Short Tandem Repeats (STRs), which are located out of pseudo-autosomal parts of the human Y chromosome and passed-down from fathers to the male offspring in a non-recombinant form, are regarded as appropriate markers for forensic purposes and evolutionary investigations. Few studies concerned the genotyping of Y chromosome short tandem repeats (Y-STRs) among the ethnic groups of the north of Iran, especially the province of Golestan which is a multiethnic region of Iran. Thus, in this work we investigated the frequency of Y-STR haplotypes among the male population from Golestan province, to el

Multiple copies of several classes of insertion sequences (IS) are found in the genome of Yersinia pestis, the causative agent of bubonic and pneumonic plague. We used the genetic instability generated by these IS to develop a method (designated 3IS-RFLP) based on the restriction fragment length polymorphism of the IS100, IS285 and IS1541 elements for studying Y. pestis strains of worldwide origin. We show that 31S-RFLP is a powerful tool to group Y. pestis isolates according to their geographical origin, and therefore that this method may be valuable for investigating the origin of new or re-

The recovery and analysis of genetic material obtained from thermally altered human bones and teeth are increasingly important to forensic investigations, especially in cases where soft-tissue identification is no longer possible. Although little is known about how these fire-related processes affect DNA degradation over time, next-generation sequencing technology in combination with traditional osteobiographical applications may provide us clues to these questions. In this study, we compare whole mitochondrial genome data generated using two different DNA extraction methods from 27 thermally

Microchimerism (MC) refers to the presence of small amounts of foreign cells or DNA in the tissues or circulation of an individual. It generally occurs through mother-fetus interaction, twin pregnancies, and intergenerational transmission. MC is influenced by genetic and environmental factors such as toxic conditions, immunological suppression, and various diseases (influenza, COVID-19, etc.). Progenitor cells transferred from the fetus to the mother through fetal MC are known to differentiate into neurons in the maternal brain. Although the relationship between these cells and the brain is no

INTRODUCTION: An increased proclivity towards violence is often associated with those diagnosed with schizophrenia (SCZ), despite contradictory findings from prior studies exploring the association between violence and SCZ. Evidence has shown that certain comorbidities, specifically the presence of a substance use disorders, can result in increased aggression in those with SCZ. Copy number variation (CNV) load has also previously been implicated in the genetic vulnerability of individuals with SCZ. For this study, we aimed to determine whether CNV load correlates with increased violence in SCZ

Hypothermia is an important cause of death in forensic pathology. For the forensic diagnosis of hypothermia, some reports point out the possibility that hypothermia without diabetes may cause ketoacidosis. In this study, we evaluated the diagnostic value of ketoacidosis in a murine model of hypothermia, using the cold stress at 4 °C for 3 or 5 hrs in genetically diabetic (BKS.Cg-+Leprdb/+Leprdb/J) mice, compared with control (BKS.Cg- Dock7m+/Dock7m+/J) mice. The core temperature decrease was larger in diabetic mice than in control mice. We observed a novel finding that ketoacidosis assessed by

Prediction of human pigmentation traits, one of the most differentiable externally visible characteristics among individuals, from biological samples represents a useful tool in the field of forensic DNA phenotyping. In spite of freckling being a relatively common pigmentation characteristic in Europeans, little is known about the genetic basis of this largely genetically determined phenotype in southern European populations. In this work, we explored the predictive capacity of eight freckle and sunlight sensitivity-related genes in 458 individuals (266 non-freckled controls and 192 freckled c

Spear guns are rarely implicated in homicidal events and are infrequently described in forensic literature, with most published reports focusing on accidental or suicidal injuries and on the surgical management of surviving victims. We report a case of homicide caused by a trident-fishing harpoon, a device designed for underwater use. The fatal injury followed a sudden verbal dispute between two individuals over a parking space on a city street. Autopsy revealed three circular entrance wounds in the precordial region arranged in an oblique pattern, corresponding to the three-pronged configurat

The biological age difference among twins is frequently an issue in studies of genetic influence on various dental features, particularly dental development. The timing of dental development is a crucial issue also for many clinicians and researchers. The aim of this study was therefore to verify within groups of twins how dental development differs, by applying Demirjian's method, Mincer's charts of development of third molars and two of Cameriere's methods for dental age estimation, which are among the most popular methods both in the clinical and the forensic scenario. The sample consisted

Violence is unfortunately a part of society. The causes of violence are not completely understood, but it involves sociological, genetic, financial, biological, and environmental factors. Drugs can cause aggression by altering the neurotransmitters dopamine, norepinephrine, gamma-aminobutyric acid (GABA), and serotonin. Specific drugs associated with aggression include alcohol, anabolic steroids, cocaine, amphetamines, sedatives, opiates, and hallucinogens. Aggression can be categorized into impulsive and predatory aggression. Drugs under certain conditions cause impulsive aggression. Sometime

BACKGROUND/OBJECTIVES: Latent fingerprints (LFs) have been a cornerstone of forensic identification through conventional friction ridge pattern analysis; however, the microbial communities they harbor remain a largely untapped source of information. Estimation of the time-since-deposition (TsDp) of LFs is still an unresolved challenge in forensic science, as existing 2D and 3D imaging methodologies provide limited temporal resolution. This study investigated whether temporal shifts in LF-associated microbiota could potentially complement dating approaches via genetic analyses.
METHODS: LFs we

Determining fetal death causes is a complex problem for the forensic pathologist. Beyond the medico-legal context, the expert must be able to evaluate the viability of the fetus at the time of death, to eliminate in-utero fetal death and to determine if the death is related to a fetal, a maternal, a placental cause, or simply related to obstetrical complications. The authors present the case of a 21-year-old woman who unexpectedly gave birth to a fetus during a party. As pregnancy was not acknowledged by the mother (regular menstrual cycles and use of hormonal contraception), no obstetrical ch

The postmortem of suddenly deceased younger adults sometimes reveals that they experienced manifest coronary arteriosclerosis. We looked at 21 cases where stenosis of the coronary arteries was at least 50%. We supplemented our postmortem findings and the results from the postmortem identification of the lipid metabolism parameters with anamnestic details. We also conducted a genetic analysis. The risk factors such as smoking and family history were relatively frequent. In most of our cases, the postmortems showed significantly deviating lipid metabolism parameters. Compared to these findings,

Forensic DNA phenotyping applies genetic and epigenetic markers to infer biogeographical ancestry, physical appearance traits, and biological age, particularly when conventional methods fail to identify a suspect or victim. DNA methylation, a key epigenetic modification, is especially valuable for age estimation and due to its tissue-specific patterns also for body fluid identification, with the latter aiding in crime scene reconstruction. Current MPS-based methylation sample analyses require time-consuming, multi-step processing, while Nanopore sequencing offers a promising alternative by ena

Freckles or ephelides are hyperpigmented spots observed on skin surface mainly in European and Asian populations. Easy recognition and external visibility make prediction of ephelides, the potentially useful target in the field of forensic DNA phenotyping. Prediction of freckles would be a step forward in sketching the physical appearance of unknown perpetrators or decomposed cadavers for the forensic DNA intelligence purposes. Freckles are especially common in people with pale skin and red hair and therefore it is expected that predisposition to freckles may partially share the genetic backgr

X-chromosome markers have been proved to be decisive both complementing and solving kinship analysis, particularly when autosomal markers are not able to produce adequate likelihood ratios between different hypothesis. On the other hand, Pereira et al., (2012) have demonstrated that 32 Insertion/Deletion (InDel) markers located on the X-Chromosome have a very important power of discrimination in human populations, being a novel tool in the forensic and population fields. So, the aim of the present work was testing the forensic and population genetic efficiency of the 32 X-InDel polymorphisms i

Vascular pathology in young adults has during the last 10 years been diagnosed more often during forensic medical examination of sudden death. Major morphological alterations are revealed in cerebral vessels, coronary vessels, and at the level of the ascending portion of the aorta. Generally, in the young age there is no stenosing atherosclerosis inducing vascular lesions and the development of complications. It was determined that connective tissue dysplasia is pathology wherein weakness of the vascular wall is genetically preconditioned, thus promoting formation of vascular aneurysms and rup

A case of sudden death associated with fatty liver and encephalopathy is described in a 4-year old white boy with medium-chain acyl-coenzyme A dehydrogenase (MCAD) deficiency. The death was caused by hypoglycemia triggered by fasting and vomiting associated with a minor viral infection. The differential diagnosis of the hepatoencephalopathy is discussed in relation to other conditions, especially Reye's syndrome. The forensic pathologist should be familiar with MCAD and other deficiencies of beta-oxidation of fatty acids as a cause of sudden unexpected death in children in order to advise pare

Radiologic comparison is a potentially reliable means of identification in forensic contexts. Most radiologic comparisons are subjective and involve a qualitative visual comparison of the degree of similarity between antemortem and postmortem images, which is insufficient for quantitatively assessing the evidentiary value of an identification. Rather than simply concluding that antemortem and postmortem radiologic comparisons appear the same in the opinion of the examiner, results should be expressed quantitatively. This bolsters conclusions by providing statistical support for the probability

In this study, we developed and validated a novel microhaplotype (MH) panel, the FGID Microhaplotype Kit, which contains 232 loci and was specifically designed for forensic kinship analysis. The performance of the panel was evaluated through rigorous testing that included sensitivity, species specificity, inhibitor resistance, uniformity, stability, accuracy and mixture deconvolution. The results showed that the kit is capable of reliably detecting all loci with minimal DNA input. It showed high species specificity for 12 non-human DNA samples and resistance to common inhibitors. In addition,
