A germin-like gene (CchGLP) cloned from geminivirus-resistant pepper (Capsicum chinense Jacq. Line BG-3821) was characterized and the enzymatic activity of the expressed protein analyzed. The predicted protein consists of 203 amino acids, similar to other germin-like proteins. A highly conserved cupin domain and typical germin boxes, one of them containing three histidines and one glutamate, are also present in CchGLP. A signal peptide was predicted in the first 18 N-terminal amino acids, as well as one putative N-glycosylation site from residues 44-47. CchGLP was expressed in E. coli and the

The class 1 pathogenesis-related (PR) proteins are thought to be involved in plant defence responses, but their molecular functions are unknown. The function of PR-1 was investigated in tobacco by generating stable PR-1a-silenced lines in which other acidic PR-1 genes (PR-1b and PR-1c) were silenced. Plants lacking extracellular PR-1s were more susceptible than wild-type plants to the oomycete Phytophthora parasitica but displayed unaffected systemic acquired resistance and developmental resistance to this pathogen. Treatment with salicylic acid up-regulates the PR-1g gene, encoding a basic pr

The systemic acquired resistance (SAR) response in Arabidopsis is characterized by the accumulation of salicylic acid (SA), expression of the pathogenesis-related (PR) genes, and enhanced resistance to virulent bacterial and oomycete pathogens. The cpr (constitutive expressor of PR genes) mutants express all three SAR phenotypes. In addition, cpr5 and cpr6 induce expression of PDF1.2, a defense-related gene associated with activation of the jasmonate/ethylene-mediated resistance pathways. cpr5 also forms spontaneous lesions. In contrast, the eds1 (enhanced disease susceptibility) mutation abol

At least 10 Dm genes conferring resistance to the oomycete downy mildew fungus Bremia lactucae map to the major resistance cluster in lettuce. We investigated the structure of this cluster in the lettuce cultivar Diana, which contains Dm3. A deletion breakpoint map of the chromosomal region flanking Dm3 was saturated with a variety of molecular markers. Several of these markers are components of a family of resistance gene candidates (RGC2) that encode a nucleotide binding site and a leucine-rich repeat region. These motifs are characteristic of plant disease resistance genes. Bacterial artifi

In Arabidopsis, loss of transcription factors Calmodulin-Binding Transcription Activators 1/2/3 (CAMTA1/2/3) results in enhanced defense responses with increased salicylic acid (SA) and N-hydroxy-pipecolic acid (NHP) biosynthesis. This is achieved through modulating the expression of master immune transcription factors SAR Deficient 1 (SARD1) and Calmodulin-Binding Protein 60-like g (CBP60g). SARD4 is an NHP biosynthesis enzyme, the camta2/3 sard4 triple mutant remains dwarfed and autoimmune. To identify positive immune regulators of the SA and NHP pathways, a forward genetic screen was perfor

BACKGROUND: Upon appropriate stimulation, plants increase their level of resistance against future pathogen attack. This phenomenon, known as induced resistance, presents an adaptive advantage due to its reduced fitness costs and its systemic and broad-spectrum nature. In Arabidopsis, different types of induced resistance have been defined based on the signaling pathways involved, particularly those dependent on salicylic acid (SA) and/or jasmonic acid (JA).
RESULTS: Here, we have assessed the implication of the transcriptional regulator OCP3 in SA- and JA-dependent induced defenses. Through

Aspergillus flavus and Aspergillus parasiticus are saprophytic fungi which can infect and contaminate preharvest and postharvest food/feed with production of aflatoxins (B1, B2, and G). They are also an opportunistic pathogen causing aspergillosis diseases of animals and humans. In this study, the volatile organic compounds (VOCs) from Streptomyces yanglinensis 3-10 were found to be able to inhibit mycelial growth, sporulation, conidial germination, and expression of aflatoxin biosynthesis genes in A. flavus and A. parasiticus in vitro. On peanut kernels, the VOCs can also reduce the disease s

BACKGROUND: Downy mildew is a destructive disease that causes severe losses in spinach (Spinacia oleracea L.) yield and quality. Nucleotide-binding site-leucine-rich repeat (NBS-LRR) genes are core regulators of plant innate immunity and play critical roles in defense against pathogen infection. However, the characteristics of NBS-LRR family genes in spinach and their specific functions in downy mildew resistance remain unclear, limiting the development of resistant spinach cultivars.
METHODS: Using available spinach genome data, we identified NBS-LRR family members via bioinformatics approac

Grapevine downy mildew, caused by Plasmopara viticola, poses significant threats to viticulture, necessitating sustainable alternatives to chemical fungicides. This study investigates the efficacy of exogenous 24-epibrassinolide (EBR), a brassinosteroid, in enhancing disease resistance across two grape cultivars: susceptible Vitis vinifera Cabernet Sauvignon and resistant Ecolly. EBR application reduced P. viticola colonization in both cultivars, suppressing hyphal growth and sporulation. In Cabernet Sauvignon, EBR delayed pathogen establishment by modulating stomatal closure and boosting anti

Glyoxalase I (GLYI) is part of the glyoxalase system; its major function is the detoxification of α-ketoaldehydes, including the potent and cytotoxic methylglyoxal (MG). Methylglyoxal disrupts mitochondrial respiration and increases production of reactive oxygen species (ROS), which also increase during pathogen infection of plant tissues; however, there have been few studies relating the glyoxalase system to the plant pathogen response. We used the promoter of VvGLYI-4 to screen the upstream transcription factors and report a NAC (NAM/ATAF/CUC) domain-containing transcription factor VvNAC72 i

Fungal exposure and sensitization are linked to asthma severity, but their clinical significance varies by host, exposure context, and disease phenotype. This review integrates epidemiological, immunological, and clinical evidence on fungal-associated asthma, distinguishing environmental exposure, allergic sensitization, and persistent airway colonization. We examine how fungal allergens, proteases, and cell-wall components engage pattern-recognition receptors, disrupt epithelial barriers, and activate overlapping type 2 and type 17 inflammatory pathways. Studies of the airway and intestinal m

Pythiosis is an emerging infectious disease caused by the oomycete Pythium insidiosum, affecting humans and animals primarily in subtropical and tropical regions. The pathogen is commonly found in swampy environments, and exposure can lead to diverse clinical manifestations. In humans, ocular and vascular infections predominate, whereas in animals, cutaneous/subcutaneous or gastrointestinal disease is more common. Medical therapy is frequently ineffective, and many patients require extensive surgical intervention. Advanced cases may progress to fatal outcomes. Therefore, early and accurate det

Potato late blight, caused by the oomycete pathogen Phytophthora infestans (P. infestans), is one of the most devastating diseases threatening global potato production. Conventional plant disease detection methods rely on a labor-intensive and time-consuming workflow and require bulky and expensive benchtop equipment, limiting their in-field applications. Here, we report a portable RPA-CRISPR/Cas12a-based diagnostic platform integrated with a polyvinyl alcohol (PVA) microneedle (MN) patch, which allows rapid in-field sampling, and smartphone-based fluorescence acquisition and analysis to detec

Muskmelons (Cucumis melo var. melo) grown in the eastern United States are affected by several foliar diseases caused by fungi and an oomycete. The objective of this survey of muskmelon production regions in South Carolina was to determine the prevalence of foliar diseases on spring crops to refine fungicide recommendations. In total, 34 fields from 21 growers in 10 counties were sampled using stratified cluster sampling. A total of 20, 20, and 40 diseased leaves were collected in each field in 2019, 2020, and 2021, respectively, along four transects forming a square of 1225 or 2,500 m2. After

The anaphase-promoting complex/cyclosome (APC/C) is a multi-subunit E3 ubiquitin ligase that regulates cell cycle progression, but its role in plant-pathogen interactions is poorly understood. Here, we demonstrate that regulation of the ubiquitin-proteasome system, involving APC/C-associated components, is critical for coordinating host cell cycle modulation and cellular homoeostasis required for proper gall formation and successful nematode parasitism. Inhibition of the 26S proteasome with MG132 severely disrupted gall morphology and nematode development, indicating that proteasome activity i

The oomycete Phytophthora capsici causes Phytophthora blight, a major constraint on global pepper production. Our previous observations indicated that pretreating plants with thiamethoxam (TMX) and imidacloprid (IMI) could reduce the incidence of pepper blight, but the underlying mechanisms remained unclear. Here, we investigated how TMX and IMI induced resistance in pepper (Capsicum frutescens) against P. capsici. Both in vitro and in vivo assays demonstrated that TMX and IMI suppressed disease, not by directly impairing pathogen virulence but by inducing systemic resistance in susceptible (C

Late blight, caused by oomycete pathogen Phytophthora infestans, is one of the most devastating diseases of potato and tomato. Plant resistance to pathogens relies on a sophisticated innate immune system, which is often targeted by pathogen-derived effectors. P. infestans secretes numerous RXLR effectors to suppress host immunity. In this study, we show that the P. infestans core effector Pi05910 functions as a virulence factor and targets the host scaffold protein Receptor for Activated C Kinase 1 (RACK1), which is identified to positively regulate potato resistance to late blight. Functional

To exploit superior microbial strains for the biocontrol of wheat crown rot, we employed high antagonistic activity and strong colonization ability as dual criteria to screen strains from the culture collection of the Agricultural Microbiology Innovation Center of Henan Province. We comprehensively evaluated the candidate strain for both biocontrol efficacy and plant growth-promoting traits, and subsequently identified it through integrated morphological, molecular biological, and genomic analyses. The results showed that Paenibacillus polymyxa YB-393 exhibited the strongest root colonization

This research aimed to identify native biocontrol bacteria capable of suppressing Phytophthora infestans in potato crops. A total of forty bacterial strains were isolated from wheat (Triticum aestivum), sunflower (Helianthus annuus), potato (Solanum tuberosum), and cotton (Gossypium herbaceum) plants collected in Sibi, Pakistan. Out of these, ten isolates showed antagonistic effects against P. infestans, with inhibition percentages ranging from 27 to 62%. The isolate WL2.3 exhibited the strongest inhibition at 62%. Furthermore, sporangia germination tests, including those using cell-free super
