Ginsenoside Rg3 Promotes Healthy Aging Through Phosphoinositide-Associated Lysosomal Regulation in Caenorhabditis elegans.

Lysosomal decline and proteostasis collapse are defining features of aging and major contributors to neurodegenerative vulnerability, yet interventions that preserve neuronal lysosomal function across aging remain poorly defined. Here, we identify 20(S)-ginsenoside Rg3, a ginseng-derived natural product, as a geroprotective small molecule in Caenorhabditis elegans. Rg3 extended lifespan and preserved key physiological outputs, including locomotion and pharyngeal pumping, indicating that Rg3 delays functional aging. Rg3 also maintained mechanosensory neuron integrity and touch sensitivity, supp

Biosynthesis of Active Ginsenoside Aglycone Analogues Using Two Oxidases Mined from Mucor spinosus.

The pharmacological activity of protopanaxadiol (PPD) can be enhanced by oxidative modification. Compared with chemical synthesis and biotransformation, synthetic biology offers a more efficient and eco-friendly approach for modifying PPD. In this study, we identified two oxidases from Mucor spinosus. MsSDR3 oxidizes C3-OH of both dammarenediol-II and PPD to a ketone, while MsCYP3 oxidizes C12-OH of PPD to a ketone and hydroxylates PPD at C7β, C15α, and C11β positions. Combining MsSDR3/MsCYP3 with the enzymes involved in ginsenoside biosynthesis, we achieved de novo biosynthesis of seven ginse

Ginsenoside Rd promotes the expression of acetylated α-tubulin and FOXJ1 in human bronchial epithelial cells.

Mucociliary clearance is an airway defense mechanism driven by the coordinated function of mucus and motile cilia. Although ciliary beat frequency (CBF) reflects the motile function of ciliated cells, the generation and maintenance of ciliated cells are also essential for sustained airway epithelial defense. Although short-term treatment with ginsenoside Rd (Rd) increases CBF, its long-term effects on the differentiation of human airway epithelial cells remain unclear. We investigated the effects of Rd on primary normal human bronchial epithelial cells cultured under air-liquid interface condi

Ginsenoside CK targets NEK7 to suppress inflammasome activation and mitigate diabetes-induced muscle atrophy.

Diabetes-induced skeletal muscle atrophy is closely linked to oxidative stress and chronic inflammation, which severely compromise quality of life. This study aimed to identify effective anti-inflammatory strategies to mitigate muscle degeneration in diabetes. We investigated the molecular mechanism of Ginsenoside CK (Gin), a major bioactive component of ginseng known for its anti-inflammatory properties. Using activity-based protein profiling, we identified never in mitosis A-related kinase 7 (NEK7) as a direct target of Gin. Mechanistically, Gin covalently binds to histidine 262 (H262) of NE

Combination of phytochemicals, including ginsenoside and curcumin, shows a synergistic effect on the recovery of radiation-induced toxicity.

Radiotherapy is commonly used to treat solid cancers located in the pelvis. A considerable number of patients experience proctitis of varying severity, even for a considerable period after radiotherapy. These side effects are often long-lasting or progressively worsen despite multiple therapeutic efforts and are a primary cause of an unexpectedly low quality of life, even after successful cancer treatment. Therefore, this study evaluated the individual and combined efficacy of ginsenoside, curcumin, butyric acid, and sucralfate compounds in treating radiation-induced proctitis. While the candi

Ginsenoside Rk1 induces autophagy-dependent apoptosis in hepatocellular carcinoma by AMPK/mTOR signaling pathway.

Hepatocellular carcinoma (HCC) is the third most lethal cancer in the world. Recent studies have shown that suppression of autophagy plays an important role in the development of HCC. Ginsenoside Rk1 is a protopanaxadiol saponin isolated from ginseng and has a significant anti-tumor effect, but its role and mechanism in HCC are still unclear. In this study, a mouse liver cancer model induced by diethylnitrosamine and carbon tetrachloride (DEN + CCl4) was employed to investigate the inhibitory effect of Rk1 on HCC. The results demonstrate that ginsenoside Rk1 effectively inhibits liver injury,

Ginsenoside Rc prevents dexamethasone-induced muscle atrophy and enhances muscle strength and motor function.

BACKGROUND: A decline in muscle mass and function can impact the health, disease vulnerability, and mortality of older adults. Prolonged use of high doses of glucocorticoids, such as dexamethasone (DEX), can cause muscle wasting and reduced strength. Ginsenoside Rc (gRc) has been shown to protect muscles by activating the PGC-1α pathway and improving mitochondrial function. The effects of gRc on muscle atrophy and function in mice are not fully understood. METHODS AND RESULTS: The study discovered that gRc prevented the DEX-induced decrease in viability of C2C12 myoblasts and myotubes. Furthe

Ginsenoside Rg1 attenuates cerebral ischemia-reperfusion injury through inhibiting the inflammatory activation of microglia.

It is recognized that the cerebral ischemia/reperfusion (I/R) injury triggers inflammatory activation of microglia and supports microglia-driven neuronal damage. Our previous studies have shown that ginsenoside Rg1 had a significant protective effect on focal cerebral I/R injury in middle cerebral artery occlusion (MCAO) rats. However, the mechanism still needs further clarification. Here, we firstly reported that ginsenoside Rg1 effectively suppressed the inflammatory activation of brain microglia cells under I/R conditions depending on the inhibition of Toll-likereceptor4 (TLR4) proteins. Ginsenoside Rb3 attenuates skin flap ischemia-reperfusion damage by inhibiting STING-IRF3 signaling.

We investigate the protective effect of ginsenoside Rb3 on skin flap microvasculature following ischemia-reperfusion (I/R) injury and its regulatory mechanism. We used a rat model of I/R injury with the right iliolumbar artery and oxidative stress model of human dermal microvascular endothelial cells. The effects of Rb3 on skin flap tissue and endothelial cell survival, STING-IRF3 pathway activation, and endothelial cell adhesion were measured. Following reperfusion, the survival rate of rat perforator flaps in the Rb3-treated group gradually increased with increasing Rb3 concentration. The tr

Generally, micro-capsule could geometrically provide higher loading capacity but also generate concerns in construction convenience, sustained release, bioaccessibility and stability. Hence there is a contradiction between loading capacity and encapsulation efficiency. Therefore, the study of the factors that decide the capsule size falling in nano or micron size with same capsu
