Phosphoproteomic Analysis of the Jejunum Tissue Response to Colostrum and Milk Feeding in Dairy Calves during the Passive Immunity Period.

Improvements in the feeding of calves are of increasing importance for the development of the dairy industry. While colostrum is essential for the health of newborn calves, knowledge of protein phosphorylation alterations in neonatal calves that are fed colostrum or mature milk is lacking. Here, mid-jejunum tissue samples were collected from calves that received colostrum or milk. Subsequently, the jejunum phosphoproteome was analyzed using a phosphopeptide enrichment method, i.e., titanium immobilized metal ion affinity chromatography, coupled with liquid chromatography-tandem mass spectromet

Changes in expression of antimicrobial peptides and Fc receptors in the small intestines of neonatal calves during the passive immunity period.

The contribution of colostrum to passive immunity transfer and intestinal protection is well known; however, the effects of colostrum intake on the expression of antimicrobial peptides (AP) and Fc receptors in the intestine of neonatal calves are unclear. Our aim was to investigate changes in the expression of AP and Fc receptor in the small intestine of calves in the first 36 h postpartum. Twenty-four Holstein bull calves were used in this study, of which 18 calves were administered 3.2 L of pooled colostrum for each calf per meal via an esophageal tube. Calves were slaughtered at 8 h (1 meal

Passive immunity to infection with Yersinia spp. mediated by anti-recombinant V antigen is dependent on polymorphism of V antigen.

The V antigen is a 37-kDa secreted polypeptide encoded on the 70-kb virulence plasmid of pathogenic Yersinia spp. Besides having regulatory functions, it is known to be a virulence factor and a protective antigen. DNA sequencing of the most common serotypes of human pathogenic Yersinia enterocolitica and Y. pseudotuberculosis revealed that two evolutionary distinct types of V antigen exist in Yersinia spp. One type is represented by Y. enterocolitica serotype 08 strains WA, WA-314, and NCTC 10938 (designated LcrV-YenO8); the other type comprises Y. pestis, Y. pseudotuberculosis, and Y. enteroc

Population dynamics in echinococcosis and cysticercosis: regulation of Taenia hydatigena and T. ovis in lambs through passively transferred immunity.

A comparison has been made of the interactions between passively transferred and actively acquired immunity in regulating populations of Taenia hydatigena and T. ovis. When ewes were grazed prior to parturition under a high infection pressure, immunity was transferred to their offspring for up to 8 weeks. A qualitative difference between the species was the destruction of larval T. ovis prior to their establishment ('pre-encystment immunity') and that of T. hydatigena after they had become established ('post-encystment immunity') in the challenged lambs. The major difference in terms of popula

Cutaneous basophil responses and immune resistance of guinea pigs to ticks: passive transfer with peritoneal exudate cells or serum.

Resistance to infestation by larval Amblyomma americanum or Rhipicephalus sanguineus ticks was transferred to naive guinea pigs with peritoneal exudate cells (PEC) or serum from donors immunized by prior infestation with homologous tick larvae. In the A. americanum system, PEC transfer induced 87% tick rejection, which was similar to the level of resistance in actively sensitized hosts. In the R. sanguineus system, PEC conferred resistance (39% rejection) that was weaker than in actively sensitized hosts (57% rejection). In both systems, immune serum conferred significant but weaker resistance

A novel HIV triple broadly neutralizing antibody (bNAb) combination-based passive immunization of infant rhesus macaques achieves durable protective plasma neutralization levels and mediates anti-viral effector functions.

To eliminate vertical HIV transmission and achieve therapy-free viral suppression among children living with HIV, novel strategies beyond antiretroviral therapy (ART) are necessary. Our group previously identified a triple broadly neutralizing antibody (bNAb) combination comprising of 3BNC117, PGDM1400 and PGT151 that mediates robust in vitro neutralization and non-neutralizing effector functions against a cross-clade panel of simian human immunodeficiency viruses (SHIVs). In this study, we evaluated the safety, pharmacokinetics, and antiviral potency of this bNAb combination in infant rhesus

Impact of 3 colostrum replacement strategies on immunoglobulin G absorption efficiency, passive immunity, health, and growth in beef calves.

OBJECTIVE: The study objectives were to calculate the apparent efficiency of immunoglobulin G (IgG) absorption in beef calves receiving complete colostrum replacement and to compare the effects of 3 colostrum replacement strategies on the apparent efficiency of IgG absorption (AEA), transfer of passive immunity (TPI), morbidity, mortality, and average daily gain (ADG). METHODS: 45 newborn beef calves were randomly assigned to 1 of 3 colostrum interventions. Group A received approximately 170 g of IgG via colostrum replacer after birth. Group B received approximately 250 g of IgG in 2 feedings

Differential polyvalent passive immune protection of egg yolk antibodies (IgY) against live and inactivated Vibrio fluvialis in fish.

Egg yolk antibodies (IgY) can be prepared in large quantities and economically, and have potential value as polyvalent passive vaccines (against multiple bacteria) in aquaculture. This study prepared live and inactivated Vibrio fluvialis IgY and immunized Carassius auratus prior to infection with V. fluvialis and Aeromonas hydrophila. The results showed that the two IgY antibodies hold effective passive protective rates against V. fluvialis and A. hydrophila in C. auratus. Further, the serum of C. auratus recognized the two bacteria in vitro, with a decrease in the bacteria content of the kidn

A new passive immune strategy based on IgY antibodies as a key element to control neonatal calf diarrhea in dairy farms.

BACKGROUND: Neonatal diarrhea remains one of the main causes of morbi-mortality in dairy calves under artificial rearing. It is often caused by infectious agents of viral, bacterial, or parasitic origin. Cows vaccination and colostrum intake by calves during the first 6 h of life are critical strategies to prevent severe diarrhea but these are still insufficient. Here we report the field evaluation of a product based on IgY antibodies against group A rotavirus (RVA), coronavirus (CoV), enterotoxigenic Escherichia coli, and Salmonella sp. This product, named IgY DNT, has been designed as a comp

Oral rice-based vaccine induces passive and active immunity against enterotoxigenic E. coli-mediated diarrhea in pigs.

Enterotoxigenic Escherichia coli (ETEC) causes severe diarrhea in both neonatal and weaned pigs. Because the cholera toxin B subunit (CTB) has a high level of amino acid identity to the ETEC heat-labile toxin (LT) B-subunit (LTB), we selected MucoRice-CTB as a vaccine candidate against ETEC-induced pig diarrhea. When pregnant sows were orally immunized with MucoRice-CTB, increased amounts of antigen-specific IgG and IgA were produced in their sera. CTB-specific IgG was secreted in the colostrum and transferred passively to the sera of suckling piglets. IgA antibodies in the colostrum and milk

[Study on the immune complex formation mechanisms of passive Heymann nephritis--role of two different renal tubular epithelial antigens].

This study was undertaken to analyze the mechanisms of immune complex formation in Heymann nephritis. We isolated two different RTE antigens by gel filtration and prepared rabbit antisera against these antigens. By the indirect immunofluorescence method using normal rat renal tissues, the 65,000 molecular weight antigen (= beta) was observed not only in RTE, but also in the glomerular epithelium, epithelium of the small intestine, liver and spleen. On the other hand, the 35,000 molecular weight antigen (= gamma) existed in RTE and epithelium of the small intestine. When rats were injected intr

Assessment of passive immunity transfer in dairy calves using integrated immunological and molecular approaches in a cow-calf dyad model.

BACKGROUND: In bovine species, the epitheliochorial placenta prevents prenatal transfer of immunoglobulins, rendering calves dependent on colostrum intake for passive immunity. Efficient passive immunity transfer (PIT) is critical for neonatal health, yet conventional assessments primarily focus on immunoglobulin quantification without evaluating structural integrity. OBJECTIVE: This pilot study aimed to comprehensively evaluate PIT in a cohort of Holstein calves using an integrated approach combining field-based diagnostic tests, quantitative protein assays, and molecular validation techniqu

Losing Less in Translation: Relating the Preclinical Evidence Base for Active and Passive Immunity to Prevention of HIV-1 Infection in Humans.

The nonhuman primate (NHP) model has been a cornerstone in the development of effective therapeutic and prophylactic interventions for HIV-1 infection. Yet, despite promising results in this key preclinical setting, major efficacy trials of HIV vaccines have failed to exhibit sufficient protection to meet their endpoint efficacy criteria. Similarly, the recent Antibody-Mediated Prevention (AMP) trials (HVTN 703, 704), which tested the ability of passive immunization with the broadly neutralizing antibody VRC01, also failed to meet predetermined overall efficacy criteria. Cumulatively, both act

Diagnostic accuracy of digital Brix and serum total protein refractometers in estimating different passive immunity levels in dairy calves.

The aim of this study was to determine the diagnostic accuracy of digital serum total protein (TP) and digital Brix refractometers in estimating different passive immunity levels (<10, <18, <25 mg/mL) in dairy calves. Blood samples were collected from 260 apparently healthy Holstein calves, aged 2-7 days. Serum IgG concentrations were measured using digital Brix and TP refractometers and the radial immunodiffusion (RID) assay, as the gold standard. Data were analyzed by a receiver operating characteristics (ROC) analysis, the area under the ROC curves (AUC) and Cohen's kappa (κ). Optimal thres

Effect of postpartum collection time and colostrum quality on passive transfer of immunity, performance, and small intestinal development in preweaning calves.

This study evaluated the effects of postpartum collection time and quality of colostrum fed to calves on the failure of passive transfer, growth, and small intestine development in the first 5 wk of life. Newborn calves (Holstein-Friesian × Jersey) were identified at birth and collected either early (E; within 12 h postpartum; n = 20) or late (L; 18-24 h postpartum; n = 20) and fed either high-quality colostrum [HQC, first milking colostrum with Brix% = 23 ± standard deviation (SD) 2] or low-quality colostrum (LQC, mixed colostrum and transition milk with Brix% = 12 ± 1) to create 4 treatments

Passive transfer of naturally acquired specific immunity against West Nile Virus to foals in a semi-feral pony herd.

Horses naturally exposed to West Nile Virus (WNV) or vaccinated against WNV develop humoral immunity thought to be protective against development of clinical disease in exposed or infected animals. No reports evaluate the efficacy of passive transfer of naturally acquired specific WNV humoral immunity from dam to foal. The purpose of this study was to investigate passive transfer of naturally acquired immunity to WNV to foals born in a herd of semi-feral ponies, not vaccinated against WNV, in an endemic area, with many dams having seroconverted because of natural exposure. Mechanisms of recovery from a generalized viral infection: mousepox. II. Passive transfer of recovery mechanisms with immune lymphoid cells.

The following passive transfer experiments evaluated the contributions of the various host responses in recovery from mousepox. (a) Immune spleen cells transferred highly efficient antiviral activity, but preinfected recipients of these cells made no detectable splenic interferon or antibody in the 24 hr interval after cell transfer. (b) Passively administered interferon was ineffective. (c) Recipients of hyperimmune serum had much more antibody than recipients of immune spleen cells but significantly less antiviral activity. (d) Immune spleen cell populations with antiviral activity contained
