Effects of nemorosone, isolated from the plant Clusia rosea, on the cell cycle and gene expression in MCF-7 BUS breast cancer cell lines.

BACKGROUND: Breast cancer is the cause of considerable morbidity and mortality in women. While estrogen receptor antagonists have been widely used in breast cancer treatment, patients have increasingly shown resistance to these agents and the identification of novel targeted therapies is therefore required. Nemorosone is the major constituent of the floral resin from Clusia rosea and belongs to the class of polycyclic polyisoprenylated benzophenones of the acylphloroglucinol group. The cytotoxicity of nemorosone in human cancer cell lines has been reported in recent years and has been related

Signaling Pathways Mediating the Cytotoxicity of (E)-4-(1-Epoxy-7,8-propenyl)phenyl Benzoate, a Phenylpropanoid from Croton velutinus, in MCF‑7 Cells.

Natural products represent an important source of candidates for breast cancer therapy. Here, we investigated the antitumor activity of (E)-4-(1-epoxy-7,8-propen)-phenylbenzoate (CV2), a phenylpropanoid isolated from Croton velutinus roots, in MCF-7 breast cancer cells and evaluated its toxicity profile in nontumor cells and zebrafish embryos. CV2 exhibited potent cytotoxic activity against MCF-7 cells (IC50= 4.63 ± 0.16 μM) and showed selectivity relative to nontumorigenic MCF-10A cells (SI = 9.69). CV2 also induced apoptosis, increased intracellular ROS levels, and modulated MAPK, NF-κB, and

Ginkgetin enhances the antitumor effect of Taxol on human breast cancer MCF-7 cells via ferroptosis mediated by the MDM2-p53-YAP1 axis.

Ginkgetin, a biflavone isolated from Ginkgo biloba, is a potent Wnt signaling inhibitor that can enhance the pharmacological effects of cisplatin on non-small cell lung cancer (NSCLC) by activating the ferroptosis pathway. However, few studies have examined the role of ginkgetin in regulating ferroptosis in breast cancer cells. In this study, ginkgetin was found to induce ferroptosis by increasing intracellular reactive oxygen species (ROS) accumulation through the downregulation of GPX4, SLC7A11, SLC40A1, and glutathione expression along with the upregulation of transferrin, glutamate, cystin

Metal thiosemicarbazonates as dual mR2 RNR and colchicine-site tubulin inhibitors: from biochemical inhibition to mitotic arrest in MCF-7 breast cancer cells.

Cancer cells exploit multiple proliferation pathways, making single-target therapies vulnerable to resistance. Here, a thiosemicarbazone-based strategy is introduced for striking two validated oncotargets, the R2 subunit of ribonucleotide reductase (RNR) and the colchicine site of tubulin, with one agent. Guided by molecular docking, quinoline-8-carboxaldehyde N4-(3,4,5-trimethoxyphenyl)thiosemicarbazone (HL) and its metal complexes [Cu II (L)(Cl 2 CHCOO)] (1), [Cu II (L)Cl] (2), [Pd II (L)Cl] (3), and [Fe III (L)2]Cl (4) were synthesized and characterized by spectroscopic, spectrometric, elec

A Novel PROTAC G9a/GLP Degrader that Inhibits, Similar to G9a siRNA, the Migration of MCF-7 Breast-Cancer Cells without Affecting Proliferation.

G9a and G9a-like protein (GLP) are histone methyltransferases that regulate epigenetics by adding methyl groups to histone H3, thereby controlling gene expression. G9a/GLP dysregulation and overexpression have been reported to cause cancer proliferation, progression, and metastasis. So far, quinazoline-based inhibitors and degraders have been frequently used as chemical tools to elucidate the role of G9a/GLP. However, quinazoline-based inhibitors exhibit toxicity in normal cells. In this context, we identified a G9a/GLP degrader (4) based on RK-701, a less-toxic G9a/GLP-selective inhibitor. Co

Improvement of Menopausal Symptoms by Beta vulgaris, Artemisia princeps, and Eleutherococcus senticosus via Estrogen Pathway Activation in MCF-7 Cells and OVX Mice.

Menopause, a natural transition marked by estrogen decline, is a growing public health concern because it impairs quality of life. Hormone replacement therapy (HRT) has been widely employed to alleviate menopausal symptoms, including osteoporosis, uterine atrophy, vaginal dryness, fat redistribution, and increased cardiovascular risk, but use is limited by adverse effects including hormonal imbalance and increased cancer risk. As potential alternatives, plant-derived phytoestrogens are actively investigated, yet validated candidate extracts remain scarce. Here we evaluated the estrogenic activ

HER4 Affects Sensitivity to Tamoxifen and Abemaciclib in Luminal Breast Cancer Cells and Restricts Tumor Growth in MCF-7-Based Humanized Tumor Mice.

The impact of the HER4 receptor on the growth and treatment of estrogen receptor-positive breast cancer is widely uncertain. Using CRISPR/Cas9 technology, we generated stable HER4 knockout variants derived from the HER4-positive MCF-7, T-47D, and ZR-75-1 breast cancer cell lines. We investigated tumor cell proliferation as well as the cellular and molecular mechanisms of tamoxifen, abemaciclib, AMG232, and NRG1 treatments as a function of HER4 in vitro. HER4 differentially affects the cellular response to tamoxifen and abemaciclib treatment. Most conspicuous is the increased sensitivity of MCF

The two ligand anions in each complex resulted in saturation of the iron coordination number and consequently the existence of these complexes as 1:1 electrolytes. As well, the iron in these complexes exhibits low-spin electronic configuration. X-ray crystallography of complex C1 indicated its triclinic crystal sy

Enhanced sensitivity to rapamycin following long-term oestrogen deprivation in MCF-7, T-47-D and ZR-75-1 human breast cancer cells.

Human breast cancer cells (MCF-7, T-47-D and ZR-75-1) can adapt to circumvent any reduced growth rate during long-term oestrogen deprivation, and this provides three model systems to investigate mechanisms of endocrine resistance in breast cancer. In this paper we report consistent differences in the effects of three growth inhibitors following long-term oestrogen deprivation in all three cell models. Long-term oestrogen deprivation of MCF-7, T-47-D and ZR-75-1 cells resulted in reduced growth inhibition by PD98059 (2-10 μg/ml), implying a loss of dependence on mitogen-activated protein kinase

Efficient suppression of secretory clusterin levels by polymer-siRNA nanocomplexes enhances ionizing radiation lethality in human MCF-7 breast cancer cells in vitro.

Small interfering RNA molecules (siRNA) hold great promise to specifically target cytoprotective factors to enhance cancer therapy. Like antisense RNA strategies, however, the use of siRNA is limited because of in vivo instability. As a first step to overcome delivery issues, a series of graft copolymers of polyethylene glycol and polyethylenimine (PEI-g-PEG) were synthesized and investigated as nontoxic carriers for delivery of siRNA targeting the signaling peptide of secretory clusterin (sCLU), a prosurvival factor that protects cells from ionizing radiation (IR) injury, as well as chemother

Effects of the pineal hormone melatonin on the anchorage-independent growth of human breast cancer cells (MCF-7) in a clonogenic culture system.

Only physiological levels of melatonin exert an antiproliferative effect on MCF-7 breast cancer cells grown in an anchorage-dependent culture system. We investigated melatonin's effect on the anchorage-independent growth of MCF-7 cells as well as the dose-response characteristics of this indoleamine under clonogenic culture conditions. Melatonin's inhibitory effect, with respect to the number and size of colonies formed, exhibit a linear dose-response curve with pharmacological concentrations producing a maximal inhibition while subphysiological levels of melatonin induce minimal inhibition. Targeting Obesity-Associated Breast Cancer: Curcumin Counteracts Leptin-Induced Tumorigenic and Metastatic Traits in MCF-7 and MDA-MB-231 Breast Cancer Cells.

Obesity-driven hyperleptinemia is a key driver of breast cancer (BC) aggressiveness; nevertheless, strategies to counteract leptin-mediated tumor progression remain limited. Curcumin, a natural polyphenol with established anti-obesity, anti-inflammatory, and anticancer properties, has emerged as a promising metabolic and tumor-modulating biomolecule, although its effects on leptin-induced oncogenic signaling remain unclear. This study evaluated Curcumin as an adjuvant nutraceutical against leptin-induced oncogenesis in MCF-7 and MDA-MB-231 cells. Cells were exposed to obesity-mimicking leptin
