Hydrogen-Deuterium Exchange and Hydroxyl Radical Footprinting for Mapping Hydrophobic Interactions of Human Bromodomain with a Small Molecule Inhibitor.

Mass spectrometry (MS)-based protein footprinting, a valuable structural tool in mapping protein-ligand interaction, has been extensively applied to protein-protein complexes, showing success in mapping large interfaces. Here, we utilized an integrated footprinting strategy incorporating both hydrogen-deuterium exchange (HDX) and hydroxyl radical footprinting (i.e., fast photochemical oxidation of proteins (FPOP)) for molecular-level characterization of the interaction of human bromodomain-containing protein 4 (BRD4) with a hydrophobic benzodiazepine inhibitor. HDX does not provide strong evid

Self-reconstruction of single copper atoms drives precise hydroxyl radicals generation for efficient water disinfection.

Antibiotic-resistant bacteria (ARB) and their genes (ARGs) pose a significant risk to public health and the ecosystem. Conventional water disinfection efficiently eliminates pathogens, however, it requires intensive chemical consumption but inefficiently removes ARGs. Herein, we propose an on-demand electrochemical platform that simultaneously eliminate both ARB and ARGs with in-situ generated hydroxyl radicals (·OH) without additional chemical inputs. Single-atom copper catalysts with CuN4 coordination structure (SA-CuN4/NC) are precisely tuned to directly convert oxygen into controllable ·OH

A sulfonate-modified cyanine-based photoacoustic probe for selective detection of hydroxyl radicals in diabetic liver injury.

Hydroxyl radicals (•OH) play a critical role in oxidative stress-related diseases, yet their real-time detection in vivo remains challenging. We developed a hydroxyl radical-responsive photoacoustic probe (OHP) by modifying IR780-SO3H with enhanced hydrophilicity (logP = -1.051) for improved biodistribution. Structural characterization confirmed the selective reduction of the conjugated system, while in vitro studies demonstrated OHP's selective and linear response to •OH with minimal interference from other ROS/RNS. In diabetic mice, OHP enabled dynamic monitoring of hepatic •OH levels, revea

In Situ Fluorescence Imaging Reveals Contribution of Cerebral Hydroxyl Radicals in Hyperhomocysteinemia-Induced Alzheimer-like Dementia.

Elevated plasma level of homocysteine, also termed as hyperhomocysteinemia, is acknowledged as a significant and independent risk factor of Alzheimer's disease. However, the mechanistic insight has not been thoroughly elucidated yet. In this work, 3,5-dihydroxybenzyloxy was explored as the unique reaction trigger and integrated into the naphthalimide fluorophore via a carbamate linker to afford a new probe for •OH imaging. •OH treatment induced aromatic hydroxylation and subsequent elimination reaction to release the caged fluorophore, accompanied with a highly specific and sensitive turn-on f

Production of Hydroxyl Radicals from Oxygenation of Simulated Acid Mine Drainage in Presence of Extracellular Polymers Substances.

The reaction mechanisms Fe(II) abiotic oxidation produce ·OH by CaCO3-induced in acid mine drainage (AMD) are well-documented, but little is known about the influence of extracellular polymeric substances (EPS) secreted by microorganisms on Fe(II) oxidation in AMD. In this study, ·OH production was experimently measured from oxygenation of simulated AMD in the presence of EPS. The cumulative ·OH increased from 56.75 to 158.70 µM within 24 h at pH 3 with the increase in EPS concentration from 0 to 12 mg/L. An appropriate pH (about 6) and EPS (6 mg/L) concentration were required for the moderate

A Ratiometric Photoacoustic Probe with a Reversible Response to Hydrogen Sulfide and Hydroxyl Radicals for Dynamic Imaging of Liver Inflammation.

Reversible imaging probes that allow for the dynamic visualization of the redox cycle between hydroxyl radical (⋅OH) and hydrogen sulfide (H2 S) are vital to probe the redox imbalance-involved pathological process in vivo. Herein, we report a reversible ratiometric photoacoustic (PA) imaging nanoprobe (1-PAIN) for the real-time imaging of ⋅OH/H2 S redox cycle in vivo. 1-PAIN displays a low PA ratio between 690 and 825 nm (PA690 /PA825 ), which significantly increases by ≈5-fold upon oxidation by ⋅OH, and is switched back to the initially low PA690 /PA825 value upon reduction by H2 S. 1-PAIN co

At the crossroad of photochemistry and radiation chemistry: formation of hydroxyl radicals in diluted aqueous solutions exposed to ultraviolet radiation.

Formation yields of ˙OH radicals were precisely determined in aqueous solutions of coumarin-3-carboxylic acid and ferrous sulfate (i.e., Fricke dosimeter) exposed to 253.7 nm radiation delivered from a continuous source. Quantum yield of ˙OH radicals was determined as ∼0.08, i.e., roughly one out of twelve photons, efficiently absorbed in UV-illuminated solutions, produced one ˙OH radical. Energetically, a water molecule should undergo a correlated action of at least two 4.9 eV photons delivering enough energy for direct H-OH dissociation (5.0-5.4 eV). We suggest a mechanism based on an intera

Superoxide dismutase enhanced the formation of hydroxyl radicals in a reaction mixture containing xanthone under UVA irradiation.

To clarify the effect of superoxide dismutase (SOD) on the formation of hydroxyl radical in a standard reaction mixture containing 15 microM of xanthone, 0.1 M of 5,5-dimethyl-1-pyrroline N-oxide (DMPO), and 45 mM of phosphate buffer (pH 7.4) under UVA irradiation, electron paramagnetic resonance (EPR) measurements were performed. SOD enhanced the formation of hydroxyl radicals. The formation of hydroxyl radicals was inhibited on the addition of catalase. The rate of hydroxyl radical formation also slowed down under a reduced oxygen concentration, whereas it was stimulated by disodium ethylene

Effect of barbiturates on hydroxyl radicals, lipid peroxidation, and hypoxic cell death in human NT2-N neurons.

BACKGROUND: Barbiturates have been shown to be neuroprotective in several animal models, but the underlying mechanisms are unknown. In this study, the authors investigated the effect of barbiturates on free radical scavenging and attempted to correlate this with their neuroprotective effects in a model of hypoxic cell death in human NT2-N neurons. METHODS: Hydroxyl radicals were generated by ascorbic acid and iron and were measured by conversion of salicylate to 2,3-dihydroxybenzoic acid. The effect of barbiturates on lipid peroxidation measured as malondialdehyde and 4-hydroxynon-2-enal was

Mazindol attenuates the 3,4-methylenedioxymethamphetamine-induced formation of hydroxyl radicals and long-term depletion of serotonin in the striatum.

The formation of hydroxyl radicals following the systemic administration of 3,4-methylenedioxymethamphetamine (MDMA) was studied in the striatum of the rat by quantifying the stable adducts of salicylic acid and D-phenylalanine, namely, 2,3-dihydroxybenzoic acid (2,3-DHBA) and p-tyrosine, respectively. The repeated administration of MDMA produced a sustained increase in the extracellular concentration of 2,3-DHBA and p-tyrosine, as well as dopamine. The MDMA-induced increase in the extracellular concentration of both dopamine and 2,3-DHBA was suppressed in rats treated with mazindol, a dopamin

Dual-channel fluorescence detection of hydroxyl radical based on red emitted carbon dots derived from imidazole cationic organic cage.

The dual-channel fluorescent sensing of hydroxyl radicals (·OH) is of great importance for environment and life. Organic cage for fluorescence sensing suffers from short excitation/emission wavelength and poor water solubility, while the application of carbon quantum dots is also hindered by the time-consuming preparation in high pressure and temperature conditions. Herein, we report a red emitted carbon dots derived from cationic organic cage for dual-channel fluorescent sensing of ·OH. The abundant flexible methylene within the cationic organic cage leads to remarkable collapse of intrinsic

A fluorescent hydrogen sulfide donor featuring hydroxyl radical responsiveness promotes diabetic wound healing through the regulation of macrophage polarization.

Hydroxyl radicals are among the most reactive and destructive reactive oxygen species, capable of inducing severe oxidative damage to critical biomolecules-including proteins, DNA, and lipids-thereby contributing to cellular dysfunction, senescence, apoptosis, and the pathogenesis of numerous diseases. In contrast, hydrogen sulfide (H2S) has emerged as a key endogenous signaling molecule with well-documented antioxidant, anti-inflammatory, and tissue-repair properties, prompting growing interest in its therapeutic applications. This study developed a novel hydroxyl radical-responsive hydrogen

Agricultural amendments enhanced the redox cycling of iron species and hydroxyl radical formation during redox fluctuation of paddy soil.

Hydroxyl radical (•OH) plays a critical role in accelerating organic contaminant attenuation during water-table decline in paddy soil, but the impacts of widely applied agricultural amendments (e.g., organic manure, rice straw, and biochar) on these processes have been rarely explored. Hence, the effects of agricultural amendments on •OH formation and pollutant degradation were examined based on field experiments. Compared with control, organic fertilizer (supplying more organic carbon (OC) and bioavailable elements that promoted Fe(II) formation by microorganisms) enhanced •OH production by 0

Tropospheric degradation of 2-fluoropropene (CH3CFCH2) initiated by hydroxyl radical: Reaction mechanisms, kinetics and atmospheric implications from DFT study.

Degradation of hydrofluoro-olefins (HFOs) with oxidants plays a significant role in the troposphere. Thus, we have investigated detail theoretical calculations of hydroxyl radical (•OH) initiated oxidation of 2-fluoropropene (CH3CFCH2) using M06-2X/6-311++G(d,p) level of theory. Here, we have considered different possible H-abstraction and OH addition for the degradation of CH3CFCH2 molecule. The potential energy analysis shows that OH-addition channels are more dominant than H-abstraction channels. The calculated reaction enthalpies (ΔrH°) and Gibbs free energies (ΔrG°) also suggest that OH-a

A comparative study on the sensitive detection of hydroxyl radical using thiol-capped CdTe and CdTe/ZnS quantum dots.

Four types of water-soluble luminescent quantum dots (QDs) whose surface was functionlaized with thioglycolic acid (TGA), 3-mercaptopropionic acid (MPA), or glutathione (GSH), were investigated for the sensitive and selective detection of hydroxyl radical (·OH) in aqueous media. It was found that the type of capping agent and QD influenced the sensitivity of the probe. The order of sensitivity of the probe was: GSH-CdTe@ZnS > MPA-CdTe@ZnS > TGA-CdTe > MPA-CdTe QDs. Under the optimum conditions, a limit of detection as low as 8.5 × 10(-8) M was obtained using GSH-CdTe@ZnS. The effects of foreig

Melatonin prevents the increase in hydroxyl radical-spin trap adduct formation caused by the addition of cisplatin in vitro.

We studied the generation of reactive oxygen species (ROS) caused by cisplatin administration and the preventive effect of melatonin, the main secretory product of the pineal gland, on the reaction in vitro using electron spin resonance spectroscopy. Cisplatin induced generation of the hydroxyl radical (OH*) in phosphate buffer in pH 7.4 as a dose-dependent manner. However, OH* was not generated in phosphate buffer containing chloride ions at concentration exceeding 120 mM. The induction of OH. production by cisplatin was completely inhibited by the addition of melatonin, but not by the additi

Non-protein-bound transition metals and hydroxyl radical generation in cerebrospinal fluid of newborn infants with hypoxic ischemic encephalopathy.

Among various hypothetical mechanisms for the in vivo production of reactive oxygen species, transition metal-catalyzed reactions in cooperation with a biologic reducing agent like ascorbic acid or superoxide may be some of the most important. In the present study, we retrospectively examined the existence of non-protein-bound metal ions, an essentially hazardous pro-oxidant form of various transition metals, and the occurrence of metal-catalyzed reactive oxygen species production in cerebrospinal fluid (CSF) of 10 infants with hypoxic ischemic encephalopathy (HIE) subsequent to perinatal asph

Differential roles of hydrogen peroxide and hydroxyl radical in cisplatin-induced cell death in renal proximal tubular epithelial cells.

Reactive oxygen species (ROS) have been suggested as important mediators of cisplatin-induced acute renal failure in vivo. However, our previous studies have shown that cisplatin-induced cell death in vitro could not be prevented by scavengers of hydrogen peroxide and hydroxyl radical in rabbit renal cortical slices. This discrepancy may be attributed to differential roles of ROS in necrotic and apoptotic cell death. We therefore examined, in this study, the roles of ROS in necrosis and apoptosis induced by cisplatin in primary cultured rabbit proximal tubule. Cisplatin induced necrosis at hig

Improved method for the estimation of hydroxyl free radical levels in vivo based on liquid chromatography with electrochemical detection.

Free radical damage to proteins, lipids, DNA and RNA has been thought to play an important role in many diseases as well as the aging process. One free radical, the hydroxyl free radical (HFR), is extremely reactive and is difficult to measure directly. HFRs were quantified by measuring the hydroxylation products 2,3- and 2,5-dihydroxybenzoic acids (DHBAs) formed as a result of the reaction between HFR and systemically administered salicylate (SAL). DHBAs and SAL concentrations were determined using RP-HPLC with dual coulometric electrode detection. The method has limits of detection of 1 pg f

Direct evidence for hydroxyl radical-induced damage to nucleic acids by chromium(VI)-derived species: implications for chromium carcinogenesis.

Reduction of Cr(VI) by NADH and NADPH has been shown to yield Cr(V) species, which have been detected by electron paramagnetic resonance (EPR) spectroscopy. The fine structure on the EPR signal of the Cr(V) species is consistent with the presence of two NAD(P)H ligands in a square-pyramidal arrangement with a single oxygen (oxo) group at the apex. Neither this species nor the initial Cr(VI) complex damage DNA components as evidenced by the lack of effect of these compounds on the optical and EPR signals of the Cr(VI) and Cr(V) species respectively. Addition of hydrogen peroxide to the Cr(V) sp
