Contamination of Engram Coactivity Networks During Forgetting.

Forgetting is a fundamental component of adaptive memory and essential for cognitive flexibility, yet its cellular basis remains unclear. Here we establish a mouse model of retroactive interference (RI) and show that post-learning novelty exploration induces active forgetting of hippocampus-dependent object location memory only within a discrete consolidation window defined by protein synthesis sensitivity. RI imposed within this window reduces engram reactivation and destabilizes a structured coactivity network formed during learning. Mice that forget retrieve reorganized engram with increase

Temporal dynamics of Purkinje cell-intrinsic excitability governs cerebellar systems consolidation.

Systems consolidation reorganizes newly encoded memories from cortical networks into downstream circuits. Although synaptic mechanisms are extensively characterized, the role of neuronal intrinsic excitability (IE) in this process remains relatively unexplored. Here, we utilized a cerebellum-dependent learning model to investigate the causal link between neuronal IE and memory consolidation. The optogenetic manipulation of Purkinje cells (PCs), the sole output of the cerebellar cortex, identified a crucial 90-min post-learning window during which PC-IE undergoes transient depression. Disruptin

Memory formation and long-term retention in humans and animals: convergence towards a transformation account of hippocampal-neocortical interactions.

Historically, the hippocampus has been viewed as a temporary memory structure. Consistent with the central premise of standard consolidation theory (SCT), a memory is initially hippocampus-dependent but, over time, it undergoes a consolidation process and eventually becoming represented in a distributed cortical network independent of the hippocampus. In this paper, we review evidence that is incompatible with each of the following essential features of SCT that are derived from its central premise: (1) Hippocampal damage reliably produces temporally graded retrograde amnesia, (2) all declarat

Scalp and hippocampal sleep correlates of memory function in drug-resistant temporal lobe epilepsy.

Seminal animal studies demonstrated the role of sleep oscillations such as cortical slow waves, thalamocortical spindles, and hippocampal ripples in memory consolidation. In humans, whether ripples are involved in sleep-related memory processes is less clear. Here, we explored the interactions between sleep oscillations (measured as traits) and general episodic memory abilities in 26 adults with drug-resistant temporal lobe epilepsy who performed scalp-intracranial electroencephalographic recordings and neuropsychological testing, including two analogous hippocampal-dependent verbal and nonver

Modification of Fear Memory by Pharmacological and Behavioural Interventions during Reconsolidation.

BACKGROUND: Dysfunctional fear responses play a central role in many mental disorders. New insights in learning and memory suggest that pharmacological and behavioural interventions during the reconsolidation of reactivated fear memories may increase the efficacy of therapeutic interventions. It has been proposed that interventions applied during reconsolidation may modify the original fear memory, and thus prevent the spontaneous recovery and reinstatement of the fear response. METHODS: We investigated whether pharmacological (propranolol) and behavioural (reappraisal, multisensory stimulati

Differential locus coeruleus-hippocampus interactions during offline states.

Patterns of locus coeruleus (LC) activity and norepinephrine (NE) release during non-rapid-eye-movement sleep suggest a critical role for the LC-NE system in offline modulation of forebrain circuits. NE transmission promotes synaptic plasticity and is required for memory consolidation, but the field has only begun to uncover how LC activity contributes to coordinated forebrain network dynamics. Hippocampal ripples, a hallmark of memory replay, are temporally coupled with thalamocortical oscillations; however, the circuit mechanisms underlying system-level consolidation across larger brain netw

Conserved Population Dynamics in the Cerebro-Cerebellar System between Waking and Sleep.

Despite the importance of the cerebellum for motor learning, and the recognized role of sleep in motor memory consolidation, surprisingly little is known about neural activity in the sleeping cerebro-cerebellar system. Here, we used wireless recording from primary motor cortex (M1) and the cerebellum in three female monkeys to examine the relationship between patterns of single-unit spiking activity observed during waking behavior and in natural sleep. Across the population of recorded units, we observed similarities in the timing of firing relative to local field potential features associated

Neural correlates and reinstatement of recent and remote memory in children and young adults.

Memory consolidation tends to be less robust in childhood than adulthood. However, little is known about the corresponding functional differences in the developing brain that may underlie age-related differences in retention of memories over time. This study examined system-level memory consolidation of object-scene associations after learning (immediate delay), one night of sleep (short delay), as well as 2 weeks (long delay) in 5- to 7-year-old children (n=49) and in young adults (n=39), as a reference group with mature consolidation systems. Particularly, we characterized how functional neu

[Application of the method of one-day learning in a Morris water maze to analyse the effects of sleep deprivation on memory trace recall 24 hours later, after learning].

The proposed model of a one-day spatial learning is of interest in research of how sleep influences the hippocamp-dependent memory consolidation. We have studied the influence of a one-day total sleep deprivation on spatial memory consolidation in hooded rats after a one-day learning in the Morris water maze according to Feldman et al. [2010] protocol. According to it rats had to find a submerged platform that was alternatively marked by a flag or completely invisible to an animal. In a previous study [Dorokhov et al., 2011] we have used another one-day learning protocol [Frick et al., 2000] a

Sodium Oxybate Alters Sleep Architecture and Reduces Emotional Selectivity in Memory-Related Responses.

Sleep supports memory consolidation. Non-rapid eye movement (NREM) and rapid eye movement (REM) sleep have been proposed to support, respectively, declarative memory consolidation and the integration of the memory affective dimension. Here, we used sodium oxybate (SXB) to modify the NREM/REM proportion during one sleep night and assessed subsequent emotional memory retrieval and related neurophysiological underpinnings. In a double-blind, randomized, cross-over study, 19 healthy young men spent two monitored nights (SXB/placebo). Before sleep, participants encoded emotional and neutral picture

Selective photostimulation of locus coeruleus terminals in hippocampal-CA1 induces LTP and enhances spatial memory in free-moving mice.

Long-term potentiation (LTP) represents a fundamental form of synaptic plasticity that underlies long-term memory. Catecholaminergic projections from the locus coeruleus (LC) to the hippocampal CA1 region modulate spatial memory consolidation; however, it remains unclear whether activity in this pathway directly induces in vivo synaptic potentiation. The present study demonstrates that selective photostimulation of tyrosine hydroxylase-positive (TH+) LC axon terminals in CA1 induces robust optogenetic LTP (oLTP) in the Schaffer collateral pathway. Induction of oLTP is associated with increased

Slow wave sleep is associated with forgetting in patients with temporal lobe epilepsy.

While time spent in slow wave sleep (SWS) after learning promotes memory consolidation in the healthy brain, it is unclear if the same benefit is obtained in patients with temporal lobe epilepsy (TLE). Interictal epileptiform discharges (IEDs) are potentiated during SWS and thus may disrupt memory consolidation processes thought to depend on hippocampal-neocortical interactions. Here, we explored the relationship between SWS, IEDs, and overnight forgetting in patients with TLE. Nineteen patients with TLE studied object-scene pairs and memory was tested across a day of wakefulness (6 hrs) and a

The critical role of sleep spindles in hippocampal-dependent memory: a pharmacology study.

An important function of sleep is the consolidation of memories, and features of sleep, such as rapid eye movement (REM) or sleep spindles, have been shown to correlate with improvements in discrete memory domains. Because of the methodological difficulties in modulating sleep, however, a causal link between specific sleep features and human memory consolidation is lacking. Here, we experimentally manipulated specific sleep features during a daytime nap via direct pharmacological intervention. Using zolpidem (Ambien), a short-acting GABAA agonist hypnotic, we show increased sleep spindle densi

Long-term verbal memory deficit and associated hippocampal alterations in 22q11.2 deletion syndrome.

Chromosome 22q11.2 deletion syndrome (22q11.2DS) is a genetic disease associated with an increased risk for schizophrenia and a specific cognitive profile. In this paper, we challenge the current view of spared verbal memory in 22q11.2DS by investigating verbal memory consolidation processes over an extended time span to further qualify the neuropsychological profile. Our hypotheses are based on brain anomalies of the medial temporal lobes consistently reported in this syndrome.Eighty-four participants (45 with 22q11.2DS), aged 8-24 years old, completed a verbal episodic memory task to investi

Sleep enhances exposure therapy.

BACKGROUND: Sleep benefits memory consolidation. Here, we tested the beneficial effect of sleep on memory consolidation following exposure psychotherapy of phobic anxiety. METHOD: A total of 40 individuals afflicted with spider phobia according to DSM-IV underwent a one-session virtual reality exposure treatment and either slept for 90 min or stayed awake afterwards. RESULTS: Sleep following exposure therapy compared with wakefulness led to better reductions in self-reported fear (p = 0.045, d = 0.47) and catastrophic spider-related cognitions (p = 0.026, d = 0.53) during approaching a live

Neural Correlates of Motor Sequence Learning and Enhanced Offline Consolidation in 7-11-Year-Old Children.

Many daily activities involve series of interrelated movements and thus the capacity to learn new motor sequences is vital for everyday functioning. Although learning new skills is especially prominent throughout childhood, remarkably few studies have examined the neural underpinnings of motor sequence learning (MSL) and memory consolidation in children. Twenty-two children (7-11 years) and 23 adults (18-30 years) underwent functional magnetic resonance imaging while completing two sessions of a MSL task, separated by a 5-h offline period of wakefulness. Analyses of the behavioral data reveale

Relational Memory Reconsolidation: A New Lens on Change Mechanisms in EFT for Couples.

This paper presents a new framework to account for change mechanisms in Emotion-Focused Therapy for Couples (EFT). Building on Lane et al.'s (2015) integrated memory model, the Relational Memory Reconsolidation model described here posits that EFT reshapes partners' relational memory structures through emotionally intense moments of expressed vulnerability and responsiveness. These vulnerability-responsiveness events allow partners to access and transform memories of past relational experiences (i.e., episodic memory) as well as associated beliefs about themselves and the relationship (i.e., s

Sleep benefits cross-dimensional binding of spatial context in episodic memory.

The active systems consolidation hypothesis predicts that sleep compared to wakefulness benefits context binding in episodic memory for retention intervals of up to 12 h. Extending previous research on sleep benefits in item-context associative memory, we investigated whether sleep also improves bound source memory for cross-dimensional context associations. In a source-monitoring experiment, we used intentionally learned pictures as items and incidentally learned horizontal and vertical screen positions as context dimensions to compare cross-dimensional binding in spatial source memory for re

Neurostimulation techniques to enhance sleep and improve cognition in aging.

Sleep plays a critical role in the process of memory consolidation. In particular, during non-rapid eye movement (NREM) slow wave sleep, slow-oscillations, spindles, hippocampal sharp wave ripples, and their phase coupling are involved in the process of transferring and consolidating information recently encoded and temporarily stored in the hippocampus into long-term memory stored in the neocortex. There is evidence that aging and neurodegenerative conditions, in particular Alzheimer's disease, are associated with changes to this transient grouping of NREM oscillations. Therefore, methods to

Embodied memory for emotional faces: Effects of cardiac cycle phases and post-learning stress on identity and expression memory.

Human learning and memory are considered 'embodied', but it is unclear which body signals affect which memory facet. We investigated whether ascending cardiac signals affect memory encoding and consolidation. Due to its relevance for social behavior, this study investigated identity and expression memory for emotional faces. To implement different conditions of ascending cardiac signals for encoding, participants viewed happy and angry faces for 150 ms time-locked either to the early (R-wave +230 ms) or late cardiac cycle phase (R-wave +530 ms), which differ in their feedback from arterial bar
