Extensive library of Ruminococcus gnavus phages revealing genomic and phenotypic diversity with novel clusters.

Ruminococcus gnavus is a prevalent member of human gut microbiota and is associated with inflammatory bowel disease. Phage-based microbiota engineering represents a promising approach for alleviating dysbiosis; however, knowledge regarding R. gnavus phages remains limited. We report the isolation and characterization of 29 novel R. gnavus phages from sewage samples. All isolates exhibited siphovirus morphology, with genome sizes ranging from 32.3 to 46.0 kbp, and represented 18 putative new genera and 24 new species. Whole-proteome clustering tools indicated that R. gnavus phages form a distin

Enhancing Learning Health Systems in Primary and Community Care: Contributions of NHS Library and Knowledge Specialists.

CONTEXT: Learning communities driving the data-knowledge-practice learning cycle are at the core of learning health systems (LHS). With the right team in place, including knowledge management expertise, LHS accelerates improvements, delivering on the healthcare quadruple aim and health equity. Within the National Health Service (NHS) in England, knowledge management and librarian expertise exist mainly within acute hospital settings. This makes it harder for those developing LHS in community health settings to benefit from this expertise. EXPERIENCE REPORT METHODS AND RESULTS: Three proof-of-

Schistosoma japonicum proteins that interact with the gynecophoral canal protein identified using a yeast two-hybrid system.

The large amount of schistosome eggs produced by mature female worms not only induce major pathological damage to the host but also lead to the transmission of schistosomiasis. Mature female schistosome worms need constant pairing contact with a male partner as male signaling is indispensable to female growth, development, and reproduction. The gynecophoral canal protein (GCP), a cell-surface glycoprotein, plays a potential role in the interaction between males and females and in stimulating female development and maturation. In this study, a yeast two-hybrid cDNA library of Schistosoma japoni

Discovery and Optimization of Novel Carbazole-Based Ferroptosis Inhibitors.

Ferroptosis is an iron-dependent form of regulated cell death that plays a critical role in the pathogenesis of multiple human diseases, making it an attractive target for therapeutic strategy. In this study, the FerrBERT model was employed to screen a small-molecule library, leading to the identification of 3-amino-9-ethylcarbazole as a potent inhibitor of RSL3-induced ferroptosis in HT22 mouse hippocampal neuronal cells (EC50 = 65.2 nM). Using this scaffold as a starting point, 50 derivatives were designed, synthesized, and systematically evaluated for their structure-activity relationships

USP26 as a hepatitis B virus-induced deubiquitinase primes hepatocellular carcinogenesis by epigenetic remodeling.

Despite recent advances in systemic therapy for hepatocellular carcinoma (HCC), the prognosis of hepatitis B virus (HBV)-induced HCC patients remains poor. By screening a sgRNA library targeting human deubiquitinases, we find that ubiquitin-specific peptidase 26 (USP26) deficiency impairs HBV-positive HCC cell proliferation. Genetically engineered murine models with Usp26 knockout confirm that Usp26 drives HCC tumorigenesis. Mechanistically, we find that the HBV-encoded protein HBx binds to the promoter and induces the production of USP26, which is an X-linked gene exclusively expressed in the

The PI3K/mTOR Inhibitor Ompalisib Suppresses Nonhomologous End Joining and Sensitizes Cancer Cells to Radio- and Chemotherapy.

As the predominant pathway for the repair of DNA double-strand breaks (DSB), non-homologous end joining (NHEJ) attenuates the efficacy of cancer treatment which relies on the introduction of DSBs, such as radiotherapy and genotoxic drugs. Identifying novel NHEJ inhibitors is of great importance for improving the therapeutic efficiency of radio- or chemotherapy. Here we miniaturized our recently developed NHEJ detecting system into a 96-well plate-based format and interrogated an FDA approved drug library containing 1732 compounds. A collection of novel hits were considered to be potential DSB

A 29-kDa merozoite protein is a prospective antigen for serological diagnosis of Babesia orientalis infection in buffaloes.

A gene encoding a 29-kDa protein of Babesia orientalis (BoP29) was isolated by immunoscreening the B. orientalis cDNA expression library using monoclonal antibodies and its nucleotide sequence of the cDNA was 971bp with an open reading frame of 750bp. The results of BLASTn and BLASTx in NCBI showed that this cDNA sequence had similarity with Babesia bovis hypothetical protein (BBOV_II005480). Western blotting showed that the recombinant protein (rBoP29) had positive seroreactivity to B. orientalis infected buffalo sera. An indirect enzyme-linked immunosorbent assay (ELISA) was developed using

From pan-active to parasite-selective antiparasitic agents: A scaffold hopping approach.

Vector-borne parasitic diseases (VBPDs) represent a major global public health concern, with human African trypanosomiasis (HAT), Chagas disease, leishmaniasis, and malaria collectively threatening millions of people, particularly in developing regions. Climate change may further influence their transmission and geographic spread, increasing the global burden. As drug resistance continues to rise, there is an urgent need for novel therapeutic agents to expand treatment options and limit disease progression. Exploiting a cell-based phenotypic approach, we had previously developed 1,3,4-oxadiazo

Human genetics guides the discovery of CARD9 inhibitors with anti-inflammatory activity.

Human genetic association studies highlight key genes involved in disease pathology, yet targets identified by these analyses often fall outside the traditional definitions of druggability. A rare truncated variant of the scaffold protein CARD9 is linked with protection from Crohn's disease, prompting us to pursue the development of inhibitors that might similarly modulate innate inflammatory responses. Using a phased approach, we first identified a ligandable site on CARD9 using a structurally diverse DNA-encoded library and defined this site in detail through X-ray crystallography. Building

Discovery and biological evaluation of HS13 as a novel covalent ligand targeting the NSD2-PWWP1 domain.

The PWWP1 domain of NSD2 is an epigenetic reader essential for the subcellular localization of NSD2 and is frequently implicated in oncogenesis, making it an attractive therapeutic target. Our previous work has identified the C294 residue of NSD2-PWWP1 as a viable covalent targeting site. In this study, we screened a library of 968 covalent compounds and identified the lead compound, 92-B7, which exhibited an IC50 of 332 μM. Subsequent structural optimization and affinity-guided evaluations led to the development of HS13, which demonstrated significantly improved potency with a Kd of 8.33 μM.

Fair and Square: Design, Synthesis and Biological Evaluations of Squaric Acid Derivatives as Novel HDAC8 Inhibitors.

Histone deacetylase 8 (HDAC8) is a clinically validated target in neuroblastoma, where isoform selective inhibition offers a strategy to suppress tumour growth while limiting off-target toxicity. Hydroxamic acids remain the dominant zinc-binding group (ZBG) in HDAC inhibitors but are also associated with metabolic instability, suboptimal pharmacokinetics and nonspecific metal chelation. In this study, we report the biological evaluation of a focussed library of 51 squaric acid derivatives as alternative, non-hydroxamic HDAC inhibitors. While a subset of these compounds has been described previ

Charting the history of premature birth in France from the 17th century to modern state-of-the art care.

AIM: To describe the concept of preterm birth in the history of medicine in France, from the 17th century up until the end of the 20th century, on the basis of old medical textbooks. METHODS: Historical manuscripts and books held in medical libraries and from our personal collection were examined. Digital copies of medical textbooks online were also studied. RESULTS: Premature deliveries were often referred to as spontaneous abortions or miscarriages, irrespective of the infant's condition at birth. The difference between growth restriction and prematurity was not known. At the end of the 18

Role of Statin in Reducing Cardiovascular Diseases in Human Immunodeficiency Virus (HIV) Patients: A Systematic Review.

Because of abnormal lipid profiles, the risk of patients living with human immunodeficiency virus (PLHIV) developing cardiovascular diseases (CVDs) is much greater than in the general population. Statins are known effective medications for reducing the risk of CVDs by influencing inflammatory mediators, endothelial function, angiogenesis, and thrombosis. However, their role in PLHIV has not been well confirmed. In this article, we aim to provide more clarity about the effects of statin in lowering CVDs in PLHIV.
