Chronic invasive fungal sinusitis: a report of two atypical cases.

The purpose of this article is to describe a chronic variant of invasive fungal sinusitis (IFS) and discuss its management. This is a retrospective review of two cases of IFS that were characterized by atypical clinical courses. Patient 1 was a 75-year-old man with noninsulin-dependent diabetes mellitus who came to us with a 5-month history of headache. Computed tomography detected an opacified left sphenoid sinus. After the man failed to respond to medical therapy, he underwent a left endoscopic sphenoidotomy. Pathologic examination revealed that septate, branching fungal hyphae had invaded t

Cell Biology of Botrytis: Live-Cell Imaging Techniques and Protein Localization Protocols.

Botrytis cinerea is a widely studied model organism for plant-fungal interactions. This chapter provides simple and easy protocols for live-cell imaging techniques and subcellular protein localization studies in B. cinerea in vitro. Using a simple microscopy setup, we describe methods for imaging spore germlings and hyphae and for studying protein dynamics in response to the antifungal compound α-tomatine. The protocols highlight the application of fluorescence microscopy to visualize sterol distribution and protein localization patterns, enabling deeper insights into fungal cell biology.

In Vitro Culture Systems to Study the Functions of Common Mycorrhizal Network in Arbuscular Mycorrhizal Symbioses.

In vitro culture systems provide a controlled platform for studying common mycorrhizal networks (CMNs) in arbuscular mycorrhizal fungi (AMF), overcoming the difficulties of pot- or field-based approaches. These systems make it possible to study with precision plant-to-plant communication through the exchange of nutrients and signaling compounds. CMNs also open up new avenues for exploring the hyphobiome by shedding light on the microbial communities living on the surface of the hyphae connecting plants. Here, we present standardized, compartmented systems establishing contaminant-free CMNs for

Invasive aspergillosis-on-chip: A quantitative treatment study of human Aspergillus fumigatus infection.

Invasive pulmonary aspergillosis is associated with a high mortality rate and poses a direct threat to immunocompromised patients. Here, we present the invasive aspergillosis-on-chip (IAC) model to investigate Aspergillus fumigatus infection in vitro. The model allows the study of the lateral growth and the invasive behaviour of fungal hyphae from the epithelium into the endothelial cell layer in an alveolus-on-chip model. We established an algorithm-based analysis pipeline for three-dimensional confocal microscopy images to visualize and quantify fungal morphology, including hyphal growth and

Interkingdom interaction between C. albicans and S. salivarius on titanium surfaces.

BACKGROUND: In oral candidiasis models, Candida albicans and Streptococcus salivarius sp. biofilms have an antagonistic relationship. Due to this, S. salivarius have been used experimentally as probiotic. However, the interaction between these microorganisms in the peri-implantitis-like microenvironment remains unknown. This study aimed to evaluate the interaction between C. albicans and S. salivarius biofilms developed on titanium surfaces, under reduced oxygen levels. METHODS: Titanium specimens were pre-conditioned with artificial saliva (1 h, 37 °C). Single-species biofilms of C. albicans

Two new species of Resupinatus (Resupinataceae, Agaricales) from China.

Within the typical saprotrophic genus Resupinatus, only 47 species are currently recognized worldwide. Most of these species are reported from Europe and North America, whereas only 12 species have been recorded from China. Species of Resupinatus are characterized by laterally or dorsally attached basidiomata, sessile or with a lateral pseudostipe, a gelatinous pileus, smooth and inamyloid basidiospores, cylindrical to coralloid cheilocystidia with apical protrusions, and pileipellis hyphae with branches or protrusions. They usually occur on dead branches and well-decayed wood in mixed conifer

Fruiting Bodies of Aspergillus in a Cavitary Lung Mass: A Rare Finding on Fine Needle Aspiration.

Endobronchial ultrasound (EBUS) guided fine needle aspiration is a modality for diagnosing lung nodules as well as fungal organisms. One of the most frequently isolated fungal organisms from immunocompromised patients or immunocompetent patients with severe COPD is Aspergillus spp., which is characterized by septate hyphae with progressive dichotomous branching. However, this finding is not entirely specific to Aspergillus spp. since it can be seen in non-Aspergillus fungi. Nonetheless, the presence of fruiting bodies, background inflammation, and in correlation with the clinical and radiologi

Candida albicans cells exhibit media specific proteomic profiles during induction of filamentation.

Candida albicans is an opportunist pathogen responsible for a broad spectrum of infections, from superficial mycosis to the systemic disease candidiasis. C. albicans has various morphological forms, including unicellular budding yeasts, filamentous pseudohyphae and true hyphae, and the ability to switch from yeast to hyphal forms is a key survival mechanism underlying the adaptation of the pathogen to the microenvironments encountered within the host. Filamentation is regulated by multiple signalling pathways and its induction in different growth media in vitro has often led to conflicting res

CAR T cells targeting Aspergillus fumigatus are effective at treating invasive pulmonary aspergillosis in preclinical models.

Aspergillus fumigatus is a ubiquitous mold that can cause severe infections in immunocompromised patients, typically manifesting as invasive pulmonary aspergillosis (IPA). Adaptive and innate immune cells that respond to A. fumigatus are present in the endogenous repertoire of patients with IPA but are infrequent and cannot be consistently isolated and expanded for adoptive immunotherapy. Therefore, we gene-engineered A. fumigatus-specific chimeric antigen receptor (Af-CAR) T cells and demonstrate their ability to confer antifungal reactivity in preclinical models in vitro and in vivo. We gene

Muriform Cells Can Reproduce by Dividing in an Athymic Murine Model of Chromoblastomycosis due to Fonsecaea pedrosoi.

Transformation of Fonsecaea pedrosoi into muriform cells enhances the resistance against phagocytosis and elimination by host immune cells, and links to the chronicity of chromoblastomycosis. Here, we aim to determine whether the muriform cells can reproduce in tissue without reverse transformation into hyphal form by using an experimental nu/nu-BALB/c mouse model of chromoblastomycosis due to F. pedrosoi. During the whole 81-day observation period, most of the hyphal inocula had transformed into muriform cells at 75 days postinoculation and maintained as this parasitic morphology till 81 days

Onychomycosis caused by Aspergillus subramanianii.

We describe a case of a 23-year-old female patient with no apparent underlying diseases. She showed a discoloration of the proximal portion of the left big toenail with paronychia. Direct microscopy revealed septate hyphae with conidiophores, and a periodic acid-Schiff-stained nail specimen revealed septate hyphae branching at angles of approximately 45°. On the basis of phylogenetic analysis, we finally arrived at the diagnosis of ungual aspergillosis caused by Aspergillus subramanianii. After p.o. administration of terbinafine and topical application of 10% efinaconazole solution, the diseas

Concomitant Aspergillus Species Infection and Squamous Cell Carcinoma Diagnosed on Pap Smear.

Concomitant infection with Aspergillus species and cervical squamous cell carcinoma in the female genital tract is a rare occurrence and attributed to the opportunistic nature of infection in the immunocompromised state due to the underlying malignancy. The contamination of smears with Aspergillus species should be excluded. The diagnosis of Aspergillus species infection along with squamous cell carcinoma was established on cervicovaginal pap smears in a 62-year-old female presented to gynecological clinic with complaints of stress urinary incontinence. Speculum examination revealed first-degr

[A case of primary pyoderma-like aspergillosis occurring in a patient with a cervical spinal cord injury].

A 65-year-old man had tetraplegia caused by a cervical spinal cord injury, and could only lie in bed with a respirator. On the 14 th day of hospitalization, a rash developed on his back. The eruption grew rapidly, and became a giant erythematous plaque with ulcer, pustules, and red papules. Direct KOH examination showed branching Aspergillus hyphae. A slide culture showed subglobose shaped vesicles with phialides. Based on these findings, the case was diagnosed as primary pyoderma-like aspergillosis caused by Aspergillus fumigatus. He was treated with bifonazole and sulfadiazine silver, and on

Paecilomyces sinusitis in an immunocompromised adult patient: case report and review.

A case of fungal sinusitis caused by Paecilomyces lilacinus that was unresponsive to amphotericin B and involved a patient with acute myeloid leukemia is described. Histologic examination of sinus tissue and periorbital bone demonstrated invasion by a fungus with septate hyphae, which was identified in culture as P. lilacinus. The isolate was resistant to amphotericin B but susceptible to itraconazole. The patient responded clinically when itraconazole was added to the treatment regimen. Invasive aspergilar infections are frequently diagnosed by histology. Other fungi such as Fusarium, Pseudal

Influence of manganese on morphology and cell wall composition of Aspergillus niger during citric acid fermentation.

Morphology and cell wall composition of Aspergillus niger were studied under conditions of manganese sufficient or deficient cultivation in an otherwise citric acid producing medium. Omission of Mn2+ (less than 10(-7) M) from the nutrient medium of Aspergillus niger results in abnormal morphological development which is characterized by increased spore swelling, and squat, bulbeous hyphae. Fractionation and analysis of manganese deficient cell walls revealed increased chitin and reduced beta-glucan contents as well as reduction of galactose containing polymers, as compared to cell walls from m

Experimental aspergillosis in rats infected via intraperitoneal and subcutaneous routes.

Normal adult rats infected via the subcutaneous (s.c.) route with viable spores of Aspergillus fumigatus develop serum antibodies, measured by passive haemagglutination and passive cutaneous anaphylaxis (PCA), to extracts of the fungus. No significant histological abnormality was detected but s.c. inoculation in cortisone-treated animals induced the formation of granulomas frequently found in lymph nodes including those of the mesentery and paratracheal group. These granulomas were associated with the presence of both spores and fungal hyphae. Intraperitoneal inoculation of A. fumigatus spores
