Food and diet were class markers in 19th-century Ireland, which became evident as nearly 1 million people, primarily the poor and destitute, died as a consequence of the notorious Great Famine of 1845 to 1852. Famine took hold after a blight (Phytophthora infestans) destroyed virtually the only means of subsistence-the potato crop-for a significant proportion of the population. This study seeks to elucidate the variability of diet in mid-19th-century Ireland through microparticle and proteomic analysis of human dental calculus samples (n = 42) from victims of the famine. The samples derive fro

Whether salicylic acid (SA) plays a role in systemic acquired resistance (SAR) signaling in potato is currently unclear because potato, unlike tobacco and Arabidopsis, contains highly elevated levels of endogenous SA. Recent studies have indicated that the SA derivative methyl salicylate (MeSA) serves as a long-distance phloem-mobile SAR signal in tobacco and Arabidopsis. Once in the distal, uninfected tissue of these plant species, MeSA must be converted into biologically active SA by the esterase activity of SA-binding protein 2 (SABP2) in tobacco or members of the AtMES family in Arabidopsi

The class 1 pathogenesis-related (PR) proteins are thought to be involved in plant defence responses, but their molecular functions are unknown. The function of PR-1 was investigated in tobacco by generating stable PR-1a-silenced lines in which other acidic PR-1 genes (PR-1b and PR-1c) were silenced. Plants lacking extracellular PR-1s were more susceptible than wild-type plants to the oomycete Phytophthora parasitica but displayed unaffected systemic acquired resistance and developmental resistance to this pathogen. Treatment with salicylic acid up-regulates the PR-1g gene, encoding a basic pr

We have previously shown that healthy potato plants respond poorly to salicylic acid (SA) for activating disease resistance against the late blight fungal pathogen Phytophthora infestans. However, SA is essential for the establishment of potato systemic acquired resistance (SAR) against P. infestans after treatment with the fungal elicitor arachidonic acid (AA). To understand the molecular mechanisms through which AA induces SA-dependent SAR in potato, we have recently studied the expression of potato class II catalase (Cat2St) in comparison with its tobacco homologue, Cat2Nt, which has previo

Eugenol is the main component of clove basil oil, mainly extracted from Syzygium aromaticum. In order to discover biorational natural product-based pesticides, 24 novel eugenol acyloxy/sulfonyloxy derivatives (3a-q and 4d,f,j-n) containing an isoxazoline ring were designed and synthesized by simultaneously introducing isoxazoline and acyloxy/sulfonyloxy pharmacophores, and their structures were characterized by 1H NMR, 13C NMR, and HRMS. The single-crystal X-ray diffraction of two titled compounds 3k and 4l usefully established the 5-substituted regiochemistry. Furthermore, we screened the bio

BACKGROUND: Citrus root weevil, Diaprepes abbreviatus is one of the most destructive pests of citrus, causing extensive root damage that predisposes trees to secondary infection with phytophthora and substantial yield losses. In the present study, we identified and functionally characterized the coatomer subunit beta gene (DaCOPB), a core component of the COPI vesicle trafficking machinery responsible for two-way transport between the endoplasmic reticulum and Golgi apparatus.
RESULTS: The full-length DaCOPB transcript was obtained from the de novo transcriptome of D. abbreviatus and exhibite

Microbial co-cultivation is employed for awakening silent biosynthetic gene clusters (BGCs) to enhance chemical diversity. However, the selection of appropriate partners for co-cultivation remains a challenge. Furthermore, competitive interactions involving the suppression of BGCs or upregulation of known, functional metabolite(s) during co-cultivation efforts is also common. Herein, we performed an alternative approach for targeted selection of the best co-cultivation pair. Eight marine sediment-derived fungi were classified as strong or weak, based on their anti-phytopathogenic potency. The

Understanding the genetic basis of pathogen susceptibility in various crop plants is crucial to increasing the stability of food, feed, and fuel production. Varietal differences in defence responses provide insights into the mechanisms of resistance and are a key resource for plant breeders. To explore the role of salicylic acid in the regulation of defence in cacao, we demonstrated that SA treatment decreased susceptibility to a pod rot pathogen, Phytophthora tropicalis in two genotypes, Scavina 6 and Imperial College Selection 1, which differ in their resistance to several agriculturally imp

Homocysteine (Hcy) is a naturally occurring intermediate metabolite formed during methionine metabolism. It has been well documented that its excess can be extremely toxic to mammalian, yeast and bacterial cells. In spite of the metabolic value of Hcy known for decades, the role of this amino acid in the plant response to stress has not been recognized yet. In the presented study, using potato plant (Solanum tuberosum L.) and Phytophthora infestans as a model system, the presence and tissue localization of Hcy in leaves was examined by an immunohistochemical method. The over-production of Hcy

Salicylic acid (SA) is a key endogenous component of local and systemic disease resistance in plants. In this study, we investigated the role of benzoic acid (BA) as precursor of SA biosynthesis in tobacco (Nicotiana tabacum cv Samsun NN) plants undergoing a hypersensitive response following infection with tobacco mosaic virus or in tobacco cell suspensions elicited with beta-megaspermin, an elicitor from Phytophthora megasperma. We found a small pool of conjugated BA in healthy leaves and untreated cell suspensions of tobacco, whereas free BA levels were barely detectable. Infection of plants

Small secreted peptides (SSPs), serving as signaling molecules for intercellular communication, play significant regulatory roles in plant growth, development, pathogen immunity, and responses to abiotic stress. Despite several SSPs, such as PIP, PSK, and PSY having been identified to participate in plant immunity, the majority of SSPs remain understudied, necessitating the exploration and identification of SSPs regulating plant immunity from vast genomic resources. Here we systematically characterized 756 putative SSPs across the genome of Nicotiana tabacum. 173 SSPs were further annotated as

Soil-borne diseases of tobacco roots and stems, caused by pathogens including Ralstonia solanacearum (bacterial wilt), Phytophthora parasitica var. nicotianae (black shank), and root-knot nematodes (Meloidogyne spp.), represent critical threats to tobacco cultivation. However, information regarding alterations in rhizosphere soil properties and microbial communities following co-infection by multiple soil-borne pathogens remains limited for tobacco-growing regions in Luxi County, Honghe Prefecture, Yunnan Province, China. This study therefore investigated rhizosphere soils collected from healt

Streptomyces species are well-known for their potential in biocontrol and plant growth promotion, with the rhizosphere serving a rich reservoir for novel isolates. In this study, a Streptomyces strain (SX92T) was isolated from the rhizosphere of healthy tobacco plants. In dual-culture assays, SX92T displayed broad-spectrum antagonistic activity against six major fungal pathogens of tobacco, with the highest inhibition (59.22%) against Phytophthora nicotianae, the causal agent of tobacco black shank. Polyphasic taxonomic characterization, combining 16S rRNA gene phylogeny, distinctive physiolog

The phytochemical diversity of plants represents a valuable reservoir for novel agrochemical agents. Our preliminary investigations identified pronounced antifungal efficacy in Laggera pterodonta extracts, although this species remains critically understudied. Herein, six eudesmane-type sesquiterpenes (1-6) were isolated from L. pterodonta and structurally characterized through spectroscopic analysis. Additionally, the antifungal activity of these compounds against six plant-pathogenic fungal species was evaluated: Phytophthora nicotianae, Fusarium oxysporum, Alternaria alternata, Gloeosporium

BACKGROUND: Global food security faces dual challenges from pathogen threats and increasing fungicide resistance, urgently necessitating the development of new pesticides that are efficient, low-toxicity, and environmentally friendly. Natural products, owing to their structural diversity and environmental compatibility, serve as important resources. Formononetin, a common isoflavone in leguminous plants, exhibits various biological activities. However, its direct antifungal efficacy is limited, leaving potential for structural optimization.
RESULTS: In this study, 20 formononetin derivatives

Taking the natural product cerbinal as the lead compound, 30 novel 5-aryl-cyclopenta[c]pyridine derivatives were designed and synthesized based on the previous bioactivity studies of the cyclopenta[c]pyridines. The modification of the position-5 of compound 2 was achieved by amination, bromination, and cross coupling using cerbinal as the raw material. The results of the bioactivity tests demonstrated that partial compounds exhibited superior activity against plant viruses compared to compound 2. Compounds 4g and 4k showed higher anti-TMV activity levels than commercial varieties of ribavirin

As a continuation of our research on the pesticide development of Polygonum orientale L., the chemical constituents of the seeds of P. orientale were systematically investigated. Eleven natural compounds (PO-1 to PO-11) were isolated from the EtOAc extract of P. orientale. Notably, compound PO-9 and its dimeric compound PO-10 were first isolated from P. orientale and possessed excellent acaricidal activity against Tetranychus cinnabarinus. With PO-9 and PO-10 as the lead compounds, two series of cinnamate derivatives were further synthesized, and their acaricidal, insecticidal, and fungicidal

How do pathogens affecting the same host interact with each other? We evaluated here the types of microbe-microbe interactions taking place between Streptomyces scabiei and Phytophthora infestans, the causative agents of common scab and late blight diseases in potato crops, respectively. Under most laboratory culture conditions tested, S. scabiei impaired or completely inhibited the growth of P. infestans by producing either soluble and/or volatile compounds. Increasing peptone levels correlated with increased inhibition of P. infestans. Comparative metabolomics showed that production of S. sc
