Annulate lamellae and single-pore complexes in human spermatogonia.

Electron microscopic examination of non-neoplastic human testes revealed cytoplasmic annulate lamellae (AL) in all cell types within the seminiferous epithelium. Short lamellar profiles with low numbers of pores were found in spermatogonia and spermatocytes, while larger stacks of AL were seen in spermatids and Sertoli cells. Stacks of lamellae were closely associated with redundant nuclear envelope during late spermiogenesis. Residual bodies shed by the developing spermatozoa contained large number of lamellar-pore complexes. One cryptorchid testis was also examined. In this case the seminife

The Central Pore of HIV-1 Capsomers Promotes Sustained Stability of the Viral Capsid.

The HIV-1 capsid, which orchestrates several key post-entry events to facilitate infection, is composed of hexamers and pentamers (capsomers) of the capsid (CA) protein. The capsomers are arranged in a closed, conical structure - the capsid - that protects the viral RNA genome and replicative enzymes reverse transcriptase (RT) and integrase (IN). Each capsomer contains a central pore lined with rings of positively charged amino acid side chains - Arg-18 (R18) and Lys-25 (K25). The R18 and K25 rings drive capsid assembly by binding the host polyanion inositol hexakisphosphate (IP6) and are prop

p97/UBXD1 Generate Ubiquitylated Proteins That Are Sequestered into Nuclear Envelope Herniations in Torsin-Deficient Cells.

DYT1 dystonia is a debilitating neurological movement disorder that arises upon Torsin ATPase deficiency. Nuclear envelope (NE) blebs that contain FG-nucleoporins (FG-Nups) and K48-linked ubiquitin are the hallmark phenotype of Torsin manipulation across disease models of DYT1 dystonia. While the aberrant deposition of FG-Nups is caused by defective nuclear pore complex assembly, the source of K48-ubiquitylated proteins inside NE blebs is not known. Here, we demonstrate that the characteristic K48-ubiquitin accumulation inside blebs requires p97 activity. This activity is highly dependent on t

A Full-Process Experimental Evaluation of Water-Blocking Mitigation in Tight Sandstones by Supercritical CO2 Shut-In Soaking Coupled with N2 Displacement.

Water blocking severely restricts gas mobility in tight sandstone reservoirs because of capillary-dominated retention of aqueous phases within micro-nanopore systems. This study establishes an integrated experimental framework to quantify the full-process evolution of water blockingfrom water invasion and entrapment to gas displacement and supercritical CO2 (SC-CO2) treatment. Vacuum-assisted water saturation, pressure-controlled N2 displacement, and SC-CO2 shut-in soaking experiments were conducted on tight sandstone cores, and multiscale characterization methods, including nuclear magnetic

The nuclear export factor CRM1 controls juxta-nuclear microtubule-dependent virus transport.

Transport of large cargo through the cytoplasm requires motor proteins and polarized filaments. Viruses that replicate in the nucleus of post-mitotic cells use microtubules and the dynein-dynactin motor to traffic to the nuclear membrane and deliver their genome through nuclear pore complexes (NPCs) into the nucleus. How virus particles (virions) or cellular cargo are transferred from microtubules to the NPC is unknown. Here, we analyzed trafficking of incoming cytoplasmic adenoviruses by single-particle tracking and super-resolution microscopy. We provide evidence for a regulatory role of CRM

Mechanisms of Water-Lock Removal by a Nanoemulsion/Mud Acid Composite System in Low-Permeability Sandstone Reservoirs: Insights from Pore-Scale Characterization in the MB Block.

Water-lock damage in low-permeability sandstone reservoirs is primarily caused by small pore-throat structures, strong capillary forces, and hydrophilic rock surfaces. Conventional treatment methods mainly target either fluid-interface modification or pore blockage problems separately. In this study, a nanoemulsion/mud acid composite system was developed to achieve a combined effect of fluid interfaces and localized mineral dissolution. Interfacial tension (IFT) measurements, contact angle analysis, constant-rate mercury intrusion (CRMI), nuclear magnetic resonance (NMR), and high-temperature

Role of the ribosomal quality control machinery in nucleocytoplasmic translocation of polyQ-expanded huntingtin exon-1.

The subcellular localization of polyQ-expanded huntingtin exon1 (Httex1) modulates polyQ toxicity in models of Huntington's disease. Using genome-wide screens in a yeast model system, we report that the ribosome quality control (RQC) machinery, recently implicated in neurodegeneration, is a key determinant for the nucleocytoplasmic distribution of Httex1-103Q. Deletion of the RQC genes, LTN1 or RQC1, caused the accumulation of Httex1-103Q in the nucleus through a process that required the CAT-tail tagging activity of Rqc2 and transport via the nuclear pore complex. We provide evidence that nuc

The dystonia-associated Torsin1A sustains CLCC1 function in membrane fusion of the nuclear envelope for NPC biogenesis.

DYT1 dystonia is an incurable movement disorder caused by a loss-of-function mutation in Torsin1A, an endoplasmic reticulum (ER)-resident AAA+ ATPase. Here, we use Drosophila and human cells to shed light on Torsins' mode of action. Fly germ cells lacking dTorsin arrest in development with defects in nuclear pore complex (NPC) biogenesis due to impaired nuclear envelope membrane fusion. We identify the conserved membrane protein Chloride Channel CLIC-like protein 1 (CLCC1) as a Torsin1A interaction partner whose absence phenocopies membrane fusion defects caused by Torsin deletion. CLCC1 is en

Muscle Pathology as a Diagnostic Clue to Allgrove Syndrome.

Allgrove or triple A syndrome is a rare autosomal recessive disorder that can present with a variable range of multi-system manifestations, including optic atrophy, cerebellar ataxia, upper and lower motoneuron signs and various neuropathic abnormalities. These cases are a diagnostic challenge, particularly when the eponymous combination of achalasia, Addisonianism and alacrima is incomplete. Therefore, it is in the differential diagnosis for multisystem conditions and should be known to pathologists who diagnose disorders of skeletal muscle. Here, we describe new findings in skeletal muscle h

Nuclear envelope disorganization in fibroblasts from lipodystrophic patients with heterozygous R482Q/W mutations in the lamin A/C gene.

Dunnigan-type familial partial lipodystrophy (FPLD), characterized by an abnormal body fat redistribution with insulin resistance, is caused by missense heterozygous mutations in A-type lamins (lamins A and C). A- and B-type lamins are ubiquitous intermediate filament proteins that polymerize at the inner face of the nuclear envelope. We have analyzed primary cultures of skin fibroblasts from three patients harboring R482Q or R482W mutations. These cells were euploid and able to cycle and divide. A subpopulation of these cells had abnormal blebbing nuclei with A-type lamins forming a periphera

Mechanisms of Podocyte Injury Due to Loss of the Nucleoporin NUP93.

KEY POINTS: Loss of NUP93 caused progressive glomerular disease in mice. Nuclear levels of podocyte transcription factors were reduced on loss of NUP93. Loss of NUP93 changed transcriptional programs in podocytes. BACKGROUND: Genetic variants in six different genes encoding nuclear pore complex (NPC) proteins cause steroid-resistant nephrotic syndrome and FSGS in pediatric patients. All affected proteins are structural components of NPCs, restricted channels across the nuclear envelope that are required for molecular exchange between nucleoplasm and cytoplasm. Although podocyte injury is a un

Virus specificity and nucleoporin requirements for MX2 activity are affected by GTPase function and capsid-CypA interactions.

Human myxovirus resistance 2 (MX2/MXB) is an interferon-induced GTPase that inhibits human immunodeficiency virus-1 (HIV-1) infection by preventing nuclear import of the viral preintegration complex. The HIV-1 capsid (CA) is the major viral determinant for sensitivity to MX2, and complex interactions between MX2, CA, nucleoporins (Nups), cyclophilin A (CypA), and other cellular proteins influence the outcome of viral infection. To explore the interactions between MX2, the viral CA, and CypA, we utilized a CRISPR-Cas9/AAV approach to generate CypA knock-out cell lines as well as cells that expr

Recurrent Increased Nuchal Translucency Led to the Identification of Novel NUP107 Variants.

Five percent of fetuses presents increased fetal nuchal translucency. It is a well-known marker for aneuploidy (T21, Turner syndrome) and a variety of monogenic syndromes such as Noonan syndrome and certain skeletal dysplasias, as well as associated with structural malformations such as congenital heart disease. Current diagnostic algorithms for increased nuchal translucency include a rapid test for aneuploidy (fluorescence in situ hybridization, FISH, or quantitative PCR), a cytogenetic analysis (karyotype or chromosomal microarray, CMA) followed by or concurrent with targeted gene panel anal

Germline NUP98 Variants in Two Siblings with a Rothmund-Thomson-Like Spectrum: Protein Functional Changes Predicted by Molecular Modeling.

Two adult siblings born to first-cousin parents presented a clinical phenotype reminiscent of Rothmund-Thomson syndrome (RTS), implying fragile hair, absent eyelashes/eyebrows, bilateral cataracts, mottled pigmentation, dental decay, hypogonadism, and osteoporosis. As the clinical suspicion was not supported by the sequencing of RECQL4, the RTS2-causative gene, whole exome sequencing was applied and disclosed the homozygous variants c.83G>A (p.Gly28Asp) and c.2624A>C (p.Glu875Ala) in the nucleoporin 98 (NUP98) gene. Though both variants affect highly conserved amino acids, the c.83G>A looked m

Alterations in the Nucleocytoplasmic Transport in Heart Transplant Rejection.

BACKGROUND: Nucleocytoplasmic transport is a crucial process for cell function. Previous studies have observed alterations in different molecules involved in it, relating them to ventricular function. However, there are no published data evaluating possible differences in the expression of these molecules in heart transplantation (HT) recipients. Our objective is to evaluate whether its levels are related to the appearance of cellular rejection (CR) during the first year after HT. METHODS: A prospective clinical cohort that included patients undergoing HT between January 2017 and January 2019

Biallelic loss of function NEK3 mutations deacetylate α-tubulin and downregulate NUP205 that predispose individuals to cilia-related abnormal cardiac left-right patterning.

Defective left-right (LR) organization involving abnormalities in cilia ultrastructure causes laterality disorders including situs inversus (SI) and heterotaxy (Htx) with the prevalence approximately 1/10,000 births. In this study, we describe two unrelated family trios with abnormal cardiac LR patterning. Through whole-exome sequencing (WES), we identified compound heterozygous mutations (c.805-1G >C; p. Nup82 functions redundantly with Nup136 in a salicylic acid-dependent defense response of Arabidopsis thaliana.

The nuclear pore complex (NPC) comprises more than 30 nucleoporins (Nups). NPC mediates macromolecular trafficking between the nucleoplasm and the cytoplasm, but specific roles of individual Nups are poorly understood in higher plants. Here, we show that the novel nucleoporin unique to angiosperm plants (designated as Nup82) functions in a salicylic acid-dependent defense in a redundant manner with Nup136, which is a component of the nuclear basket in the NPC. Arabidopsis thaliana Nup82 had a similar amino acid sequence to the N-terminal half of Nup136 and a Nup82-GFP fusion was localized on t

Structures and metal-binding properties of Helicobacter pylori neutrophil-activating protein with a di-nuclear ferroxidase center.

Helicobacter pylori causes severe diseases, such as chronic gastritis, peptic ulcers, and stomach cancers. H. pylori neutrophil-activating protein (HP-NAP) is an iron storage protein that forms a dodecameric shell, promotes the adhesion of neutrophils to endothelial cells, and induces the production of reactive oxygen radicals. HP-NAP belongs to the DNA-protecting proteins under starved conditions (Dps) family, which has significant structural similarities to the dodecameric ferritin family. The crystal structures of the apo form and metal-ion bound forms, such as iron, zinc, and cadmium, of H

Investigating the Alteration of Sandstone Pore System and Rock Features by Role of Weighting Materials.

Drilled formations are commonly invaded by drilling fluids during the drilling operations, and as a result, the rock pore system will have alterations that consequently alter the rock properties. The objective of this study is to investigate the impact of the most commonly used weighting materials in water-based mud (WBM) on the Berea Buff sandstone pore system and rock characteristics. Rock-mud interaction was imposed by using a customized high-pressure high-temperature filtration test cell under 300 psi differential pressure and 200 °F temperature to simulate downhole conditions during drill
