Contextualizing the Utility of Polygenic Risk Scores using Absolute Risk Models in Diverse Ancestry Populations.

Polygenic risk scores (PRSs) are emerging as powerful tools for quantifying inherited risk for common diseases and, in some cases, are approaching clinical implementation. A major concern for PRS implementation is their limited accuracy in non-European populations, particularly in those of African ancestry. However, past evaluations have focused on metrics such as relative risk or AUC, which do not capture background risk arising from contextual factors. We introduce a novel measure of variable importance, the conditional average derivative estimator (CADE), to evaluate PRS utility across dive

"Colorectal Cancer Care Quality in a Developing Country: Insights from a Comparison of Teaching and Non-teaching Hospitals in Iran".

BACKGROUND: Our study represents the first effort in the Eastern Mediterranean Region to identify disparities in the quality of colorectal cancer (CRC) care in Iran. METHODS: We established a collaborative registry program for non-metastatic CRC patients to evaluate survival rates between teaching cancer centers (TCCs) and a high-volume, non-teaching, non-cancer center (NTNC). The study included a diverse patient population and considered various factors such as cancer stage, margin involvement, adherence to guidelines for adjuvant and neoadjuvant treatments, emergency surgeries, socioeconomi

Cancer risks and mortality in heterozygous ATM mutation carriers.

BACKGROUND: Homozygous or compound heterozygous mutations in the ATM gene are the principal cause of ataxia telangiectasia (A-T). Several studies have suggested that heterozygous carriers of ATM mutations are at increased risk of breast cancer and perhaps of other cancers, but the precise risk is uncertain. METHODS: Cancer incidence and mortality information for 1160 relatives of 169 UK A-T patients (including 247 obligate carriers) was obtained through the National Health Service Central Registry. Relative risks (RRs) of cancer in carriers, allowing for genotype uncertainty, were estimated w

National and Subnational Burden of Female Breast Cancer in Iran from 2010 to 2021.

Background and Objectives: Female breast cancer (FBC) is an increasing public health concern in Iran, with notable geographic disparities that necessitate comprehensive burden assessments at national and provincial levels. This study presented the national and subnational burden and changes in FBC burden from 2010 to 2021 in Iran in comparison with global data. Materials and Methods: The GBD (2021) data on female BC were extracted from the Global Health Data Exchange (GHDx) query tool. Age-standardized incidence, deaths, prevalence, and adjusted years of life with disabilities (DALYs) rates (p

Five-Year Overall Survival of Interval Breast Cancers is Better than Non- Interval Cancers from Korean Breast Cancer Registry.

Objective: Interval breast cancer (IC) is a limitation of breast cancer screening. We investigated data from a large scaled breast cancer dataset of patients with breast cancer who underwent breast cancer screening in order to recapitulate the overall survival (OS) of patients with ICs compared to those with non-ICs. Methods: A total of 27,141 patients in the Korean breast cancer registry with breast cancer who had ever participated in biannual national breast cancer screening programs between 2009 and 2013 were enrolled. We compared the social, pregnancy-associated, and pathologic characteris

Non-communicable disease care for persons living with HIV in Peru: A national physician cross-sectional study.

Non-communicable diseases (NCDs) are a significant cause of morbidity and mortality for the aging HIV population worldwide. In Peru, no data exists on how providers address NCDs for persons living with HIV (PLWH). This study examines HIV physician confidence and current management practices for NCDs for PLWH in Peru. We recruited public-sector HIV physicians via Peru's National HIV, STI and Hepatitis Program's (NHSTIHP) physician registry and by program coordinator referral. Participants completed a telephone survey encompassing seven NCDs [hyperlipidemia, hypertension, diabetes, osteoporosis,

Updated population-based review of carcinoid tumors.

OBJECTIVE: To determine the population-based incidence, anatomic distribution, and survival rates of gastrointestinal carcinoid tumors. BACKGROUND: Carcinoid tumors arise from neuroendocrine cells and may develop in almost any organ. Many textbooks and articles represent single institution studies and report varying incidence rates, anatomic distribution of tumors, and patient survival rates. Population-based statistics remain largely unknown. METHODS: Data was obtained from the National Cancer Institute Surveillance, Epidemiology, and End Results program (1973 to 1997). Incidence rates, dis

Survival by sex and HIV status in patients with anal cancer in the USA between 2001 and 2019: a retrospective cohort study.

BACKGROUND: The risk of anal cancer is increased among people with HIV, particularly men who have sex with men. Estimating survival by HIV status and sex and identifying groups at high risk is crucial for documenting prognostic differences between populations. We aimed to compare all-cause and anal cancer-specific survival in patients with anal cancer with and without HIV, stratified by sex, and to identify predictors of survival, stratified by HIV status. METHODS: In this retrospective cohort study, we used data from the HIV/AIDS Cancer Match Study of 13 population-based HIV and cancer regis

Long-term exposure to wildfire-related PM2·5 and overall survival of older patients with lung cancer: a retrospective, registry-based cohort study.

BACKGROUND: Fine particulate matter (PM2·5) has been identified as a key risk factor for lung cancer incidence and survival. PM2·5 from wildfire smoke is an increasingly important contributor to total PM2·5 in the USA, driven by climate change, and potentially more toxic than non-wildfire PM2·5 due to its smaller particle size and higher toxic contents. However, the effects of wildfire-related PM2·5 on the overall survival of patients with lung cancer remain unclear. METHODS: This retrospective cohort study included patients aged 65 years or older with histologically confirmed primary small-c

Emerging cancer trends among young adults in the USA: analysis of a population-based cancer registry.

BACKGROUND: Cancer trends in young adults, often under 50 years, reflect recent changes in carcinogenic exposures, which could foreshadow the future overall disease burden. Previous studies reported an increase in early onset colorectal cancer, which could partly reflect the obesity epidemic. We examined age-specific contemporary incidence trends in the USA for 30 common cancers, including 12 obesity-related cancers. METHODS: We obtained incidence data for invasive cancers among people aged 25-84 years diagnosed from Jan 1, 1995, to Dec 31, 2014, for 25 population-based state registries in th

Burden of human papillomavirus infections and related diseases in the extended Middle East and North Africa region.

In this chapter, we present the available information on the burden of human papillomavirus (HPV)-related cancers (cancers of the cervix, anogenital cancers, and cancers of the oral cavity and pharynx), as well as available data on the prevalence and distribution of HPV types in women with and without cervical cancer, for the countries of the Extended Middle East and North Africa region (EMENA). The EMENA region is characterized by societies that share similar cultures and religions and that are considered to have a more conservative sexual behaviour compared to Western societies. The incidenc

The First 6 Years' Experiences of a National Centralized Offspring Surveillance Setting for Dutch Children Prenatally Exposed to Maternal Cancer to Inform Future International Practice: Protocol for a Demographic Review of Referred Families and Key Lessons Learned.

BACKGROUND: Cancer during pregnancy is a rare and significant life-changing event affecting approximately 1 in 1000-2000 pregnancies. With increasing maternal age and broader application of prenatal screening programs such as the Dutch Non-Invasive Prenatal Testing, incidental detection of maternal cancer is becoming more frequent. Advancements in safe treatment options during pregnancy, supported by the International Network on Cancer Infertility and Pregnancy (INCIP), have led to fewer pregnancy terminations. Consequently, more children are exposed to chemotherapy and other cancer treatments

Emerging cancer disease burden in a rural sub-Saharan African population: northeast Nigeria in focus.

INTRODUCTION: Sub-Saharan Africa (SSA) is plagued by myriads of diseases, mostly infectious; but cancer disease burden is rising among non-communicable diseases. Nigeria has a high burden of cancer, however its remote underserved culturally-conserved populations have been understudied, a gap this study sought to fill. METHODS: This was a cross-sectional multi-institutional descriptive study of histologically diagnosed cancers over a four-year period (January 2019-December 2022) archived in the Departments of Pathology and Cancer Registries of six tertiary hospitals in the northeast of Nigeria

Incidence of suicide in persons with cancer.

PURPOSE: The purpose of this study was to characterize suicide rates among patients with cancer in the United States and identify patient and disease characteristics associated with higher suicide rates. Prior studies, mostly in Europe, have suggested that patients with cancer may be at increased risk for suicide, but large cohort studies comparing patients with cancer with the general population have not been performed in the United States. METHODS: Patients in the study were residents of geographic areas served by the Surveillance, Epidemiology, and End Results (SEER) program who were diagn

Frequency and natural history of inherited bone marrow failure syndromes: the Israeli Inherited Bone Marrow Failure Registry.

BACKGROUND: Inherited bone marrow failure syndromes are rare genetic disorders characterized by bone marrow failure, congenital anomalies, and cancer predisposition. Available single disease registries provide reliable information regarding natural history, efficacy and side effects of treatments, and contribute to the discovery of the causative genes. However, these registries could not shed light on the true incidence of the various syndromes. We, therefore, established an Israeli national registry in order to investigate the relative frequency of each of these syndromes and their complicati

National Brain Tumour Registry of China (NBTRC) statistical report of primary brain tumours diagnosed in China in years 2019-2020.

BACKGROUND: The lack of a well-designed brain tumour registry with standardized pathological diagnoses in underdeveloped countries hinders the ability to compare epidemiologic data across the globe. The National Brain Tumour Registry of China (NBTRC), created in January 2018, is the first multi-hospital-based brain tumour registry in China. Patient data reported to the NBTRC in years 2019-2020 were assessed. METHODS: Tumour pathology was based on the 2016 World Health Organization (WHO) classification of tumours of the central nervous system and ICD-O-3. The anatomical site was coded per the

Mortality Rate and Years of Life Lost Due to Breast and Gynecologic Cancers in Southern Iran 2004-2019: A Population-Based Study.

BACKGROUND: There is an increase in the incidence of breast and gynecologic cancers in Iran during the last three decades. Literature is inadequate about the Years of Life Lost (YLL) attributed to these cancers in Iran. METHODS: Mortality data due to these cancers amongst females aged >20 yr from 2004 to 2019 was acquired from "Electronic Death Registry System", Fars Province, Iran. Using local health centers' databases and national census reports from 1996 to 2016, the age-standardized incidence rate (ASR) per 100,000 people were estimated. YLLs were calculated using the WHO's 2015 "YLL temp

Decreasing trend in mortality of chronic myelogenous leukemia patients after introduction of imatinib in Japan and the U.S.

PURPOSE: Although the impact of imatinib in improving survival outcomes in chronic myelogenous leukemia (CML) patients has been widely reported, its impact on mortality from CML has not been evaluated. A survival benefit demonstrated in clinical trials does not simply translate to a decrease in mortality. To evaluate the impact of imatinib on the public health, we estimated the age-standardized mortality rate of CML patients in Japan and the U.S. using vital statistics data for Japan and data from the Surveillance, Epidemiology, and End Results (SEER) Program of the National Cancer Institute f

Breast Cancer Mortality After Implementation of Organized Population-Based Breast Cancer Screening in Norway.

BACKGROUND: We estimated breast cancer (BC) mortality reduction associated with invitations to a nationwide population-based screening program and with changes in treatment. MATERIALS AND METHODS: BreastScreen Norway started in 1996 and became nationwide in 2005. It invites women aged 50-69 years to biennial mammographic screening. We retrieved individual-level data for 1 340 333 women from national registries. During 1996-2014 (screening window), women contributed person-years in noninvited and invited periods.
