Protein expression profiles of 10 isolates of Mycobacterium paratuberculosis, M avium 18 (formerly M paratuberculosis 18), and 1 isolate each of M avium serotype 2, M avium serotype 8, and M bovis BCG were examined. Protein expression profiles of M paratuberculosis and M avium were similar. However, two-dimensional gel analysis of [35S]methionine-labeled cellular proteins resolved 4 proteins, with molecular mass of 28,000, 32,000, 32,000, and 42,000 daltons, which were expressed in greater amounts in M paratuberculosis than in M avium. Two proteins, with molecular mass of 43,000 and 60,000 dal

Measurement of secreted interferon (IFN)-gamma has proven to be a valuable tool for the detection of animals infected with mycobacterial pathogens, including Mycobacterium avium subsp. paratuberculosis. Previous reports have suggested that tuberculin skin testing can influence the performance of the IFN-gamma assay. In the present study, healthy noninfected cows, and cows subclinically and clinically infected with M. paratuberculosis were administered an intradermal injection of johnin purified protein derivative (JPPD) and effects on secreted and intracellular IFN-gamma were observed. Intrade

BACKGROUND: The development of novel antibiotics to treat multidrug-resistant (MDR) tuberculosis is time-consuming and expensive. Multiple immune modulators, immune suppressants, anti-inflammatories, and growth enhancers, and vitamins A and D, inhibit Mycobacterium avium subspecies paratuberculosis (MAP) in culture. We studied the culture inhibition of Mycobacterium tuberculosis complex by these agents.
METHODS: Biosafety level two M. tuberculosis complex (ATCC 19015 and ATCC 25177) was studied in radiometric Bactec or MGIT culture. Agents evaluated included clofazimine, methotrexate, 6-merca

Understanding the pathogenesis of paratuberculosis (PTB), caused by Mycobacterium avium subspecies paratuberculosis (MAP), is essential in the management of this economically important disease. MAP strains are known to differ genotypically, which could contribute to differences in strain virulence and disease outcomes. The behaviour of MAP inside the host and the mechanisms utilized by MAP to overcome the host defence system are not entirely understood. This study aimed at evaluating the potential virulence of different MAP isolates from cattle in Uganda. This was achieved by analysing the cyt

Mycobacterium avium subspecies paratuberculosis (MAP) is the causative agent of Johne's Disease, a chronic granulomatous enteritis of ruminants. MAP establishes an infection in the host via the small intestine. This requires the bacterium to adhere to, and be internalised by, cells of the intestinal tract. The effector molecules expressed by MAP for this purpose remain to be fully identified and understood. Mammalian cell entry (mce) proteins have been shown to enable other Mycobacterial species to attach to and invade host epithelial cells. Here, we have expressed Mce1A, Mce1D, Mce3C and Mce4

Paratuberculosis (Johne's disease) is a chronic enteric infection of ruminants caused by Mycobacterium avium subsp. paratuberculosis (MAP), leading to significant economic losses in livestock production. While the solute carrier family 11 member 1 (SLC11A1) gene has been implicated in resistance to intracellular pathogens in several species, its role in ovine paratuberculosis remains largely uncharacterized. The present study investigated whether polymorphic variation in the SLC11A1 3' untranslated region (3'UTR) (GT)n microsatellite is associated with resistance or susceptibility to MAP infec

Mycobacterium avium subspecies paratuberculosis (MAP) causes chronic inflammation of the intestines in humans, ruminants, and other species. It is the causative agent of Johne's disease in cattle, and has been implicated as the causative agent of Crohn's disease in humans. To date, no quantitative microbial risk assessment (QMRA) for MAP utilizing a dose-response function exists. The objective of this study is to develop a nested dose-response model for infection from oral exposure to MAP utilizing data from the peer-reviewed literature. Four studies amenable to dose-response modeling were ide

Paratuberculosis is a chronic infectious disease of ruminants caused by Mycobacterium avium subsp. paratuberculosis (MAP). MAP is transmitted primarily through ingestion of contaminated feces, milk, or water and establishes persistent infection within intestinal macrophages by evading host immune defenses. Typical lesions include granulomatous enteritis, lymphangitis, and lymphadenitis. Lymphangitis mimicking arteritis has not been reported in bovine paratuberculosis, to our knowledge. Here, we describe a case of paratuberculosis in a 2-y-old Angus cow that was found dead. Gross changes consis

Paratuberculosis (PTBC) is a chronic granulomatous enteritis of ruminants caused by Mycobacterium avium subsp. paratuberculosis (Map). Although PTBC intrauterine transmission has been described in cattle, sheep, and deer, data in dairy goats are limited. The present study investigated intrauterine Map infection in 32 pregnant Saanen goats from a herd with high PTBC prevalence in Jujuy, Argentina. Maternal and fetal tissues were analyzed using a range of methods, including bacteriological culture, IS900 PCR, histopathology, and serology. All goats were positive to the intradermal test, and 75%

This study evaluated the efficacy of four different culture media in cultivating Mycobacterium avium subspecies paratuberculosis (MAP). Additionally, it examined the extraction of surface antigens through the suspension of harvested bacteria in ethanol, followed by vortex agitation. The potential of the extracted antigens in the agar gel immunodiffusion test (AGID) was evaluated by altering the loading pattern. This modification involved adding varying concentrations of antigens to peripheral wells and comparing the results to the conventional technique, which typically places a single antigen

INTRODUCTION: Paratuberculosis is a granulomatous intestinal infection that affects ruminant animals worldwide. The disease is often detected when most animals are already infected due to the long incubation period and the high transmissibility of the infectious agent. The lack of a comprehensive method to diagnose Paratuberculosis is a global challenge. Therefore, a non-destructive, fast, and cost-effective diagnostic method for early detection of Paratuberculosis is crucial.
METHODS: Near-infrared spectroscopy (NIRS) and Aquaphotomics have the potential to diagnose the disease by detecting

Extracellular microRNAs (miRNAs) are detectable in the peripheral blood and have been touted as potential biomarkers for a range of maladies. The presence and biomarker potential of miRNAs in other biofluids has been less thoroughly explored, particularly in the veterinary realm. Faecal miRNAs are a case in point; while they have been identified largely in rodents and humans, they have not been reported in cattle but may have prognostic or diagnostic value for Johne's Disease (JD) in cattle, a chronic granulomatous inflammation of the ileum caused by Mycobacterium avium subspecies paratubercul

Pathogenic mycobacteria such as Mycobacterium tuberculosis are capable of utilising cholesterol as a primary carbon-based energy source in vitro but there has been little research examining the significance of cholesterol in vivo. Johne's disease is a chronic enteric disease of ruminants caused by Mycobacterium avium subspecies paratuberculosis (MAP). This study sought to evaluate the levels of total serum cholesterol in the host following exposure to MAP. Blood samples were collected from both sheep and cattle prior to experimental challenge with MAP and at monthly intervals post-challenge. T

Vaccination is the most cost effective control measure for Johne's disease caused by Mycobacterium avium subspecies paratuberculosis (MAP) but currently available whole cell killed formulations have limited efficacy and are incompatible with the diagnosis of bovine tuberculosis by tuberculin skin test. We have evaluated the utility of a viral delivery regimen of non-replicative human Adenovirus 5 and Modified Vaccinia virus Ankara recombinant for early entry MAP specific antigens (HAV) to show protection against challenge in a calf model and extensively screened for differential immunological

The gastrointestinal tract contains a complex mix of microorganisms. Therefore, the finding of a particular microbe in the gastrointestinal tract or stools of a patient with intestinal disease does not necessarily indicate that the clinical state resulted from the presence of this organism. For this reason, extending from epidemiological association to causality is particularly challenging for gastrointestinal diseases. Most established agents of acute bacterial gastroenteritis (eg, Escherichia coli, Campylobacter jejuni) have been shown to fulfil Koch's postulates of causality in human volunt

BACKGROUND: Paratuberculosis is a common disease in ruminants, causing economic losses in livestock farming, and a relationship between the exposure to its causative agent Mycobacterium avium subsp. paratuberculosis, and Crohn's disease in humans is discussed. Despite this, only a minority of cattle farmers have enroled in voluntary control programmes in most countries. Therefore, this study aimed at investigating the farmer's opinion on paratuberculosis and their motivations to participate in a control programme. The objective was to identify different groups among farmers regarding their mot

BACKGROUND: Among the non-tuberculous mycobacteria, Mycobacterium (M.) avium are important pathogens for humans and/or animals. Currently, there are four M. avium subspecies: subsp. hominissuis (Mah), subsp. paratuberculosis (Map), subsp. avium (Maa), and subsp. silvaticum (Mas). While sufficient data is available for the first three mentioned, only few reports exist on the isolation, epidemiology and even less on the genetic equipment of Mas.
RESULTS: Here, Mas was isolated from an Egyptian goose that died of avian tuberculosis. Subspecies identification was based on the presence of IS901 an

Defects in macrophage function have been implicated in the establishment of Crohn's disease (CD). However, the response of macrophages from CD patients to live bacteria, particularly Mycobacterium avium subsp. paratuberculosis (MAP), has not been addressed. Considering MAP has long been associated to CD, our objective was to assess whether macrophages from CD patients showed impaired inflammatory response to infection by MAP comparing to M. avium subsp. avium (MA) and other live intestinal commensal bacteria. Human peripheral blood monocyte-derived macrophages were obtained from CD patients, u
