[Therapy-induced fluorosis--damage or goal?].

Report on 3 female patients who, after treatment of an osteoporosis with sodium fluoride, have developed a skeletal fluorosis. The proof was performed radiologically, histologically and by means of fluoride analysis in the bone ash. The diagnosis was made accidentally. Complaints relating to this are not mentioned. Osteosclerosis after fluoride therapy is no iatrogenically damage for the patient, but the aim of the therapy for the prevention of osteopenic fractures.

Disodium cromoglycate in the treatment of systemic mastocytosis involving only bone.

A 50-year-old woman with a 5-year history of low back pain and osteosclerosis was diagnosed as having systemic mastocytosis. She had no cutaneous or gastrointestinal manifestations of this disease. She was successfully treated with oral disodium cromoglycate. This is the first reported case of systemic mastocytosis involving only bone which responded to treatment with oral disodium cromoglycate.

Bovine fluorosis in Sweden.

Fluorotic lesions have been studied in cows and calves of SRB breed on farms belonging to 2 agricultural companies. From the one company (HP) in Dalecarlia 3 calves, 4 heifers and 2 cows were examined and from the other (B) in Södermanland 12 bull-calves. The submitted material consisted of a carcase from 1 dead calf and skull, metacarpus and in some cases kidneys from slaughtered animals. The fluorine content of feed and water samples was determined. In material from HP extensive enamel defects and strong wear were found in the deciduous incisors. Fluorotic lesions in calves have not been des

Oral Lesions in Pycnodysostosis Syndrome: Eleven Years of Follow-Up.

Pycnodysostosis is a rare genetic disease that is characterized by osteosclerosis, short stature, and bone fragility. There are not cases of gnathic bones lesions reported on the international literature. This study aims to describe a clinical case of a 10-year-old girl with pycnodysostosis syndrome and an uncommon association with 4 distinct lesions (dentigerous cyst, central giant cell lesions, and 2 fibro-osseous lesions).

Marble bone disease: a review of osteopetrosis and its oral health implications for dentists.

Osteopetrosis is one cause of osteosclerosis and may result in such serious oral complications as osteomyelitis and exposed necrotic bone. Dentists should be aware of patients with the disease because of its effect on osteoclast function, which results in impaired wound healing. The purpose of this paper is to review the causes, pathogenesis and differential diagnosis of osteopetrosis and to provide guidance to dentists on the management of patients with osteopetrosis.

[Special radiology of the hand in renal osteopathy].

The different radiographical abnormalities of the hand with renal osteodystrophy are demonstrated. The soft tissue immersion low-energy X-ray technique allows early detection of macrostructural changes of spongy and compact bone, irregularities of the cortex, defects and pseudocysts, osteosclerosis as well as calcifications in the soft tissue and arterio sclerosis of patients with chronic renal failure. Possible erroneous interpretations will be discussed.

Osteoarthritis associated with osteopetrosis treated by total knee arthroplasty. Report of a case.

Osteopetrosis is due to a defect in osteoclastic cell function and results in osteosclerosis and progressive obliteration of the marrow spaces. Early onset osteoarthritis is associated with osteopetrosis. The authors describe a case of early onset osteoarthritis associated with osteopetrosis treated by total knee arthroplasty. The marblelike quality of osteopetrotic bone makes surgical treatment of these patients technically challenging and requires modification of standard surgical technique.

Osteopetrosis: Gene-based nosology and significance Dysosteosclerosis.

Dysosteosclerosis (DSS) refers to skeletal dysplasias that radiographically feature focal appendicular osteosclerosis with variable platyspondyly. Genetic heterogeneity is increasingly reported for the DSS phenotype and now involves mutations of SLC29A3, TNFRSF11A, TCIRG1, LRRK1, and CSF1R. Typical radiological findings are widened radiolucent long bones with thin cortices yet dense irregular metaphyses, flattened vertebral bodies, dense ribs, and multiple fractures. However, the radiographic features of DSS evolve, and the metaphyseal and/or appendicular osteosclerosis variably fades with inc

Long-term survival in infantile malignant autosomal recessive osteopetrosis secondary to homozygous p.Arg526Gln mutation in CLCN7.

Infantile malignant autosomal recessive osteopetrosis (ARO; OMIM 259700) has been reported to be associated with mutations in TCIRG1, CLCN7, or OSTM1. ARO caused by homozygous (or compound heterozygous) mutations in CLCN7, as described here, is usually diagnosed at birth or early in infancy due to generalized osteosclerosis and severe hematologic deficits. The maximal life expectancy of patients with ARO in the absence of bone marrow transplantation is thought to be 10 years. We report on a 25-year-old Thai man who is affected with ARO. Clinical features include proportionate short stature, vi

Case Report - Pyknodysostosis.

Pyknodysostosis is an extremely rare lysosomal storage disease of the bone. Osteosclerosis of the skeleton is due to a decrease of the bone turnover. A plethora of clinical and radiological findings are observed. Patients are short statured with acroosteolysis and dense bones and develop multiple spontaneous bone fractures. A female patient presented with frontal bossing, small and thick fingers and toes. The classical clinical and radiological findings confirmed it as pyknodysostosis.

Cytogenic investigations on leucocytes of cattle intoxicated with fluoride.

Cattle which grazed in the vicinity of a plant manufacturing enamel and which displayed signs of chronic fluoride poisoning such as osteosclerosis and mottled enamel or dental fluorosis were investigated for the presence of structural chromatid and chromosome aberrations. Venous blood was incubated for 48 h, and 100 cells were analyzed for each animal. No statistical difference in the incidence of such anomalies was observed between the controls and the intoxicated animals.

A novel FAM20C mutation causes a rare form of neonatal lethal Raine syndrome.

Raine syndrome is a rare, autosomal recessive, osteosclerotic bone dysplasia due to pathogenic variants in FAM20C. The clinical phenotype is characterized by generalized osteosclerosis affecting all bones, cerebral calcifications, and craniofacial dysmorphism. Most cases present during the neonatal period with early lethality due to pulmonary hypoplasia and respiratory compromise while only few affected individuals have been reported to survive into adulthood. FAM20C is a ubiquitously expressed protein kinase that contains five functional domains including a catalytic domain, a binding pocket

Biochemical evidence of disturbed bone metabolism and calcium homeostasis in two types of autosomal dominant osteopetrosis.

Biochemical markers of bone resorption and bone formation were measured in 14 patients with autosomal dominant osteopetrosis, and compared with age- and sex-matched controls. There were eight patients with the radiological type I characterized by diffuse, symmetrical osteosclerosis with pronounced sclerosis of the skull and enlarged thickness of the cranial vault, and six patients with type II characterized by diffuse, symmetrical osteosclerosis, "Rugger-Jersey spine" and "endobones" (bone within a bone) in the pelvis. Serum levels of alkaline phosphatase and osteocalcin in types I and II did

Dialysis osteodystrophy. A study involving 94 patients.

Dynamic skeletal histomorphometry was performed in 94 unselected patients receiving maintenance dialysis for chronic renal failure. An attempt was made to correlate the results with the clinical, biochemical and radiological findings. Skeletal histology was abnormal in each case. Hyperparathyroidism was present as the only abnormality in 18 patients and osteomalacia in 26; 50 patients showed both abnormalities. Osteomalacia, in contrast to hyperparathyroidism, increased in prevalence and severity with the duration of dialysis and with bone aluminum content. The majority of patients had histolo

The radiographic spectrum of renal osteodystrophy.

Chronic renal failure often results in bone changes, collectively known as renal osteodystrophy. These changes include osteitis fibrosa, osteosclerosis, soft tissue calcifications, osteomalacia (in adults) and rickets (in children). Early recognition of renal osteodystrophy allows more aggressive clinical control of serum calcium and phosphate levels. Intervention may spare the patient late complications such as fractures, pain and loss of skeletal function.

Pycnodysostosis Revealed by Recurrent Fractures: Report of Two Cases.

Pycnodysostosis is a rare autosomal recessive skeletal dysplasia caused by cathepsin K deficiency and characterized by osteosclerosis and bone fragility. Recurrent low-trauma fractures and delayed healing represent major orthopedic challenges, particularly when medullary canal narrowing limits surgical options. We report the case of two brothers born to consanguineous parents who presented with short stature, characteristic craniofacial dysmorphism, acro-osteolysis, and recurrent fractures after minor trauma. The older sibling had the more severe skeletal phenotype, with 14 fractures involving

Recurrent petit mal seizures in Erdheim-Chester disease mimicking an intra-axial brain tumor: illustrative case.

BACKGROUND: Erdheim-Chester disease (ECD) is a rare non-Langerhans cell histiocytosis characterized histologically by foamy histiocytes and Touton giant cells in a background of fibrosis. Bone pain with long bone osteosclerosis is highly specific for ECD. Central nervous system involvement is rare, although dural, hypothalamic, cerebellar, brainstem, and sellar region involvement has been described. OBSERVATIONS: A 59-year-old man with a history of ureteral obstruction, medically managed petit mal seizures, and a left temporal lesion followed with serial magnetic resonance imaging (MRI) prese

Myelofibrosis osteoclasts are clonal and functionally impaired.

Bone marrow (BM) sclerosis is commonly found in patients with late-stage myelofibrosis (MF). Because osteoclasts (OCs) and osteoblasts play a key role in bone remodeling, and MF monocytes, the OC precursors, are derived from the neoplastic clone, we wondered whether decreased OC numbers or impairment in their osteolytic function affects the development of osteosclerosis. Analysis of BM biopsies from 50 MF patients showed increased numbers of multinucleated tartrate-resistant acid phosphatase (TRAP)/cathepsin K+ OCs expressing phosphorylated Janus kinase 2 (JAK2). Randomly microdissected TRAP+

Skeletal Involvement in Systemic Mastocytosis: Pathophysiology, Clinical Management, Standards of Care, and Novel Therapeutic Strategies.

Systemic mastocytosis comprises a group of clonal mast cell disorders characterized by multisystem involvement. Bone involvement represents a major source of morbidity, particularly in young men affected by indolent systemic mastocytosis. This review provides an integrated and up-to-date overview of SM-related bone disease. We dissect the dual and context-dependent role of mast cells in bone remodeling, detailing how they promote osteoclastogenesis, suppress osteoblast function, and, in advanced disease, drive osteosclerosis. We critically appraise available treatments, including classic anti-

TBXAS1 deficiency: a novel monogenic cause of chronic nonbacterial osteomyelitis responsive to COX inhibitors.

BACKGROUND: Chronic nonbacterial osteomyelitis (CNO) is an autoinflammatory bone disease characterized by sterile bone inflammation. While monogenic causes are rare and typically present with early-onset, distinct skin or lytic bone lesions, the contribution of COX-pathway genes remains underexplored. TBXAS1 gene variants are classically associated with Ghosal hematodiaphyseal dysplasia (GHDD), which causes osteosclerosis and hematologic abnormalities. RESEARCH DESIGN AND METHODS: In this single-center, cross-sectional study, whole-exome sequencing first revealed the TBXAS1 variants in the in
