Systemic Inonotus sp. Infection in a dog.

A 3-years-old male golden retriever was presented for decreased activity (lethargy), anorexia, and titubation. Superficial lymph nodes were enlarged, and arrhythmia and tachycardia were auscultated. Fungal hyphae-like structures were detected in the biopsy samples from an enlarged lymph node and spleen. Nucleotide sequence of the internal transcribed spacer region of the fungi amplified by PCR was highly homologous to that of Inonotus pachyphloeus. The dog was treated with antifungal agents such as itraconazole, fluconazole, and voriconazole. Clinical signs resolved for 325 days but the dog di

Hair and nail relationship.

Hair and nails are often stated to have much in common in relation to their origin, anatomical structures, and common involvement in many diseases. Hair and nails are predominantly epithelial structures derived from primitive epidermis and made up of keratinous fibrils embedded in a sulfur-rich matrix. It was first noted early in the 20th century that the nail unit was comparable in several respects to a hair follicle sectioned longitudinally and laid on on its side. The epithelial components of hair follicle and nail apparatus are differentiated epidermal structures that may be involved joint

Toward a Cytological Classification of Nasal Biofilms in Upper Airway Inflammation: Microscopic Patterns, Clinical Meaning, and Therapeutic Implications.

Background/Objectives: Nasal biofilms are increasingly recognized as contributors to chronic and recurrent upper airway inflammation, yet their cytological interpretation is still often reduced to a binary presence/absence judgment, underestimating their morphological, microbial, and inflammatory heterogeneity. This Perspective proposes a cytological classification framework for nasal biofilms based on microscopic morphology, degree of organization, bacterial and fungal composition, host inflammatory response, and therapeutic implications. Methods: This conceptual framework integrates standard

Fungal Δ9-fatty acid desaturase: a unique enzyme at the core of lipid metabolism in Aspergillus fumigatus and a promising target for the search for antifungal strategies.

Aspergillus fumigatus, Candida albicans, and Cryptococcus neoformans are the leading fungal pathogens that cause life-threatening deep mycosis, posing significant challenges to immunocompromised patients and increasing healthcare costs worldwide. Lipid metabolism is crucial for the growth and development of all organisms. Increasing evidence highlights that complex structural lipids in the fungal cell membrane emerge as important factors involved in cell signaling, stress response, and immune recognition. Membrane fluidity is primarily regulated by the ratio of saturated and unsaturated fatty

Effects of developmental stage-driven fungal community shifts on biomass and metabolite accumulation in Gastrodia elata.

BACKGROUND: Fungal communities play crucial roles in plant development and metabolite accumulation, especially in fully mycoheterotrophic medicinal plants like Gastrodia elata. While the importance of fungal symbiosis in G. elata is recognized, how fungal community dynamics evolve across its entire growth cycle and how they influence biomass and bioactive compound accumulation remain largely unclear. RESULTS: High-throughput sequencing combined with multi-omics analyses revealed that developmental progression significantly shapes fungal diversity and composition, thereby influencing biomass a

Results of large penetrating keratoplasty in microbial keratitis.

Extensive corneal disease secondary to microbial keratitis can result in frank or impending corneal perforation requiring a large penetrating keratoplasty. In an 8-year period, 26 penetrating keratoplasties with recipient beds of > or = 9.5 mm were performed on 22 eyes: 11 for bacterial keratitis, 10 for fungal keratitis, and one for a mixed bacterial and fungal keratitis. The graft failed in 18 of 19 eyes (94.7%), with a median time to failure of 12.9 weeks in bacterial keratitis and 4.0 weeks in fungal keratitis. After large keratoplasty, 17 of 20 eyes (85.0%) maintained the structural integ

Extracellular vesicles from diverse fungal pathogens induce species-specific and endocytosis-dependent immunomodulation.

Microbial pathogens generate extracellular vesicles (EVs) for intercellular communication and quorum sensing. Microbial EVs also induce inflammatory pathways within host innate immune cells. We previously demonstrated that EVs secreted by Candida albicans trigger type I interferon signaling in host cells specifically via the cGAS-STING innate immune signaling pathway. Here, we show that despite sharing similar properties of morphology and internal DNA content, the interactions between EVs and the innate immune system differ according to the parental fungal species. EVs secreted by C. albicans,

Novel neogala-series glycosphingolipids with a terminal glucose residue from the fungus Mariannaea elegans.

Glycosphingolipids (GSLs) are essential membrane components of eukaryotic cells. Recently, a new type of fungal neogala-series GSL was identified in aureobasidin A-resistant fungi. In this study, we analyzed GSLs from four pathogenic fungal strains belonging to the order Hypocreales, and found that Mariannaea elegans contained both acidic GSLs and neutral GSLs with mono- and di-saccharides. The structures of the neutral GSLs of M. elegans were determined by compositional sugar, fatty acid, and sphingoid analyses by GC/MS, MALDI time-of-flight/MS, and 1H NMR. The ceramide moiety of Glcβ1-Cer co

Structure-Guided Discovery of the Novel Covalent Allosteric Site and Covalent Inhibitors of Fructose-1,6-Bisphosphate Aldolase to Overcome the Azole Resistance of Candidiasis.

Fructose-1,6-bisphosphate aldolase (FBA) represents an attractive new antifungal target. Here, we employed a structure-based optimization strategy to discover a novel covalent binding site (C292 site) and the first-in-class covalent allosteric inhibitors of FBA from Candida albicans (CaFBA). Site-directed mutagenesis, liquid chromatography-mass spectrometry, and the crystallographic structures of APO-CaFBA, CaFBA-G3P, and C157S-2a4 revealed that S268 is an essential pharmacophore for the catalytic activity of CaFBA, and L288 is an allosteric regulation switch for CaFBA. Furthermore, most of th

Antigen complex of Coccidioides immitis which elicits a precipitin antibody response in patients.

The occurrence in patients of elevated levels of immunoglobulin M (IgM) precipitin antibody to Coccidioides immitis antigens, which are commonly detected by the immunodiffusion-tube precipitin (TP) assay, is suggestive of primary nondisseminating coccidioidomycosis. We previously demonstrated that the concanavalin A-bound mycelial culture filtrate plus lysate preparation is a source of at least two TP antibody-reactive antigens (TP-Ags), which were identified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis as 120- and 110-kDa fractions. Evidence is presented here that the crude fi

Protease-Mediated Dual Action Against Rice Blast: Suppression of Magnaporthe oryzae Ergosterol Biosynthesis Pathway and Enhancement of Rice Disease Resistance by Bacillus amyloliquefaciens SN16-1.

Rice blast, caused by the fungal pathogen Magnaporthe oryzae, poses a severe threat to global rice production. While biological control represents a promising sustainable strategy, the molecular mechanisms by which biocontrol agents suppress M. oryzae remain poorly understood. Our findings elucidate a novel mechanism of bacterial biocontrol, wherein a secreted bacterial protease modulates fungal signalling pathways to disrupt its environmental fitness and pathogenic structures, while enhancing host resistance to blast disease. We demonstrate that the Bacillus amyloliquefaciens SN16-1 exhibits

Advances in 3D-printed transdermal microneedle patches for antifungal therapy: Current scenario and challenges.

Skin fungal infection and systemic fungal infections are highly prevalent, causing significant healthcare burden. Addressing the pervasive and sometimes life-threatening nature of fungal infections has spurred extensive research into novel therapeutic strategies, particularly focusing on effective drug formulations and innovative delivery routes. Among this transdermal drug delivery (TDD) using microneedles (MNs) is a promising approach. 3D-printed MN patches are composed of arrays of tiny, needle-like structures engineered to facilitate the delivery of therapeutic agents by breaching the stra

Recent Progress in Research on Mitochondrion-Targeted Antifungal Drugs: a Review.

Fungal infections, which commonly occur in immunocompromised patients, can cause high morbidity and mortality. Antifungal agents act by disrupting the cell membrane, inhibiting nucleic acid synthesis and function, or inhibiting β-1,3-glucan synthase. Because the incidences of life-threatening fungal infections and antifungal drug resistance are continuously increasing, there is an urgent need for the development of new antifungal agents with novel mechanisms of action. Recent studies have focused on mitochondrial components as potential therapeutic drug targets, owing to their important roles

Binding of the wheat germ lectin to Cryptococcus neoformans chitooligomers affects multiple mechanisms required for fungal pathogenesis.

The principal capsular component of Cryptococcus neoformans, glucuronoxylomannan (GXM), interacts with surface glycans, including chitin-like oligomers. Although the role of GXM in cryptococcal infection has been well explored, there is no information on how chitooligomers affect fungal pathogenesis. In this study, surface chitooligomers of C. neoformans were blocked through the use of the wheat germ lectin (WGA) and the effects on animal pathogenesis, interaction with host cells, fungal growth and capsule formation were analyzed. Treatment of C. neoformans cells with WGA followed by infection

[The first case of persistent vaginitis due to Aspergillus protuberus in an immunocompetent patient].

The vast majority of vaginal fungal infections are caused by Candida species. However, vaginitis cases caused by molds are extremely rare. Aspergillus protuberus is previously known as a member of Aspergillus section Versicolores which can cause opportunistic infections in immunocompromised patients, however it has recently been described as a seperate species. Although the members of Aspergillus section Versicolores have been isolated rarely in cases of pulmonary infections, eye infections, otomycosis, osteomyelitis and onycomycoses, to the best of our knowledge, there is no published case of

An epigenetic modifier enhances the generation of anti-phytopathogenic compounds from the endophytic fungus Chaetomium globosporum of Euphorbia humifusa.

Endophytic fungi are striking resources rich in bioactive structures with agrochemical significance. In order to maximize the opportunity of search for bioactive compounds, chemical epigenetic manipulation was introduced to enhance the structural diversity of the fungal products, and an UPLC-ESIMS and bioassay-guided separation was used to detect novel bioactive metabolites. Consequently, four previously undescribed compounds including two cyclopentenones (globosporins A and B) and two monoterpenoid indole alkaloids (globosporines C and D), as well as three known compounds, were isolated from
