Tooth development in a scincid lizard, Chalcides viridanus (Squamata), with particular attention to enamel formation.

Comparative analysis of tooth development in the main vertebrate lineages is needed to determine the various evolutionary routes leading to current dentition in living vertebrates. We have used light, scanning and transmission electron microscopy to study tooth morphology and the main stages of tooth development in the scincid lizard, Chalcides viridanus, viz., from late embryos to 6-year-old specimens of a laboratory-bred colony, and from early initiation stages to complete differentiation and attachment, including resorption and enamel formation. In C. viridanus, all teeth of a jaw have a si

Oral effects of steroid therapy in a patient with systemic lupus erythematosus: report of case.

Steroids are used extensively in modern medicine and dentistry for their anti-inflammatory and immunosuppressive properties. However, this class of drugs can lead to adverse effects on other organs. In this case, developmental and functional disturbances of the primary and permanent dentition are seen. The patient's dental development and pharmacological history suggest prednisone as the offending agent.

Tooth eruption: the regulation of a localized, bilaterally symmetrical metabolic event in alveolar bone.

Tooth eruption is a complicated process by which developing teeth are moved within the jaws to their functional position. The usual model chosen to study this process, the erupted rodent incisor, differs both structurally and functionally from the human dentition and conclusions drawn from these studies are not directly applicable to tooth eruption in human beings. We have studied the eruption of developing permanent premolars in dogs and present evidence by scanning electron microscopy for regional differences in metabolic activities on bone surfaces of the crypt during eruption. We review ev

Hyperdontia in children below four years of age: a radiographic study.

Sixty-six children (forty-one boys and twenty-five girls) were evaluated for differences in characteristics of premaxillary hyperdontia between the primary and permanent dentitions. The study population had forty-five supernumerary teeth in their primary dentitions and fifty supernumerary teeth in their permanent dentitions. Eighty percent of the primary supernumerary teeth were located in the lateral incisor regions, in proper alignment within the dental arch. Sixty-two percent of the supernumerary teeth in the permanent dentitions were situated in the central incisor regions; 90 percent were

Comparative evaluation of calretinin immunohistochemical expression in calcifying odontogenic cyst and ameloblastoma.

BACKGROUND: Calretinin is a 29 kilodalton (KDa) calcium-binding protein that is expressed in normal and tumoral tissues. The expression of calretinin has been shown in the dental epithelium during odontogenesis and in different odontogenic cysts and tumors such as ameloblastoma. Since the epithelium of calcifying odontogenic cyst (COC) is similar to ameloblastoma and in both lesions, an arrangement of loose cells similar to stellate reticulum is seen, we aimed to investigate the comparative expression of calretinin in COC and ameloblastoma. METHODS: In this observational-analytical study, 11

Genetic Variants in KIF7 May Contribute to Supernumerary Tooth Formation.

OBJECTIVE: Kinesin Family Member 7 (KIF7) regulates ciliary integrity. Cilia-mediated sonic hedgehog signalling regulates tooth number, position, and morphology. The objective of the study was to investigate whether KIF7 variants could contribute to the formation of supernumerary teeth. METHODS: Whole-exome sequencing was performed on 153 individuals from 130 families with isolated supernumerary tooth types. Immunohistochemistry of Kif7 expression during early odontogenesis was performed in mouse embryos. Mutant KIF7 protein models were generated. RESULTS: We identified 4 rare missense varia

Absence of melatonin during development impairs craniofacial and dental onset in rats.

OBJECTIVE: Herein, we evaluated pinealectomy-induced melatonin absence to determine its effects on craniofacial and dental development in the offspring. DESIGN: Female Wistar rats in three groups, i.e., intact pregnant rats, pinealectomized pregnant rats (PINX), and pinealectomized pregnant rats subjected to oral melatonin replacement therapy, were crossed 30 days after surgery. The heads of 7-day-old pups were harvested for cephalometric and histological analyses, and maxillae and incisors were collected for mRNA expression analysis. RESULTS: The PINX pups exhibited a reduction in neurocran

The Notch-mediated circuitry in the evolution and generation of new cell lineages: the tooth model.

The Notch pathway is an ancient, evolutionary conserved intercellular signaling mechanism that is involved in cell fate specification and proper embryonic development. The Jagged2 gene, which encodes a ligand for the Notch family of receptors, is expressed from the earliest stages of odontogenesis in epithelial cells that will later generate the enamel-producing ameloblasts. Homozygous Jagged2 mutant mice exhibit abnormal tooth morphology and impaired enamel deposition. Enamel composition and structure in mammals are tightly linked to the enamel organ that represents an evolutionary unit forme

Clinicopathologic and Molecular Characteristics of Familial Cherubism with Associated Odontogenic Tumorous Proliferations.

Cherubism is a rare autosomal dominant condition affecting the jaws and caused by mutations in the gene encoding for the adapter protein SH3BP2 that maps to chromosome 4p16.3. Cherubism is characterized by symmetrically developing bone lesions in the maxilla and mandible. The lesions have been radiographically and histopathologically well-described. Here, we present a family with cherubism with two of its members featuring odontogenic tumorous proliferations in association with persistent central giant cell lesions (CGCL). Specifically, the proband, a 25-year-old male, developed a radiolucent

Distribution of Cathepsin K in Late Stage of Tooth Germ Development and Its Function in Degrading Enamel Matrix Proteins in Mouse.

Cathepsin K (CTSK) is a member of cysteine proteinase family, and is predominantly expressed in osteoclastsfor degradationof bone matrix proteins. Given the similarity in physical properties of bone and dental mineralized tissues, including enamel, dentin and cementum, CTSK is likely to take part in mineralization process during odontogenesis. On the other hand, patients with pycnodysostosis caused by mutations of the CTSK gene displayedmultipledental abnormalities, such as hypoplasia of the enamel, obliterated pulp chambers, hypercementosis and periodontal disease. Thereforeitis necessary to

18-year follow-up of dens invaginatus: retrograde endodontic treatment.

INTRODUCTION: Dens invaginatus is a rare developmental anomaly that occurs during odontogenesis, with a higher prevalence in the Caucasian race and no significant sex predilection; it can be bilateral. This anomaly occurs in approximately 1.26% of the teeth, and the maxillary lateral incisor is most often affected. METHODS: This case report describes a female patient with dens invaginatus in the maxillary right conoid lateral incisor. The patient presented to Araçatuba School of Dentistry, Universidade Estadual Paulista, São Paulo, Brazil, in 1995 with an acute periapical abscess in the palat

The dentition: the outcomes of morphogenesis leading to variations of tooth number, size and shape.

The clinical importance of variations of tooth number, size and shape is seen in many dental disciplines. Early diagnosis allows optimal patient management and treatment planning, with intervention at an appropriate time to prevent complications in development and so reduce later treatment need. Understanding the process of dental morphogenesis and the variations in outcomes is an important contribution to the multidisciplinary clinical team approach to treatment. Tooth number, size and shape are determined during the initiation and morphogenetic stages of odontogenesis. The molecular evidence

Tooth-bone morphogenesis during postnatal stages of mouse first molar development.

The first mouse molar (M1) is the most common model for odontogenesis, with research particularly focused on prenatal development. However, the functional dentition forms postnatally, when the histogenesis and morphogenesis of the tooth is completed, the roots form and the tooth physically anchors into the jaw. In this work, M1 was studied from birth to eruption, assessing morphogenesis, proliferation and apoptosis, and correlating these with remodeling of the surrounding bony tissue. The M1 completed crown formation between postnatal (P) days 0-2, and the development of the tooth root was ini

Genetic basis of non syndromic hypodontia: a DNA investigation performed on three couples of monozygotic twins about PAX9 mutation.

INTRODUCTION: Hypodontia, agenesis of one or of more teeth, is a common developmental dental anomaly. To date, over 200 candidate genes have been demonstrated to be active in tooth development. The genes Pax9 plays an important role in the initial stage of odontogenesis. Mutations of Pax9 are associated with autosomal dominant forms of oligodontia, the agenesis of more than six teeth and occasionally of premolars (MIM 604625) in humans. The aim of the present study was to screen the candidate gene causing the non syndromic hypodontia, with agenesis of upper third molars and upper lateral incis

The matricellular protein periostin is required for sost inhibition and the anabolic response to mechanical loading and physical activity.

Periostin (gene Postn) is a secreted extracellular matrix protein involved in cell recruitment and adhesion and plays an important role in odontogenesis. In bone, periostin is preferentially expressed in the periosteum, but its functional significance remains unclear. We investigated Postn(-/-) mice and their wild type littermates to elucidate the role of periostin in the skeletal response to moderate physical activity and direct axial compression of the tibia. Furthermore, we administered a sclerostin-blocking antibody to these mice in order to demonstrate the influence of sustained Sost expr

Cranial suture biology and dental development: genetic and clinical perspectives.

Premature fusion of the calvarial bones at the sutures, or craniosynostosis (CS), is a relatively common birth defect (1:2000-3000) frequently associated with limb deformity. Patients with CS may present oral defects, such as cleft soft palate, hypodontia, hyperdontia, and delayed tooth eruption, but also unusual associations of major dental anomalies such as taurodontism, microdontia, multiple dens invaginatus, and dentin dysplasia. The list of genes that are involved in CS includes those coding for the different fibroblast growth factor receptors and a ligand of ephrin receptors, but also ge
