Since its production in the 1980s, ivermectin (IVM) has been used indiscriminately and the selection pressure to which bovine gastrointestinal nematodes have been exposed has been intense, resulting in considerable economic losses due to parasitic resistance. One possibility for the control of resistant parasites is the use of P-glycoprotein (P-gp) modulators, because one of the main biochemical changes in ivermectin-resistant parasites is the increased activity of membrane proteins responsible for the efflux of drugs and xenobiotics. This study aimed to evaluate the in vitro effect of eight P

Gastrointestinal nematode (GIN) infections are a common constraint to small ruminant industry throughout the world, and among those, haemonchosis has its own significance. Control of GIN primarily relies on the use of anthelmintics, but this approach has become less reliable due to the development of resistance in GINs against commonly used anthelmintics and an increased consumer demand for environmentally friendly animal products. These issues have stimulated investigations to find alternative sustainable control strategies, which are less reliant on anthelmintic input. One of such strategies

This study was carried out to determine the effects of Acacia karoo and Acacia nilotica diets on Haemonchus contortus infections in goats. Twenty-four Boer goats of mixed sex (live weight 17-22 kg) were randomly assigned to four treatment groups, namely: A. nilotica (AN) group, A. karoo (AK) group, control infected with H. contortus (HC) group and the non-infected control (NHC) group. Animals in the AN, AK and HC groups were orally infected with a single dose of 3000 HC third stage larvae. The AN and AK groups had dried leaves of AN and AK, respectively, included in their basal diet at a rate

Faecal samples were obtained from sheep and goats before, during and after the rainy season at three locations in south-west Mauritania. Several animals were also necropsied at the same time. Haemonchus contortus was the most prevalent worm. Infection by digestive-tract strongyles and Strongyloides papillosus was always very light (prevalence less than 20%). Sheep were more heavily infected than goats but animals under 1 year of age were not infected by digestive-tract strongyles. It is likely that young small ruminants became infected during the rainy season and that the parasites so acquired

AIM: To explore associations between phenotypic traits and polymorphisms in the DRB1 and GALNT6 gene in Nellore, Deccani and Kenguri sheep naturally infected with Haemonchus contortus.
MATERIALS AND METHODS: Blood and faecal samples were collected to evaluate fecal worm egg counts (FEC), packed cell volume (PCV), hemoglobin (Hb), eosinophilia and for DNA isolation.
RESULTS: Animals were grouped into susceptible and resistant groups based on EPG counts. FEC and circulating eosinophilia were higher in a susceptible group. Log FEC was negatively correlated (P < 0.01) with PCV, and Hb estimates.

Suppression and modulation of the host immune response to parasitic nematodes have been extensively studied. In the present study, we cloned and produced recombinant electron transfer flavoprotein α subunit (ETFα) protein from Haemonchus contortus (rHCETFα), a parasitic nematode of small ruminants, and studied the effect of this protein on modulating the immune response of goat peripheral blood mononuclear cells (PBMCs) and monocytes. Immunohistochemical tests verified that the HCETFα protein was localized mainly in the intestinal wall and on the body surface of worms. Immunoblot analysis reve

The aim of the present study was to evaluate the effects of treatment with different anthelmintic compounds on the productivity of naturally infected calves and the economic viability of these treatments within extensive breeding systems employing different nutritional strategies after weaning. For this purpose, 4 farms with 42-60 calves naturally infected with gastrointestinal nematodes were selected. The calves were distributed into 6 groups (7-10 animals each) per farm and treated with ivermectin 1%, ivermectin 3.15%, eprinomectin 5%, levamisole 7.5%, albendazole 15%, and control group (no

The potential of the chitin synthesis inhibitor diflubenzuron (DFB) to alter the development of the parasitic nematodes (Ascaris suum and Haemonchus contortus was investigated. DFB given orally (10 mg kg-1 per day for 30 days) to sheep inoculated with H. contortus infective larvae did not prevent the establishment of adults or affect fecal egg output. However, there was a significant (greater than 90%) decrease in the number of infective larvae recovered from fecal cultures derived from lambs harboring H. contortus adults that were treated with DFB. DFB did not affect egg hatching. Oral admini

The pattern of faecal egg counts after infection with 10,000 Haemonchus contortus larvae was similar within but not between, four pairs of monozygous twin sheep. Transfer of whole lymph or washed lymph cells from three immune donor sheep to their identical co-twin recipients reduced the susceptibility of the recipients to challenge with 10,000 larvae as measured by faecal egg counts. Cells from a nonimmune donor did not have this effect. In a final experiment, washed cells from an immune triplet sheep, which was actively responding to a repeated challenge infection, transferred a secondary loc

BACKGROUND: Trehalose-6-phosphate synthase (HcTPS) and trehalose-6-phosphate phosphatase (HcGOB) are key enzymes for trehalose synthesis in Haemonchus contortus. In addition, previous studies have also demonstrated that HcTPS and HcGOB can regulate the function of host immune cells in vitro, and are important immunosuppressive molecules. Therefore, this study evaluated the potential of HcTPS and HcGOB as vaccine candidates through in vitro and in vivo experiments.
METHODS: To evaluate the inhibitory effects of polyclonal antibodies on egg hatching and larval development, anti-rHcTPS and anti-

BACKGROUND: Haemonchus contortus (H. contortus) is one of the most important parasites that cause huge economic losses to small ruminant industry worldwide. Effective prognosis and treatment depend upon the early diagnosis of H. contortus infection. To date, no widely-approved methods for the identification of prepatent H. contortus infection are available to identify prepatent H. contortus infection properly. The aim of this study was to evaluate the diagnostic potential of recombinant cold shock H. contortus protein (rHc-CS) during early and late infections of H. contortus in goat.

Excretory and secretory products (ESPs) are released by the parasites during Haemonchus contortus (H. contortus) infection. In this study, Tropomyosin (TpMy), one of these ESPs was used to develop western blotting and optimized Enzyme Linked immunosorbent assay (ELISA) for detection of H. contortus during early infection in goat. Microscopic examination was performed parallel for comparison. Recombinant tropomyosin protein was purified successfully. Western blotting results revealed that anti-recombinant H. contortus Tropomyosin (rHc-TpMy) antibodies could recognize the natural proteinand rHc-

Haemonchus populations were collected from cattle from mid-western and eastern Southern US (four and six populations, respectively) to determine the relative prevalence of Haemonchus contortus and Haemonchus placei and the frequency of the three isotype-1 β-tubulin polymorphisms associated with benzimidazole resistance. A minimum of 32 individual adult worms were genotyped at position 24 of the rDNA ITS-2 for each population to determine species identity (296 worms in total). One population from Georgia was identified as 100% H. contortus with the remaining nine populations identified as 100%

Haemonchus contortus is the most prevalent nematode in Brazil. The objective of this study was to select 6 populations of H. contortus of known or suspected benzimidazole resistance status and characterize these using quantitative real-time polymerase chain reaction (qPCR) for single nucleotide polymorphisms (SNPs) F200Y, F167Y and E198A in the β-tubulin isotype 1 gene. qPCR was performed using DNA from a pool of 10 adult male H. contortus from a single animal per farm. Faecal egg count reduction test (FECRT) and egg hatch test (EHT) were used to determine the resistance status. Samples were o

OBJECTIVE: To investigate correlates of resistance to GI nematode infection in Nigerian West African dwarf (WAD) sheep.
METHODS: Thirty three sheep were randomly assigned to two groups, A (n=27) which were used for experimental infections, and B (n=6) which served as uninfected control. Each infected animal received weekly escalating infections with infective larvae (60%Haemonchus contortus (H. contortus) and 40%Trichostrongylus colubriformis (T. colubriformis) for 4 weeks. The responses of all the infected and control sheep were assessed by faecal egg count (FEC), worm burden (Wb), packed ce

A pharmaco-parasitological assessment of four different albendazole (ABZ) formulations was carried out in lambs infected with multiple resistant gastrointestinal (GI) nematodes. The comparative drug systemic exposure profiles (ABZ sulphoxide plasma concentrations) and anthelmintic efficacies (clinical endpoint measured through the faecal nematode eggs reduction counts) were determined for a reference formulation (RF) and three different test (T1, T2, T3) generic ABZ preparations. Fifty (50) Corriedale lambs naturally infected with multiple resistant GI nematodes were allocated into five experi

From autumn (April) to spring (November), groups of pregnant and dry Merino ewes grazed pasture contaminated with infective larvae of Haemonchus contortus, Ostertagia circumcincta and Trichostrongylus colubriformis. A periparturient rise in faecal egg counts occurred in the pregnant ewes, following the maturation in early spring of arrested fourth-stage larvae of H. contortus and failure of pregnant ewes to expel the resulting adult worms. Peak egg counts were seen in September, just before lambing. In dry ewes, egg counts were slightly elevated at the time of the rise in the pregnant ewes, bu

Th17 cells play important roles in anti-infective responses. The 15 kDa excretory/secretory protein of Haemonchus contortus (HcES-15) has been identified as a promising immune-protective antigen against H. contortus infection capable of up-regulating IL-17, IL-4 and IL-10 production. To obtain the peptides that primarily induce the Th17 immune response, we amplified and expressed the peptides ES15-1, ES15-2 and ES15-3 from HcES-15. In vitro studies demonstrated that ES15-1 stimulated transcriptional activation of the STAT3/RORγt signaling pathway and induced IL-17 production in goat peripheral
