The maternal-placental-fetal interface: Adaptations of the HPA axis and immune mediators following maternal stress and prenatal alcohol exposure.

This review addresses underlying physiological, cellular, and molecular factors that alter the developing fetal brain stress circuits and responses of the hypothalamic-pituitary-adrenal (HPA) axis caused by maternal stress and prenatal alcohol exposure (PAE). An emphasis is placed on the contribution of the placenta following maternal stress separately, and as a co-occurrence with PAE. Altered fetal HPA axis ultimately results in dysregulation of the brain stress-response system long after birth and possibly lifelong. Additional consideration of the role of placentally-derived endocrine and se

Association of maternal exposure to Superstorm Sandy and maternal cannabis use with development of psychopathology among offspring: the Stress in Pregnancy Study.

BACKGROUND: Early-life adverse experiences can elevate the magnitude of the risk of developmental psychopathology, but the potential synergistic effects of multiple factors have not been well studied. AIMS: To determine whether prenatal exposures to maternal stress (Superstorm Sandy) and maternal cannabis use synergistically alter the risk of developmental psychopathology. METHOD: The study included 163 children (53.4% girls), longitudinally tracked (ages 2-5 years) in relation to the effects of two early-life adverse exposures (Superstorm Sandy and maternal cannabis use). Offspring were gro

Fetal response to maternal exposures of environmental chemicals: Utility of a four-cell human feto-maternal interface organ-on-chip.

Epidemiological studies suggest that maternal exposures to environmental pollutants may be linked to spontaneous preterm birth. Mechanistic studies are needed to provide support to these hypotheses; however, existing in vitro models do not replicate human feto-maternal barriers beyond placenta. The recently developed four-cell Feto-Maternal interface Organ-On-Chip (FMi-OOC) model enables studies of chemical effects that are critically important for maintaining full-term pregnancy. We tested four environmental pollutants that have been associated with preterm birth - dichlorodiphenyltrichloroet

Maternal pre-pregnancy BMI influences the associations between bisphenol and phthalate exposures and maternal weight changes and fat accumulation.

BACKGROUND: Bisphenols and phthalates are two classes of endocrine-disrupting chemicals (EDCs) thought to influence weight and adiposity. Limited research has investigated their influence on maternal weight changes, and no prior work has examined maternal fat mass. We examined the associations between exposure to these chemicals during pregnancy and multiple maternal weight and fat mass outcomes. METHODS: This study included a sample of 318 women enrolled in a Canadian prospective pregnancy cohort. Second trimester urinary concentrations of 2 bisphenols and 12 phthalate metabolites were quant

Susceptible window identification of the relationship between maternal ozone exposure and preterm birth.

Previous studies have focused on the effects of ozone (O3) exposure and preterm birth (PTB), but the findings are contentious and the susceptible window for O3 exposure during pregnancy remains inconclusive. Here, we pooled the current evidence to explore the relationship between maternal O3 exposure and PTB and further identified the susceptible exposure windows. We pooled a meta-analysis of 17 eligible studies by searching Embase, PubMed and Web of Science through 9 September 2024. The odds ratio (OR) and the corresponding 95% confidence intervals (CIs) were extracted for analysis. The tests

Maternal exposure to titanium dioxide nanoparticles disrupts ultrasonic vocalization development in mouse offspring.

BACKGROUND: Early neurodevelopment is a critical period during which environmental exposures can have lasting effects on brain function and behavior. One key indicator of early neurodevelopmental integrity in rodents is the production of neonatal ultrasonic vocalizations (USVs), which are essential for maternal-offspring communication. Given the widespread use of titanium dioxide nanoparticles (TiO2NPs) in food and consumer products, there is growing concern that perinatal exposure to these particles may interfere with normal neurodevelopment. However, the effects of TiO2NPs exposure on USV pr

In-utero exposure to maternal hyperinsulinemia impacts glucose homeostasis in male offspring transiently.

Fetal in-utero exposures are known to impact the long-term metabolic health of offspring. Insulin is the primary treatment to manage glucose concentrations in mothers with gestational diabetes. However, adverse offspring outcomes persist, such as macrosomia and increased risk of early obesity in offspring with potential metabolic consequences in adulthood. Here, we developed two novel mouse models to investigate the effects of maternal insulin on offspring health. In the first model, RIP-βAkt dams exhibited hyperinsulinemia, normoglycemia, and insulin resistance without obesity. Their wild-typ

Connecting the Dots: Mass Media, Maternal Exposure, and Child Health Outcomes in Benin.

INTRODUCTION: Maternal exposure to mass media has been linked to various health outcomes, but its influence on childhood morbidity and mortality in Benin remains unexplored. This study examines the relationship between maternal mass media exposure and two critical child health indicators: morbidity and mortality. METHODS: This secondary analysis of the 2017/2018 Benin Demographic and Health Survey (BDHS) included 13,851 women. Childhood morbidity (fever, cough, and diarrhea) and mortality were analyzed as outcomes. Bivariable and multivariable logistic regression and Cox regression models wer

Maternal ozone exposure and differential cardiometabolic outcomes in aging male and female offspring.

Fetal development is a vulnerable life stage for exposure to environmental stressors. Gestational exposure to ozone (O3) has been shown to compromise fetal growth, predisposing offspring to increased risk of pulmonary and metabolic dysfunction later in life. However, the cardiovascular consequences of maternal O3 exposure on offspring health remain uncharacterized. Herein, pregnant Long-Evans rats were exposed to 0.8 ppm O3 for 4 h each day on gestation day (GD) 5 and 6. Following weaning, male and female offspring were monitored for cardiometabolic health for up to ∼5 months of age. Although

Associations between weekly maternal exposure to ambient particulate matter and congenital heart disease.

BACKGROUND: The association between maternal exposure to air pollution and congenital heart diseases (CHDs) has garnered increasing attention. However, investigations into critical weekly specific exposure windows for CHDs remain limited. This study evaluated the relationship between maternal ambient particulate matter exposure throughout the entire pregnancy period and the risk of CHDs, as well as explored critical weekly specific exposure windows. METHOD: Based on the Jiangsu Maternal and Child Health Monitoring Manual and the Environmental Health Department of Suzhou CDC, 19,914 pregnant w

Maternal organophosphate pesticide exposure in relation to birthweight: Modification by placenta transporter genotype.

Exposure to organophosphate pesticides (OPPs) has been linked to adverse birth outcomes, including low birthweight. Maternal biomarkers are commonly used as proxies for fetal exposure, but fetal exposure also depends on placental transport mechanisms. In 240 mother-newborn pairs, we explored how genetic variation in membrane transporters influences the association between maternal OPP concentrations and birthweight. Single nucleotide polymorphisms (SNPs) in the OAT4/SLC22A11 and OATP2B1/SLCO2B1 membrane transporters modified the relationship between OPP exposure and birthweight-for-gestational

Maternal prenatal mercury exposure from rice and its association with newborn neurobehavioral development.

Methylmercury (MeHg) exposure is a global environmental concern. Rice-derived MeHg exposure poses a potential neurodevelopmental risk in inland populations with low fish intake, yet epidemiological evidence remains scarce. This representative mother-infant cohort study was conducted in a mercury mining area in China, involving 420 mother-infant pairs, of which 206 were from mercury-exposed regions and 214 were from control regions. It was first found that the median total mercury and methylmercury levels in rice from mercury-exposed areas (11.19 ng/g and 5.07 ng/g) were significantly higher th

Association between Maternal Lead Exposure from Seafood Consumption and Neurodevelopment: A Systematic Review.

Despite the essential nutrients, maternal seafood consumption during pregnancy or lactation (PL) is also a potential source of toxins, including lead (Pb). The association between exposure to Pb from seafood during PL on children's neurodevelopment is uncertain. This systematic review assessed the association of exposure to Pb from seafood during PL on children's neurodevelopment. Embase, PubMed, CENTRAL, and PsycINFO were searched for English-language peer-reviewed articles. Two independent reviewers screened at title, abstract, and full-text levels. Lead Toxicity and Maternal Exposure: Characterisation of Alveolar Bone Changes on Offspring Rats.

Lead poisoning is a global public health concern. Maternal exposure during intrauterine and lactational periods can present a higher susceptibility of harm to the offspring. Thus, pregnant female Wistar rats (Rattus norvegicus) were randomly divided in two experimental groups: control group and Lead group. The animals were exposed to 50 mg/kg of Lead Acetate daily for 42 days (21 days of gestational period + 21 days of lactational period). After the exposure period, the mandibles of the offspring were collected for lead quantification, Raman spectroscopy analysis, micro-CT, morphometric e hist

Urinary metabolite biomarkers of pregnancy complications associated with maternal exposure to particulate matter.

Particulate matter 2.5 (PM2.5) is associated with reproductive health and adverse pregnancy outcomes. However, studies evaluating biological markers of PM2.5 are lacking, and identifying biomarkers for estimating prenatal exposure to prevent pregnancy complications is essential. Therefore, we aimed to explore urine metabolites that are easy to measure as biomarkers of exposure. In this matched case-control study based on the PM2.5 exposure, 30 high PM2.5 group (>15 μg/m3) and 30 low PM2.5 group (<15 μg/m3) were selected from air pollution on pregnancy outcome (APPO) cohort study. Prenatal exposure to Benzo[a]pyrene affects maternal-fetal outcomes via placental apoptosis.

Prenatal exposure to Benzo[a]pyrene (BaP) has been suggested to increase the risk of adverse pregnancy outcomes. However, the role of placental apoptosis on BaP reproductive toxicity is poorly understood. We conducted a maternal animal model of C57BL/6 wild-type (WT) and transformation-related protein 53 (Trp53) heterozygous knockout (p53KO) mice, as well as a nested case-control study involving 83 women with PB and 82 term birth from a birth cohort on prenatal exposure to BaP and preterm birth (PB). Pregnant WT and p53KO mice were randomly allocated to BaP treatment and control groups, intrap

Maternal particulate matter exposure and gestational diabetes mellitus: a population-based cohort study.

BACKGROUND: Gestational diabetes mellitus prevalence is steadily increasing worldwide, posing a significant threat to the short-term and long-term health of both mother and offspring. Because particulate matter air pollution has been reported to affect glucose metabolism, it was suggested that maternal particulate matter exposure may be associated with the development of gestational diabetes mellitus; however, the evidence is limited and inconsistent. OBJECTIVE: This study aimed to determine the association between maternal exposure to particulate matter of diameter ≤2.5 µm and of diameter of

Maternal consumption and perinatal exposure to non-nutritive sweeteners: should we be concerned?

The context for this review is the rapid increase in the use of non-nutritive sweeteners (NNSs) instead of sugar in foods and beverages, a situation so prevalent in some countries that consumers are finding it increasingly challenging to access foods without NNSs. The benefits of consuming NNSs on obesity and diabetes are now being questioned, and studies have shown that they may exert physiological activities, sometimes independently of sweet taste receptor stimulation. Few studies, limited mainly to North American and European countries, have described the consumption of NNSs by pregnant or
