The Role of NLRP1, AIM2 and MEFV Inflammasomes in the High-Intensity Interval Training of Individuals With Obesity.

Obesity is a chronic disease associated with systemic inflammation caused by excess visceral fat and pro-inflammatory cytokines such as IL-1β and IL-6. Inflammasomes-particularly those involving genes such as NLRP1, AIM2 and MEFV-play a key role in this process. High-intensity interval training (HIIT) can counteract this inflammation; however, it remains unclear how HIIT modulates inflammasome gene expression in obesity. This study investigated whether HIIT can alter the expression of genes related to the inflammasomes NLRP1, AIM2 and MEFV in obese individuals. The results showed that, after 8

Jatrorrhizine attenuates inflammatory response in Helicobacter pylori-induced gastritis by suppressing NLRP3 inflammasomes and NF-κB signaling pathway.

Helicobacter pylori (H. pylori) was demonstrated to induce gastric mucosa inflammation in chronic atrophic gastritis (CAG). Jatrorrhizine possesses numerous biological properties, including antibacterial and anti-inflammatory effects. The present study was aimed at investigating whether Jatrorrhizine inhibits H. pylori-induced CAG and elucidating the underlying potential mechanisms. The rats were orally inoculated with H. pylori to establish CAG model. Fourteen days after Jatrorrhizine treatment, we measured the rat's body weight as well as rat's food intake. Besides, alcian blue and periodic

Influence of Pyroptosis Induced by NOD-Like Receptor Pyrin Domain-Containing 3 Inflammasomes on Viral Myocarditis in Children.

PURPOSE: Viral myocarditis (VMC) is a leading cause of heart failure and dilated cardiomyopathy in children. Pyroptosis, an inflammatory form of programmed cell death mediated by NOD-like receptor pyrin domain-containing 3 (NLRP3) inflammasomes, has been implicated in myocardial injury. However, the diagnostic significance of combined gene and protein expression analysis remains unclear. This study aimed to clarify the role of NLRP3 inflammasome-mediated pyroptosis in pediatric VMC and assess whether integrated gene-protein profiling could enhance diagnostic prediction. Vaporization of perfluorocarbon attenuates sea-water-drowning-induced acute lung injury by deactivating the NLRP3 inflammasomes in canines.

Seawater-drowning-induced acute lung injury (SD-ALI) is a life-threatening disorder characterized by increased alveolar-capillary permeability, an excessive inflammatory response, and refractory hypoxemia. Perfluorocarbons (PFCs) are biocompatible compounds that are chemically and biologically inert and lack toxicity as oxygen carriers, which could reduce lung injury in vitro and in vivo. The aim of our study was to explore whether the vaporization of PFCs could reduce the severity of SD-ALI in canines and investigate the underlying mechanisms. Eighteen beagle dogs were randomly divided into t

A Review on Inflammasomes and Immune Checkpoints in Pre-Eclampsia Complicated with Tuberculosis and Human Immune Deficiency Virus.

The current review evaluates how inflammasomes and immune checkpoints are regulated in pre-eclampsia (PE) associated with tuberculosis (TB) and Human Immune Deficiency Virus (HIV). Studies indicate that inflammasomes such as (NRLP3, NEK7, and AIM2) and immune checkpoints such as (CLT4, PD-1, TIM3, and LAG-3) are dysregulated in TB- and HIV-infected individuals, and also in pre-eclamptic pregnancies, which explains why pregnant women who are either infected with TB or HIV have an increased risk of developing PE. Evidence suggests that inhibition of inflammasomes and immune checkpoints may assis

Inflammasomes in placental explants of women with preeclampsia cultured with monosodium urate may be modulated by vitamin D.

OBJECTIVES: Preeclampsia (PE) is an important syndrome of gestation characterized by placental and systemic inflammation. High plasma concentration of uric acid are frequently associated with inflammation and endothelial dysfunction and may contribute to PE pathogenesis. This study aimed to evaluate the vitamin D (VD) immunomodulatory effect on the NLRP1/NLRP3 inflammasomes in placental explants from preeclamptic (PE) and normotensive (NT) pregnant women. STUDY DESIGN: Placental explants from 10 late-onset PE (LOPE), 10 early-onset PE (EOPE), and 10 NT pregnant women were cultured with or wit

Human NAIP/NLRC4 and NLRP3 inflammasomes detect Salmonella type III secretion system activities to restrict intracellular bacterial replication.

Salmonella enterica serovar Typhimurium is a Gram-negative pathogen that uses two distinct type III secretion systems (T3SSs), termed Salmonella pathogenicity island (SPI)-1 and SPI-2, to deliver virulence factors into the host cell. The SPI-1 T3SS enables Salmonella to invade host cells, while the SPI-2 T3SS facilitates Salmonella's intracellular survival. In mice, a family of cytosolic immune sensors, including NAIP1, NAIP2, and NAIP5/6, recognizes the SPI-1 T3SS needle, inner rod, and flagellin proteins, respectively. Ligand recognition triggers assembly of the NAIP/NLRC4 inflammasome, whic

X-shaped DNA potentiates therapeutic efficacy in colitis-associated colon cancer through dual activation of TLR9 and inflammasomes.

BACKGROUND: Immunotherapy has been extensively pursed as a promising strategy for the treatment of cancer. Pattern-recognition receptors (PRRs) play important roles in triggering activation of innate and adaptive immunity. Therefore, agents that stimulate PRRs could be useful for cancer immunotherapy. We developed two kinds of X-shaped double-stranded oligodeoxynucleotides (X-DNA), a single unit of X-DNA (XS-DNA) composed of four strands of DNA and a ligated X-DNA complex (XL-DNA) formed by crosslinking each XS-DNA to the other, and investigated if they had immunostimulatory activity and could

α-Methylene-γ-butyrolactone 3 inhibits the activation of NLRP3 inflammasome in sepsis by blocking both the priming and inflammasome assembly.

Excessive NLRP3 inflammasome activation is implicated in pathologies like sepsis. While the histone acetyltransferase inhibitor α-methylene-γ-butyrolactone 3 (MB-3) is typically studied in cancer and epigenetic research, its anti-inflammatory potential remains largely unexplored. This study investigated the effect of MB-3 on NLRP3 inflammasome activation in both cellular and animal models. Results demonstrated that MB-3 treatment significantly downregulated NLRP3-associated inflammatory cytokines and proteins in vitro and in vivo. Mechanistic studies revealed that MB-3 acts by dual inhibition

NLRP3 Inflammasome's Activation in Acute and Chronic Brain Diseases-An Update on Pathogenetic Mechanisms and Therapeutic Perspectives with Respect to Other Inflammasomes.

Increasingly prevalent acute and chronic human brain diseases are scourges for the elderly. Besides the lack of therapies, these ailments share a neuroinflammation that is triggered/sustained by different innate immunity-related protein oligomers called inflammasomes. Relevant neuroinflammation players such as microglia/monocytes typically exhibit a strong NLRP3 inflammasome activation. Hence the idea that NLRP3 suppression might solve neurodegenerative ailments. Here we review the recent Literature about this topic. First, we update conditions and mechanisms, including RNAs, extracellular ves

Molecular mechanisms of NLR proteins in inflammasome activation.

Nucleotide-binding domain and leucine-rich repeat-containing proteins (NLRs) are central mediators of inflammasome signaling, activating caspase-1 to drive pyroptotic cell death and pro-inflammatory responses. Although NLR activation is generally thought to involve conformational rearrangements that oligomerize their N- or C-terminal death domains to nucleate ASC or pro-caspase-1 oligomerization, the molecular mechanisms that enable these transitions vary substantially among family members. Here, we highlight recent structural advances that reveal how distinct NLRs are activated and assemble i

L-Arginine inhibits NLRP3 inflammasome activation.

Nutritional status and metabolic homeostasis are intricately associated with the inflammatory response. However, the direct mechanistic link between nutrition, metabolism and inflammation remains unclear. Here, we show that the amino acid L-arginine (L-Arg) is a potent endogenous inhibitor of the NLRP3 inflammasome, a central component of the inflammatory signalling pathway. We show that L-Arg binds directly to the Asp31 site of the NLRP3 protein, blocking its interaction with the adaptor ASC and subsequent inflammasome assembly and activation. L-Arg supplementation inhibits the NLRP3 inflamma

Current understanding of the NLRC4 Inflammasome in autoinflammation and enterocolitis.

The NLRC4 inflammasome is a cytosolic immune sensor that detects bacterial virulence structures and rapidly initiates inflammatory responses. While tightly regulated NLRC4 activation is essential for host defense, genetic mutations that enhance NLRC4 signaling can drive severe autoinflammatory disease. Studies in patients and genetically engineered mouse models have begun to reveal how dysregulated NLRC4 activation disrupts intestinal and systemic immune homeostasis. This review summarizes current insights into NLRC4-associated autoinflammatory diseases, with particular emphasis on the mechani

Sensing the spectrum: diverse stimuli for the NLRP3 inflammasome.

The NLRP3 inflammasome is a central inflammatory signaling pathway in host defense, cancer, metabolic disorders, and neurodegenerative diseases. Unlike other pattern-recognition receptors, NLRP3 senses perturbations in organelle integrity and ion homeostasis, enabling its activation by a remarkably broad spectrum of pathogen-derived and cellular stress signals. In this review, we summarize recent advances that have expanded our understanding of the diverse classes of NLRP3 stimuli and highlight breakthroughs in elucidating the molecular mechanisms underlying both infection-driven and sterile i

The dynamic and heterogeneous structure of the non-canonical inflammasome.

Inflammasomes are high-molecular-weight complexes that play integral roles in the innate immune system, triggering an inflammatory cascade to protect against cellular stresses such as pathogenic bacteria. Both canonical and non-canonical inflammasomes have been described in the literature and detailed structural studies of many components of the more complex and larger canonical versions have been reported. In contrast, corresponding structures of the non-canonical inflammasome have not emerged even though it consists of only two components: lipopolysaccharide (LPS) from Gram-negative bacteria

Structure-Activity Relationship for Inflammasome Inhibition by Thiomuscimol.

Inflammasomes are central mediators of innate immune defense but can also drive pathological inflammation and pyroptotic cell death in numerous diseases. While several small-molecule inhibitors have been described, many selectively target individual inflammasomes or act through the adaptor protein ASC, leaving ASC-independent pathways unaffected. We previously identified thiomuscimol as a broad-spectrum inflammasome inhibitor that blocks both ASC-dependent and ASC-independent activation, although the structural basis for this activity remains unclear. Here, we examined the structure-activity r

Targeting inflammasome, IL-1β, and coagulation in melioidosis.

Burkholderia pseudomallei is a Gram-negative bacterium that causes melioidosis, a disease endemic in Southeast Asia and Northern Australia and increasingly detected in other tropical regions. B. pseudomallei is resistant to many antibiotics and no vaccine for melioidosis is currently available. The disease, particularly its pneumonic form, has high mortality, often due to sepsis. Accumulating evidence from our and other groups has shown that inflammasome and toll-like receptors (TLR) activation during B. pseudomallei lung infection causes an excessive inflammatory response that becomes deleter

Inflammasome targeting for periodontitis prevention is sex dependent.

Inflammasome initiates inflammation via the maturation of interleukin-1 beta (IL-1β). Periodontitis is a prevalent, male-biased disease characterized by inflammation-driven bone loss, yet the mechanism(s) of this sex bias is unknown. This study explored whether enhanced inflammasome represents a causal mechanism for this bias. Analyses of three separate human studies (>6,200 samples) show that males have significantly higher IL-1β in the gingival crevicular fluid than females during health and periodontitis. This pattern is experimentally reproduced with different versions of the ligature-indu

Hyperactivity of the non-canonical inflammasome in SPG11 and SPG48.

BACKGROUND: Hereditary spastic paraplegia (HSP) denotes a heterogeneous group of neurodegenerative spastic gait disorders. Variants in SPG11 cause the most common autosomal recessive HSP also known as SPG11. The gene product Spatacsin interacts with the adaptor protein complex 5 (AP5). Because neurodegeneration in SPG11 is accompanied by marked neuro-inflammation, we hypothesised that Spatacsin may play a cell-autonomous role in pro-inflammatory cells. METHODS: Inflammasome activation was assessed in primary microglia and bone-marrow-derived-macrophages (BMDMs) from wild-type, Spg11, and Ap5z

MAP Kinase Signaling at the Crossroads of Inflammasome Activation.

Inflammasomes are crucial mediators of both antimicrobial host defense and inflammatory pathology, requiring stringent regulation at multiple levels. This review explores the pivotal role of mitogen-activated protein kinase (MAPK) signaling in modulating inflammasome activation through various regulatory mechanisms. We detail recent advances in understanding MAPK-mediated regulation of NLRP3 inflammasome priming, licensing and activation, with emphasis on MAPK-induced activator protein-1 (AP-1) signaling in NLRP3 priming, ERK1 and JNK in NLRP3 licensing, and TAK1 in connecting death receptor s
