Interaction of native and aggregated albumin with DMPC bilayers.

The study of protein-lipid interaction offers interesting insights into the mutual alterations determined in the formation of the supramolecular complex. It gains even more interest, not only in basic research but also in biomedical and biomaterial applications, when protein aggregation and fibril formation are involved. In this study, the reciprocal influence of human serum albumin (HSA), in both the native and the thermally aggregated state, and dimyristoylphosphatidylcholine (DMPC) bilayers is investigated by combining UV-Vis scattering, attenuated total reflection Fourier transform infrare

Pro-inflammatory protein S100A9 alters membrane organization by dispersing ordered domains.

Pro-inflammatory, calcium-binding protein S100A9 is localized in the cytoplasm of many cells and regulates several intracellular and extracellular processes. S100A9 is involved in neuroinflammation associated with the pathogenesis of Alzheimer's disease (AD). The number of studies on the impact of S100A9 in co-aggregation processes with amyloid-like proteins is increasing. However, there is still a lack of data on how this protein interacts with lipid membranes. We employed atomic force microscopy (AFM), dynamic light scattering (DLS), and fluorescence measurements (Laurdan and Thioflavin-T) t

Membrane texture induced by specific protein binding and receptor clustering: active roles for lipids in cellular function.

Biological membranes are complex, self-organized structures that define boundaries and compartmentalize space in living matter. Composed of a wide variety of lipid and protein molecules, these responsive surfaces mediate transmembrane signaling and material transport within the cell and with its environment. It is well known that lipid membrane properties change as a function of composition and phase state, and that protein-lipid interactions can induce changes in the membrane's properties and biochemical response. Here, molecular level changes in lipid organization induced by multivalent toxi

Comparing the lipid membrane affinity and permeation of drug-like acids: the intriguing effects of cholesterol and charged lipids.

PURPOSE: Lipid bilayers regulate the passage of solutes into and between cellular compartments. A general prerequisite for this passage is the partitioning of the solute into the bilayer. We investigated the relationship between bilayer partitioning and permeation of three drug-like acids in liposomal systems consisting of phosphatidylcholine alone or mixed with cholesterol or charged lipids. MATERIALS AND METHODS: Bilayer partitioning was determined by equilibrium dialysis. Bilayer permeation was studied with a luminescence assay which is based on the energy transfer of the permeant to intra

Membrane Architecture-Dependent Photophysics of Conjugated Oligoelectrolytes Distinguishes Extracellular Vesicles From Lipoproteins.

Lipid-bound nanoparticles (LBNs), including extracellular vesicles and lipoproteins, are characterized by distinct membrane architectures that modulate biological functions. However, optical probes capable of directly distinguishing lipid bilayers from monolayers remain limited. Here, we establish conjugated oligoelectrolytes (COEs) as molecular transducers of lipid membrane architecture capable of converting differences in supramolecular organization into distinct optical signatures. Using model systems, we show that COEs intercalate across lipid bilayers but preferentially adsorb onto lipid

Imaging Artificial Membranes Using High-Speed Atomic Force Microscopy.

Supported lipid bilayers represent a very attractive way to mimic biological membranes, especially to investigate molecular mechanisms associated with the lateral segregation of membrane components. Observation of these model membranes with high-speed atomic force microscopy (HS-AFM) allows the capture of both topography and dynamics of membrane components, with a spatial resolution in the nanometer range and image capture time of less than 1 s. In this context, we have developed new protocols adapted for HS-AFM to form supported lipid bilayers on small mica disks using the vesicle fusion or L

Concentration-dependent mechanism of the binding behavior of ibuprofen to the cell membrane: A molecular dynamic simulation study.

Ibuprofen is a commonly used drug for treating headaches, pain, and fever. The lipid bilayer is the primary and most important interface for drugs to interact with biological systems. However, the molecular interactions between ibuprofen and the cell membrane are not well understood. Our findings suggest that the interactions between ibuprofen and the bilayer involve multiple steps and depend on the concentration of the drug. At low concentrations of ibuprofen, it can bind to the surface of the lipid bilayer. The electrostatic and vdW energies of IBU-lipid at 0 ns of the simulation were -22.5

Control of the morphology of lipid layers by substrate surface chemistry.

In this study, surface coatings were used to control the morphology of the deposited lipid layers during vesicle spreading, i.e., to control if liposomes self-assemble on a surface into a supported lipid bilayer or a supported vesicular layer. The influence of the properties of the surface coating on formation of the deposited lipid layer was studied with quartz crystal microbalance and two-wavelength multiparametric surface plasmon resonance techniques. Control of lipid self-assembly on the surface was achieved by two different types of soft substrate materials, i.e., dextran and thiolated po

Amyloidogenic SARS-CoV-2 Spike Protein-Derived Peptides Form Oligomers and Selectively Damage Lipid Membranes.

Several peptides derived from the SARS-CoV-2 spike protein (S-protein) are amyloidogenic, suggesting a potential role in COVID-19 disease-associated pathogenesis. Many disease-related amyloids form a variety of intermediate oligomeric aggregates and directly damage lipid membranes. Gain of toxic function is commonly observed for oligomeric and fibrillar aggregates of amyloid peptides. Therefore, the ability of four S-protein-derived peptides (s192, s258, s601, and s1166) to form oligomers and fibrils was investigated. Additionally, their ability to bind and damage lipid membranes was also dete

Curvature enhances binding and aggregation of huntingtin at lipid membranes.

Huntington disease (HD) is a genetic neurodegenerative disease caused by an expanded polyglutamine (polyQ) domain in the first exon of the huntingtin (Htt) protein, facilitating its aggregation. Htt interacts with a variety of membraneous structures within the cell, and the first 17 amino acids (Nt17) of Htt directly flanking the polyQ domain comprise an amphiphathic α-helix (AH) lipid-binding domain. AHs are also known to detect membrane curvature. To determine if Htt exon 1 preferentially binds curved membranes, in situ atomic force microscopy (AFM) studies were performed. Supported lipid bi

Interactions between Beta-2-Glycoprotein-1 and Phospholipid Bilayer-A Molecular Dynamic Study.

This study aims to investigate the interactions appearing when the beta-2-glycoprotein-1 binds to a lipid bilayer. The inter- and intra-molecular forces acting between the two macromolecular systems have been investigated using a molecular dynamics simulation method. The importance of water bridges has also been addressed. Additionally, the viscoelastic response of the bilayer has been studied. In detail, the (saturated-chain) 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) and (unsaturated-chain) 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoethanolamine (POPE) bilayers have been chosen to test

Accumulation and separation of membrane-bound proteins using hydrodynamic forces.

The separation of molecules residing in the cell membrane remains a largely unsolved problem in the fields of bioscience and biotechnology. We demonstrate how hydrodynamic forces can be used to both accumulate and separate membrane-bound proteins in their native state. A supported lipid bilayer (SLB) was formed inside a microfluidic channel with the two proteins streptavidin (SA) and cholera toxin (CT) coupled to receptors in the lipid bilayer. The anchored proteins were first driven toward the edge of the lipid bilayer by hydrodynamic forces from a flowing liquid above the SLB, resulting in t

Insights into the Destabilizing Effect of Fluorinated Ethanol on Biomimetic Membranes.

The ability of alcohols to perturb the structure and function of membrane bilayers and membrane proteins has made them indispensable in the pharmaceutical and biochemical industry. In the present study, we delineate the bilayer-modifying potency of trifluoroethanol (TFE), a fluorinated analogue of widely used ethanol (EtOH), toward biomimetic lipid membranes composed of pure 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (POPC) and POPC/cholesterol (POPC/CHOL) lipids using experimental techniques and atomistic molecular dynamics simulations. Vesicle size and membrane composition control monomer transfer pathways in multicomponent lipid vesicles.

Lipid exchange between populations of non-fusing protocells, mediated by monomer diffusion through the aqueous phase, is a key process underlying protocell growth and compositional equilibration. While most theoretical descriptions consider single-component membranes with constant desorption rates, the influence of vesicle size and lipid composition on lipid-transfer dynamics remains poorly understood. Here, we develop a kinetic model for lipid exchange in multicomponent protocell populations that incorporates lipid species with different desorption rates and composition-dependent membrane pac

Can NO-indomethacin counteract the topical gastric toxicity induced by indomethacin interactions with phospholipid bilayers?

Nitric oxide (NO)-releasing nonsteroidal anti-inflammatory drugs (NSAIDs) have been developed to overcome the gastrointestinal and cardiovascular toxicity of NSAIDs, by chemically associating a NO-releasing moiety with commercial NSAIDs. Since increasing evidence supports that NSAIDs toxicity is related to their topical actions in membrane lipids, this work aims to evaluate the impact of adding a NO-releasing moiety to parent NSAIDs regarding their effect on lipid bilayers. Thus, the interactions of NO-indomethacin and indomethacin (parent drug) with 1,2-dimyristoyl-sn-glycero-3-phosphocholine

Supported Planar Mammalian Membranes as Models of in Vivo Cell Surface Architectures.

Emerging technologies use cell plasma membrane vesicles or "blebs" as an intermediate to form molecularly complete, planar cell surface mimetics that are compatible with a variety of characterization tools and microscopy methods. This approach enables direct incorporation of membrane proteins into supported lipid bilayers without using detergents and reconstitution and preserves native lipids and membrane species. Such a system can be advantageous as in vitro models of in vivo cell surfaces for study of the roles of membrane proteins as drug targets in drug delivery, host-pathogen interactions

Interplay of electrostatics and lipid packing determines the binding of charged polymer coated nanoparticles to model membranes.

Understanding of nanoparticle-membrane interactions is useful for various applications of nanoparticles like drug delivery and imaging. Here we report on the studies of interaction between hydrophilic charged polymer coated semiconductor quantum dot nanoparticles with model lipid membranes. Atomic force microscopy and X-ray reflectivity measurements suggest that cationic nanoparticles bind and penetrate bilayers of zwitterionic lipids. Penetration and binding depend on the extent of lipid packing and result in the disruption of the lipid bilayer accompanied by enhanced lipid diffusion. On the

Lipid packing and local geometry influence septin curvature sensing.

Septins assemble into scaffolds that direct cell growth and morphology that are often localized to the plasma membrane. While septins preferentially bind convex membranes via amphipathic helices, their assembly on varied geometries in cells suggests additional localization cues. We tested the hypothesis that lipid composition directs septin assembly through lipid packing properties. Lipid mixtures varying in lipid packing were designed by molecular dynamics simulations and incorporated onto supported lipid bilayers to measure septin adsorption in vitro. Septins strongly favor loosely-packed, d

Structure and function of the influenza A M2 proton channel.

The M2 protein of influenza A viruses forms a tetrameric pH-activated proton-selective channel that is targeted by the amantadine class of antiviral drugs. Its ion channel function has been extensively studied by electrophysiology and mutagenesis; however, the molecular mechanism of proton transport is still elusive, and the mechanism of inhibition by amantadine is controversial. We review the functional data on proton channel activity, molecular dynamics simulations of the proton conduction mechanism, and high-resolution structural and dynamical information of this membrane protein in lipid b

Understanding the Mechanical Properties of Ultradeformable Liposomes Using Molecular Dynamics Simulations.

Improving drug delivery efficiency to solid tumor sites is a central challenge in anticancer therapeutic research. Our previous experimental study (Guo et al., Nat. Commun. 2018, 9, 130) showed that soft, elastic liposomes had increased uptake and accumulation in cancer cells and tumors in vitro and in vivo respectively, relative to rigid particles. As a first step toward understanding how liposomes' molecular structure and composition modulates their elasticity, we performed all-atom and coarse-grained classical molecular dynamics (MD) simulations of lipid bilayers formed by mixing a long-tai
