Current approaches to cyanotoxin risk assessment and risk management around the globe.

Toxic cyanobacteria became more widely recognized as a potential health hazard in the 1990s, and in 1998 the World Health Organization (WHO) first published a provisional Guideline Value of 1 μg L-1 for microcystin-LR in drinking-water. In this publication we compare risk assessment and risk management of toxic cyanobacteria in 17 countries across all five continents. We focus on the three main (oral) exposure vehicles to cyanotoxins: drinking-water, water related recreational and freshwater seafood. Most countries have implemented the provisional WHO Guideline Value, some as legally binding s

Cyanopeptide Co-Production Dynamics beyond Mirocystins and Effects of Growth Stages and Nutrient Availability.

Intensified cyanobacterial bloom events are of increasing global concern because of adverse effects associated with the release of bioactive compounds, including toxic cyanopeptides. Cyanobacteria can produce a variety of cyanopeptides, yet our knowledge about their abundance and co-production remains limited. We applied a suspect-screening approach, including 700 structurally known cyanopeptides, and identified 11 cyanopeptides in Microcystis aeruginosa and 17 in Dolichospermum flos-aquae. Total cyanopeptide concentrations ranged from high nmol to μmol gdry-1 with slightly higher cell quotas

Dual-frequency ultrasonic oxidation of cyanobacterial toxins (MC-LR and MC-RR) at drinking water resources: Assessment of analytical methods and ultrasonic reactor configuration.

Ultrasonic oxidation provides the degradation of a wide range of water pollutants to the final products defined as carbon dioxide, short-chain organic acids, and inorganic ions, typically less toxic and favorable to biodegradation. In this study, it was investigated the application of novel ultrasonic reactor that allows the several combinations of low (20 kHz and 40 kHz) and high frequency ultrasonic piezoceramic transducer (578 kHz, 862 kHz and 1142 kHz) to degrade two main cyanobacterial toxins, Microcystin-RR (MC-RR) and Microcystin-LR (MC-LR). A plate transducer operating at different fre

Global transcriptional responses of the toxic cyanobacterium, Microcystis aeruginosa, to nitrogen stress, phosphorus stress, and growth on organic matter.

Whole transcriptome shotgun sequencing (RNA-seq) was used to assess the transcriptomic response of the toxic cyanobacterium Microcystis aeruginosa during growth with low levels of dissolved inorganic nitrogen (low N), low levels of dissolved inorganic phosphorus (low P), and in the presence of high levels of high molecular weight dissolved organic matter (HMWDOM). Under low N, one third of the genome was differentially expressed, with significant increases in transcripts observed among genes within the nir operon, urea transport genes (urtBCDE), and amino acid transporters while significant de

Are known cyanotoxins involved in the toxicity of picoplanktonic and filamentous North Atlantic marine cyanobacteria?

Eight marine cyanobacteria strains of the genera Cyanobium, Leptolyngbya, Oscillatoria, Phormidium, and Synechococcus were isolated from rocky beaches along the Atlantic Portuguese central coast and tested for ecotoxicity. Strains were identified by morphological characteristics and by the amplification and sequentiation of the 16S rDNA. Bioactivity of dichloromethane, methanol and aqueous extracts was assessed by the Artemia salina bioassay. Peptide toxin production was screened by matrix assisted laser desorption/ionization time of flight mass spectrometry. Molecular analysis of the genes in

The influence of hydrological conditions on phytoplankton community structure and cyanopeptide concentration in dammed lowland river.

Study results show continuous summer-autumn dominance of toxic cyanobacteria in plankton not only in the strongly eutrophicated lowland Siemianówka reservoir, but also along 130 km of Narew river below the lake. The negative effects of eutrophication of the reservoir reach far outside its boundaries. One of the symptoms of eutrophication was a mass development of cyanobacteria, including the toxin-producing Planktothrix agardhii. In the reservoir, the biomass of the species strongly correlated with the concentration of microcystins. Redundancy analysis (RDA) identified seven environmental vari

Blood pressure and hepatocellular effects of the cyclic heptapeptide toxin produced by the freshwater cyanobacterium (blue-green alga) Microcystis aeruginosa strain PCC-7820.

Laboratory rats and mice were used to investigate the hepatotoxicity caused by the cyclic heptapeptide (mol. wt 994) termed microcystin-LR. Microcystin-LR (also known as cyanoginosin-LR) is produced by the freshwater cyanobacterium (blue-green alga) M. aeruginosa strain PCC-7820. In time course histopathology studies with mice significant liver damage, with an absence of pulmonary emboli, were observed after 15 min. Pulmonary emboli did not appear until 1 hr. In rats, significant liver damage and the presence of occasional emboli were observed at 20 min. Pulmonary emboli did not contain fibrin

Senescence-like phenotype in post-mitotic cells of mice entering middle age.

The expanding footprint of global aquaculture has pushed production systems into regions with unknown or limited baseline information about associated hazards and risks. Oyster (Crassostrea virginica) aquaculture, for example, has expanded throughout euhaline coastal sounds across the southeastern United States yet there exists limited information on phycotoxin levels in oysters cultured in these environments. Here we investigated domoic acid and microcystin accumulation in cultured oysters at three coastal sites in North Carolina, with all sites varying in surrounding land use. Oyster deploym

Searching for Optimal Substitute Habitats for Plants by Biological Experiments-A Case Study of the Endangered Species Aldrovanda vesiculosa L. (Droseraceae).

The selection of appropriate locations for the reintroduction of endangered plant species is an important process, because it usually influences the success of the conservation. The aim of this study was to select the optimal substitute habitats for Aldrovanda vesiculosa, taking into account the influence of physical-chemical factors (light intensity, temperature, pH, concentration of dissolved forms of nitrogen and cyanobacterial toxin microcystin-LR) on the efficiency of plant growth. Water analysis and field observations of the habitats of six lakes in Eastern Poland typified as potential s

Acclimation temperature influences phage susceptibility in a toxin-producing strain of Microcystis aeruginosa.

Microcystis aeruginosa is a freshwater cyanobacterium known for its role in the formation of harmful algal blooms. However, less well characterized are the host-phage interactions that occur in these toxic blooms. Here, we describe the relationship between a microcystin-producing strain of M. aeruginosa, NIES-298, and a lytic bacteriophage Ma-LMM01. In this study, we explored how host pre-acclimation to different temperature conditions influenced sensitivity to subsequent infection by Ma-LMM01. We focused on 19°C, a cooler temperature often associated with microcystin-producing blooms in sprin

Eutrophic urban ponds suffer from cyanobacterial blooms: Dutch examples.

Ponds play an important role in urban areas. However, cyanobacterial blooms counteract the societal need for a good water quality and pose serious health risks for citizens and pets. To provide insight into the extent and possible causes of cyanobacterial problems in urban ponds, we conducted a survey on cyanobacterial blooms and studied three ponds in detail. Among 3,500 urban ponds in the urbanized Dutch province of North Brabant, 125 showed cyanobacterial blooms in the period 2009-2012. This covered 79% of all locations registered for cyanobacterial blooms, despite the fact that urban ponds

Hepatopathy following consumption of a commercially available blue-green algae dietary supplement in a dog.

BACKGROUND: Dietary supplement use in both human and animals to augment overall health continues to increase and represents a potential health risk due to the lack of safety regulations imposed on the manufacturers. Because there are no requirements for demonstrating safety and efficacy prior to marketing, dietary supplements may contain potentially toxic contaminants such as hepatotoxic microcystins produced by several species of blue-green algae. CASE PRESENTATION: An 11-year-old female spayed 8.95 kg Pug dog was initially presented for poor appetite, lethargy polyuria, polydipsia, and an i

Recent Progress and Applications of Thermal Lens Spectrometry and Photothermal Beam Deflection Techniques in Environmental Sensing.

This paper presents recent development and applications of thermal lens microscopy (TLM) and beam deflection spectrometry (BDS) for the analysis of water samples and sea ice. Coupling of TLM detection to a microfluidic system for flow injection analysis (μFIA) enables the detection of microcystin-LR in waters with a four samples/min throughput (in triplicate injections) and provides an LOD of 0.08 µg/L which is 12-times lower than the MCL for microcystin-LR in water. μFIA-TLM was also applied for the determination of total Fe and Fe(II) in 3 µL samples of synthetic cloudwater. The LODs were fo

Identification of Microcystis aeruginosa Peptides Responsible for Allergic Sensitization and Characterization of Functional Interactions between Cyanobacterial Toxins and Immunogenic Peptides.

BACKGROUND: The cyanobacterium species Microcystis aeruginosa produces microcystin and an array of diverse metabolites believed responsible for their toxicity and/or immunogenicity. Previously, chronic rhinitis patients were demonstrated to elicit a specific IgE response to nontoxic strains of M. aeruginosa by skin-prick testing, indicating that cyanobacteria allergenicity resides in a non-toxin-producing component of the organism. OBJECTIVES: We sought to identify and characterize M. aeruginosa peptide(s) responsible for allergic sensitization in susceptible individuals, and we investigated

Elucidation of Taste- and Odor-Producing Bacteria and Toxigenic Cyanobacteria in a Midwestern Drinking Water Supply Reservoir by Shotgun Metagenomic Analysis.

UNLABELLED: While commonplace in clinical settings, DNA-based assays for identification or enumeration of drinking water pathogens and other biological contaminants remain widely unadopted by the monitoring community. In this study, shotgun metagenomics was used to identify taste-and-odor producers and toxin-producing cyanobacteria over a 2-year period in a drinking water reservoir. The sequencing data implicated several cyanobacteria, including Anabaena spp., Microcystis spp., and an unresolved member of the order Oscillatoriales as the likely principal producers of geosmin, microcystin, and

Down-regulation of iron/zinc ion transport and toxin synthesis in Microcystis aeruginosa exposed to 5,4'-dihydroxyflavone.

Flavonoids, common natural polyphenolic compounds from plants, have been proposed as highly effective and safe algicides. However, the molecular mechanism of flavonoids inhibiting Microcystis aeruginosa remains unclear. This study aims in exploring the global transcriptional changes and molecular docking in cyanobacterial cells in response to flavonoids. Transcriptomic analysis revealed that 5,4'-dihydroxyflavone (DHF) primarily affected the genes transcription of iron and zinc ion transport, resulting in the blockage of transport for iron (II), iron (III) and zinc (II), which eventually led t

Elevated CO2-mediated climate warming favors protozoan's top-down effect on controlling toxic Microcystis.

Under temperature and CO2 level rising, the dominance of toxic cyanobacteria in primary producers is continuously increasing the risks of water safety and hindering functions of aquatic ecosystems. Thus, it is necessary to evaluate the efficiency of algal control measures under climate warming. Based on highly efficient control of cyanobacteria by protozoan reported in previous studies, this study aimed to investigate top-down effect of protozoan Paramecium on toxic Microcystis under CO2-mediated climate warming. Results showed that Microcystis removal by Paramecium was mainly affected by Micr
