Neocortical Expansion: An Attempt toward Relating Phylogeny and Ontogeny.

The neocortex is the most characteristic feature of the human brain. On gross inspection, its convoluted surfaces can be seen to have overgrown and covered most other brain structures. In the functional sphere, it is to the neocortex that we attribute those behaviors assumed to be most uniquely human such as cognition and linguistic behavior. This essay is an attempt to understand how this structure expanded during the course of mammalian evolution. At present, any attempt must be more speculative than definitive, but it is offered in the hope that it will generate more discussion on a topic t

Short-term changes in NADPH-diaphorase reactivity in rat brain following perinatal asphyxia. Neuroprotective effects of cold treatment.

Perinatal asphyxia (PA) produces changes in nitric oxide synthase (NOS) activity in neuronal and endothelial cells of the striatum and neocortex. The changes were examined using a histochemical NADPH-diaphorase (NADPH-d) staining method. Newborn rats were exposed to severe PA at 37 degrees C and other groups were subjected to severe PA under hypothermic condition (15 degrees C) for 20 or 100 min, respectively. Quantitative image analysis was performed on the striatum and neocortex in order to count cell number of reactive neurons and to compare the pattern of staining between the different gro

Diagnostic value of diffusion-weighted brain magnetic resonance imaging in patients with sporadic Creutzfeldt-Jakob disease: a systematic review and meta-analysis.

OBJECTIVE: To evaluate the diagnostic yield and performance of DWI in patients with sporadic CJD (sCJD). METHODS: A systematic literature search of the MEDLINE and EMBASE databases was performed, since their inception up to July 28, 2020. Pooled diagnostic yield of diffusion-weighted imaging was calculated using DerSimonian-Laird random-effects model. Pooled diagnostic performance of DWI (sensitivity, specificity, and area under the curve) in diagnosing sCJD among patients with rapidly progressive dementia was calculated using a bivariate random-effects model. Subgroup analysis and meta-regre

Neocortical tissue recovery in severe congenital obstructive hydrocephalus after intraventricular administration of bone marrow-derived mesenchymal stem cells.

BACKGROUND: In obstructive congenital hydrocephalus, cerebrospinal fluid accumulation is associated with high intracranial pressure and the presence of periventricular edema, ischemia/hypoxia, damage of the white matter, and glial reactions in the neocortex. The viability and short time effects of a therapy based on bone marrow-derived mesenchymal stem cells (BM-MSC) have been evaluated in such pathological conditions in the hyh mouse model. METHODS: BM-MSC obtained from mice expressing fluorescent mRFP1 protein were injected into the lateral ventricle of hydrocephalic hyh mice at the moment

[Effect of intrauterine exposure to ethanol on synthesis of various phospholipids in the rat brain during the postnatal period].

Male rats, whose mothers were given ethanol during pregnancy, were injected inorganic P32 into lateral brain ventricles. Some animals during 1 hour before decapitation were subjected to stress. Phosphatidylethanolamine, phosphatidylcholine and phosphatidylserine were isolated from neocortex and hippocamp. Prenatal alcohol treatment led to 30% inorganic P32 incorporation increase into neocortex phosphatidylcholine. Stress was followed by phosphatidylcholine synthesis level decrease in neocortex by 13% and in hippocamp by 26%. Amplitude of phospholipid synthesis alterations increased after both

[Cortical efferents in different areas of the cat orbitofrontal cortex].

The efferental relations of different parts of the cat orbitofrontal cortex with other parts of the neocortex are not equal. As to the distribution in different zones of the neocortex they differ in the quantitative and spatial characteristics. The number of efferents originating from the orbitofrontal cortex increases from the medial part of the proreal gyrus to its dorsolateral parts and anterior part of the orbital gyrus. Thus the frontal part of the orbital gyrus has the greatest number of the efferental relations with other parts of the neocortex including the projection visual cortex. Th

Visual read of [F-18]florquinitau PET that includes and extends beyond the mesial temporal lobe is associated with increased plasma pTau217 and cognitive decline in a cohort that is enriched with risk for Alzheimer's disease.

INTRODUCTION: Patterns of signal from tau positron emission tomography (tau-PET) confined to the medial temporal lobe (MTL) or extended into the neocortex may be relevant for Alzheimer's disease (AD) research if they are linked to differential biomarker levels and cognitive decline. METHODS: Visual assessment of Tau-PET [F-18]florquinitau (FQT) exams from 728 initially non-demented older adults yielded four uptake groups: tau-negative (T-), MTL-only (T+MTL), neocortex-only (T+Neo), or both (T+MTL&Neo). Mixed effects models assessed group differences in retrospective cognitive and plasma pTau2

Human neocortical development as a basis to understand mechanisms underlying neurodevelopmental disabilities in extremely preterm infants.

AIM: Recent advances in perinatal and neonatal medicine have resulted in marked improvements in the survival rates of extremely preterm infants (born before 28 gestational weeks) around the world, and Japan is among the countries with the highest reported survival rates of extremely preterm infants. However, it remains a major concern that many survivors develop neurodevelopmental disabilities, including cognitive dysfunctions and neurodevelopmental disorders later in life. In order to understand the pathophysiological mechanisms underlying the neurodevelopmental disabilities observed in the s

Effect of current conduction for local epileptiform discharges in patients with temporal lobe epilepsy.

OBJECTIVES: The effects of current conduction were researched to confirm that it can decrease focal epileptogenicity in patients with temporal lobe epilepsy (TLE). METHODS: Data from 13 patients with mesial TLE were collected. After no less than two habitual seizures were captured during stereo-electroencephalogram monitoring, current conduction was measured in the hippocampus to a homemade, zero potential circuit board. The interictal spike, ripple, fast ripple, and ictal epileptogenicity index (EI) changes were analyzed in the hippocampus, amygdala, and anterior and middle temporal neocorte

Relation between clinical findings and progression of cerebral cortical pathology in MM1-type sporadic Creutzfeldt-Jakob disease: proposed staging of cerebral cortical pathology.

In our pathologic observation of the cerebral cortex including the neocortex, hippocampus, and limbic cortex in 43 Japanese patients with MM1-type sporadic Creutzfeldt-Jakob disease, the earliest pathologic finding was spongiform change and next was gliosis. Subsequently, neuropil rarefaction appeared, followed by neuron loss. On the basis of these observations, we propose the following cortical pathologic staging: Stage I, spongiform change; Stage II, hypertrophic astrocytosis; Stage III, neuropil rarefaction; Stage IV, neuron loss; Stage V, status spongiosus; and Stage VI, large cavity forma

Isoflurane causes neocortical but not hippocampal-dependent memory impairment in mice.

BACKGROUND: The aim of this study is to investigate the effect of general anaesthesia induced by isoflurane with buprenorphine on hippocampus-dependent and neocortex-dependent memory, respectively, in mice, and in addition, to compare the effects of such anaesthesia on these memory processes with the effects induced by lipopolysaccharide (LPS) administration on the same memory processes. METHODS: To assess hippocampus-dependent memory, isoflurane (for 15 min) after buprenorphine injection, or LPS 100 μg/kg (intraperitoneally) was administered 24 h before or after fear conditioning. Spreading depolarizations in the rat endothelin-1 model of focal cerebellar ischemia.

Focal brain ischemia is best studied in neocortex and striatum. Both show highly vulnerable neurons and high susceptibility to spreading depolarization (SD). Therefore, it has been hypothesized that these two variables generally correlate. However, this hypothesis is contradicted by findings in cerebellar cortex, which contains highly vulnerable neurons to ischemia, the Purkinje cells, but is said to be less susceptible to SD. Here, we found in the rat cerebellar cortex that elevated K+ induced a long-lasting depolarizing event superimposed with SDs. Cerebellar SDs resembled those in neocortex

Parallel pathways from motor and somatosensory cortex for controlling whisker movements in mice.

Mice can gather tactile sensory information by actively moving their whiskers to palpate objects in their immediate surroundings. Whisker sensory perception therefore requires integration of sensory and motor information, which occurs prominently in the neocortex. The signalling pathways from the neocortex for controlling whisker movements are currently poorly understood in mice. Here, we delineate two pathways, one originating from primary whisker somatosensory cortex (wS1) and the other from whisker motor cortex (wM1), that control qualitatively distinct movements of contralateral whiskers.

Suppressor of Fused Is Critical for Maintenance of Neuronal Progenitor Identity during Corticogenesis.

Proper lineage progression and diversification of neural progenitor cells (NPCs) ensures the generation of projection neuron (PN) subtypes in the mammalian neocortex. Here, we show that Suppressor of Fused (Sufu) controls PN specification by maintaining the identity of NPCs in the embryonic neocortex. Deletion of Sufu in NPCs of the E10.5 mouse neocortex led to improper specification of progenitors and a reduction in intermediate progenitors (IPs) during corticogenesis. We found that Sufu deletion resulted in unstable Gli2 and Gli3 activity, leading to the ectopic activation of Sonic hedgehog

Periventricular heterotopias display cortical degenerative neuropathology.

This report describes beta-amyloid plaques in periventricular heterotopias in two elderly women. The neuropathologic features of the heterotopias recapitulated those of nearby neocortex but were distinct from those of adjacent caudate nucleus. In response to the factors important in plaque and tangle formation, the heterotopias biologically behave like neocortex and unlike adjacent striatum, further supporting the morphologic evidence that these tissues represent heterotopic cortex.

Status marmoratus in fetal cortical transplants.

Adult rats received unilateral transplants of either embryonic day-17 fetal neocortex or striatum. Sixty-two percent of the fetal neocortical transplants grafted into the striatum, and 25% of these neocortical transplants placed in host neocortex had regions of hypermyelination, whereas none of the striatal transplants showed such an effect. There are similarities between these neuropathologic findings and status marmoratus in humans and animals with perinatal hypoxic injuries.

Induction of NGFI-B mRNA following contextual fear conditioning and its blockade by diazepam.

Expression of the immediate-early gene, NGFI-B (nerve growth factor inducible gene B), was examined in the amygdala, hippocampus, and neocortex following contextual fear conditioning. Rats were either handled, placed within the testing context without receiving the footshock, received a footshock immediately upon placement within the context, or received a footshock after a 3-min delay (delayed-shock). Only the delayed-shock group displayed a fear response (freezing) in the post-shock period and in a retention test 24 h after fear conditioning. Expression of NGFI-B mRNA was increased in the do

Evolutionary origins of the reptilian brain: the question of putative homologues of dorsal ventricular ridge. An overview and proposal.

The reptilian brain is characterized by a structure that bulges into the lateral ventricle, called dorsal ventricular ridge (DVR). The DVR was originally considered to be a part of the basal ganglia, although more recent studies indicate that it may correspond to the dorsal part of the hemisphere. The anterior portion of the DVR has several connectional and functional similarities with parts of the mammalian neocortex, for which reason it has been claimed that the two structures can be considered as homologues. In this article I review the evidence supporting and refuting homology of the DVR w

Molecular and anatomical signatures of sleep deprivation in the mouse brain.

Sleep deprivation (SD) leads to a suite of cognitive and behavioral impairments, and yet the molecular consequences of SD in the brain are poorly understood. Using a systematic immediate-early gene (IEG) mapping to detect neuronal activation, the consequences of SD were mapped primarily to forebrain regions. SD was found to both induce and suppress IEG expression (and thus neuronal activity) in subregions of neocortex, striatum, and other brain regions. Laser microdissection and cDNA microarrays were used to identify the molecular consequences of SD in seven brain regions. In situ hybridizatio

Staging of cytoskeletal and beta-amyloid changes in human isocortex reveals biphasic synaptic protein response during progression of Alzheimer's disease.

We have examined the relationships between dementia, loss of synaptic proteins, changes in the cytoskeleton, and deposition of beta-amyloid plaques in the neocortex in a clinicopathologically staged epidemiological cohort using a combination of biochemical and morphometric techniques. We report that loss of synaptic proteins is a late-stage phenomenon, occurring only at Braak stages 5 and 6, or at moderate to severe clinical grades of dementia. Loss of synaptic proteins was seen only after the emergence of the full spectrum of tau and beta-amyloid pathology in the neocortex at stage 4, but not
