Analysis of autoinducer-2 quorum sensing in Yersinia pestis.

The autoinducer-2 (AI-2) quorum-sensing system has been linked to diverse phenotypes and regulatory changes in pathogenic bacteria. In the present study, we performed a molecular and biochemical characterization of the AI-2 system in Yersinia pestis, the causative agent of plague. In strain CO92, the AI-2 signal is produced in a luxS-dependent manner, reaching maximal levels of 2.5 μM in the late logarithmic growth phase, and both wild-type and pigmentation (pgm) mutant strains made equivalent levels of AI-2. Strain CO92 possesses a chromosomal lsr locus encoding factors involved in the bindin

Voices of Eukaryotic Microbes: Chemical Communication Via Quorum Sensing.

Quorum sensing (QS) is a cell-cell communication mechanism mediated by secreted hormone-like signaling molecules that operates in both Gram-positive and Gram-negative bacteria, driving coordinated alterations in gene expression once a critical cell density is reached. In these prokaryotic systems, bacteria produce, release, detect, and respond to small autoinducers, such as acyl-homoserine lactones in Gram-negative bacteria, oligopeptides in Gram-positive bacteria, and the universal autoinducer-2, to regulate community behaviors including biofilm formation, virulence factor production, and str

Mitigating bio-clogging in subsurface flow constructed wetlands via quorum quenching: Performance evaluation and mechanistic insights.

Subsurface flow constructed wetlands (SSFCWs) have been widely applied for sewage wastewater remediation due to the efficient utilization of wetland space and considerable improvement in treatment capacity. However, bio-clogging driven by the accumulation of extracellular polymeric substances (EPS) severely restricts their longevity and hydraulic performance. To address this issue, two quorum quenching (QQ) strategies based on Rhodococcus sp. BH4 and porcine kidney acylase I were developed to alleviate bio-clogging in SSFCWs. Compared with the control, the two QQ treatments increased hydraulic

Lactones as promising biofilm inhibitors: disrupting bacterial communication for Next-Gen therapies.

The rise of antibiotic-resistant bacteria and biofilm-related infections presents a significant challenge in modern medicine, necessitating the development of novel therapeutic strategies. Lactones, compounds that can mimic N-acyl homoserine lactones (AHLs), have the potential to disrupt quorum sensing (QS) in pathogenic biofilm-forming bacteria. In this study, we evaluated the antibacterial and antibiofilm properties of five different lactones-γ-caprolactone, γ-decalactone, δ-decalactone, γ-octalactone, and γ-methyl decalactone-along with a lab-produced lactone from newly isolated strains, ag

Cholinergic neurotransmission links solitary chemosensory cells to nasal inflammation.

Solitary chemosensory cells (SCCs) of the nasal cavity are specialized epithelial chemosensors that respond to irritants through the canonical taste transduction cascade involving Gα-gustducin and transient receptor potential melastatin 5. When stimulated, SCCs trigger peptidergic nociceptive (or pain) nerve fibers, causing an alteration of the respiratory rate indicative of trigeminal activation. Direct chemical excitation of trigeminal pain fibers by capsaicin evokes neurogenic inflammation in the surrounding epithelium. In the current study, we test whether activation of nasal SCCs can trig

Early prediction of bronchopulmonary dysplasia by urinary metabolomics: a case-control study.

OBJECTIVE: Bronchopulmonary dysplasia (BPD), the most frequent complication of extreme preterm birth, lacks not only of a comprehensive definition but also of effective treatments and predictive tools. Metabolomics is a valuable tool to unravel the underlying pathogenetic pathways of diseases and identify possible early markers. The objective of this study was to find metabolic signatures of subsequent BPD development, defined and stratified as per Jobe and Bancalari 2001 NHICD Consensus. METHODS: In this observational case-control study, we initially enrolled 161 very preterm unmatched infan

The Cyclic Adenosine Monophosphate Phosphodiesterase CpdA Modulates cAMP Homeostasis, Stress Adaptability, Biofilm Formation, Motility, Quorum Sensing and Antibiotic Resistance of Aeromonas veronii.

Aeromonas veronii, a Gram-negative pathogen, is ubiquitous in nature and infects broad hosts, which becomes a serious threat to the public health. The cyclic adenosine monophosphate (cAMP), precisely modulated by cyclic nucleotide phosphodiesterase (PDE), regulates key cellular processes in bacteria. However, the specific function and regulatory role of PDE in A. veronii remain unclear. In this study, a Class III PDE gene named as cpdA in A. veronii WL-3 was identified for the first time, which shared 39.18%-49.19% identity with homologs from other bacteria and also contained the conserved met

Application of GC-MS technique for the determination of homocysteine thiolactone in human urine.

It is well established that homocysteine thiolactone (HTL) is associated with some health disorders, including cardiovascular diseases. HTL is a by-product of sulfur metabolic cycle. So far, its presence has been confirmed in human plasma and urine. It has been also shown that a vast majority of HTL is removed from human body through kidney. Thus, the aim of the current investigations has been the identification, separation and quantification of HTL in urine samples. For the first time a cheap, reliable and robust GC-MS method was developed for the determination of HTL in human urine in the fo

Functional characterization of the quorum sensing regulator RsaL in the plant-beneficial strain Pseudomonas putida WCS358.

In many bacteria, quorum sensing (QS) systems rely on a signal receptor and a synthase producing N-acyl-homoserine lactone(s) as the signal molecule(s). In some species, the rsaL gene, located between the signal receptor and synthase genes, encodes a repressor limiting signal synthase expression and hence signal molecule production. Here we investigate the molecular mechanism of action of the RsaL protein in the plant growth-promoting rhizobacterium Pseudomonas putida WCS358 (RsaL(WCS)). In P. putida WCS358, RsaL(WCS) displayed a strong repressive effect on the promoter of the QS signal syntha

Regulation of interbacterial interactions between Pseudomonas aeruginosa and Stenotrophomonas maltophilia by pqs quorum sensing.

Pseudomonas aeruginosa, an opportunistic pathogen, uses a trio of quorum sensing (QS) systems to regulate the production of some virulence factors. Two of these, the las and rhl systems, involve acyl-homoserine lactone signals; the third, called pqs, primarily uses the signal 2-heptyl-3-hydroxy-4(1H)-quinolone ("PQS"). We aimed to identify how interbacterial interactions are regulated between P. aeruginosa and Stenotrophomonas maltophilia, which co-occur in the airways of people with cystic fibrosis. We explored P. aeruginosa and S. maltophilia interactions using a co-culture model. S. maltoph

Coumarins Disrupt Cell-Cell Communication and Virulence in Priority Pathogens: Targeting the PQS Signalling System in Pseudomonas aeruginosa.

Cell-to-cell communication in microbial systems is known for its vital role in cellular signalling and gene expression. A specific form termed Quorum Sensing (QS) has received considerable attention since its discovery in the marine symbiont Aliivibrio fischeri. QS-controlled microbial functions are associated with bacterial virulence, pathogenicity, host-microbe interactions, and biofilm development. Interference in these signalling systems can modulate microbial virulence and pathogenicity, and microbial infection caused by drug-resistant pathogens. Plant-derived phytochemicals are considere

Phosphorus stress and spatial confinement lower the quorum-sensing activation threshold for phenazine production in Pseudomonas synxantha.

Bacteria often coordinate collective behaviors such as biofilm formation and secondary metabolite production through quorum sensing (QS), a regulatory system traditionally linked to high cell density. However, in environments such as soil, where microbial populations are spatially fragmented, sparse, nutrient-limited, and subject to mass transport, the mechanisms that enable QS-dependent processes remain incompletely understood. Here, we investigate the regulation of a secreted redox-active metabolite, phenazine-1-carboxylic acid (PCA), in Pseudomonas synxantha 2-79, a model rhizobacterium, un

Insights into the suppression of filamentous bulking by quorum quenching in the activated sludge process with low dissolved oxygen: Endogenous quorum quenching mediated multi-enzymatic degradation of AHLs and carbon-flux redistribution.

Low dissolved oxygen (DO) operation can substantially reduce aeration energy consumption in activated sludge systems but often induces filamentous sludge bulking dominated by Sphaerotilus natans (S. natans). This study isolated endogenous quorum quenching (QQ) bacteria from activated sludge and evaluated their potential to control low-DO bulking. Three endogenous QQ strains were newly isolated, among which Pseudochrobactrum sp. (P. sp.) showed excellent environmental adaptability and broad-spectrum N-acyl homoserine lactone degradation through multi-enzyme synergy, quenching C6-HSL, C8-HSL, an

Application of novel quorum quenching bacteria to mitigate biofouling in antibiotic-stressed membrane bioreactors: Performances and mechanisms.

Quorum quenching (QQ)-based strategies are efficient for biofouling control. However, the feasibility of using QQ bacteria in antibiotic-stressed membrane bioreactors (MBRs) remains unknown. In this study, we isolated three novel QQ strains (Bacillus sp. QX01 and QX03, Delftia sp. QX14) from the activated sludge of an actual MBR. They can degrade 11 N-acyl-homoserine lactones (AHLs) with high efficiencies and rates through intracellular QQ pathways involving putative acylases and lactonases. Running two lab-scale MBRs, we found that introducing antibiotics (sulfamethoxazole, azithromycin, and
