Maintenance of effective control of Simulium damnosum in the face of insecticide resistance.

The control of onchocerciasis through reduction of its vector Simulium damnosum depends on highly effective insecticide treatments directed against the larvae. As these treatments must be applied weekly over wide areas for many years, ideal conditions for the development of resistance have been created. Resistance to organophosphate compounds has occurred. To maintain effective vector control, an intense operational research effort has been necessary. Close and constant monitoring of susceptibility, combined with cytotaxonomic identification of the tested larvae, has revealed the pattern of sp

In vitro extracellular replication of Wolbachia endobacteria.

Obligate intracellular endobacteria of the genus Wolbachia are widespread in arthropods and several filarial nematodes. Control programs for vector-borne diseases (dengue, Zika, malaria) and anti-filarial therapy with antibiotics are based on this important endosymbiont. Investigating Wolbachia, however, is impeded by the need for host cells. In this study, the requirements for Wolbachia wAlbB growth in a host cell-free in vitro culture system were characterized via qPCRs. A cell lysate fraction from Aedes albopictus C6/36 insect cells containing cell membranes and medium with fetal bovine ser

The whole genome sequence of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann), reveals insights into the biology and adaptive evolution of a highly invasive pest species.

BACKGROUND: The Mediterranean fruit fly (medfly), Ceratitis capitata, is a major destructive insect pest due to its broad host range, which includes hundreds of fruits and vegetables. It exhibits a unique ability to invade and adapt to ecological niches throughout tropical and subtropical regions of the world, though medfly infestations have been prevented and controlled by the sterile insect technique (SIT) as part of integrated pest management programs (IPMs). The genetic analysis and manipulation of medfly has been subject to intensive study in an effort to improve SIT efficacy and other as

Reassessing ecdysteroidogenic cells from the cell membrane receptors' perspective.

Ecdysteroids secreted by the prothoracic gland (PG) cells of insects control the developmental timing of their immature life stages. These cells have been historically considered as carrying out a single function in insects, namely the biochemical conversion of cholesterol to ecdysteroids and their secretion. A growing body of evidence shows that PG cells receive multiple cues during insect development so we tested the hypothesis that they carry out more than just one function in insects. We characterised the molecular nature and developmental profiles of cell membrane receptors in PG cells of

Pesticide resistance: can we make it a renewable resource?

Negative cross-resistance (NCR) occurs when a mutant allele confers (i) resistance to one toxic chemical and (ii) hyper-susceptibility to another. Sequential deployment of NCR toxins is useful for insect control in few situations (Pittendrigh et al., 2000). Using Monte Carlo simulations, we investigated the concurrent use of a pair of NCR toxins to control a hypothetical insect pest population. When the toxins killed more heterozygotes than homozygotes, the resistance allele became either extremely common or rare depending on starting allelic frequency. If the NCR toxins did not kill the two h

Modulation of insect Ca(v) channels by peptidic spider toxins.

Insects have a much smaller repertoire of voltage-gated calcium (Ca(V)) channels than vertebrates. Drosophila melanogaster harbors only a single ortholog of each of the vertebrate Ca(V)1, Ca(V)2, and Ca(V)3 subtypes, although its basal inventory is expanded by alternative splicing and editing of Ca(V) channel transcripts. Nevertheless, there appears to be little functional plasticity within this limited panel of insect Ca(V) channels, since severe loss-of-function mutations in genes encoding the pore-forming alpha1 subunits in Drosophila are embryonic lethal. Since the primary role of spider v

Edward F. Knipling: titan and driving force in ecologically selective area-wide pest management.

Few entomologists have impacted insect pest management as profoundly as Edward F Knipling (1909-2000). During WWII, Knipling and his colleagues developed highly effective measures to protect both military personnel and civilian populations from major arthropod-borne diseases. The sterile insect technique was Knipling's conception, and he successfully guided its development and use against the screwworm, Cochliomyia hominivorax Coquerel, and against various other pests. He inspired and guided the development of a wide range of ecologically selective methods of insect detection and suppression.

Immune tolerance of vector beetle to its partner plant parasitic nematode modulated by its insect parasitic nematode.

Immune response of insect vectors to transmitted pathogens or insect hosts against parasites are well studied, whereas the mechanism of tripartite interactions remains elusive. In this study, we investigated the immune interactions of the vector beetle Monochamus alternatus ( Ma) to the devastating plant parasitic nematode Bursaphelenchus xylophilus ( Bx) and the insect parasitic nematode Howardula phyllotretae ( Hp). We report the unique immune mechanism by which the vector beetle tolerates many devastating Bx in its trachea, yet that immune tolerance is compromised by the parasitic nematode

Paraborg: Paramedic Cyborg Insects for In Situ Emergency Drug-Delivery.

Over the past two decades, cyborg insects have emerged as a promising alternative to artificial insect-scale mobile robots, owing to their low power consumption, resilient biological bodies with naturally integrated sensors, agile locomotion, and self-adaptability. However, most studies have focused primarily on autonomous control and hardware integration for search-oriented urban search-and-rescue operations following natural disasters such as earthquakes. This raises a crucial next-step challenge: beyond locating survivors, what life-saving interventions can cyborg insects perform? Here, we

Use of shaking treatments and preharvest sprays of pyrethroid insecticides to reduce risk of yellowjackets and other insects on Christmas trees imported into Hawaii.

Insects are commonly found by Hawaii's quarantine inspectors on Christmas trees imported from the Pacific Northwest. To reduce the risk of importing yellowjacket (Vespula spp.) queens and other insects, an inspection and tree shaking certification program was begun in 1990. From 1993 to 2006, the annual percentage of shipped containers rated by Hawaii quarantine inspectors as moderately or highly infested with insects was significantly higher for manually shaken trees than for mechanically shaken trees. Between 1993 and 2001, 343 insect species in total were recovered from Christmas trees. Liv

AaIT: from neurotoxin to insecticide.

AaIT is a single chain neurotoxic polypeptide derived from the venom of the Buthid scorpion Androctonus australis Hector, composed of 70 amino acids cross-linked by four disulfide bridges. Its strict selectivity for insects has been documented by toxicity, electrophysiological and ligand receptor binding assays. These last have shown that various insect neuronal membranes possess a single class of non-interacting AaIT binding sites of high affinity (K(D) = 1-3(n)M) and low capacity (0.5-2.0 pmol/mg prot.). The fast excitatory paralysis induced by AaIT is a result of a presynaptic effect, namel

Can the leafhopper microbiome unlock new strategies for its control?

Leafhoppers (Hemiptera: Cicadellidae) are significant agricultural pests worldwide, causing direct feeding injury and transmitting plant pathogens. Conventional management still relies heavily on insecticides, but resistance development, non-target effects, and environmental concerns increasingly limit their effectiveness. Recent progress in leveraging insect microbiomes for sustainable pest control, mostly in well-studied groups such as mosquitoes, whiteflies, and aphids, suggests that symbiotic manipulation could offer new tools. Whether such strategies can be developed for leafhoppers remai

Reviewing the female accessory glands of insects: Where they come from, what they do, and where they are going.

Female accessory glands (AGs) are structures associated with the reproductive system of many arthropods. In insects, the glands are mostly related to oviposition by secreting egg coatings and adhesives. Nevertheless, the AGs are versatile concerning their function, and show a high diversity of chemical compounds. Here we aim to review the AGs in insects, highlighting their distribution among the orders, origins, location, chemical nature of the secretions, development, endocrine control, and morphological features. We also propose dividing the glands into epidermal and mesodermal, according to

Design, Synthesis, Antifeedant Activity, and Mechanistic Research of Aconitine Derivatives.

For decades, natural product-derived insect repellents have been a major focus of research. Aconitine, a diterpene alkaloid isolated from Aconitum plants, exhibits not only significant antifeedant activity but also potent toxicological effects against certain insects. In this study, 48 novel aconitine derivatives were synthesized via esterification of the C3-hydroxyl group. Among them, compound A4 demonstrated optimum antifeedant activity with an AFC50 value of 4.91 µg/cm2, surpassing that of the parent compound aconitine (AFC50 = 11.65 µg/cm2) and the positive control Azadirachtin (AFC50 = 5.

Barrier or breach? Assessing swine housing features for mosquito threats.

Effective mosquito control is critical in swine production to reduce disease transmission and prevent mechanical damage. However, current biosecurity measures on swine premises primarily target microbial pathogens, often overlooking the importance of excluding insects, particularly mosquitoes. While vector-borne disease transmission is the primary concern, mosquito infestations also contribute to mechanical damage, leading to secondary infections, stress-related production losses, and compromised animal welfare. Mitigation efforts aimed at mosquitoes can also have broader benefits by reducing
