Functional consequences of embryonic neocortex transplanted to rats with prefrontal cortex lesions.

In four experiments we reexamined the recent report by Labbe, Firl, Mufson, and Stein (1983) that fetal cortical tissue transplanted to an aspirative prefrontal cortical cavity in rats can ameliorate the learning impairments induced by the aspirative lesions. Healthy surviving grafts from young (E16) embryonic donors had no immediate effects on the rats' impairments in T-maze alternation, spatial navigation in the Morris swimming pool task, or locomotor activity, and they produced even greater impairments than the lesions alone when all three tests were conducted after longer (3-5 month) survi

Contribution of the medial prefrontal cortex to performance on the Traveling Salesperson Problem in rats.

Cognitive impairments, including memory loss, disrupted spatial navigation, and poor executive function, are devastating features of many neurological conditions. Effective research into the causes and treatments of such disorders requires knowledge of the neurological mechanisms that underlie these cognitive processes. Our lab used a naturalistic spatial foraging task, known as the Traveling Salesperson Problem (TSP), to examine these processes in rodents. Previous studies from our lab have found that rats with hippocampal lesions or medial entorhinal cortex lesions were impaired on measures

Interhemispheric Metabolic Effects of Transcranial Infrared Laser Stimulation of the Prefrontal Cortex.

Photobiomodulation is the use of low-power, non-ionizing light to stimulate cellular processes. Transcranial infrared laser stimulation (TILS) is a method for prefrontal photobiomodulation that produces dose-dependent metabolic effects in vivo. However, interhemispheric differences in metabolic responses to unilateral right versus left TILS are unknown. Investigating interhemispheric effects of TILS is important because right-left differences are observed with other forms of transcranial stimulation, including magnetic and electrical stimulation. TILS penetration was first characterized using

The Role of Prefrontal and Posterior Parietal Cortex in Generating Multiple Step Saccades.

While multiple step saccades (MSS) are occasionally reported in the healthy population, they are more evident in patients with Parkinson's disease (PD). Therefore, MSS has been suggested as a biological marker for the diagnosis of PD. However, the lack of clarity on the neural mechanism underlying the generation of MSS largely impedes their application in the clinic. We have proposed recently that MSS are triggered by the discrepancy between desired and executed saccades. Accordingly, brain regions involved in saccadic planning and execution might play a role in the generation of MSS. To test

Outbalanced: The cross-cortical effects of prefrontal neuromodulation in posterior parietal cortex.

Cognitive phenomena such as the Spatial-Numerical Association of Response Codes (SNARC) effect can arise in the fronto-parietal cortical network. Prior neuromodulation studies with cathodal transcranial direct current stimulation (tDCS) over the left prefrontal cortex (PFC) reduced the SNARC effect. Prior neuroimaging studies with functional near-infrared spectroscopy (fNIRS), however, showed signatures of the SNARC effect in the posterior parietal cortex (PPC). In this study, we investigated the distant neural effect of prefrontal neuromodulation on hemodynamic activity in the parietal cortex

The Role of Basolateral Amygdala and Medial Prefrontal Cortex in Fear: A Systematic Review.

Fear is a primary adaptive response to potential threats. It triggers a complex cascade of physiological, cognitive, and behavioral changes that prepare an organism to cope with dangerous situations. The basolateral amygdala (BLA) and medial prefrontal cortex (mPFC) are both linked to adaptation, the generation of strong emotions, and decision-making. In this systematic review, we aimed to analyze recent studies of the connections between the BLA and mPFC in the context of their neuroanatomy, cellular composition, micro-circuitry, and involvement in fear. The role of the medial prefrontal cortex in risk and time preferences: A tDCS study.

Risk and time preferences are fundamental determinants of economic decision making. Although medial prefrontal cortex (mPFC) activity has been associated with valuation across these domains, causal evidence that focal mPFC perturbation alters both risky and intertemporal choices remains limited. In this research, we applied anodal, cathodal, or sham transcranial direct current stimulation (tDCS) to mPFC and administered a battery of risky lotteries and a Convex Time Budget task that jointly elicits risk and intertemporal trade-offs. Our results indicate that individuals who received cathodal s

Observation on prefrontal cortex activation in patients with Parkinson's disease: a fNIRS study.

BACKGROUND: Patients with Parkinson's disease (PD) commonly experience difficulties when performing a second task while walking. The mechanisms underlying dual-task walking deficits remain poorly understood. In previous studies the second tasks were often simplistic, typically comprising questions from standardized cognitive assessments. Additionally, existing fNIRS studies comparing PD patients and healthy controls have reported inconsistent findings, limiting our understanding of prefrontal cortex (PFC) contributions to cognitive-motor integration. METHODS: Forty-two healthy older adults (1

A dataset profiling the multiomic landscape of the prefrontal cortex in amyotrophic lateral sclerosis.

Amyotrophic lateral sclerosis (ALS) is the most common motor neuron disease, which still lacks effective disease-modifying therapies. Similar to other neurodegenerative disorders, such as Alzheimer and Parkinson disease, ALS pathology is presumed to propagate over time, originating from the motor cortex and spreading to other cortical regions. Exploring early disease stages is crucial to understand the causative molecular changes underlying the pathology. For this, we sampled human postmortem prefrontal cortex (PFC) tissue from Brodmann area 6, an area that exhibits only moderate pathology at

The dorsomedial prefrontal cortex promotes self-control by inhibiting the egocentric perspective.

The dorsomedial prefrontal cortex (dmPFC) plays a crucial role in social cognitive functions, including perspective-taking. Although perspective-taking has been linked to self-control, the mechanism by which the dmPFC might facilitate self-control remains unclear. Using the multimodal neuroimaging dataset from the Human Connectome Project (Study 1, N =978 adults), we established a reliable association between the dmPFC and self-control, as measured by discounting rate-the tendency to prefer smaller, immediate rewards over larger, delayed ones. Experiments (Study 2, N = 36 adults) involving hig

Stress-Induced Tinnitus in a Rat Model: Transcriptomics of the Prefrontal Cortex and Hippocampus.

OBJECTIVES: The molecular mechanisms by which stress leads to the development of tinnitus are not yet well understood. This study aimed to identify brain changes in a stress-induced tinnitus (ST) animal model through transcriptome analysis of the prefrontal lobe and hippocampus. METHODS: Twenty Sprague-Dawley rats were subjected to restraint stress for 2 h. Following the gap prepulse inhibition of the acoustic startle (GPIAS) reflex test to assess tinnitus development, the prefrontal lobes and hippocampi of the brains were harvested from 15 rats: five with evident tinnitus (ST), five with not

The role of prefrontal cortex and temporoparietal junction in interpersonal comfort and emotional approach.

Our perception of physical distance to individuals and stimuli is influenced by our mental distance and relatedness. The present study aimed to investigate the role of the dorsolateral prefrontal cortex (dlPFC), ventromedial prefrontal cortex (vmPFC), and right temporoparietal junction (rTPJ) in interpersonal comfortable distance and approach behaviors towards emotional stimuli. Twenty healthy volunteers received brain stimulation in four separate sessions with a one-week interval, including anodal left dlPFC, anodal right vmPFC, anodal rTPJ, and sham condition, with an extracranial return ele

When brain stimulation backfires: the effects of prefrontal cortex stimulation on impulsivity.

Transcranial direct current stimulation (tDCS) can sometimes cause the opposite of its intended effect. These reverse effects may be related in part to individual differences in personality and neurochemistry. Previous studies have demonstrated that dopamine levels can impact the effects of tDCS. In the present study, 124 healthy participants took the UPPS impulsive behavior scale. Participants then underwent a single, randomized anodal or sham tDCS session on the prefrontal cortex. While the effects of tDCS were still active, they performed the Stop Signal Task, a measure of state impulsivity

Distinct roles of the medial prefrontal cortex in advantageous and disadvantageous inequity aversion.

Human beings have a strong preference for the fair distribution of resources in situations of both advantageous and disadvantageous inequity. Neuroimaging studies have shown that the process of advantageous and disadvantageous inequity aversion involves distinct brain regions. However, little is known about the causal roles of the dorsal medial prefrontal cortex (dmPFC) in these two types of inequity aversion. To clarify the roles of the dmPFC in both types of inequity aversion, 70 subjects were recruited and randomly assigned to two anodal transcranial direct current stimulation (tDCS) groups

Sex differences in the behavioral inhibition system and ventromedial prefrontal cortex connectivity.

The reinforcement sensitivity theory proposes brain-behavioral systems that underlie individual differences in sensitivity to punishment and reward. Such trait sensitivity is assessed using the behavioral inhibition/activation system (BIS/BAS) scales. Recent studies have reported sex-linked neuroanatomical correlates of the BIS/BAS, especially in the regions belonging to the valuation and salience networks that are associated with the representation of subjective value (SV), whereas less effort has been focused on investigating the neurofunctional aspects associated with sex differences in the

Differential expression of NPAS4 in the dorsolateral prefrontal cortex following opioid overdose.

BACKGROUND: Although preclinical models reveal the neurobiological pathways altered through opioid abuse, comprehensive assessments of gene expression in human brain samples are needed. Moreover, less is known about gene expression in response to fatal overdose. The primary goal of the present study was to compare gene expression in the dorsolateral prefrontal cortex (DLPFC) between brain samples of individuals who died of acute opioid intoxication and group-matched controls. METHODS: Postmortem tissue samples of the DLPFC from 153 deceased individuals (Mage  = 35.4; 62% male; 77% European an

Causal Role of the Dorsolateral Prefrontal Cortex in Belief Updating under Uncertainty.

Adaptive performance in uncertain environments depends on the ability to continuously update internal beliefs about environmental states. Recent correlative evidence suggests that a frontoparietal network including the dorsolateral prefrontal cortex (dlPFC) supports belief updating under uncertainty, but whether the dlPFC serves a "causal" role in this process is currently not clear. To elucidate its contribution, we leveraged transcranial direct current stimulation (tDCS) over the right dlPFC, while 91 participants performed an incentivized belief-updating task. Participants also underwent a

Abnormal expression of post-synaptic proteins in prefrontal cortex of patients with schizophrenia.

There is converging evidence of dendritic spine dysfunction in schizophrenia. In the present study we hypothesized that the expression of key proteins involved in dendritic spine development and stability may be affected in schizophrenia. Postmortem frontal cortex (BA6) from patients with schizophrenia, major depressive disorder, bipolar disorder and healthy controls was processed for glutamate post-synaptic fraction extraction and post-synaptic density purification. Protein expression of the post-synaptic fraction and the post-synaptic density was assessed using immunoprecipitation and Wester

Gene expression profiles underlying aggressive behavior in the prefrontal cortex of cattle.

BACKGROUND: Aggressive behavior is an ancient and conserved trait, habitual for most animals in order to eat, protect themselves, compete for mating and defend their territories. Genetic factors have been shown to play an important role in the development of aggression both in animals and humans, displaying moderate to high heritability estimates. Although such types of behaviors have been studied in different animal models, the molecular architecture of aggressiveness remains poorly understood. This study compared gene expression profiles of 16 prefrontal cortex (PFC) samples from aggressive

Prefrontal Cortex Activity and Gait in Parkinson's Disease With Cholinergic and Dopaminergic Therapy.

Degradation of striatal dopamine in Parkinson's disease (PD) may initially be supplemented by increased cognitive control mediated by cholinergic mechanisms. Shift to cognitive control of walking can be quantified by prefrontal cortex activation. Levodopa improves certain aspects of gait and worsens others, and cholinergic augmentation influence on gait and prefrontal cortex activity remains unclear. This study examined dopaminergic and cholinergic influence on gait and prefrontal cortex activity while walking in PD. A single-site, randomized, double-blind crossover trial examined effects of l
