Effects of the location of distal histidine in the reaction of myoglobin with hydrogen peroxide.

To clarify how the location of distal histidine affects the activation process of H2O2 by heme proteins, we have characterized reactions with H2O2 for the L29H/H64L and F43H/H64L mutants of sperm whale myoglobin (Mb), designed to locate the histidine farther from the heme iron. Whereas the L29H/H64L double substitution retarded the reaction with H2O2, an 11-fold rate increase versus wild-type Mb was observed for the F43H/H64L mutant. The Vmax values for 1-electron oxidations by the myoglobins correlate well with the varied reactivities with H2O2. The functions of the distal histidine as a gene

Investigation of the effect of hydrogen peroxide on the chromosomes of young and elderly individuals.

Active oxygen species have been considered to be responsible for the aging process and for the induction and initiation of neoplastic processes. The effect of hydrogen peroxide, an active oxygen species, was investigated in the chromosomes of three young women (20-21 years of age) and of three elderly women (73-79 years of age) in a culture medium favorable to the appearance of folate-sensitive fragile sites. Hydrogen peroxide at a final concentration of 5 X 10(-6) during the final hours of culture caused a significant increase in hypodiploidy and structural aberrations, chromatid gaps in part

Ethylene participates in strigolactone-triggered stomatal closure via Gα protein-activited hydrogen peroxide and hydrogen sulfide synthesis in Arabidopsis.

The signaling cascade of strigolactone (SL)-regulated stomatal closure in Arabidopsis thaliana was investigated using pharmacological assays, fluorescence microscopy, spectrophotometry, gas chromatography, RT-PCR, and qRT-PCR. In wild-type plants, GR24 (a synthetic strigolactone analog)-induced stomatal closure was significantly inhibited by ethylene biosynthesis/perception inhibitors or Gα inhibitor pertussis toxin (PTX). GR24 obviously promoted closure in eto1-1, cGα1 and wGα1 mutants, but failed to do so in mutants etr1-1, etr1-3, gpa1-1 and gpa1-2. GR24 also upregulated 1-aminocyclopropane

Requirement of hydrogen peroxide generation in TGF-beta 1 signal transduction in human lung fibroblast cells: involvement of hydrogen peroxide and Ca2+ in TGF-beta 1-induced IL-6 expression.

Stimulation of human lung fibroblast cells with TGF-beta1 resulted in a transient burst of reactive oxygen species with maximal increase at 5 min after treatment. This reactive oxygen species increase was inhibited by the antioxidant, N-acetyl-l -cysteine (NAC). TGF-beta1 treatment stimulated IL-6 gene expression and protein synthesis in human lung fibroblast cells. Antioxidants including NAC, glutathione, and catalase reduced TGF-beta1-induced IL-6 gene expression, and direct H2O2 treatment induced IL-6 expression in a dose-dependent manner. NAC also reduced TGF-beta1-induced AP-1 binding act

Engineering a Hydrogen Peroxide-Activated Hydrogen Sulfide Donor-Based Fluorescent Agent for Integrated Diagnosis and Therapy of Chronic Wounds.

Diabetic chronic wounds are one of the most severe complications of diabetes mellitus, which are pathologically characterized by persistent oxidative stress-inflammatory cascades. Thus, it is vital for effective diagnosis and treatment of diabetic chronic wounds by targeting the critical pathological oxidative stress feature in the tissue microenvironment. Herein, regarding the overexpressed hydrogen peroxide (H2O2) at the oxidative stress sites, we innovatively developed a H2O2-activated hydrogen sulfide (H2S) donor-based near-infrared fluorescent theragnostic agent (NDTA) to achieve synergis

Scanning electron microscopy study of dental enamel surface exposed to 35% hydrogen peroxide: alone, with saliva, and with 10% carbamide peroxide.

Several vital bleaching systems have been introduced in response to the demand in esthetic dentistry. The active agents are commonly hydrogen peroxide or carbamide peroxide used in at-home or in-office techniques. Although generally positive results have been reported concerning the whitening ability of these agents, concerns still remain as to their effects on dental tissues. The purpose of this investigation was to evaluate the effect of these bleaching agents on the enamel surface morphology. Twelve extracted teeth were used according to three experimental protocols. In experimental protoco

Integration of hydrogen peroxide sensing and detoxification by the C. elegans nervous system.

Hydrogen peroxide (H2O2) is the most common reactive chemical threat organisms face. Here, we map the neural circuit that drives chemotactic escape from environmental H2O2 in the nematode C. elegans. Twenty-four neuron classes with sensory endings at the mouth and nose respond to H2O2, with dynamics encoding stimulus intensity and exposure history. Sensing begins when H2O2 oxidizes the peroxidatic and resolving cysteines of the cytosolic peroxiredoxin PRDX-2, which relays this oxidative signal to cysteines on the LITE-1 and GUR-3 ion channels, triggering calcium influx in sensory neurons that

Predictive Modeling of Bacterial Inactivation With Hydrogen Peroxide Over a Cobalt Ferrite Catalyst.

Hydrogen peroxide activated by a cobalt ferrite catalyst generates hydroxyl radicals that effectively inactivate bacteria. A continuous-flow packed-bed reactor containing cobalt ferrite was developed and tested for bacterial disinfection. The inactivation kinetics were highly nonlinear. The classical Weibull model accurately described the initial decline in bacterial counts at low H2O2 concentration (3 mM) but lost accuracy at longer contact times and higher concentrations (6-10 mM). To improve prediction, 10 machine learning algorithms were evaluated. Gradient boosting (GB) and random forest

MOFs-Derived Integrated Hierarchical Carbon Electrode for Hydrogen Peroxide Synthesis in Acidic Media.

Current research on electrosynthesis of hydrogen peroxide mainly focuses on optimizing intrinsic active sites, while overlooking the constraints imposed by mass transport limitations and weak oxygen adsorption capacity during the reaction. Herein, we developed an integrated hierarchically porous carbon electrode composed of MOF-derived porous carbon sheets arrayed on carbon cloth and epitaxially grown carbon nanotube networks, with highly active Co-Nx sites generated in situ. During the synthesis, pure water was initially employed as the solvent to promote the ordered growth of sheet-like MOFs

High-quality hydrogen peroxide production through electrochemical reduction of anthraquinone in ambient environment.

At present, the traditional anthraquinone method is the main method for producing high-quality and high-concentration H2O2, but it involves the use of H2, resulting in high carbon emissions and certain risks. The double-electron oxygen reduction reaction has become a more environmentally friendly alternative method for producing H2O2. However, at high overpotentials, the production and conversion rates of H2O2 are still limited. Here, we report an electrochemical method that can convert H2O and formaldehyde into high-concentration H2O2 and high-value formate salts with high product purity unde

A selective near-infrared fluorescent probe for detecting hydrogen peroxide in Alzheimer's disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder with growing incidence among younger populations, highlighting the urgent need for early and accurate detection of AD-related biomarkers. Excessive production of reactive oxygen species (ROS), particularly hydrogen peroxide (H2O2), induces oxidative stress, leading to neuronal damage and death, which implicates H2O2 as a potential biomarker for AD. In this study, leveraging the advantages of fluorescence imaging, we developed a novel near-infrared (NIR) fluorescent probe, LDMD-B, capable of selectively detecting H2O2 through

Impact of bleaching on white spot lesions: hydrogen peroxide permeability and color alteration.

OBJECTIVES: To assess the penetration of hydrogen peroxide (HP) into the pulp chamber and the bleaching efficacy on teeth with white spot lesions (WSLs) compared to sound teeth, when subjected to both in-office and at-home bleaching treatments. MATERIALS AND METHODS: Forty premolars were allocated into four groups (n = 10 each): sound teeth and teeth with artificially induced WSLs, both subjected to at-home bleaching (16% carbamide peroxide) and in-office bleaching (35% hydrogen peroxide). WSLs were induced in half of the specimens using a 14-day pH-cycling protocol alternating between demine

Electrochemical sensing of hydrogen peroxide using SnO2-SA nanoparticles with enhanced catalytic activity.

Accurate quantification of hydrogen peroxide (H2O2) is essential for biomedical analysis and process monitoring, yet metal-oxide sensors often suffer from limited active-site exposure and sluggish charge transport. Here we report a sodium-alginate (SA)-templated route to porous SnO2 nanoparticles (SnO2-SA) that integrates high crystallinity with abundant surface oxygen vacancies and a permeable mesoporous network. When drop-cast on a glassy carbon electrode (GCE), the SnO2-SA enables non-enzymatic H2O2 detection with a wide linear range of 0.02-2.8 mM, high sensitivity of 381.12 μA mM-1 cm-2,

Management and Therapeutic Considerations of Hydrogen Peroxide-Induced Necrotizing Gastroenteritis: A Case Report.

Necrotizing gastroenteritis is a rare but severe condition characterized by extensive inflammation and mucosal necrosis of the gastrointestinal tract. It is most commonly seen in neonates, infants, and immunocompromised individuals. We present a rare case of a 46-year-old female patient who developed necrotizing gastroenteritis following accidental ingestion of 3% hydrogen peroxide, manifesting as three days of epigastric pain, heartburn, and recurrent emesis. This report details the patient's clinical presentation, diagnostic evaluation, therapeutic strategies, and management course. The case

Glycerol metabolism contributes to competition by oral streptococci through production of hydrogen peroxide.

UNLABELLED: As a biological byproduct from both humans and microbes, glycerol's contribution to microbial homeostasis in the oral cavity remains understudied. In this study, we examined glycerol metabolism by Streptococcus sanguinis, a commensal associated with oral health. Genetic mutants of glucose-PTS enzyme II (manL), glycerol metabolism (glp and dha pathways), and transcriptional regulators were characterized with regard to glycerol catabolism, growth, production of hydrogen peroxide (H2O2), transcription, and competition with Streptococcus mutans. Biochemical assays identified the glp pa

Scinderin Promotes Hydrogen Peroxide-induced Lens Epithelial Cell Injury in Age-related Cataract.

BACKGROUND: Scinderin (SCIN) is a calcium-dependent protein implicated in cell growth and apoptosis by regulating actin cleavage and capping. In this study, we investigated the role of SCIN in hydrogen peroxide-induced lens epithelial cell (LEC) injury related to age-related cataract (ARC). METHODS: Anterior lens capsules from ARC patients were collected to examine SCIN expression levels. Immortalized human LEC cell line SRA01/04 and lens capsules freshly isolated from mice were induced by H2O2 to mimic the oxidative stress in ARC. The role of SCIN was investigated by gain-of-function (overex

Evidence for endogenous hydrogen peroxide production by E. coli fatty acyl-CoA dehydrogenase.

Aerobic organisms continuously generate internal superoxide and hydrogen peroxide, which can damage enzymes and impair growth. To avoid this problem cells maintain high levels of superoxide dismutases, catalases, and peroxidases. Surprisingly, we do not know the primary sources of these reactive oxygen species (ROS) in living cells. However, in vitro studies have shown that flavoenzymes can inadvertently transfer electrons to oxygen. Therefore, it seems plausible that substantial ROS may be generated when large metabolic fluxes flow through flavoproteins. Such a situation may arise during the

Hydrogen peroxide is not generated intracellularly in human neural spheroids during ischemia-reperfusion.

Reactive oxygen species (ROS) are considered a primary source of damage during ischemic stroke. However, the precise timing of ROS production (during hypoxia or reperfusion) remains unclear. Cellular 3D spheroids are often proposed as an optimal alternative to both 2D cell cultures and animal models in modeling disease conditions. Here we report live imaging of hydrogen peroxide dynamics during the acute phase of hypoxia and reperfusion in human iPSC-derived neural spheroids, stably expressing fluorescent biosensor HyPer7. Contrary to previous reports, we did not observe a hydrogen peroxide pr

Hydrogen peroxide attenuates rhinovirus-induced anti-viral interferon secretion in sinonasal epithelial cells.

BACKGROUND: Altered innate defense mechanisms, including an imbalance between oxidants and antioxidants release, have been implicated in the pathogenesis of chronic rhinosinusitis (CRS). The aim of this study is to investigate whether oxidative stress may attenuate the secretion of anti-viral interferons in human sinonasal mucosa. METHODS: The levels of H2O2 in nasal secretion were increased in patients with CRS with nasal polyps, compared with that of CRS patients without nasal polyps and control subjects. Normal sinonasal epithelial cells derived from healthy subjects were cultured under an
