Tonsillar homing of Epstein-Barr virus-specific CD8+ T cells and the virus-host balance.

Patients with infectious mononucleosis (IM) undergoing primary EBV infection show large expansions of EBV-specific CD8+ T cells in the blood. While latent infection of the B cell pool is quickly controlled, virus shedding from lytically infected cells in the oropharynx remains high for several months. We therefore studied how responses localize to the tonsil, a major target site for EBV, during primary infection and persistence. In acute IM, EBV-specific effectors were poorly represented among CD8+ T cells in tonsil compared with blood, coincident with absence of the CCR7 lymphoid homing marke

Lower protein concentration in GCF from patients with periodontitis: an indicator of host-specific inflammatory reaction.

To study the local inflammatory reaction associated with gingivitis and periodontitis, the protein concentrations were measured in gingival crevicular fluid (GCF) from 3 types of site: (i) inflamed sites in patients with gingivitis alone; (ii) inflamed sites with or (iii) without tissue destruction in patients with periodontitis. The GCF was sampled with paper strips and the protein concentration was measured with the Bradford protein assay. Higher protein concentrations in GCF were found at inflamed sites in patients with gingivitis alone than at clinically similar sites without tissue destru

Enterotoxigenic Bacteroides fragilis Induces Host Genotype-Specific Colonic Epithelial and Immune Responses in Mice.

BACKGROUND: Colorectal cancer pathogenesis involves complex interactions between multiple risk factors including somatic mutations and microbial dysbiosis. A number of individual microbiota members have been implicated in colorectal cancer, including enterotoxigenic strains of Bacteroides fragilis (ETBF). Enterotoxigenic Bacteroides fragilis promotes inflammation in mouse models, which has been mechanistically linked to colon tumorigenesis. We hypothesized that ETBF would promote distinct patterns of colonic damage and inflammation in mice expressing different oncogenic mutations. METHODS: Mi

Viral Comorbidities Remodel Host Transcriptome and Redox Signaling in an NADPH Oxidase Isoform-Specific Manner.

Viral comorbidities elicit complex host responses by activating redox-sensitive signaling pathways, prominently those regulated by NADPH oxidase (Nox) enzymes. Nox are critical components of host defense, generating reactive oxygen species (ROS) that modulate key cellular signaling cascades. Under normal physiological conditions, Nox activity is tightly controlled; however, viral infections frequently disrupt this regulation, leading to aberrant upregulation of specific Nox isoforms. Elevated expression of individual Nox enzymes has been observed in infections such as influenza A and hepatitis

Sex-Specific Signatures of Circulating Protein and Cellular Host Responses Predicting COVID-19 Severity.

BACKGROUND/OBJECTIVES: Although COVID-19 is generally more severe in males, data on sex-specific differences in the predictive value of commonly used inflammatory biomarkers remain limited. The study aimed to evaluate the sex-specific prognostic performance of selected biomarkers during the Alpha variant wave. METHODS: In single-center study, univariate and multivariable regressions analyses, along with receiver operating characteristic curve (ROC) analyses, were performed to assess the association of acute-phase proteins, cytokines, and white blood cell indices (at admission and 7 days later

Transcriptomic Dissection of Bothrops moojeni Venom Reveals Fraction-Specific Modulation of Host Cellular Pathways.

Snake venom is a remarkably complex cocktail of bioactive molecules capable of hijacking diverse host physiological processes, yet how individual venom components drive these cellular responses remains a bit of a black box. To map these dynamics, we ran a comparative transcriptomic analysis on human osteoclastogenic cultures, exposing them continuously to crude Bothrops moojeni venom and its high (HMM) and low (LMM) molecular mass fractions throughout differentiation. This allowed us to capture the cumulative transcriptional shifts that unfold across the entire lifecycle of osteoclast developm

Strain-specific variation in virulence and host-immune modulation across the Cryptococcus gattii species complex.

Cryptococcus gattii is an environmental yeast capable of causing severe infections in immunocompetent hosts. Its virulence mechanisms remain less characterized than those of C. neoformans. This study aimed to evaluate the expression of major virulence factors and the immunomodulatory potential of eight C. gattii strains representing five genotypes and multiple sequence types (STs). Virulence traits-including capsule thickness, melanin and laccase production, phospholipase, hemolysin, urease, and DNase activities-were assessed using standardized in vitro assays. In vivo pathogenicity was evalua

Host-specific platelet-activating factor acetylhydrolase selectively remodels diacylglycerophospholipids to control schistosome development.

BACKGROUND: Schistosomiasis, one of the most prevalent neglected tropical diseases, is treated exclusively with praziquantel, which targets only adult worms. High reinfection rates and the potential emergence of drug resistance emphasize the need for a deeper understanding of host-parasite developmental biology for innovative therapeutic targets. Most studies have focused on parasite-derived immunomodulators and proteolytic enzymes essential for host invasion, while the contribution of host-derived factors to parasite development has remained largely unexplored. Here, we identify and functiona

Specific host factors determine resistance in a North American crayfish to the crayfish plague, Aphanomyces astaci.

The crayfish plague is caused by the oomycete Aphanomyces astaci with North American crayfish (for example Pacifastacus leniusculus and Procambarus clarkii) serving as carriers and vectors for this pathogen. This poses a constant threat to native crayfish in Europe, Asia, South America and Australia, which all are highly susceptible to this pathogen. In this study we now show how the symbiotic balance between the pathogen and its host are maintained at the molecular level. The host factors involved in this balance between the pathogen, A. astaci and the host, P. leniusculus, are one glycine-ri

Organ-specific Toxocara canis larvae migration and host immune response in experimentally infected mice.

We investigated organ specific Toxocara canis larval migration in mice infected with T. canis larvae. We observed the worm burden and systemic immune responses. Three groups of BALB/c mice (n=5 each) were orally administered 1,000 T. canis 2nd stage larvae to induce larva migrans. Mice were sacrificed at 1, 3, and 5 weeks post-infection. Liver, lung, brain, and eye tissues were collected. Tissue from 2 mice per group was digested for larval count, while the remaining 3 mice underwent histological analysis. Blood hematology and serology were evaluated and compared to that in a control uninfecte

Exploring the second intermediate hosts and morphology of human- and cat-specific Opisthorchis viverrini-like populations.

Infection by the zoonotic fish-borne trematode, Opisthorchis viverrini, remains a crucial health issue in Thailand and neighboring countries. Recently, molecular analysis revealed two populations of putative O. viverrini: one found primarily in human hosts ("human-specific" population) and the other primarily in cats ("cat-specific" population). It is unclear how the infective stages (metacercariae) of these different populations circulate among definitive and reservoir hosts in nature. To gain an insight into this, mitochondrial cox1 and nad1 gene sequences of metacercariae from fish intermed

Structural insights in cell-type specific evolution of intra-host diversity by SARS-CoV-2.

As the global burden of SARS-CoV-2 infections escalates, so does the evolution of viral variants with increased transmissibility and pathology. In addition to this entrenched diversity, RNA viruses can also display genetic diversity within single infected hosts with co-existing viral variants evolving differently in distinct cell types. The BriSΔ variant, originally identified as a viral subpopulation from SARS-CoV-2 isolate hCoV-19/England/02/2020, comprises in the spike an eight amino-acid deletion encompassing a furin recognition motif and S1/S2 cleavage site. We elucidate the structure, fu

Bortezomib Eliminates Persistent Chlamydia trachomatis Infection through Rapid and Specific Host Cell Apoptosis.

Chlamydia trachomatis, a parasitic intracellular bacterium, is a major human pathogen that causes millions of trachoma, sexually transmitted infections, and pneumonia cases worldwide. Previously, peptidomimetic inhibitors consisting of a hydrophobic dipeptide derivative exhibited significant inhibitory effects against chlamydial growth. Based on this finding, this study showed that both bortezomib (BTZ) and ixazomib (IXA), anticancer drugs characterized by proteasome inhibitors, have intensive inhibitory activity against Chlamydia. Both BTZ and IXA consisted of hydrophobic dipeptide derivative

Human norovirus exhibits strain-specific sensitivity to host interferon pathways in human intestinal enteroids.

Human noroviruses (HuNoVs) are the leading cause of viral gastroenteritis worldwide; yet currently, no vaccines or FDA-approved antiviral drugs are available to counter these pathogens. To understand HuNoV biology and the epithelial response to infection, we performed transcriptomic analyses, RT-qPCR, CRISPR-Cas9 modification of human intestinal enteroid (HIE) cultures, and functional studies with two virus strains (a pandemic GII.4 and a bile acid-dependent GII.3 strain). We identified a predominant type III interferon (IFN)-mediated innate response to HuNoV infection. Replication of both str

Epigenetic dysregulation of host gene expression in Toxoplasma infection with specific reference to dopamine and amyloid pathways.

Recent interest has focussed on the influence of infectious disease organisms on the host epigenome. Toxoplasma gondii infection acquired congenitally or in early life is associated with severe ocular and brain developmental anomalies, while persistent asymptomatic infection is a proposed risk factor for neurodegenerative and psychiatric disorders, including Parkinson's and Alzheimer's Diseases, and schizophrenia. Genome-wide analysis of the host methylome and transcriptome following T. gondii infection in a retinal cell line identified genes (132, 186 and 128 genes at 2, 6 and 24 h post-infec

Stage-Specific Changes in Plasmodium Metabolism Required for Differentiation and Adaptation to Different Host and Vector Environments.

Malaria parasites (Plasmodium spp.) encounter markedly different (nutritional) environments during their complex life cycles in the mosquito and human hosts. Adaptation to these different host niches is associated with a dramatic rewiring of metabolism, from a highly glycolytic metabolism in the asexual blood stages to increased dependence on tricarboxylic acid (TCA) metabolism in mosquito stages. Here we have used stable isotope labelling, targeted metabolomics and reverse genetics to map stage-specific changes in Plasmodium berghei carbon metabolism and determine the functional significance

The Vibrio cholerae ToxR Regulon Encodes Host-Specific Chemotaxis Proteins that Function in Intestinal Colonization.

Virulence gene regulation in Vibrio cholerae is under the control of the ToxR-ToxT regulatory cascade. Chemotaxis and net motility have been shown to influence the infectivity of Vibrio cholerae. V. cholerae toxR mutants do not synthesize proteins required for chemotaxis towards mucus. The inability of the toxR mutant strain to recognize and swim towards mucus is due to their failure to synthesize AcfB, a methyl-accepting chemotaxis protein. AcfB has previously been shown to be involved in intestinal colonization using the infant mouse model of cholera infection. Wild type V. cholerae recogniz

Multi-locus sequence analysis reveals host specific association between Bartonella washoensis and squirrels.

To clarify phylogenetic relationships and genetic diversity among Bartonella washoensis strains obtained from squirrels, multi-locus sequence analysis (MLSA) with the 16S rRNA, ftsZ, gltA, groEL, ribC, and rpoB genes was applied for 20 strains of B. washoensis isolated from five genera of squirrels (Tamias, Tamiasciurus, Glaucomys, Sciurus, and Spermophilus) within the family Sciuridae. Sequence similarities in the concatenated sequences of B. washoensis strains from squirrels of different genera ranged from 94.7% (Sciurus vs. Spermophilus) to 98.4% (Tamiasciurus vs. Glaucomys).
