DBD-FISH Using Specific Chromosomal Region Probes for the Study of Cervical Carcinoma Progression.

Genomic instability is an important biomarker in the progression of cervical carcinoma. DBD-FISH (DNA breakage detection-fluorescence in situ hybridization) is a sensitive method that detects strand breaks, alkali-labile sites, and incomplete DNA excision repair in cells of the cervical epithelium. This technique integrates the microgel immersion of cells from a vaginal lesion scraping and the DNA unwinding treatment with the capacity of FISH integrated into digital image analysis. Cells captured within an agarose matrix are lysed and submerged in an alkaline unwinding solution that generates

Chirality-Encoded Biomaterials Regulate Local and Systemic Immune Responses in Transplantation.

Biomaterial-based immune modulation offers an opportunity to achieve localized graft acceptance without systemic immunosuppression. Here, we demonstrate that molecular chirality within microporous annealed particle (MAP) hydrogels governs innate and adaptive immune responses in skin transplantation and is associated with modulation of alloimmune outcomes. By engineering injectable MAP scaffolds composed of microgels crosslinked with L- or D-peptides, we show that chirality directs early antigen-presenting cell (APC) activation and trafficking in draining lymph nodes, leading to distinct T cell

Microporous Immune-Isolating Capsule with Improved Diffusion for Restored Dynamic Bidirectional Hormone Signaling in a Murine Model of Premature Ovarian Insufficiency.

Pediatric cancer survivors treated with gonadotoxic chemotherapy or radiation face lifelong premature ovarian insufficiency (POI), leading to elevated risk of cardiovascular disease, osteoporosis, and metabolic dysfunction. Pharmacological hormone replacement therapy (HRT) cannot replicate the pulsatile, bidirectional signaling of the hypothalamic-pituitary-gonadal (HPG) axis, leaving a critical therapeutic gap. Immune-isolating hydrogel capsules offer a promising strategy for the implantation of donor ovarian tissue without immunosuppression yet they require optimization for human application

Development of a Synthetic 3D Platform for Compartmentalized Kidney In Vitro Disease Modeling.

3D in vitro tissue and disease models have emerged as an important tool for diagnostic and therapeutic screenings, as they offer a closer approximation toward native environments than traditional 2D cell culture. Kidney disease modeling in particular has progressed to using induced pluripotent stem cells (iPSCs) and microfluidic platforms to replicate the complex microenvironment of the kidney. However, current models lack mature tissue development, scalability, tunability, and spatial organization. In this study, a fully synthetic, 3D kidney disease platform that addresses these challenges is

A Multifunctional Electrochemical Biosensor Based on Near-Infrared Light-Responsive Hydrogel for In Vivo Recording and Modulation.

Monitoring and regulating neural signals are crucial for the management of neurological disorders. Herein, we constructed a near-infrared light (NIR)-responsive device for controllable recording and modulation of neural signals based on enzyme-loaded microgels/multiwalled carbon nanotubes (MWCNTs) composite-modified microelectrodes. The embedded MWCNTs within the composites can convert NIR light into heat via a photothermal effect. Consequently, on one hand, the enzyme-catalyzed reactions on the electrode surface can be reversibly modulated by NIR light. Under NIR irradiation, the contraction

Helical-Like Assembly of Nateglinide as Coating for Oral Delivery of Insulin and Their Synergistic Prevention of Diabetes Mellitus.

Oral delivery of antidiabetic active components promises to free millions of people from daily suffering who require routine injections. However, oral insulin (Ins) and other short-acting compounds such as nateglinide (NG) in harsh gastrointestinal tract still face great challenging, including low bioavailability, and rapid elimination. In this study, inspired by the self-assembly of phenylalanine-based peptides in nature, it is showed that NG a small phenylalanine derivative, assembles into left-handed helical nanofibers in the presence of Ca2+ . These helical NG nanofibers functioned as a co

Facile Microfluidic Fabrication of Biocompatible Hydrogel Microspheres in a Novel Microfluidic Device.

Poly(ethylene glycol) diacrylate (PEGDA) microgels with tuneable size and porosity find applications as extracellular matrix mimics for tissue-engineering scaffolds, biosensors, and drug carriers. Monodispersed PEGDA microgels were produced by modular droplet microfluidics using the dispersed phase with 49-99 wt% PEGDA, 1 wt% Darocur 2959, and 0-50 wt% water, while the continuous phase was 3.5 wt% silicone-based surfactant dissolved in silicone oil. Pure PEGDA droplets were fully cured within 60 s at the UV light intensity of 75 mW/cm2. The droplets with higher water content required more time

Phenol release from pNIPAM hydrogels: scaling molecular dynamics simulations with dynamical density functional theory.

We employed molecular dynamic simulations (MD) and the Bennett's acceptance ratio method to compute the free energy of transfer, ΔGtrans, of phenol, methane, and 5-fluorouracil (5-FU), between bulk water and water-pNIPAM mixtures of different polymer volume fractions, ϕp. For this purpose, we first calculate the solvation free energies in both media to obtain ΔGtrans. Phenol and 5-FU (a medication used to treat cancer) attach to the pNIPAM surface so that they show negative values of ΔGtrans irrespective of temperature (above or below the lower critical solution temperature of pNIPAM, Tc). [Genotoxic effect on human mucous membrane biopsies of the upper aerodigestive tract].

BACKGROUND: In numerous epidemiologic studies, environmental and occupational substances such as sodium dichromate (Na2Cr2O7), benzo[a]pyren (B(a)P), and N'nitroso-diethanolamine (NDELA) have been shown to be of potential carcinogenic risk on human epithelial cells in the upper aerodigestive tract. METHODS: Using the alkaline microgel electrophoresis technique (comet assay). mucosal cells isolated from biopsies of the upper aerodigestive tract (nose, paranasal sinuses, mouth, pharynx, larynx, and tonsils) were used to analyze target sites for different genotoxic substances and specific sensit

Drying of Soft Colloidal Films.

Thin films made of deformable micro- and nano-units, such as biological membranes, polymer interfaces, and particle-laden liquid surfaces, exhibit a complex behavior during drying, with consequences for various applications like wound healing, coating technologies, and additive manufacturing. Studying the drying dynamics and structural changes of soft colloidal films thus holds the potential to yield valuable insights to achieve improvements for applications. In this study, interfacial monolayers of core-shell (CS) microgels with varying degrees of softness are employed as model systems and to

Conjugation of IL-33 to Microporous Annealed Particle Scaffolds Enhances Type 2-Like Immune Responses In Vitro and In Vivo.

The inflammatory foreign body response (FBR) is the main driver of biomaterial implant failure. Current strategies to mitigate the onset of a FBR include modification of the implant surface, release of anti-inflammatory drugs, and cell-scale implant porosity. The microporous annealed particle (MAP) scaffold platform is an injectable, porous biomaterial composed of individual microgels, which are annealed in situ to provide a structurally stable scaffold with cell-scale microporosity. MAP scaffold does not induce a discernible foreign body response in vivo and, therefore, can be used a "blank c

Synthesis of biocompatible sterically-stabilized poly(2-(methacryloyloxy)ethyl phosphorylcholine) latexes via dispersion polymerization in alcohol/water mixtures.

Poly(2-(methacryloyloxy)ethyl phosphorylcholine) (PMPC) is soluble in either 2-propanol or water but becomes insoluble in certain alcohol-rich 2-propanol/water mixtures. We have exploited this unusual cononsolvency behavior in order to prepare new biocompatible sterically stabilized PMPC latexes via nonaqueous dispersion polymerization in 2-propanol/water mixtures. All polymerizations were conducted in the presence of monomethoxy-capped poly(ethylene glycol) methacrylate (PEGMA) as a reactive stabilizer, with some formulations including ethylene glycol dimethacrylate (EGDMA) as a cross-linker.

[The physical compatibility of ketamine and diazepam in infusion solutions].

Physical compatibilities of infusion solutions containing ketamine and diazepam or midazolam in different concentrations were studied by visual examination and by light-scattering measurements. The isotonic infusion solutions used were fructose 5%, glucose 5%, and NaCl 0.9%. The concentration of additive were between 500 and 600 mg ketamine, 60 and 100 mg diazepam and 30 and 60 mg midazolam (Dormicum) per liter intravenous solution. During a period of 24 h the solutions remained colorless without demixing, precipitation, or changes in pH. The light-scattering measurements were performed immedi

Elucidating the combinatorial effect of substrate stiffness and surface viscoelasticity on cellular phenotype.

Cells maintain tensional homeostasis by monitoring the mechanics of their microenvironment. In order to understand this mechanotransduction phenomenon, hydrogel materials have been developed with either controllable linear elastic or viscoelastic properties. Native biological tissues, and biomaterials used for medical purposes, often have complex mechanical properties. However, due to the difficulty in completely decoupling the elastic and viscous components of hydrogel materials, the effect of complex composite materials on cellular responses has largely gone unreported. Here, we characterize

Order-disorder transition in soft and deformable particle assembly with dynamic size-dispersity in two dimensions.

Soft and deformable objects are widespread in natural and synthetic systems, including micellar domains, microgel particles, foams, and biological cells. Understanding their phase behavior at high concentrations is crucial for controlling long-range order. Here, we employ a Voronoi-based model to study the packing of deformable particles in two dimensions under thermal fluctuations. Particles are represented as interconnected polygons, with the system energy comprising penalties for deviations in area and perimeter from preferred values. The strengths of these penalties capture two key feature

Hydroxybutyl chitosan-based thermosensitive hydrogel enhanced osteoporotic bone regeneration through sustained release of alendronate and BMP-2.

Osteoporotic patients face an elevated risk of fractures and bone defects due to impaired repair mechanisms stemming from imbalanced osteoclast and osteoblast activity. In this context, we present an injectable thermo-responsive hydrogel/microgel platform based on Hydroxybutyl Chitosan, which exhibits long-term anti-osteoporotic and osteogenic efficacy aimed at enhancing bone regeneration in osteoporotic conditions. The hydrogel demonstrated excellent thermo-responsive properties, injectability, and an optimal degradation rate. It facilitated the sustained release of anti-osteoclastogenic alen

Enhanced Elastic Multifunctional Dual-Network Hydrogel Microspheres for the Treatment of Intervertebral Disc Degeneration through Inflammation Modulation and Apoptosis Inhibition.

Modulating the local inflammatory environment and restoring nucleus pulposus cell (NPC) function are essential strategies for mitigating IVDD. In this study, a microgel-based delivery system for microRNA therapeutics, termed POCM@MCCP (PMCCP), was developed. The system employs dual-network hydrogel microspheres composed of chitosan, citric acid, and poly(vinyl alcohol) (CCP), further functionalized with metal-phenolic networks (MPNs) formed from strontium ions (Sr2+) and epigallocatechin gallate (EGCG). This microgel enables dynamic, stimulus-responsive loading of phenylboronic acid-modified o

Bioorthogonally reinforced injectable granular hydrogels synergizing ECM mimicry with microporosity for skin tissue engineering.

Designing injectable biomaterials that simultaneously recapitulate extracellular matrix (ECM) composition while maintaining interconnected microporosity remains a central challenge in regenerative scaffolds. Granular hydrogels offer unique opportunities to address this limitation because their jammed microgel architecture inherently supports injectability and cell-accessible porosity. Here, we report a bioorthogonally reinforced ECM-mimetic granular hydrogel platform assembled from complementary gelatin and hyaluronic acid-derived microparticles that anneal through dynamic hydrazone coupling.

Cross-Hierarchical Analysis of Self-Oscillating Polymer Materials Through Activation Energy.

In chemically driven soft materials, polymer architecture can shape how chemical reactions are transduced into material-level dynamic behavior. However, the relationship between polymer architecture and the resulting dynamics remains insufficiently understood. Here, using Arrhenius analysis, we perform a cross-hierarchical comparison of the temperature dependence of Belousov-Zhabotinsky (BZ) reaction-driven oscillatory dynamics across three structurally related systems: molecular Ru(bpy)3Cl2, a linear self-oscillating polymer (SOP), and a self-oscillating microgel (SOMG). In these systems, Ru(

Mild photothermal-responsive hydrogel with H2Se delivery for anti-infection and microenvironment remodeling to bone regeneration in diabetic osteomyelitis.

Diabetic osteomyelitis, exacerbated by a hyperglycemic microenvironment, leads to increased bacterial infections and bone tissue destruction, raising the risk of amputation. Current antibiotic therapies are limited in effectiveness due to rising antibiotic resistance and biofilm barrier. In response, we developed a microgel-based hydrogel system that delivers H2Se gas combined with mild photothermal therapy (MPTT) to achieve integrated treatment for infection control, anti-inflammation, and osteogenesis. This system utilizes Fe3O4 nanoparticles as photothermal-responsive carriers to encapsulat
