Detection of Mycobacterium avium subsp. paratuberculosis in Bovine Faeces Using a Rhodamine B Hydrazone Immunosensor on Antibody-Coated Nitrocellulose Membranes.

Rapid, inexpensive and reliable detection of Mycobacterium avium subsp. paratuberculosis (MAP) is essential for the control of Johne's disease. This study developed a rhodamine B hydrazone nanoparticle-based immunosensor coupled with antibody-coated nitrocellulose membranes (IMS-CNM) for the detection of MAP in bovine faecal samples. Anti-MAP antibodies were purified from hyperimmune donkey and rabbit sera. Purified donkey IgG was immobilized on nitrocellulose membranes as the capture antibody, whereas rabbit anti-MAP IgG was conjugated to rhodamine B hydrazone nanoparticles using EDC-NHS chem

Roles of mdh and MAP1981c in Mycobacterium avium subsp. paratuberculosis intracellular survival within bovine monocyte-derived macrophages via CRISPR interference.

Mycobacterium avium subsp. paratuberculosis (MAP) is the causative agent of Johne's disease, a chronic enteritis in ruminants, and is capable of persisting within macrophages despite the activation of host immune defenses. Although this intracellular persistence is a key determinant of MAP pathogenicity, the bacterial factors and host responses that regulate this process remain poorly understood. In this study, we established the first CRISPR interference (CRISPRi) platform applied to bovine monocyte-derived macrophages (MDM) to evaluate the functions of MAP genes involved in intracellular sur

Multi-peptide subunit vaccination induced IFN-γ but lacked efficacy against Mycobacterium avium subsp. paratuberculosis in a goat challenge trial.

Paratuberculosis is a chronic debilitating disease in ruminants caused by infection with Mycobacterium avium subsp. paratuberculosis (MAP). In endemic herds, animals are infected shortly after birth with no effective control measures beyond hygiene and sanitation programs. Current vaccines are only moderately effective and may interfere with surveillance and diagnosis of tuberculosis caused by M. bovis. In the current vaccination trial, we studied the efficacy of a vaccine against MAP in goats based on multiple 15-18 mer immunogenic peptides that are specific to MAP to not interfere with the d

Short Chain Fatty Acids Lower Inflammation and Restore Intestinal Integrity and Function Markers in Mycobacterium paratuberculosis-Infection In Vitro Model.

Background: Infection with Mycobacterium avium paratuberculosis (MAP) is closely associated with Crohn's disease (CD) development, where excessive inflammation and marked intestinal damage are observed. Objectives: In this study, the role of short chain fatty acids, including propionic acid (PPA) and butyric acid (BA), was evaluated in an in vitro model, mimicking CD characteristics. Methods: MAP-infected THP-1 macrophages were treated with 1 mM and 10 mM of PPA or BA, and the conditioned media was co-cultured in Caco-2 cells. Results: Both PPA and BA caused an M2 shift with significant downre

Discovery of Mycobacterium avium subsp. paratuberculosis Lytic Phages with Extensive Host Range Across Rapid- and Slow-Growing Pathogenic Mycobacterial Species.

BACKGROUND/OBJECTIVES: Developing interventions for Johne's disease, which focuses on controlling Mycobacterium avium subsp. paratuberculosis (MAP) in contaminated environments by treating infected cows and preventing transmission from diseased animals, is a critical priority. Bacteriophage (phage) therapy, an emerging biological intervention, offers a promising alternative for the treatment and management of MAP infections. METHODS: In this study, we generated an MAP-specific lytic phage library aimed at characterizing the therapeutic potential of phages under environmental and biological co

Isolation of Mycobacterium avium subspecies paratuberculosis from muscle and peripheral lymph nodes using acid-pepsin digest prior to BACTEC culture.

Meat has received little attention regarding human exposure to Mycobacterium avium subsp. paratuberculosis, a possible infectious trigger of Crohn's disease. Meat has less contamination with other organisms than gut tissues, facilitating modifications to existing decontamination protocols prior to BACTEC culture that could increase analytical sensitivity. Using spiked meat samples we trialled enzymatic and chemical digestion techniques to concentrate larger starting samples, and modifications to existing clinical mycobacteriological decontamination protocols. An acid-pepsin digestion method us

Humoral immune responses of type 1 diabetes patients to Mycobacterium avium subsp. paratuberculosis lend support to the infectious trigger hypothesis.

Mycobacterium avium subsp. paratuberculosis is a zoonotic pathogen whose association with Crohn's disease in humans is under scrutiny. The objective of this work was to investigate its association with other chronic diseases such as type 1 diabetes mellitus (T1DM), where the involvement of a persistent pathogen such as M. avium subsp. paratuberculosis could be the trigger. For this purpose, 59 diabetic patients and 59 healthy controls were investigated for the presence of antibodies against two recombinant proteins of M. avium subsp. paratuberculosis and the whole-cell lysate. Extremely signif

Evaluation of three ELISA assays for detection of Mycobacterium avium subsp. paratuberculosis antibodies in serum of sika deer (Cervus nippon).

ELISA-based assay testing offers a practical, high-throughput approach for early detection of Mycobacterium avium subsp. paratuberculosis (MAP), the causative agent of Johne's disease (JD), particularly for identifying subclinically infected animals within a herd. As no ELISA has been developed specifically for sika deer, the aim of this study was to compare the diagnostic performance of two commercial ELISA assays (ID.VET and IDEXX) designed for cattle, sheep and goats, and the Paralisa™ assay developed for red deer. Serum and faecal samples (n = 105) were collected from 36 clinically affecte

Contamination of alpine pastures by Mycobacterium avium subsp. paratuberculosis: Evaluation of cattle and red deer contribution through environmental fecal samples.

Paratuberculosis is a chronic enteritis of ruminants (Bovidae, Cervidae) caused by Mycobacterium avium subsp. paratuberculosis (MAP). The prevalence of shedding animals, together with fecal MAP loads, is critical to the biocontamination of the environment. Surveillance for paratuberculosis has been introduced within the frame of European Health Law (Regulation EU 2016/429) both in livestock and wild ruminants. Semi-extensive and extensive livestock production may determine exposure to MAP infection and intraspecies and interspecies transmission on pasture. In the present study MAP prevalence a

Exploration of Solanum xanthocarpum Schrad. & Wendl. against Mycobacterium avium Subspecies paratuberculosis and Assessment of Its Immunomodulatory and Anti-Inflammatory Potential.

Mycobacterium avium subspecies paratuberculosis (MAP), being a dairy-borne pathogen, resistant of pasteurization and other sterilization techniques, is a major cause for development of inflammatory bowel disorders such as Johne's disease (JD) in dairy animals and Crohn's Disease (CD) in humans, for which no therapy is available to date. In the absence of effective therapy or a vaccine, management of CD has been accomplished by removal of the affected intestines. However, usually, even after removal of 2/3 of the intestine, CD reoccurs. Hence, there exists a need to develop an alternative thera

Systematic Assessment of Mycobacterium avium Subspecies Paratuberculosis Infections from 1911-2019: A Growth Analysis of Association with Human Autoimmune Diseases.

Mycobacterium avium subsp. paratuberculosis (MAP) is an understudied pathogen worldwide with continuous implications in human autoimmune diseases (ADs). The awareness of MAP appears to be low in many places and its research is at infant stage in many countries. The lack of worldwide coverage of the MAP research landscape calls for urgent research attention and prioritization. This present study aimed to assess MAP global research productivity with an emphasis on its implications in ADs via bibliometric and growth analytic frameworks from authors, countries, institutions, international, discipl

A PNA-assisted magnetic bead-confined CbAgo-CRISPR/Cas12a cascade for amplification-free detection of Mycobacterium avium subsp. paratuberculosis F57 DNA.

Mycobacterium avium subsp. paratuberculosis (MAP), the causative agent of Johne's disease in ruminants, has attracted increasing attention because of its potential zoonotic relevance. Sensitive molecular detection of MAP typically relies on nucleic acid amplification to overcome low pathogen abundance; however, amplification may generate nonspecific products and false-positive results. Here, targeting MAP-specific F57 double-stranded DNA, we developed a peptide nucleic acid (PNA)-assisted, magnetic bead-confined Clostridium butyricum Argonaute (CbAgo)-CRISPR/Cas12a cascade electrochemiluminesc

Systematic evaluation and optimization of TaqMan qPCR assays targeting F57, ISMAP02, and IS900 for multiplex detection of Mycobacterium avium subsp. paratuberculosis.

This study aimed to develop a multiplex quantitative PCR (qPCR) assay for the detection of Mycobacterium avium subsp. paratuberculosis (MAP), the etiological agent of paratuberculosis disease, a chronic and endemic infectious disease affecting ruminant livestock worldwide. Infected animals may remain asymptomatic for years while intermittently shedding MAP into their environment through feces, contributing to ongoing transmission. To develop a robust multiplex qPCR assay, we reviewed all TaqMan qPCR studies published since 1990 and selected 18 primer-probe combinations targeting the MAP-specif

Evidence for transfer of maternal antigen specific cellular immunity against Mycobacterium avium ssp. paratuberculosis via colostrum in a goat twin model.

Colostrum intake is one of the most important factors in neonatal health in ruminants, mainly because of its unique immunological properties. Both in practice as well as in research, the attention of lactogenic immunity is focused on the importance of colostral antibodies and less attention is given to the functional role of maternal cells in colostrum. Here we study the transfer of maternal leukocytes via colostrum and the functionality in goat kids. In experiment 1, twenty twin pairs of goat kids from dams previously immunized with an inactivated Mycobacterium avium subsp. paratuberculosis (
