustaining and enhancing crop productivity. This study compared the effects of two-year (2023–2024) winter cultivation of Morchella esculenta (ME) and fallow (CK) on summer maize leaf functional traits, phyllosphere microbial community composition, and their potential implications for maize health-related traits. The resu

Immobilization of l-asparaginase on oxidized bacterial cellulose to enhance stability and cytotoxic activity.

l-asparaginase (l-ASNase) is a key therapeutic enzyme used to treat acute lymphoblastic leukemia (ALL). However, its use against solid tumors cells is limited due to its instability, rapid inactivation, and resistance mechanisms; thus, developing novel carriers that can improve l-ASNase stability while preserving its antitumor activity is crucial. In this study, oxidized BC (OBC) films were used for the first time as covalent carriers for l-ASNase from Erwinia carotovora (EwA) to achieve prolonged action against cancer cells. Oxidizing BC films with sodium periodate (NaIO₄) or 2,2,6,6-tetramet

Characterization and Evaluation of the Anti-proliferative Activity and Hypersensitivity of L-Asparaginase from Trichosporon asahii Isolate ChL11 and Candida palmioleophila Isolate JK12.

BACKGROUND: L-Asparaginase is a crucial enzyme to treat Acute Lymphoblastic Leukemia (ALL), as it depletes L-asparagine, an essential amino acid for cancer cell survival. However, its clinical use is often restricted due to hypersensitivity reactions. This study examined the anti-proliferative effects and hypersensitivity of fungal L-asparaginases (L-ASNases) from Trichosporon asahii isolate ChL11 (TaIChL11 L-ASNase) and Candida palmioleophila isolate JK12 (CpIJK12 L-ASNase). METHODS: The enzymes were produced and purified through ammonium sulfate precipitation, dialysis, and Sephadex G-100 c

Synthesis and biological activity of novel hydantoin cyclohexyl sulfonamide derivatives as potential antimicrobial agents in agriculture.

BACKGROUND: Plant disease is one of the most serious problems in agriculture that can damage crops. Chemical fungicides are widely used to control plant diseases, but have led to resistance and a series of environmental problems. It is, therefore, necessary to develop highly effective and eco-friendly antimicrobial compounds with novel structures. RESULTS: A series of novel hydantoin cyclohexyl sulfonamide derivatives were synthesized through an intramolecular condensation reaction. The bioassay results indicated that a majority of the title compounds displayed potent inhibitory activity agai

Clinical Utility of Pegaspargase in Children, Adolescents and Young Adult Patients with Acute Lymphoblastic Leukemia: A Review.

Acute lymphoblastic leukemia (ALL) is a heterogenous hematological malignancy representing 25% of all cancers in children less than 15 years of age. Significant improvements in survival and cure rates have been made over the past four decades in pediatric ALL treatment. Asparaginases, derived from Escherichia coli and Erwinia chrysanthemi, have become a critical component of ALL therapy since the 1960s. Asparaginases cause depletion of serum asparagine, leading to deprivation of this critical amino acid for protein synthesis, and hence limit survival of lymphoblasts. Pegaspargase, a conjugate

Prolonged versus standard native E. coli asparaginase therapy in childhood acute lymphoblastic leukemia and non-Hodgkin lymphoma: final results of the EORTC-CLG randomized phase III trial 58951.

Asparaginase is an essential component of combination chemotherapy for childhood acute lymphoblastic leukemia and non-Hodgkin lymphoma. The value of asparaginase was further addressed in a group of non-very high-risk patients by comparing prolonged (long-asparaginase) versus standard (short-asparaginase) native E. coli asparaginase treatment in a randomized part of the phase III 58951 trial of the European Organization for Research and Treatment of Cancer Children's Leukemia Group. The main endpoint was disease-free survival. Overall, 1,552 patients were randomly assigned to long-asparaginase

Wolbachia-mediated antibacterial protection and immune gene regulation in Drosophila.

The outcome of microbial infection of insects is dependent not only on interactions between the host and pathogen, but also on the interactions between microbes that co-infect the host. Recently the maternally inherited endosymbiotic bacteria Wolbachia has been shown to protect insects from a range of microbial and eukaryotic pathogens. Mosquitoes experimentally infected with Wolbachia have upregulated immune responses and are protected from a number of pathogens including viruses, bacteria, Plasmodium and filarial nematodes. It has been hypothesised that immune upregulation underpins Wolbachi

Endophytic bacterial communities of field-grown potato plants and their plant-growth-promoting and antagonistic abilities.

To study the effect of plant growth on potato-associated bacteria, the composition and properties of bacteria colonizing the endosphere of field-grown potato were analyzed by a multiphasic approach. The occurrence and diversity of potato-associated bacteria were monitored by a cultivation-independent approach, using terminal restriction fragment length polymorphism analysis of 16S rDNA. The patterns obtained revealed a high heterogeneity of community composition and suggested the existence of plant-specific communities. However, endophytic populations correlated to a certain extent with plant

Cefotaxime prevents microbial contamination and improves microspore embryogenesis in wheat and triticale.

Cefotaxime (100 mg/l) mitigate occasional gram negative bacterial contamination in wheat and triticale microspore culture and most importantly it increases cell growth and green plant production. Isolated microspore culture is a promising option to rapidly fix the product of meiotic recombination of F1 hybrids, in the process of varietal development. Clean culture and high embryogenesis rate are essential to commercial triticale and wheat microspore cultures. So, this study investigated (1) contaminants from isolated microspores cultures, (2) two antibiotics to control bacterial growth, and (3

Novel L-Asparaginases from the human gut microbiome: Genome mining, biochemical characterization, and in vitro anti-leukemic activity.

L-asparaginase is essential for acute lymphoblastic leukemia treatment; however, current Escherichia coli and Erwinia chrysanthemi formulations face significant limitations, including immunogenicity, glutaminase-associated toxicity, and short plasma half-life. The human gut microbiome represents an unexplored reservoir of therapeutic enzymes that may offer superior biocompatibility due to host-commensal co-evolution. We employed a systematic genome-mining approach to screen human gut metagenomic data for novel L-asparaginase candidates. Five candidate enzymes from the genera Bacteroides, Rumin

Extracts ofHylotelephiumerythrostictum (miq.) H. Ohba ameliorate intestinal injury by scavenging ROS and inhibiting multiple signaling pathways in Drosophila.

BACKGROUND: The intestinal epithelial barrier is the first line of defense against pathogens and noxious substances entering the body from the outside world. Through proliferation and differentiation, intestinal stem cells play vital roles in tissue regeneration, repair, and the maintenance of intestinal homeostasis. Inflammatory bowel disease (IBD) is caused by the disruption of intestinal homeostasis through the invasion of toxic compounds and pathogenic microorganisms. Hylotelephium erythrostictum (Miq.) H. Genomic diversity and organization of complex polysaccharide biosynthesis clusters in the genus Dickeya.

The pectinolytic genus Dickeya (formerly Erwinia chrysanthemi) comprises numerous pathogenic species which cause diseases in various crops and ornamental plants across the globe. Their pathogenicity is governed by complex multi-factorial processes of adaptive virulence gene regulation. Extracellular polysaccharides and lipopolysaccharides present on bacterial envelope surface play a significant role in the virulence of phytopathogenic bacteria. However, very little is known about the genomic location, diversity, and organization of the polysaccharide and lipopolysaccharide biosynthetic gene cl
